Molecular Identification and Sequence Analysis of Coat Protein Gene of Ornithogalum mosaic virus Isolated from Iris Plant

  • A potyvirus was isolated from cultivated Iris plants showing leaf streak mosaic symptom. Reverse transcription and polymerase chain reaction (RT-PCR) product of 1 kb long which encoded partial nuclear inclusion B and N-terminal region of viral coat protein (CP) genes for potyviruses was successfully amplified with a set of potyvirus-specific degenerate primers with viral RNA samples from the infected leaves: The RT-PCR product was cloned into the plasmid vector and its nucleotide sequences were determined. The nucleotide sequence of a CDNA clone revealed that the virus was an isolate of Ornithogalum moseic virus (OrMV) based on BLAST search analysis and was denoted as OrMV Korean isolate (OrMV-Ky). To further characterize the CP gene of the virus, a pair of OrMV-specific primers was designed and used for amplification of the entire CP gene of OrMV-Kr, The virus was easily and reliably detected from virus-infected Iris leaves by using the RT-PCR with the set of virus-specific primers. The RT-PCR product of the CP gene of the virus was cloned and its sequences were determined from selected recombinant CDNA clones. Sequence analysis revealed that the CP of OrMV-Kr consisted of 762 nucleotides, which encoded 253 amino acid residues. The CP of OrMV-Ky has 94.1-98.0% amino acid sequence identities (20 amino acid alterations) with that of other three isolates of OrMV, Two NT rich potential N-glycosylation motif sequences, NCTS and NWTM, and a DAC triple box responsible for aphid transmission were conserved in CPs of all the strains of OrMV. The virus has 58.5-86.2% amino acid sequence identities with that of other 16 potyviruses, indicating OrMV to be a distinct species of the genus. OrMV-Ky was the most related with Pterostylia virus Yin the phylogenetic tree analysis of CP at the amino acid level. This is the first report on the occurrence of OrMV in Iris plants in Korea. Data in this study indicate that OrMV is found in cultivated Iris plants, and may have mixed infection of OrMV and Iris severe mosaic virus in Korea.

Nutritional Properties of Yakju Brewed with Natural Plants (천연식물류를 첨가한 약주의 영양학적 특성)

  • Nutritional characteristics and antioxidation effects of Yakju brewed with natural plants(fine tree leaves, bamboo leaves, ginger, cinnamon and licorice root) were tested. With regard to the contents of phenolic compound, Yakju brewed with natural plants was about 1.7 times higher than Yakju which were not brewed with natural plants. In Yakju brewed with natural plants, lactose content was the highest but maltose was not detected. Minerals were detected in the order of P>K>Mg>Ca>Fe and the content of Fe and Na had a little changes during its 30 days storage. Amino acid contents were getting lower during storage. They were about 28% lower compared 3 days with 30 days storage. Electron donating ability of Yakju and its diluents were tested by DPPH. It was found that the electron donating ability of Yakju was lower as the dilution ratio got higher and as the storage terms got longer. Also, Yakju brewed with natural plants has 95.7∼96.7% of electron donating ability during 30 days storage without regard to its dilution ratio. Superoxide dismutase(SOD) like activity was in inverse proportion to dilution ratio and storage days. The highest SOD like activity was shown 51.5% and 53.9% in 3 days storage Yakju brewed with natural plants and in its 2 times diluents, respectively.

  • This Camptotheca acuminata Decne. (CA), belonging to Nyssaceae, is a deciduous tree. and has been used as Traditional Chinese medicine since ancient times. The CA produces camptothecin a natural indole alkaloid, and reported to have anti-cancer effects. But the studies on biological activities of CA leaves are insufficient. Therefore, this study confirmed various biological activities such as antioxidant, antidiabetic, anticancer, antiinflammatory and metabolism analysis by HPLC-MS/MS of CA leaves. The $RC_{50}$ values of DPPH radical scavenging activity of ethyl acetate fraction, n-Butanol fraction, methanol extraction, water fraction and n-Hexane fraction were $12.23{\pm}0.01$, $15.93{\pm}0.42$, $55.12{\pm}0.45$, $56.29{\pm}4.15$ and $427.29{\pm}6.13ug/mL$, respectively. The $IC_{50}$ values of ${\alpha}$-glucosidase inhibitory activity of ethyl acetate fraction, n-Butanol fraction, methanol extraction, n-Hexane fraction and water fraction were $24.29{\pm}0.14$, $47.86{\pm}0.45$, $54.23{\pm}1.21$ $466.76{\pm}2.21$ and $623.91{\pm}9.67ug/mL$, respectively. The nitric oxide release activity of n-Hexane fraction, methanol extraction, ethyl acetate fraction, water fraction and n-Butanol fraction were $31.49{\pm}5.74$, $29.79{\pm}0.71$, $26.89{\pm}0.71$, $8.24{\pm}5.83$ and $7.75{\pm}4.08%$ at 25 ug/mL, respectively. The anti-cancer activity of n-Hexane fraction, methanol extraction, ethyl acetate fraction, water fraction and n-Butanol fraction were $31.49{\pm}5.74$, $29.79{\pm}0.71$, $26.89{\pm}0.71$, $8.24{\pm}5.83$ and $7.75{\pm}4.08%$ at 25 ug/mL, respectively. The ethyl acetate fraction activities showed higher biological activities than other fractions. Thus, Additional studies were conducted using ethyl acetate fraction. Metabolite analysis was performed using a LCMS-8040 triple quadrupole mass spectrometer. As a result, Five compounds (1-5) were identified in the ethyl acetate fraction of the CA leave. The identification of these compounds was generated by the analysis of fragmentation methods of the negative and positive ion modes. Five compounds were identified as gallic acid (1), chlorogenic acid (2), isoquercetin (3), astragalin (4) and camptothecin (5). These results suggest that the CA leave can be used for functional materials.

Studies on Growth and Biomass Production of Deciduous Tree Seedlings under Different tight Environment (광환경 차이에 의한 활엽수종 유묘의 생장과 물질생산에 관한 연구)

  • The study was carried out to determine the growth and biomass production of deciduous trees including Betula pendula, Fraxinus rhynchophylla, Cornus controversa subjected to artificial shade treatment of three levels in nursery field. The results were as follows; The height growth of seedlings was no difference subjected to artificial shade treatment. The root collar diameters of the relative growth rates of seedlings grown in full sun showed 29-32% as compared with those subjected to the shade treatment of 7-10% light intensities of full sun. Corrus controversa of the root collar diameters of the relative growth rates were better in the seedlings grown in 26-34% light intensities of full sun. Total dry mass including the dry mass of leaves, shoot and root were as a whole decreased with shade treatment. The ratio of the dry mass of leaves and stem increased the dry mass of root. T/R ratio of the seedlings increased by decreasing the relative light intensity. And the T/R ratio of 7-10% light intensities of full sun was ranged from 1.00~2.27 were greater in the full sun light was ranged from 0.51~l.13. Light intensity by artificial shade treatment decreased in deciduous trees when compared on the whole, it showed tendency that SLA increases, increased that seeing resemblant tendency in LAR and LWR and changed of light intensity is strong, it increased that showed difference as statistical.

Antimicrobial Activities of Ethanol Extracts from Korean Bamboo Culms and Leaves (국내산 대나무 줄기와 잎의 에탄올 추출물의 항균활성)

  • Antimicrobial activities in 70% ethanol extracts of Korean bamboo trees were investigated. Among the antimicrobial activities against ten major strains causing food poisening, Wangdae (Phyllostachys bambusoides S. et Z.) showed the strongest activity among five major bamboo trees cultivated in Korea. Although all extracts showed relatively strong antimicrobial activities against Staphylococcus aureus and Streptoccus mutans, the extracts of bamboo culms showed stronger activities than that of bamboo leaves. MICs (minimal inhibitory concentration) of the extracts of Wangdae culm against Staphylococcus aureus and Streptococcus mutans were over $10\;and\;20\;{\mu}L/disc$, respectively. The extract of Wangdae culm inhibited the growth of Staphylococcus aureus and Streptococcus mutans at over 0.5% concentration, and almost inhibited the growth of these strains at 1% level. The inhibitory effect of the extract of Wangdae culm was not destroyed by heating at 60, 80, and $100^{\circ}C$ for 60 min and at $121^{\circ}C$ for 15 min, suggesting that the component in the bamboo tree was very heat-stable. These results indicate that 70% ethanol extract of Wangdae culm could be useful as a natural antimicrobial agent.

First Report of Tomato Spotted Wilt Virus in Oxypetalum coeruleum in Korea (옥시페탈룸에서 발생한 토마토반점위조바이러스 국내 첫 보고)

  • Oxypetalum coeruleum, commonly known as Tweedia, is a perennial herbaceous plant of the Apocynaceae family native to southern Brazil and Uruguay. Tweedia plants are grown as one of the most popular ornamental flowers for floral arrangement in Korea. In May 2021, several tweedia plants in a single greenhouse in Gimje, Jeollabuk-do were found to show virus-like symptoms including necrotic rings, vein-clearing, chlorotic mottle, and mosaic on the leaves, and necrosis on the stems. Here, we have identified tomato spotted wilt virus (TSWV) in symptomatic tweedia leaves by applying high-throughput RNA sequencing. In the result, a single infection by TSWV was verified without mixed infections of different virus species. To confirm the presence of TSWV, a reverse transcription polymerase chain reaction was performed with a specific primer set to the N gene of TSWV. The complete genomic sequence of L, M, and S segments of TSWV 'Oxy' isolate were determined and deposited in GenBank under accession numbers LC671525, LC671638, and LC671639, respectively. In the phylogenetic tree analysis by maximum likelihood method, 'Oxy' isolate showed a high relationship with TSWV 'Gumi' isolate from Gerbera jamesonii in Gyeongsangbuk-do, Korea; for all three RNA segments. To our knowledge, this is the first report of TSWV infection of O. coeruleum in Korea.

A New Method for Cultivation of Sclerotium of Grifola umbellata

  • Sclerotia of Grifola umbellata were cultivated by two methods such as burying and root inoculation methods. The sclerotia of G. umbellata produced by the burying method were $6.0{\sim}6.8{\times}3.4{\sim}4.6{\times}1.8{\sim}1.9cm$(Width$\times$Length$\times$Thickness) in size and $17.3{\sim}19.6g$ in weight, respectively. Their increase rate was $1.10{\times}1.12$ times. On the other hand, the sclerotia cultivated by the root inoculation method were $18.3{\sim}31.5{\times}12.5{\sim}26.4{\times}3.1{\sim}3.7cm(W{\times}L{\times}T)$ in size and $219.1{\sim}576.6g$ in weight, respectively. Their growth increment was $11.18{\sim}39.77$ times. The rhizomorphs of Armillaria mellea were developed with a high density under fallen leaves layer covering cultivation site, and distributed mainly between soil surface and soil depth of about 10 cm as well as colonized prominently on the inoculated wood logs. Fungal interaction between G. umbellata and A. mellea were observed mainly in the stage of white sclerotium of G. umbellata. The sclerotia of G. umbellata which were developed newly and harvested in the root inoculation method were twined with root hairs of host tree and rhizomorphs of A. mellea. The sclerotia of G. umbellata decomposing root hairs of host tree were confirmed through SEM examination. Physiochemical characteristics of soil in all cultivation sites had no significant differences. Soil pH were in the range of pH $3.98{\sim}4.40$. Organic matters were the range of $17.97{\sim}23.86%$ and moisture contents of soil were $12.00{\sim}18.20%$. Soil temperatures showed $12.9{\sim}13.8^{\circ}C$ in November and $22.0{\sim}23.9^{\circ}C$ in August, respectively. In conclusion, the root inoculation method seems to be a practical method for cultivating sclerotia of G. umbellata due to its many advantages such as simplicity of inoculation process, shortening of cultivation periods and facility of harvest.

Defense Strategies against Herbivory of Five Species of the Genus Viburnum (가막살나무속 5 수종의 초식에 대한 방어전략)

  • To explore on the defense strategies against hervivory of five Viburnum species, morphological characteristics of the leaf, leaf domatia structure and the number, herbivores insects and mites on the leaves, collected from the trees growing in Wonju-si, Pyeungchang-gun, Taebaek-si, Taean-gun, Bonghwa-gun, Sancheung-gun and Jinju-si were investigated from May 2009 to October 2011. Domatia of V. carlesii reveals pocket type, these of V. burejaeticum, V. dilatatum and V. wrightii reveal tuft type, and that of V. odoratissimum var. awabuki reveals pouch type. Domatia number per leaf proves the highest figures, 24.0/leaf for V. burejaeticum, and the lowest, 4.9/leaf for V. carlesii. Leaf surface trichomes of four Viburnum species except for V. odoratissimum var. awabuki are mainly stellate and hispid on the leaf-blade and veins, and dense stellate trichomes on the upper leaf surface of V. carlesii, V. burejaeticum, and V. dilatatum are observed till late growing season. Extrafloral nectaries(EFN) on the terminal veins of five Viburnum species are observed. Pellucid dots on the lower leaf surface of V. dilatatum and V. wrightii might be a good defense strategies against herbivores. We observe many predatory mites eating nectar on large pellucid dots of V. dilatatum. Predatory mites number per leaf proved highly significant differences among tree species, and mean of mites number was highest values 8.1/leaf for V. burejaeticum, and the lowest 2.6/leaf for V. odoratissimum var. awabuki. These results indicate that temperate broadleave trees develop various defense strategies against herbivores for survival. The more studies on the defense strategies against herbivores of tree species might be needed for sustainable forest ecosystem.

Injury Responses of Landscape Woody Plants to Air Pollutants - Malondialdehyde content - (조경수목(造景樹木)의 대기오염물질(大氣汚染物質)에 대한 피해반응(被害反應)(III) - Malondialdehyde 함량(含量)을 중심으로 -)

  • This study was conducted to investigate sensitivity of woody plants growing in urban and industrial regions of Seoul and Taejon, Korea. Malondialdehyde(MDA) contents were analyzed in tree foliage of Pinus densiflora, Pinus koraiensis, Ginkgo biloba, Liriodendron tulipifera and Platanus occidentalis. In addition, MDA contents were analyzed in the foliage of tree seedlings, i.e. Pinus densiflora. Pinus koraiensis, Ginkgo biloba and Liriodendron tulipifera, with the fumigation of $SO_2$ in gas chamber 4 hours a day for six days. MDA contents of leaves in Ginkgo biloba, Platanus occidentalis and Liriodendron tulipifera in polluted regions were higher than those in unpolluted region and among them Liriodendron tulipifera had the highest. MDA contents of Pinus densiflora and Pinus koraiensis increased with the increase of needle age. MDA contents of Liriodendron tulipifera seedlings were increased with the higher concentrations of $SO_2$ but MDA contents in other seedlings showed no changes with $SO_2$ treatment concentrations. MDA contents in all species were increased with the passage of exposure day. Especially. Liriodendron tulipifera had higher MDA content than other species. In Liriodendron tulipifera the MDA production increased with the passage of exposure day until the fourth day after that decreased.

Disease Management in Road Trees and Pepper Plants by Foliar Application of Bacillus spp. (Bacillus spp. 엽면살포에 의한 가로수 및 고추의 병 방제)

  • Out of plant-associated bacteria, certain plant growth-promoting bacteria (PGPB) have been reported to increase plant growth and productivity and to elicit induced resistance against plant pathogens. In this study, our objective was to broaden the range of applications of leaf-colonizing PGPB for foliar parts of road tress and pepper. Total 1,056 isolates of endospore-forming bacteria from tree phylloplanes were collected and evaluated for the enzymatic activities including protease, lipase, and chitinase and antifungal capacities against two fungal pathogens, Colletotrichum graminicola and Botrytis cinerea. Fourteen isolates classified as members of the bacilli group displayed the capacity to colonize pepper leaves after spraying inoculation. Three strains, 5B6, 8D4, and 8G12, and the mixtures were employed to evaluate growth promotion, yield increase and defence responses under field condition. Additionally, foliar application of bacterial preparation was applied to the road tress in Yuseong, Daejeon, South Korea, resulted in increase of chlorophyll contents and leaf thickness, compared with non-treated control. The foliar application of microbial preparation reduced brown shot-hole disease of Prunus serrulata L. and advanced leaf abscission in Ginkgo biloba L. Collectively, our results suggest that leaf-colonizing bacteria provide potential microbial agents to increase the performance of woody plants such as tree and pepper through spray application.
