Antioxidant Activities of Amaranth (Amaranthus spp. L.) Flower Extracts (아마란스 꽃 추출물의 항산화에 관한 연구)

  • This study investigates the free radical-scavenging activities of Amaranth (Amaranthus spp. L.) red and purple flower extracts. The methanol and hot water extracts of flower are being evaluated for its total polyphenol and flavonoid contents, scavenging activities by the DPPH and ABTS analysis, SOD-like activity, and inhibition activities of superoxide radical on the HL-60 cells and nitric oxide of the RAW 264.7 cells. The PFM (purple flower extracted with MeOH) showed the highest total phenolic and flavonoid content, 606.95 mg GAE/100 g and 254.69 mg CE/100 g, respectively. Amongst the scavenging activities of the DPPH radicals, PFM($RC_{50}=155.06{\mu}g/m{\ell}$) is the highest of all the samples. The ABTS radical-scavenging activity is also highest for PFM (53.16%) at the $250{\mu}g/m{\ell}$ concentration. But, the SOD-like activity of the PFW (purple flower extracted with hot water) increases more than 3 folds of the PFM. In the leukemia HL-60 cell, the PFM shows strongly inhibited superoxide radical generations at a concentration of $200{\mu}g/m{\ell}$ at 72.34%, which increases with 1.79 folds more than the RFW (red flower extracted with hot water). The inhibition activity of nitric oxide in Raw 264.7 cells is the highest for PMF (46.90%) at a $250{\mu}g/m{\ell}$ concentration. In conclusion, PMF show the highest flavonoid contents and the most powerful free radical-scavenging activity. Our results suggest that the increase of antioxidant activities depend on flavonoid contents. Thus, Amaranth flower can be useful for natural antioxidant compounds.

Antioxidant and Physicochemical Changes in Salvia plebeia R. Br. after Hot-air Drying and Blanching (열풍건조 및 데침 처리에 따른 배암차즈기(Salvia plebeia R. Br.)의 항산화 및 이화학적 특성 변화)

  • The aim of this study was to investigate physicochemical changes in Salvia plebeia R. Br. (SPA) upon hot-air drying and blanching. After hot-air drying and blanching, total polyphenol and flavonoid contents were reduced from 96.64 mg and 48.40 mg gallic acid equivalent/g to 29.70 mg and 22.10 mg quercetin equivalent/g, respectively. DPPH radical scavenging activities at $25{\mu}g/mL$ of SPA were 94.5% for ascorbic acid, 84.3% for hot-air dried SPA, and 59.7% for blanched SPA and there was no significant difference between those of hot-air dried SPA and ascorbic acid as a positive control. Total sugar contents were 7.187% and 6.104% for hot-air dried SPA and blanched SPA, respectively. During the blanching process, sucrose and maltose contents decreased, whereas glucose and fructose contents increased. Glucose and fructose were converted into citric acid in blanched SPA, whereas sucrose and maltose were converted into tartaric acid, malic acid, and succinic acid. Fourteen kinds of amino acids were found, but methionine, lysine, glycine, histidine, and cysteine were absent. The proportion of essential amino acids was 61.76% upon hot-air drying, which was a relatively high amount. In addition, hot-air drying resulted in 1.40 mg/100 g of norvaline as well as 39.00 mg/100 g of GABA, which are non-amino acids. Therefore, Salvia plebeia R. Br. can be used not only as vegetables but also as highly useful and various health functional foods with antioxidant effects and excellent nutrition.

Quality Characteristics of Yakju (Korean Traditional Rice Wine) Added with Bamboo Ethanol Extract (대나무 알코올추출액을 첨가한 약주의 품질특성)

  • This study was carried out to develop Yakju, a new kind of Korean traditional rice wine added with bamboo ethanol extract and characterized by containing various health benefits. The wine with bamboo ethanol extract was fermented by adding ginger and pear to improve flavor and quality. Several quality characteristics and physiological functionalities of Yakju were investigated. When bamboo ethanol extract was added to Yakju, the pH level decreased compared to that of the control. However, total acidity and soluble solid levels were higher than those of the control. The alcohol content of Yakju added with bamboo ethanol extract was slightly higher than that of the control. In examining the colors of Yakju containing bamboo ethanol extract, L value was lower than that of the control, whereas b value and brownness were higher. Various organic acids such as citric acid, malic acid, succinic acid, lactic acid, and acetic acid were determined, and the main organic acid among them was lactic acid. Various free sugars such as sorbitol, glucose, and fructose were detected and glucose was the most abundant. Total polyphenol content of Yakju added with bamboo ethanol extract was lower than that of the control. Further, electron-donating ability ranged from 23.8~40.0%, which was higher than that of the control. Nitrite scavenging ability of Yakju added with bamboo ethanol extract ranged from 15.2~36.4%, which was higher than that of the control at pH 6.0. Sensory evaluation of Yakju showed that bamboo ethanol extract+ginger as well as bamboo ethanol extract+ginger+pear treatment significantly increased flavor, taste, and overall acceptance compared to the others.

Antioxidant and Anti-Proliferative Activities of Oats under Different Solvent Extraction Conditions (추출용매별 귀리의 항산화 및 암세포 증식 억제 활성)

  • The objective of this study was to determine the antioxidant and anti-proliferative activities of methanol, ethanol, acetone, and ethyl acetate extracts from oats (Avena sativa L.). Total polyphenol contents of extracts were analyzed by Folin-Ciocalteu assay. The antioxidant activities of extracts were determined by 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities and reducing power. The anti-proliferative activities of colon (HCT116), lung (NCI-H460), and breast (MCF7) cancer cells were investigated. Among solvents, methanol extract showed the highest amount of total polyphenols, which was 8.2 mg gallic acid equivalents/g residue. High levels of ABTS radical [12.1 mg Trolox equivalent antioxidant capacity (TEAC)/g residue] and DPPH radical (4.4 mg TEAC/g residue) scavenging activity and reducing power ($A_{700}=0.39$) were found in methanol extracts. Moreover, methanol extracts indicated higher anti-proliferative activities against HCT116 (69.5%), NCI-H460 (75.2%), and MCF7 (84.8%) cells compared with other extracts. The results show that methanol was the best solvent for extraction of antioxidant and anti-proliferative compounds from oats. Moreover, notable antioxidant and anti-proliferative activities of oats could have significant health benefits.

Development of Functional Vinegar by Using Cucumbers (오이를 이용한 기능성 식초 음료 개발)

  • This study was performed to develop functional vinegar by using cucumbers through two stages of fermentation. The alcohol content was maximized (7.8%) after 6-days of alcohol fermentation at $25^{\circ}C$ by adjusting the initial sugar concentration to $15^{\circ}Brix$, and vinegar with an acidity of 5.8% was obtained after 12-days of acetic acid fermentation at $30^{\circ}C$. The major sugars in the produced vinegar were glucose and fructose, which were present in concentrations of 3,067.26 and 395.73 mg%, respectively. The major organic acids were acetic acid and succinic acid, which were present in concentrations of 4,410.5 and 841.11 mg%, respectively. The total free amino acid content of the cucumber vinegar was 181.45 ${\mu}g/mL$ and citrulline, valine, aspartic acid, asparagine, and ornithine were the major amino acids. The inorganic components included various alkaline elements, such as K, Ca, and Mg. In addition, experimental methods to assess the DPPH and $ABTS^+$ radical-scavenging ability, reducing power, and ${\beta}$-carotene bleaching activity showed that the cucumber vinegar had strong antioxidant properties. The total polyphenol content, which are the major components responsible for the antioxidant activities of the cucumber vinegar, was 40.14 mg/100 mL. The cucumber vinegar showed significantly higher hepatic aldehyde dehydrogenase activity when compared to the alcoholic control (negative) and the marketing drink (positive), resulting in decreased plasma acetaldehyde concentrations in rats. These results demonstrate that cucumber vinegar possesses antioxidant properties and holds great promise for use in preventing hangovers.

Antioxidant Activity and Anti-hyperglycemic Activity of Medicinal Herbal Extracts According to Extraction Methods (약용식물의 추출방법에 따른 항산화 및 항당뇨 활성)

  • Korean traditional medicinal herbs have been reported to possess antioxidant and anti-hyperglycemic activities. We tested the antioxidant and anti-hyperglycemic activities of 6 kinds of medicinal herbs: Angelica gigas N., Poria cocos, Mori radicis Cortex, Mori folium, Aralia elata Cortex, and Panax ginseng, prepared as hot water, ethanol, and sonication extracts. The antioxidant activities of the extracts were examined by performing total polyphenol, total flavonoid, and ${\alpha}$,${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) assays. For M. folium, the ethanol extract showed the strongest effects in DPPH radical scavenging activity among the three extraction methods. In addition, sonication extracts of M. radicis Cortex and M. folium showed the highest inhibitory activities for ${\alpha}$-glucosidase among the different extracts. The ethanol extracts of M. folium had the highest inhibition effects against ${\alpha}$-amylase. A direct correlation between antioxidant and anti-hyperglycemic inhibition activity was found in the ethanol and sonication extracts. From the results, it is considered that these six medicinal herbal extracts have antioxidative, anti-hyperglycemic, and correlation effects based on different extraction methods.

Antioxidant activities of hot water extract of Syneilesis palmata root and aerial part (우산나물 뿌리와 지상부 열수 추출물의 항산화 활성)

  • This study was performed in order to investigate the antioxidant properties of hot water extract from the root and aerial part of the Syneilesis palmata in respect to its potential use as food, cosmetics material, or medicinal resource. The results showed that the S. palmata root hot water extract (RHW) possessed a higher content of total flavonoid compounds (4.58 mg/g) and total polyphenol compounds (59.11 mg/g). The SOD-like activities of the RHW and APHW were 23.74% and 21.61%, respectively, at a concentration $2,000{\mu}g/mL$. In the nitrite scavenging ability of a $2,000{\mu}g/mL$ concentration, the RHW showed 63.06% (pH 1.2) and 47.16% (pH 3.0). The $IC_{50}$ values of the nitrite scavenging abilities were $99.93{\mu}g/mL$ (ascorbic acid), $1,150.85{\mu}g/mL$ (RHW), and $1,610.25{\mu}g/mL$ (APHW). The $IC_{50}$ values of DPPH free radical scavenging abilities were $99.87{\mu}g/mL$ (RHW) and $118.29{\mu}g/mL$ (APHW). The inhibition values ($IC_{50}$) of xanthine oxidase were $139.62{\mu}g/mL$ (RHW) and $111.11{\mu}g/mL$ (APHW). In all of the experiments, the S. palmata root hot water extracts have higher activities than the aerial hot water extract, except for the xanthine oxidase inhibitory activity. These results suggest that the S. palmata is a potentially useful antioxidant source for the development of functional nutraceuticals, cosmetics and medicine.

Effect of black garlic on antioxidant activity and amino acids composition in Cheonggukjang (흑마늘 첨가에 따른 청국장의 항산화 및 아미노산의 변화)

  • The antioxidant activity and amino acid composition of various Cheonggukjang extracts, such as the water extract from Cheonggukjang (CWE), the alcohol extract from Cheonggukjang (CEE), the water extract from Cheonggukjang with black garlic (BWE) and the alcohol extract from Cheonggukjang with black garlic (BEE), were examined to investigate the effect of black garlic on the sensory quality and functional properties of Cheonggukjang. The total polyphenol contents of various extracts were 7.03 mg/100 g (BWE), 3.64 mg/100 g (CWE), 2.88 mg/100 g (BEE) and 0.81 mg/100 g (CEE). The radical scavenging activity of the DPPH radical was highest in BWE (91.83%), followed by BEE (37.35%), CWE (25.54%) and CEE (14.80%), in that order. The SOD-like activity was highest in BWE (20.20%), followed by BEE (9.22%), CWE (7.91%) and CEE (6.45%). The thiobarbituric acid reactive substances (TBARS) were highest in BWE (35.18%), followed by BEE (28.33%), CWE (17.40%) and CEE ((14.93%). The total amino acid content of Cheonggukjang (CC) was higher than that of Cheonggukjang with black garlic (CCWB), but the essential amino acid content of CCWB (43.18%) was higher than that of CC (42.27%). The 27 kinds of free amino acid were found in CC, but only 23 kinds were found in CCWB. The L-lysine content was highest (9.23%) in CC, and the L-phenylalanine content was highest (23.14%) in CCWB. The free amino acids (L-threonine, L-serine, L-sarcosine, L-proline, L-alanine, L-valine and D,L-${\beta}$-aminoisobutyric acid) were found in CC but not in CCWB. The ${\gamma}$-amino-n-butyric acid (GABA) was found in CCWB but not in CC. These results suggest that the addition of black garlic has beneficial effects on the functionality of Cheonggukjang without decreasing its sensory characteristics.

Phytochemical compounds and quality characteristics of spray-dried powders with the blanching condition and selected forming agents from pressed extracts of Ligularia fischeri leaves (블랜칭 처리 및 부형제 종류에 따른 곰취 착즙액 분무건조 분말의 phytochemical 성분 및 품질특성)

  • This study was performed to determine the effects of the blanching condition (immersion ratio 1:15 (w/v) for 3 min at $95^{\circ}C$, and solution containing 1% sodium chloride) and selected forming agents (dextrin DE=10, dextrin DE=20, ${\beta}$-cyclodextrin; each forming agents added 5%) on the phytochemical compounds and quality characteristics of Ligularia fischeri leaves. The moisture was not affected by the forming agent. The color of a, b and chroma values were low in the blanching treatment groups and were significantly lowest with ${\beta}$-cyclodextrin (CD). The polyphenol and flavonoid contents in the blanching treatment groups were higher than those in the non-blanching-treatment group. The ascorbic acid content was higher in the non-blanching-treatment group and was significantly highest in the group treated with dextrin (DE=10) whereas the blanching treatment groups showed lower dehydroascorbic acid content than the non-blanching-treatment group. The water absorption was higher in the non-blanching-treatment group and was significantly highest in the group treated with CD. The water solubility in the blanching treatment groups treated with dextrin (DE=20) and CD was higher than that in the blanching treatment group treated with DE=10. The total chlorophyll and chlorophyll a and b contents were high in the blanching treatment group treated with CD, and for the total carotenoid contents, the same tendency as that seen with the chlorophyll content was observed. With regard to the particle diameter, those in the blanching treatment groups were lower than that in the non-blanching-treatment group and was significantly lowest in the blanching treatment groups treated with DE=20 and CD. The result of SEM observation showed that the spray-dried powders in blanching treatment groups treated with the DE=20 and CD forming agents had uniform particle distribution.

Antioxidant activity and physicochemical composition of fermented Vigna angularis using Bacillus subtilis KCCM 11965P (Bacillus subtilis KCCM 11965P를 이용한 붉은팥 발효물의 항산화 활성 및 이화학성분 분석)

  • Health of human intestine has deteriorated due to excessive stress and western diet. In this study, Vigna angularis was fermented by Bacillus subtilis KCCM11965P in order to develop prebiotic resource for improving bowel movement. The contents of ash and crude protein were $3.35{\pm}0.04%$ and $21.1{\pm}0.19%$ respectively. Vigna angularis extract (1, 3, 5%) were incubated with 3% (v/v) Bacillus subtilis KCCM 11965P for 0, 24, 48, and 72 h. Total bacterial numbers showed that the combination of 3% powdered seeds and 72 h incubation time was optimum condition for this experiment. Total polyphenol content increased from $0.18{\pm}0.010mg/mL$ in pre-incubation to $0.23{\pm}0.007mg/mL$ in post-incubation with the condition mentioned above. DPPH radical scavenging activity also increased from $36.1{\pm}6.0%$ to $63.6{\pm}5.2%$. Analysis of protease activity showed $2.69{\pm}0.003unit/mL$ in combination of 5% powdered seeds and 72 h incubation time. Amylase activity increased from $1.0{\pm}0.1unit/mL$ in pre-incubation to $26.0{\pm}0.2unit/mL$ in post-incubation. The analysis of free amino acids after incubation with Bacillus subtilis KCCM 11965P showed that leucine increased from 5.22 mg/L to 67.59 mg/L and tyrosine, one of non-essential amino acid also increased 10.08 mg/L to 259.35 mg/L by incubation with 5% powdered seeds. Most of organic acid were reduced by incubation for 72 h. These results suggest that Vigna angularis could be utilized most as a prebiotic resources.
