To develope skin-whitening or therapeutic agents against hyperpigmentation, aqueous extract from Amomum xanthioides (AEAX) was evaluated for melanogenesis inhibitory activity in B16F10 melanoma cell. The treatment with AEAX at the 0.5 and 1.0 mg/ml level significantly inhibits the biosynthesis of melanin compared with untreated control. The AEAX-treated cells at the 1.0 mg/ml level were more efficient than commercial arbutin at 0.1 mg/ml. The tyrosinase activity also significantly decreased in AEAX-treated cells at the 0.5 and 1.0 mg/ml level. The Western analyses confirmed the significantly decreased expression of tyrosinase and tyrosinase-related protein-1 by AEAX treatment. These results indicate that AEAX may contribute to the inhibition of melanin biosynthesis through regulating tyrosinase activity and expression and serve as a new candidate in the design of new skin-whitening or therapeutic agents.
Melanogenesis is induced mainly by ultraviolet radiation of sunlight and ${\alpha}$-Melanocyte stimulation hormone (${\alpha}$-MSH) which binds to a specific G protein coupled receptor. ${\alpha}$-MSH and cAMP-elevating agents are known to melanin syntheisis and dendrite outgrowth. The purpose of this study was to investigate the mechanism of melanogenesis inhibition in B16/F10 cells by ethanol extract of Artemisiae Capillaris Herba. In the present study, ${\alpha}$-MSH led to a stimulation of melanin synthesis that appeared to result from an increased tyrosinase activity and melanin content. However, the ethanol extract of Artemisiae Capillaris Herba inhibited the ${\alpha}$-MSH-induced tyrosinase activity and melanin content. In control conditions, B16/F10 cells displayed a fibroblastic appearance while ${\alpha}$-MSH treatment promoted the emergence of small and numerous dendrites from the plasma membrane. The ethanol extract of Artemisiae Capillaris Herba abolished the ${\alpha}$-MSH-induced dendricity. Regarding protein levels of the melanogenic enzymes, the amounts of tyrosinase were increased after incubation with ${\alpha}$-MSH. The treatment of Artemisiae Capillaris Herba ethanol extract decreased the ${\alpha}$-MSH expression levels of tyrosinase. Based on these findings, it is likely that the ethanol extract of Artemisiae Capillaris Herba exerts its depigmenting effects in B16/F10 cells through the suppression of tyrosinase expression, which are key enzymes for melanogenesis.
Piper methysticum 뿌리의 에탄올 추출물은 멜라닌 세포 자극 호르몬 활성화 B16 흑색종 세포에서 멜라닌 생성을 억제하는 것으로 알려져 왔다. 이 추출물로부터 분리한 flavokawain B (FKB)와 flavokawain C (FKC)는 항멜라닌 생성 활성을 기반으로 멜라닌 생성을 억제하는 것으로 밝혀져 있다. 본 연구는 melan-a 세포에서의 멜라닌 합성에 대한 FKB 및 FKC의 억제 및 그 과정을 알아보고자 하였다. FKB와 FKC는 각각 10 μM, 5 μM 농도에서 melan-a 세포의 멜라닌 생성을 억제하였다. 그러나 FKB와 FKC 모두 세포 외 타이로시네이즈 활성은 억제하지 않았다. FKB는 타이로시네이즈 (tyrosinase, Tyr), 타이로시네이즈 연관 단백질 1 (tyrosinaserelated protein 1, TRP-1)과 2(tyrosinase-related protein 2, TRP-2), microphthalmia-associated transcription factor (MITF)의 단백질 발현량을 감소시켰으며 Tyr와 TRP-1의 mRNA 발현량을 감소시켰다. FKC는 TRP-2와 MITF의 단백질 발현량을 감소시켰으며 Tyr와 TRP-1의 mRNA 발현량을 감소시켰다. Tyr와 TRP-1의 감소된 발현은 주요 멜라닌 생성 단백질을 조절하는 MITF발현 감소로 인한 것임을 예상할 수 있었다. 다만 MITF의 mRNA 발현량은 FKB와 FKC 처리에 의해 변하지 않았기 때문에, MITF의 분해를 조절한다고 알려진 세포 외 신호 조절 키나아제(ERK)/AKT 인산화에 대한 FKB와 FKC의 영향을 확인하였다. FKB와 FKC는 AKT의 인산화를 증가시키지는 않았지만 ERK1/2의 인산화를 유의미하게 증가시켰다. 이러한 결과는 FKB와 FKC가 다양한 색소 침착 증상들에 대한 잠재적인 미백제로 도움이 될 수 있음을 말해준다.
The purpose of this study was to investigate the anti-melanogenic effects of the extracts from Decaschistia intermedia craib (EDI). In this study, we examined the effects of EDI on mushroom tyrosinase activity in in vitro, melanin contents, and expression levels of mRNA and proteins of melanogenesis-related genes in B16F10 melanoma cells. The treatment of EDI significantly decreased both tyrosinase activity and melanin contents in B16F10 cells with dose-dependent manner. In addition, we found that the expression of mRNA or proteins of melanogenic proteins, such as, a-melanocyte-stimulating hormone (a-MSH)-induced microphthalmia associated transcription factor (MITF), tyrosinase, tyrosinase related protein-1 (TRP-1), and TRP-2 was significantly downregulated with dose-dependent manner in the EDI-treated B16F10 cells compared to controls. Our results suggest that the EDI inhibits cellular melanogenesis through downregulation of a-MSH-stimulated melanin synthesis. Thus EDI may potentially be an effective whitening agent.
본 연구는 B16F10 melanoma 세포를 사용하여 도인승기탕의 70% EtOH와 물 추출물의 멜라닌 생합성, tyrosinase 활성, western blotting으로 측정하였다. 도인승기탕 추출물은 농도 의존적으로 멜라닌 생합성과 tyrosinase활성을 저해하였다. 그 결과 도인승기탕 70% 에탄올 추출물이 멜라닌 합성을 40%, tyrosinase는 51% 저해효과를 나타내었다. Western blot을 이용하여 B16F10 melanoma 세포 내에 tyrosinase, TRP-1, TRP-2, MITF의 발현을 억제하는 효과를 관찰하였다. 이상의 결과에 따라 도인승기탕의 70% 에탄올 추출물은 미백 소재로서 가능성을 가지는 것으로 나타났다.
To date, research on the regulation of melanogenesis has focused on factors which affect tyrosinase, the rate-limiting enzyme in the melanogenic pathway, by searching for chemicals which competitively inhibit tyrosinase function. Many types of tyrosinase inhibitors have been developed, but no satisfactory results have been made clinically until now, To find a new whitening agent, which effectively inhibits melanogenesis, we synthesized several compounds and selected compounds by cell-based assay system. Finally, 3, 4, 5-trimethoxy cinnamaie thymol ester(TCTE, Melasolv) was selected and the effects of TCTE on melanogenesis were investigated. Treatment of mouse-derived melanocyte melan-a cells with TCTE results in a marked down-regulation of tyrosinase activity. 80% decrease of tyrosinase activity occurs with 30uM TCTE treatment for 72 hours without affecting cell growth. The inhibition of tyrosinase activity is dose-dependent and melanin content was also decreased to 40%. From the in vitro tyrosinase assay using cell extract, TCTE does not act as a direct inhibitor of the enzyme. Treatment of melan-a cultures with TCTE blocks the increase in tyrosinase activity by either forskolin, 3-isobutyl-1-methtyl-xanthine. TCTE decreased the expression of tyrosinase, TRP-1 without effects on TRP-2 protein expression through the down regulation of tyrosinase and TRP-1 mRNA. From the results of cAMP immunoassays, intracellular levels of the cyclin nucleotide are unaffected in cells treated with TCTE. The inhibitory effects of melanin synthesis were also shown in reconstitute human epidermis model by topical application. These findings suggest that TCTE can be used for studying the regulation of melanogenesis and depigmenting agent.
Commelina communis Ledeb is a widely used medication for the treatment of antidiabetic, antioxidant and hypoglycemic agent in Korea. Alpha-melanocyte stimulating hormone (${\alpha}$-MSH) is a major factor to stimulate melanin synthesis in the skin. The purposes of this study was to investigate the inhibitory effects of extract from Commelina communis Ledeb (ECC) on ${\alpha}$-MSH-stimulated melanogenesis in B16F10 cells. ECC suppressed melanin synthesis and intracellular tyrosinase activity in B16F10 cells or ${\alpha}$-MSH-induced B16F10 cells in a dose dependent manner. In study on the melanogenic protein expressions, it had especially influence on expressions of tyrosinase and tyrosinase-related protein (TRP-1). Tyrosinase and TRP-1 expressions were gradually decreased in a dose-dependent. Additionally, the extract also decreased the ${\alpha}$-MSH-induced over-expression of tyrosinase and TRP-1. This results show that the anti-melanogenic activity of ECC is correlated with the suppression of tyrosinase and TRP-1 protein expressions in B16F10 cells.
Three ergosterol derivatives, ergosta-4,6,8(14),22-tetraen-3-one (1), ergosta-7,24(28)-dien-3-ol (2), and 5,8-epidioxyergosta-6,22-dien-3-ol(3) were isolated from the fruit body of Phellinus pini. Their structures were based on spectroscopic methods including IR, MS, and NMR (1D and 2D). These compounds were evaluated for their activity to decrease melanin production in ${\alpha}$-MSH (melanocyte stimulating hormone) activated B16F10 cells. Compound 1, 2, and 3 reduced melanin content in a dose-dependent manner at concentrations of 5~15 uM. They also suppressed the tyrosinase expression of protein and m-RNA level dose dependently by western blot analysis and RT-PCR experiment in B16F10 murine melanoma cells.

Therefore, the present study was conducted to evaluate the effects of myricetin on the melanogenesis in human melanoma (HM₃KO) cells. The cells were treated for 5 days with myricetin at several concentrations (10 - 100 μg/ml). Treatment with myricetin dose-dependently suppressed cell growth in HM₃KO cells, But melanin formation was markedly increased by myricetin as a dose dependent manner. Myricetin did not affect to tyrosinase activity, which is a key enzyme for melanogenesis, but significantly increased the level of tyrosinase protein expression, These results suggest that myricetin stimulate melanin synthesis of human melanoma cells through the modification of tyrosinase protein expression.
