고밀도 지단백 콜레스테롤과 베타 3-아드레날린성 수용체 유전자 변이와의 관련성 (Association of β3-Adrenergic Receptor Polymorphisms and High-Density Lipoprotein Cholesterol)

  • 지방분해와 열생산에 관여한다고 알려진 ADRB3 유전자의 염기서열 분석을 통하여 한국인에서 호발하는 유전자 다형성 부위를 먼저 확인한 후 이 유전자 다형성들과 HDL-C와의 연관성에 대하여 조사하고자 2006년 5월에서 12월 사이에 부산지역의 일개 대학병원에서 건강진단을 받은 991명을 대상으로 신장, 체중, 체질량지수, 허리둘레, 고밀도 지단백 콜레스테롤, 중성지방, 공복 혈당을 측정하였으며, 대상자들의 혈액에서 DNA를 분리하여 ADRB3 유전자에서 흔히 발생하는 유전자다형성 부위를 확인하였다. 연구결과 한국인에서 ADRB3 유전자의 intron2 +3893T>C의 변이를 처음으로 발견하였으며 열성 대립형질의 발현빈도는 0.164이었다. Exon1의 +188T>C와 intron2의 +3893T>C의 열성 대립형질인 C형이 있을 경우 HDL-C의 농도가 낮았다. 따라서 ADRB 유전자 다형성은 HDL-C과 관련이 있을 것으로 생각된다.

Effects of cranberry powder on serum lipid profiles and biomarkers of oxidative stress in rats fed an atherogenic diet

  • This study investigated that the antioxidative effect of freeze-dried cranberry powder against protein and lipid oxidation and ameliorative effect of serum lipid profile in rat fed atherogenic diet. Six weeks old male Sprague-Dawley rats were divided into the following four groups: normal diet group with 5% com oil(control), atherogenic diet group with 5% com oil, 10% lard, 1% cholesterol, and 0.5% sodium cholate(HFC), atherogenic plus 2% cranberry powder diet group(HFC+C2), and atherogenic plus 5% cranberry powder diet group(HFC+C5), and respective diet and water were fed daily for 6 weeks. After the experimental period, the serum lipid profile, such as total cholesterol, HDL-cholesterol, LDL-cholesterol, and triglyceride, ferric reducing ability of plasma(FRAP), plasma phenolics content, superoxide dismutase(SOD) activity, serum protein carbonyl and thiobarbituric acid reactive substances(TBARS) levels were examined. Total phenolic compound and total flavonoid levels in freeze-dried cranberry powder were 9.94 mg/g and 8.12 mg/g, respectively. Serum total cholesterol and LDL-cholesterol levels were not significantly different for cranberry powder treatment, but serum HDL-cholesterol level was significantly increased in HFC+C5 group compared with HFC group. Plasma FRAP value tended to be increased by cranberry powder treatment though there was no significant difference. Plasma total phenol concentrations and SOD activities were not significantly different among all groups. Serum protein carbonyl and TBARS levels were significantly decreased in HFC+C5 group compared with HFC group. Overall results suggested that freeze-dried cranberry powder might have the serum lipid improving effect, as well as anti oxidative effect demonstrated by its protective effect against protein and lipid oxidation.

녹차가루 식이가 당뇨쥐의 혈당 및 혈중 지질 농도에 미치는 영향 (Effects of Green Tea Powder Supplementation on Blood Glucose and Lipid Concentrations in Rats with Streptozotocin-induced Diabetes)

  • The study examined the effects of dietary green tea powder supplementation on blood glucose, and plasma and liver lipid concentrations in diabetic rats. Twenty-five male Sprague-Dawley rats (body weight $200{\pm}5\;g$) were divided into two groups (diabetic and non-diabetic), which were each randomly divided into two subgroups that were fed a control and 1% green tea powder-supplemented diet. Serum and liver lipid concentrations were measured by established techniques. Low density lipoprotein-cholesterol (LDL-C) was calculated from an established equation. Body weight gain and feed efficiency ratio were lower in diabetic rats than in non-diabetic rats regardless of diet. There were no differences in weight gain in diabetic and non-diabetic rats consuming the control and green tea powder-supplemented diets. The levels of fasting plasma glucose, serum total cholesterol, triglyceride, LDL-C and atherogenic index of diabetic rats were significantly higher than that of non-diabetic rats. Conversely, the levels of high density lipoprotein-cholesterol (HDL-C) of diabetic rats was significantly lower than that of non-diabetic rats. Fasting plasma glucose, serum total cholesterol, triglyceride, LDL-C and atherogenic index were significantly lower in diabetic rats fed the green tea powder diet than in rats fed the control diet, and HDL-C was significantly higher in rats fed the green tea powder diet than in rats fed the control diet. The content of liver total cholesterol and triglyceride of diabetic rats were significantly higher than that of non-diabetic rats. Liver total cholesterol and triglyceride were significantly lower in diabetic rats fed green tea powder-supplemented diet than in rats fed the control diet. It is concluded that green tea powder supplementation positively influences blood glucose and lipid metabolism in diabetic rats. The present study, although not directly applicable to humans, may have some implications for individuals who habitually consume green tea powder.

칼로리 급여량 제한이 OLETF 쥐의 혈당 및 혈청 지질량에 미치는 영향 (Effects of Caloric Restriction on Blood Glucose and Serum Lipid Levels of OLETF Rats)

  • To determine the effects of caloric restriction on obese type 2 diabetes we measured body blood glucose and serum lipid level in dietary restricted Otsuka Long Evans Tokushima Fatty (OLETF) rats. OLETF rats (obese diabetic rats) and Long Evans Tokushima Otsuka (LETO) rats (control rats) were grouped into 2 groups; control (free feed) group and 30% caloric restricted (30% CR) group. At 24 weeks of age the 30% CR animals were provided a diet at a level of 30% less food (by weight) than control rats consumed during the previous week. Blood glucose levels and serum triglyceride, total cholesterol, and high density lipoprotein (HDL)-cholesterol levels of CR rats were determined every 2 weeks for 8 weeks total. Blood glucose, triglyceride and total cholesterol levels of OLETF rats were significantly higher compared to LETO rats. In OLETF rats, the blood glucose levels were decreased to 61% by 8 weeks in the 30% CR compared to the non-CR control group, but changes of blood glucose levels were not observed in LETO rats during the 8weeks in the 30% CR. The serum triglyceride levels of OLETF rats were decreased significantly in the 30% CR but no change in the serum of LETO rats was found. The total cholesterol level was not changed by dietary restriction in LETO rats, but significant changes were observed in OLETF rats by 30% dietary restriction. HDL-cholesterol levels were also increased by dietary restriction in both LETO and OLETF rats. These results suggested that elevated blood glucose, triglyceride and total cholesterol levels in diabetes II patients may be reduced by caloric restriction.

한국 중년의 고밀도지단백(HDL) 콜레스테롤과 관련요인 (The High-density lipoprotein (HDL) cholesterol and related factors in Korean middle-aged)

  • 이 연구는 40-59세의 중년들이 갖고 있는 여러 가지 건강행태 중에서 고밀도지단백(HDL) 콜레스테롤에 영향을 명확히 미치는 건강행태를 파악함으로써 한국 중년의 건강증진에 도움이 되는 기초자료를 제공되고자 한다. 국민건강영양조사의 제4기 3차년도(2009), 제5기의 1차년도(2010), 2차년도(2011), 3차년도(2012), 제6기의 1차년도(2013) 등 총 5개년의 원시데이터를 합하여 사용하여 7,406명을 최종분석 대상자로 선정하였다. 고밀도지단백 콜레스테롤을 40mg/dl 미만과 40mg/dl 이상으로 두 집단으로 구분했다. 다중 로지스틱 회귀분석을 시행한 결과, 여자보다 남자일 경우 OR=3.916배, 비만 유병이 저체중에서 정상일 경우 OR=3.439배, 비만일 경우 OR=7.336배, 음주자일 때 OR=1.629배, 흡연자일 때 OR=1.498배, 중등도 신체활동 미실천일 시 OR=1.426배, 걷기 미실천일 시 OR=1.264배, 탄수화물을 과소 섭취할 때보다 적정 섭취할 때 OR=1.510배, 과다 섭취할 때 OR=1.787배 고밀도지단백(HDL) 콜레스테롤이 40mg/dl 이상에서 40mg/dl 미만으로 될 확률이 증가하였다. 한국 중년의 고밀도지단백(HDL) 콜레스테롤 수치를 높여 적절한 건강을 유지하기 위해서는 유산소 운동, 금연, 절절한 음주, 비만 해소, 건강한 식습관과 건강한 식품 선택으로 인한 외식이 이루어져야 한다. 이를 위해서는 지역사회 내에서 지속적인 홍보와 교육이 필요하며, 건강행태를 생활 속에서 실천할 수 있도록 하는 사회적 환경이 구축되어야 할 것이다.

산마늘추출물이 과산화지질급여 비만쥐의 지질강하, 항산화효과 및 염증매개물질의 생산에 미치는 영향 (Effects of Allium victorials Extract on Lowing Lipid, Anti-oxidation and Concentration of Inflammatory Mediators in Rats Fed High Oxidized Fat)

  • 본 연구는 산마늘추출물이 과산화지질을 급여한 비만쥐의 지질강하, 항산화효과 및 염증매개물질의 생산에 미치는 영향을 검토했다. 그 결과 혈장 FFA, TG, total cholesterol 및 LDL-cholesterol 농도는 산마늘추출물 처리군 들에서 감소했으며, 혈장HDL-cholesterol 농도는 산마늘추출물 처리군 들에서 증가했다. 간장 내 total cholesterol 농도 및 TG 농도는 산마늘추출물 처리군 들에서 감소하는 경향을 나타내었다. 혈장 및 간장의 TBARS 농도는 산마늘추출물 처리군 모두가 대조군보다 낮은 경향을 보였다. 간장 GSH-Px, SOD 및 CAT활성치모두가 산마늘추출물 처리군 들에서 증가하는 경향을 보였다. 혈장 NO, Ceruloplasmin 및 ${\alpha}1$-acid glycoprotein 농도는 산마늘추출물 투여군 들이 대조군보다 낮은 경향을 나타내었다. 이와 같은 결과는 산마늘추출물에 지질강하, 항산화 및 항염증작용에 효과를 나타내는 기능성물질이 내재하고 있음을 시사한다.

청각의 지질강하 및 항산화효과 (Effects of Cheunggak (Codium fragile) on lowering Lipid and Antioxidant)

  • 청각 추출액이 비만을 유도한 흰쥐의 지질강하 및 항산화효과에 미치는 영향을 검토했다. 각 처리군 별 유리지방산의 농도는 청각 처리군 모두가 대조군보다 낮은 값을 보였다. Triglyceride 량은 대조군과 비교하여 청각 처리군이 유의하게 낮은 값을 보였으며, 청각 첨가량이 증가함에 따라 하락했다. 혈액 내 total cholesterol 및 LDL-cholesterol농도는 청각 처리군 모두에서 감소하는 경향을 보였다. 그러나 HDL-cholesterol량은 대조군을 비롯하여 전 처리군에서 유의한 차이를 나타내지 않았다. 간장 내 total cholesterol과 triglyceride량은 청각 처리군 모두에서 하락하는 경향을 보여 정상군의 수준을 유지하였다. 혈장 및 간장의 TBARS 농도는 청각처리군 모두가 하락하는 경향을 보였으며, 정상군의 수준과 유사한 수치를 나타내었다. GSH-Px의 활성치는 청각 처리량이 증가함에 따라 증가하는 경향을 보여, 200mg/kg 처리군에서는 정상군의 활성치와 유의한 차이를 나타내지 않았다. 그러나 SOD 활성치는 청각처리에 관계없이 정상군을 비롯한 전 처리군에서 유의한 차이를 나타내지 않았다. 또한 CAT의 활성치는 청각 처리군에서 다소 증가하는 경향을 보였으나, 대조군과 유의한 차이를 나타내지는 않았다.

항산화성 비타민 보충 급여가 흡연자의 혈중 지질 양상에 미치는 영향 (Effect of Antioxidant Vitamin Supplementation on Plasma Lipid Pattern in Smoking College Men)

  • Cigarette smoking is a well known risk factor for cardiovascular disease and has negative effects on blood lipid and lipoprotein . Some of the associations between smoking and chronic disease can be attributed to the less healthful lifestyles of smokers. A large body of epidemiologic evidence suggests inverse relationships between ischemic heart disease and plasma vitamin C and E concentrations . Smokers have lower plasma concentrations of these vitamins than do nonsmokers. Smokers therefore need antioxidant vitamin supplementation. The purpose of this study was to investigate the effect vitamin supplementation on plasma lipid patterns in smoking college men. 24subjects were divided into 3 groups of which were the vitamin C supplementation group (n=8), the vitamin E supplementation group(n=8) and the vitamin C+E supplementation group(n=8). The vitamin C supplementation group consumed 500mg of ascorbic acid, the vitamin E supplementation group consumed 200IU of D-$\alpha$-tocopherol, and the vitamin C+E supplementation group consumed 500mg of ascorbic acid+ 200IU of D-$\alpha$-tocopherol for 4 weeks. We examined the plasma lipid patterns before and after the vitamins were supplemented. The results obtained were as follows ; In the vitamin C supplementation group, the concentration of total cholesterol decreased significantly and HDL-cholesterol increased significantly with the supplementation of vitamin. In the vitamin E and vitamin C+E supplementation groups, however, there were no significant differences observed with the supplementation of vitamin. Concentration of plasma LDL, triglyceride, free fatty acid were not significantly affected by the supplementation of vitamin in all groups. In terms of plasma fatty acid composition, the concentrations of saturated fatty acid were not significantly affected by the supplementation of vitamin in all groups. The concentrations of palmitoleic acid, arachidonic acid, and docosahexaenoic acid, however, significantly increased in the vitamin E supplementation group(p<0.05). The concentration of plasma linoleic acid significantly increased in the vitamin C+ E supplementation group)(p<0.05). The results of this study show that antioxidant vitamin supplementation in smokers has a tendency to decrease coronary heart disease risk in view of the plasma total cholestrol and HDL-cholesterol concentrations of the vitamin C supplementation group and fatty acid concentration of the vitamin E supplementation group.

아마인유와 해바라기 종자유의 혼합급이가 식이성 고지혈증 흰쥐의 혈청 지질 성분에 미치는 영향 (Effects of Feeding the Mixture of Linseed and Sunflower Seed Oil on the Lipid Components of Serum in Dietary Hyperlipidemic Rats)

  • This study was carried out to investigate the effect of the feeding mixture of linseed oil, rich in n-3 PUFA and the sunflower seed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hyperlipidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter and cholesterol for 3 weeks, then they were fed with the diet containing lard 3.0 % and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil and antihyperlipidemic drugs for 2 weeks. Analysis of the lipid component of the serum showed following results. Concentration of total cholesterol in serum was significantly lower in the other groups except group 2 (lard 3.0 % + olive oil 12.0%) than in the control group , especially in groups 3 (cholestyramine 2.0%) and 5 (lard 3.0% + linseed oil 12.0%). HDL-C and the ratio of HDL-C to total cholesterol concentration were higher in groups 1, 3, 5 and 9 (lard 3.0% + sunflower seed oil 12.0%) , while the atherosclerotic index was low in groups3, 5. Concentrations of free cholesterol and choesteryl ester in serum were significantly lower in the other groups except group 2 than in the control group, especially lower in grop 5. Concentration of LDL was significantly higher in group 2 while it was remarkably lower in groups 3 and 5, while those of triglyceride in groups in 3 and 4 (lipraroid) were significantly higher than in the control groups. From the data on concentration of total choelsterol , HDL-cholesterol, LDL, phospholipid and triglyceride in serum, the results suggested that the feeding mixed with 3.0% lard and 12.0% linseed oil or 2.0% cholestyramine were most effective for the improvement of the serum lipids.

Apo E 유전자 변이형이 혈청지질 및 유리아미노산 농도에 미치는 영향 (Effects of Apo E Polymorphism on the Plasma Lipid Profiles and Free Amino Acids in Korean Women)

  • Apo E polymorphism(e2, e3, e4) was among the first reported genetic polymorphism that explained part of the normal variation in plasma cholesterol concentrations. Among 62 normolipidemic healthy females, aged 19 up to 22 years, the relative frequencies of E3/3 was 0.806(n=50), E3/2 was 0.081(n=5), E3/4 allele was 0.113(n=7), and no E2/2, E2/4 and E4/4 were found. Based on the five samples of E2 allele, five subjects were randomly selected by E3 and E4 groups for the study of effects of apo E polymorphism on the distribution of serum lipid and amino acids profiles. No differences in the anthro pometric data among apo E isomers were found, otherwise the pulsation was higher in E4 than that in the others. There were no differences in plasma total HDL , HDL3 , HDL2 & LDL cholesterol, and apo A I concentrations. However, phenotype means significantly rank E2>E3>E4 allele in average TG levels(p=0.014), and rank E4>E3>E2 in total cholesterol levels(p=0.011). Atherogenic index(AI) such as lipoproteins was significantly increased in E2 & E4 than that in E3(p=0.045). Subjects with E3/2 allele had significantly higher concentrations of glutamine, phosphoserine and taurine, while subjects with E3/4 allele showed significantly lower concentrations of arginine and am inobutyrate and elevated level of phosphoserine in plasma com pared to those of E3/3 allele. Higher level of plasma taurine in subjects with E3/2 or E3/4 allele appears to be related to the elevated level of plasma total and LDL cholesterol concentrations compared to those of E3/3 allele.
