Production Properties on Extracellular Protease from Chryseobacterium Novel Strain JK1 (Chryseobacterium 속 신종세균 JK1의 세포외 단백질분해효소 생산특성)

  • A novel Chryseobacterium sp. JK1 strain producing extracellular protease had been isolated from soil. The largest clear zones were observed on nutrient agar plates supplemented with 1% skim milk at $30-35^{\circ}C$ along with the growth of Chryseobacterium sp. JK1. The cell growth of JK1 strain was maximal at 24 h and maximum protease activity was reached up to 560 unit/ml at the stationary phase in liquid culture. In the presence of maltose, glucose or mannitol in Nutrient broth, cells grew well, but protease were produced poorly with lower production yields of 64-77% than in NB broth only. Similarly, the addition of skim milk, beef extract, yeast extract, malt extract or tryptone showed good growth and poor enzyme production. On the contrary, the addition of $(NH_4)_2HPO_4$ or $(NH_4)_2SO_4$ gave poor growth and good enzyme production of 121-146%.

  • Proteases are degradative enzymes which hydrolyze a peptide bond between amino acids and they are abundantly applied to commercial field. In order to investigate optimal medium compositions of carbon and nitrogen source for enzyme production, modified STY medium containing 0.15% yeast extract were used as basal medium. When galactose was used as carbon source, enzyme activity showed 1.3 higher than that of glucose. For nitrogen source addition of casamino acids to basal medium in place of tryptone showed lowest activity, whereas addition of malt extract showed maximal activity. The optimum temperature and pH of extracellular protease were found to be $35^{\circ}C$ an pH 8.5.

Characterization and optimum production condition of extracellular protease from Pseudoalteromonas donghaensis HJ51 (Pseudoalteromonas donghaensis HJ51의 체외 단백질 분해효소 특성과 생산 조건)

  • Pseudoalteromonas donghaensis HJ51, isolated from the East Sea, has been reported as a novel strain to produce extracellular protease. Crude supernatant was used to determine optimal activity and optimal production conditions for the enzyme. It was found that the optimal temperature and pH of the protease were $40^{\circ}C$ and pH 7.5-10.5, respectively. The enzyme activity was kept to 88% at the pH 11. In metal requirement analysis, the enzyme exhibited the highest activity when 10 mM $Fe^{3+}$ was supplied. While supplementation of additional carbon sources used in study showed no positive effect on cell growth and enzyme activity, the addition of beef extract, tryptone, or casamino acids instead of peptone of PY-ASW containing 1% glucose increased enzyme production to 21, 7, 4%, respectively. Taken together these properties, the enzyme produced from P. donghaensis HJ51 can be applied to the industries that require protease activity under alkaline pH and low temperature.

  • To reducing offensive odor form compost UIW-6 mutant obtained by UV treatment from sulfur-oxidizing bacteria, Thiobacillus sp. IW. The UIW-6 mutant was found 1.6 times faster in growth than the parent strain on thiosulfate medium (TM) at 36 h after incubation. Initial pH, temperature and agitation for the optimum growth of UIW-6 were 6.5, $35^{\circ}C$ and 200 rpm, respectively. The UIW-6 mutant growth was two times higher than parent strain at 6 h culture in TM liquid medium containing 50 mM sodium thiosulfate. The UIW-6 mutant used fructose and sucrose as carbon sources and yeast extract> tryptone> peptone as nitrogen ones. It was found that the growth of UIW-6 was increased in addition of 0.2% yeast extract.

Optimization of Culture Conditions for toe Production of Chitinase (Chitinase 생성을 위한 배did 조건 최적화)

  • Chitinase producing microorganism, Serratia marcescens KY, was isolated from seashore mud around Beobseongpo in Chunnam province by selective enrichment culture. As the colloidal chitin concentration increased, chitinase production was increased. But chitinase production with addition of other carbon sources (glucose, fructose, galactose, maltose, sucrose, starch) was decreased. The effect of nitrogen sources on the chitinase production with serratia marcescens KY was as fellows. The opitimum mineral concentration for chitinase production was K$_2$HPO$_4$ 0.2 g/L and MgSO$_4$ 0.20 ∼ 0.25 g/L, respectively. The effect of nitrogen sources on chitinase production by Serratia marcescens KY was increased as follows, tryptone > yeast extract > beef extract > asparagine.

Properties of Protease from Aeromonas hydrophila AM-28 Isolated from Soil (토양에서 분리된 Aeromonas hydrophila AM-28이 생산하는 단백질 가수분해효소의 특성)

  • A bacterial strain NO. AM-28, showing proteolytic activity against defatted soybean was isolated from domestic soil. The isolated strain was identified as Aeromonas hydrophila by both the biochemical tests using API kit and the analysis of cellular fatty acid profile with MIDI system. The protease production from A. hydrophila AM-28 was highly enhanced when it was cultivated in the medium containing glycerol as a carbon source, tryptone or $(NH_4)_2HPO_4$ as a nitrogen source, and $CaCl_2$ as a mineral source. The optimal pH and temperature for the enzyme was 8.0 and $65^{\circ}C$, respectively. The enzyme was stable up to $55^{\circ}C$ and at pH values ranging from 7.0 to 13.0. The enzyme activity was inhibited by phenylmethylsulfonyl fluoride and EDTA, indicating that serine residue and metal ions be involved in enzyme activity.

Optimization of a Medium for the Production of Cellulase by Bacillus subtilis NC1 Using Response Surface Methodology (반응 표면 분석법을 사용한 Bacillus subtilis NC1 유래 cellulase 생산 배지 최적화)

  • Previously, cellulase and xylanase producing microorganism, Bacillus subtilis NC1, was isolated from soil. Based on the 16S rRNA gene sequence and API 50 CHL test the strain was identified as Bacillus subtilis, and named as B. subtilis NC1. We cloned and sequenced the genes for cellulase and xylanase. Plus, the deduced amino acid sequences from the genes of cellulase and xylanase were determined and were also identified as glycosyl hydrolases family (GH) 5 and 30, respectively. In this study to optimize the medium parameters for cellulase production by B. subtilis NC1 the RSM (response surface methodology) based on CCD (central composite design) model was performed. Three factors, tryptone, yeast extract, and NaCl, for N or C source were investigated. The cellulase activity was measured with a carboxylmethyl cellulose (CMC) plate and the 3,5-dinitrosalicylic acid (DNS) methods. The coefficient of determination (R2) for the model was 0.960, and the probability value (p=0.0001) of the regression model was highly significant. Based on the RSM, the optimum conditions for cellulase production by B. subtilis NC1 were predicted to be tryptone of 2.5%, yeast extract of 0.5%, and NaCl of 1.0%. Through the model verification, cellulase activity of Bacillus subtilis NC1 increased from 0.5 to 0.62 U/ml (24%) compared to the original medium.

Optimization of the Indole-3-Acetic Acid Production Medium of Pantoea agglomerans SRCM 119864 using Response Surface Methodology (반응표면분석법을 활용한 Pantoea agglomerans SRCM 119864의 Indole-3-acetic acid 생산 배지 최적화)

  • In this study, we optimized the composition of the indole-3-acetic acid (IAA) production medium using response surface methodology on Pantoea agglomerans SRCM 119864 isolated from soil. IAA-producing P. aglomerans SRCM 119864 was identified by 16S rRNA gene sequencing. There are 11 intermediate components known to affect IAA production, hence the effect of each component on IAA production was investigated using a Plackett-Burman design (PBD). Based on the PBD, sucrose, tryptone, and sodium chloride were selected as the main factors that enhanced the IAA production at optimal L-tryptophan concentration. The predicted maximum IAA production (64.34 mg/l) was obtained for a concentration of sucrose of 13.38 g/l, of tryptone of 18.34 g/l, of sodium chloride of 9.71 g/l, and of L-tryptophan of 6.25 g/l using a the hybrid design experimental model. In the experiment, the nutrient broth medium supplemented with 0.1% L-tryptophan as the basal medium produced 45.24 mg/l of IAA, whereas the optimized medium produced 65.40 mg/l of IAA, resulting in a 44.56% increase in efficiency. It was confirmed that the IAA production of the designed optimal composition medium was very similar to the predicted IAA production. The statistical significance and suitability of the experimental model were verified through analysis of variance (ANOVA). Therefore, in this study, we determined the optimal growth medium concentration for the maximum production of IAA, which can contribute to sustainable agriculture and increase crop yield.

Effect of nitrogen sources on the dextran production by Leuconostoc mensenteroids NRRL-B512F

  • The dextran prodution by Leuconostoc mensenteroids NRRL-B512F was studied in a synthetic medium from sucrose as a sole carbon source. Especially, effect of nitrogen source was treated and compared in this study. In oder to maximize the cell growth and dextran produtivity through fermentation two nitrogen sources, yeast extract and tryptone, were used with various concentrations. At the end of fermentation, when the concentration of yeast extract was 9g/L we can obtain the maximum dry cell weight(14.1g/L), dextran dry weight(25.4g/L) and productivity(1.4g/L ${\cdot}$ hr).

Isolation of Bacillus sp. AIR-5 PRoducing Maltopentaose Forming Amylase and Optimization of Maltopentaose Production (Maltopentaose 생산 균의 분리 및 생산 조건 연구)

  • We isolated a bacterium that produces an extracellular maltopentaose(G5)-forming amylase from amylose and soluble starch. The bacterium was identified and assigned as a Bacillus sp. AIR-5. The amylase did not hydrolyze maltose, maltotriose, maltotetraose or maltopentaose. Optimum medium composition for maltopentaose production in flask culture was 2%(w/v) soluble starch, 0.4%(w/v) tryptone, 0.5%(w/v) NaCl, 0.5%(w/v) K$_2$HPO$_4$, and 3 mM CaCl$_2$at pH 8.0, 28$^{\circ}C$. The highest yield for maltopentaose production in this condition was 6.45 g/L and was 32.55% of theoretical yield.
