Glucose-6-phosphate dehydrogenase를 이용한 Moina macrocopa의 중금속 독성 검정 (Heave Metal Toxicity Test in Moina macrocopa with Glucose-6-phosphate Dehydrogenase Activity)

  • A rapid, inexpensive enzymatic method is proposed for indirect water quality testing in terms of heavy metal toxicity. The activity of glucose-6-phosphate dehydrogenase was applied for heavy metal toxicity test as an effective criterion in water quality. The toxicity of Pb (lead) and Cd (cadmium) for water flea, Moina macrocopa, were evaluated for 2-8 days with variables of mobilization ability. And the reproduction impairment of Moina macrocopa were investigated as the parameter of chronic toxicity test for Pb and Cd. As a result, the EC$_{50}$ for immobilization of Moina macrocopa were Pb and Cd were 1.6749 and 0.4683, respectively. The values of reproductive impairment to Moina macrocopa for Pb and Cd were 9.5938 and 8.3264 in EC$_{50}$ A significant alteration of G6PDH (Glucose-6-phosphate dehydrogenase) activity of Moina macrocopa was observed when Cd and Pb were treated in media. The results obtained indicate that G6PDH activity of Moina macrocopa can be used as an indicative parameter in aquatic toxicity tests for heavy metals.als.

  • We developed and evaluated a method of short-term acute toxicity testing based on the feeding behavior of Ceriodaphnia dubia. In prior toxicity tests, neonates of C. dubia were hatched and cultivated with the addition of yeast only for the preparation of the transparent daphnid's gut. Scenedesmus subspicatus was supplied as food after 1 to 6 h of exposure to toxicants. The effects of 1-h and 6-h exposure time on test sensitivity did not significantly differ. A comparison of the short-term l-h acute toxicity test developed in this study to the standard 48-h acute toxicity test using heavy metals, cyanide, and pentachlorophenol indicated that the 1-h test provided an acceptable sensitivity level in toxicity testing of C. dubia..

  • Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) are fully flurorinated organic compounds which are highly persistent in environment and accumulated in organism. These chemicals are released to the environment at their manufacture, during their use in industrial and consumer applications and from disposal after their use. The purpose of this study was to determine the effects of PFOA and PFOS on the freshwater flea (Moina macrocopa). Acute toxicity test and chronic toxicity test were performed for 2 days and 10 days, respectively. Acute toxicity was assessed on the basis of mortality, while chronic toxicity was assessed by fecundity as well as mortality. The acute toxicity studies on PFOA and PFOS showed that the values of $LC_{50}$ were $73.9\;mg/L\;and\;27.7\;mg/L$ respectively. In the chronic toxicity test, fecundity was reduced significantly at 24.1 mg/L of PFOA and 9.3 mg/L of PFOS, respectively. Conclusively, the results of this work suggest that Moina macrocopa could be a suitable model organism for screening and assessing of environmental pollutants in water.

폐광산 배수와 퇴적물의 중금속 오염과 생물독성 평가 (Assessment of Heavy Metal Contamination and Biological Toxicity of Mine Drainages and Sediments from Abandoned Mines)

  • Heavy metal contamination and biological toxicity of mine drainages and sediments in abandoned mines were studied. Mine drainages had pH at a range of 2.94~7.86, and contained heavy metals at a toxic level. For coal mines, toxicity of mine drainage to Daphnia magna was attributable to acidic pH. In addition to the low pH, suspended heavy metals such as Zn and Cu contributed to toxicity of mine drainages at abandoned metalliferous mines. All mine sediments studied in this work showed biological toxicity to Chironomus riparius, having mortality at a range of 15~60%. However, its relationship with physicochemical properties including heavy metal content of the sediments was not statistically explained. Exceptionally clay ($< 2{\mu}m$ particle) content was negatively correlated with the biological toxicity for sediment samples collected at the same abandoned mines.

Ibuprofen Increases the Hepatotoxicity of Ethanol through Potentiating Oxidative Stress

  • Over 30 million prescriptions of NSAIDs (non-steroidal anti-inflammatory drugs) are issued every year. Considering that these drugs are available without a prescription as over the counter (OTC) drugs, their use will be astronomical. With the increasing use of NSAIDs, their adverse effects are drawing attention. Especially, stomach bleeding, kidney toxicity, liver toxicity, and neurological toxicity are reported as common. Ibuprofen, one of the extensively used NSAIDs along with aspirin, can also induce liver toxicity, but few studies are addressing this point. Here we examined the liver toxicity of ibuprofen and investigated whether co-exposure to ethanol can manifest synergistic effects. We employed 2D and 3D cultured human hepatoma cells, HepG2 to examine the synergistic hepatotoxicity of ibuprofen and alcohol concerning cell viability, morphology, and histology of 3D spheroids. As a result, ibuprofen and alcohol provoked synergistic hepatotoxicity against hepatocytes, and their toxicity increased prominently in 3D culture upon extended exposure. Oxidative stress appeared to be the mechanisms underlying the synergistic toxicity of ibuprofen and alcohol as evidenced by increased production of ROS and expression of the endogenous antioxidant system. Collectively, this study has demonstrated that ibuprofen and EtOH can induce synergistic hepatotoxicity, providing a line of evidence for caution against the use of ibuprofen in combination with alcohol.

Clinical Characteristics and Treatment of Immune-Related Adverse Events of Immune Checkpoint Inhibitors

  • Immune checkpoint inhibitors (ICIs) have been changing the paradigm of cancer treatment. However, immune-related adverse effects (irAEs) have also increased with the exponential increase in the use of ICIs. ICIs can break up the immunologic homeostasis and reduce T-cell tolerance. Therefore, inhibition of immune checkpoint can lead to the activation of autoreactive T-cells, resulting in various irAEs similar to autoimmune diseases. Gastrointestinal toxicity, endocrine toxicity, and dermatologic toxicity are common side effects. Neurotoxicity, cardiotoxicity, and pulmonary toxicity are relatively rare but can be fatal. ICI-related gastrointestinal toxicity, dermatologic toxicity, and hypophysitis are more common with anti- CTLA-4 agents. ICI-related pulmonary toxicity, thyroid dysfunction, and myasthenia gravis are more common with PD-1/PD-L1 inhibitors. Treatment with systemic steroids is the principal strategy against irAEs. The use of immune-modulatory agents should be considered in case of no response to the steroid therapy. Treatment under the supervision of multidisciplinary specialists is also essential, because the symptoms and treatments of irAEs could involve many organs. Thus, this review focuses on the mechanism, clinical presentation, incidence, and treatment of various irAEs.

  • The following is experimental result of selecting soil bacteria showing toxicity against Root-knot nematode (Meloidogyne hapla). Out of 286 strains isolated from soil, one(NC67) showing toxicity against M.hapla is selected The selected strain(NC67) is identified of B. thuringiensis subsp. indiana. It proved out that the toxic maerial against M. hapla produce by NC67 strain is an exotoxin. The result of examining the existence of the extercellular toxicity product by the toxic strain(NC67) by usign activated carbon column chromatography, Dowex 50W column chromatography and TLC of silical gel etc. proved out that it is a single material.

연명피 사포닌의 독성 및 용혈작용 (Toxicity and Hemolytic Activitiy of Saponin Isolated from Sapindus mukorossi)

  • Saponin isolated from Sapindus mukorossi Gaertn has been shown to contain a strong anti-inflammatory activity. In this study, several pharmacological properties such as acute toxicity, local irritation and hemolytic activity of Sapindus saponin and its genin component, hederagenirl, were examined. The acute toxicity of Sapindus saponin was very low. Estimated from the LD$_{50}$ values, it showed much weaker toxicity in oral administration than in intraperitoneal injection. Hederagenin gave a very high LD$_{50}$ value even in intraperitoneal injection. Sapindus saponin showed a potent local irritation after topical application, whereas hederagenin did a very weak local irritation. Sapindus saponin also gave a high hemolytic activity.

Carbosulfan과 Phenthoate의 송사리(Oryzias latipes, Medaka)에 대한 독성시험 (Toxicity Test of Carbosulfan and Phenthoate on Killifish)

  • 우리나라에서 널리 사용되고 있는 벼재배용 살충제인 carbosulfan과 phenthoate의 담수어에 미치는 급 만성적 영향을 알아보고자 송사리(Oryzias latipes, Medaka)에 대한 급성독성시험(Acute Toxicity Test)과 생육초기 독성시험(Early-life Stage Toxicity Test)을 실시하였다. 급성독성시험 결과 carbosulfan과 phenthoate의 송사리에 대한 96시간 $LC_{50}$은 0.102, 0.167 mg/L로 비슷한 감수성을 보였다. 생육초기 독성시험 결과 carbosulfan의 경우 부화율은 80%이상, 부화기간은 $9\sim10$일로 나타나 처리구간 유의성이 없었다. 처리농도 0.042 mg/L이상에서는 부화후 생존율이 통계적으로 유의한(p<0.05) 차이를 보였으며, 척추가 휘어지는 형태적으로 비정상적인 결과가 관찰되었다. 시험 종료후 생존한 송사리의 전장 및 체중 변화는 농도 상관성을 보였으며, 특히 0.017 mg/L이상에서는 대조구에 비해 유의한(p<0.05) 감소를 나타내었다. 따라서 이런 결과를 근거로 한 무영향농도(NOEC), 최저영향농도(LOEC), 그리고 최대허용농도(MATC)는 각각 0.0067, 0.017, 0.011 mg/L으로 산출하였다. phenthoate 처리구의 부화율과 부화기간은 처리 농도간 유의성이 없었으나, 부화후 생존율은 처리농도 0.076 mg/L 이상에서 유의한(p<0.05) 차이를 보였다. 또한 처리농도 0.076 mg/L 이상에서는 척추기형과 유영이상을 보이는 이상어가 나타났다. 그러나 시험 종료후 송사리 전장 및 체중의 변화는 통계적인 유의한 차이가 없었으며, 농도 상관성도 보이지 않았다. 따라서 이런 결과를 종합하여 NOEC, LOEC, 그리고 MATC는 각각 0.011, 0.029, 0.018 mg/L으로 결정되었다.
