Liver enzyme elevations, as an indicator of liver function, are widely associated with metabolic diseases. Genome-wide population-based association studies have identified a genetic susceptibility to liver enzyme elevations and their related traits; however, the genetic architecture in childhood remains largely unknown. We performed a genome-wide association study to identify new genetic loci for liver enzyme levels in a Korean childhood cohort (n = 484). We observed three novel loci (rs4949718, rs80311637, and rs596406) that were multiply associated with elevated levels of alanine transaminase and aspartate transaminase. Although there are some limitations, including genetic power, additional replication and functional characterization will support the clarity on the genetic contribution that the ST6GALNAC3, ADAMTS9, and CELF2 genes have in childhood liver function.
This study was performed to evaluate the biological activity and protective effect of Cassia tora ethanol extracts against D-galactosamine induced hepatotoxicity in rats. Male Sprague-Dawley rats were grouped into normal group, D-galactosamine treated group(control), D-galactosamine plus 0.25% Cassia tora extracts treated group and D-galactosamine plus 0.5% Cassia tora extracts treated group. Normal and control group were fed control diet and Cassia tora extracts treated groups were fed experimental diets containing 0.25% or 0.5% Cassia tora ethanol extracts for 5 weeks. Body weight gain and liver weight of rats were not significantly different between groups. Cholesterol and triglyceride concentrations in serum and liver were significantly lower in rats treated only with D-galactosamine compared to normal group, and improved in Cassia tora extracts supplemented rats. D-galactosamine treatment significantly increased serum aspartate transaminase, alanine transaminase, gamma glutamyl transferase and alkaline phosphatase, however, the activities of aspartate transaminase and alanine transaminase were significantly decreased in Cassia tora extracts supplemented rats when compared with D-galactosamine treated control group. Cassia tora extracts significantly suppressed the D-galactosamine-induced elevation of liver thiobarbituric acid reactive substances(TBARS) contents. Superoxide dismutase activity was decreased by D-galactosamine treatment, however by the supplementation of Cassia tora ethanol extracts, significantly increased in dose-dependent manner. Glutathione peroxidase activity in rats fed diets containing Cassia tora extracts was decreased compared to control. Based on these results, we concluded that Cassia tora ethanol extracts may prevents the D-galactosamine-induced hepatotoxicity probably via an antioxidant mechanism.
Hepatoprotective effects of polysaccharides extracted from liquid cultured mycelia were screened by measuring the glutamic pyruvate transaminase activity of the primary cultured rat hepatocytes intoxicated with carbon tetrachloride. Sixty among 75 isolates of higher fungi showed to be hepatoprotective effect, and these were 13 of Ganoderma lucidum, 5 of Lentinus edodes, 1 of Pleurotus ostereatus, 4 of Coriolus versicolor, 2 of Lyophyllum spp., 7 of Grifora frondosa, 3 of Agaricus spp., 14 of Schizophyllum commune and 11 of Cordyceps spp.. Especially, 10 isolates, Ganoderma lucidum IY003 and IY009, Lentinus edodes IY103, Lyophyllum sp. IY402, Agaricus sp. IY701 and IY703, Schizophyllum commune IY804, IY810 and IY818, Cordyceps sp. IY902, were indicated below 80% of glutamic pyruvate transaminase activity.
In order to determine the influence of low atmospheric pressure on serum glutamic oxaloacetic transaminase (SGOT) and serum lactic dehydrogenase (SLDH) activities of rats, blood samples were collected from laboratory-conditioned male rats of the Sprague-Dawley strain which were randomly grouped into control and the experimental subjected to a series of one hour-exposure a day to low atmospheric pressure of 500 and 380mmHg up to the the time of 15 day. Results obtained indicated that decompression caused marked alterations in SGOT and SLDH levels when compared to that of the control. The trend of increases or decreases in these enzyme levels were similar in both 500 and 380mmHg exposed rats although the changes were greater in the latter group. Thus, generally all the experimental rats showed temporary steady state to low atmospheric pressure. Changes in enzymatic contents depended on the intensity and extent of the environmental stress under study. The lower the atmospheric pressure the greater is the effect on these serum enzyme levels.

A derivatization method in mass spectrometry for small molecular analysis was developed to solve the problems of volatility of many analytes, difficult ionization of analytes, and undiscriminating isobaric analytes. This derivatization method, oximation of $\alpha$-ketoacid, in the transaminase reaction leads to change of mass difference between amine reactants and ketoacid products. In addition, regardless of the kinds of ketoacid, the linear relationship between the peak intensity and its concentration (from 1 mM to 10 mM) shows that quantitative analysis of the conversion of the reaction can be executed by the analysis of peak intensity of the corresponding oximated ketoacids. Furthermore, this method can be used for identifying transaminase as well as determining its substrate specificity
Effect of Quartz porphyry (QP) on clinical chmical parameters in male common finch by feeding the basal diet (Control group) containing 3.0% QP (QP group) for 14 days was studied. Body weight gain, feed intake, feed efficiency, the relative weight (mg/10 g body wight) of liver and heart were not significantly different in the both groups. However, the relative weights of kidney and gizzard in the QP group were higher and lower than in the control group, respectively. The contents of triglyceride, cholesterol, total protein, glucose and albumin, and the activity of glutamic pyruvic transaminase (GPT) in serum were not significantly different in the both groups. The activities of alkaline phosphatase (ALP) and glutamic oxaloacetic transaminase (GOT) in serum were lower and higher in the QP group than in the control group.

Mesorhizobium sp. LUK, which utilizes 3-amino-3-phenylpropionic acid as the sole source of nitrogen with high enantioselectivity (E(S)>100), was isolated using enrichment culture. The enzyme involved in the utilization of (S)-3-amino-3-phenylpropionic acid was confirmed to be a transaminase and was purified by 235-folds with a specific activity of 0.72 U/mg. The molecular weight of the purified protein was ca. 47 kDa and the active enzyme was determined as a dimer on gel filtration chromatography. The N-terminal sequence was obtained from the purified protein. Spontaneous decarboxylation of produced ${\beta}-keto$ acids was observed during the chiral resolution of 3-amino-3-phenylpropionic acid.

Glutamate decarboxylase catalyzes the conversion of glutamate to gamma-aminobutyric acid (GABA), contributing to pH homeostasis through proton consumption. The reaction is the first step toward the GABA shunt. To date, the enzymes involved in the glutamate metabolism of Photobacterium damselae subsp. piscicida have not been elucidated. In this study, an open reading frame of P. damselae subsp. piscicida, showing homology to the glutamate decarboxylase or putative pyridoxal-dependent aspartate 1-decarboxylase genes, was isolated and cloned into an expression vector to produce the recombinant enzyme. Preliminary gas chromatography-mass spectrometry characterization of the purified recombinant enzyme revealed that it catalyzed not only the decarboxylation of glutamate but also the transamination of GABA. This enzyme of P. damselae subsp. piscicida could be bifunctional, combining decarboxylase and transaminase activities in a single polypeptide chain.

GABA transaminase is inactivated by preincubation with p$^1$, p$^2$-bis(5'-pyridoxal) diphosphate at pH 7.0. The inactivation under pseudo-first order conditions proceeds at a slow rate (K$\_$obs/=0.035 min$\^$-1/). The degree of labeling of the enzyme by p$^1$, p$^2$-bis(5'-pyridoxal) diphosphate was determined by absorption spectroscopy, The blocking of 2 lysyl residues/dimer is needed for inactivation of the transaminase. The time course of the reaction is significantly affected by the substrate ${\alpha}$-ketoglutarate, which afforded complete protection against the loss of the catalytic activity. Whereas cofator pyridoxal phosphate failed to prevent the inactivation of the enzyme. Therefore, it is postulated that binding of ${\alpha}$-ketoglutarate tn lysyl residues is the major factor contributing to stabilization of the catalytic site and bifuctional reagent p$^1$, p$^2$bis(5'-pyridoxal) diphosphate blocks lysyl residues other than those involved in the binding of the cofactor.
We have developed a polyculture container which is preferable for rearing of abalones and sea cucumbers in East Sea. To test the rearing capacity of the polyculture container, 50, 75 and 100 sea cucumbers ranging from the body length of 7-9 cm were cultured in three containers including 500 abalones with the shell length of about 5 cm, respectively. It was revealed that preferable density for sea cucumbers was 52-72 individuals in the polyculture container. Glutamate oxaloacetate transaminase and glutamate pyruvate transaminase were analyzed with Reflotron kit to investigate the health degree of abalones in two polyculture containers including 300 and 500 abalones with the shell length of about 5 cm. Glutamate oxaloacetate transaminase has been an important tool to know myocardial infarction, disease of liver, and destruction of muscle. Next experiment was conducted to determine the effects on growth of abalones and sea cucumbers in polyculture container. Experimental findings, RNA/DHA ratio, DNA and RNA contents (ug/mg) were not significantly different among all groups. The results imply that the method of polyculture can be rearing with sea cucumber without growth retardation of abalone. Production ability between polyculture container and the container used in south sea were carried out using suspended culture method during 8months. It was revealed that abalones and sea cucumbers are faster growing in polyculture container than in container used in south sea. Therefore, polyculture container is considered more appropriate for the abalones and sea cucumbers culture in East Sea.
