Regulatory Dendritic Cells Induced by Mesenchymal Stem Cells Ameliorate Dextran Sodium Sulfate-Induced Chronic Colitis in Mice

  • Background/Aims: Regulatory dendritic cells (rDCs), which can be induced by mesenchymal stem cells (MSCs), play an important role in inducing and maintaining homeostasis of regulatory T cells and exhibit anti-inflammatory functions. In this study, we investigated whether MSCs could differentiate DCs into rDCs and compared the therapeutic effects of rDCs and MSCs on dextran sodium sulfate (DSS)-induced chronic colitis mice. Methods: Immature DCs (imDCs) and lipopolysaccharide (LPS)-treated mature DCs (mDCs) were co-cultured with MSCs for 48 hours, and then the profiles of surface markers and cytokines and regulatory roles of these DCs for primary splenocytes were analyzed. In addition, the therapeutic effects of MSCs and DCs co-cultured with MSCs were compared in chronic colitis mice. Results: After co-culture of imDCs (MSC-DCs) or LPS-treated mDCs (LPS+MSC-DCs) with MSCs, the expression of CD11c, CD80, CD86, interleukin 6 (IL-6), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and interferon-${\gamma}$ (IFN-${\gamma}$), was decreased, but that of CD11b, IL-10, and transforming growth factor-${\beta}$ (TGF-${\beta}$) was increased. Furthermore, MSC-DCs and LPS+MSC-DCs induced the expression of CD4, CD25, and Foxp3 in primary splenocytes isolated from mice. In DSS-induced colitis mice, MSCs and MSC-DCs increased colon length, body weight, and survival rate and induced histological improvement. Moreover, in the colon tissues, the expression of IL-6, TNF-${\alpha}$, and IFN-${\gamma}$ decreased, but that of IL-10, TGF-${\beta}$, and Foxp3 increased in the MSC- and MSC-DC-injected groups. Conclusions: Our data suggest that MSCs differentiate DCs into rDCs, which ameliorate chronic colitis. Thus, rDCs stimulated by MSCs may be therapeutically useful for the treatment of chronic inflammatory diseases.

헤스페리딘/PLGA 필름에서 망막색소상피세포의 부착과 증식거동 (Adhesion and Proliferation Behavior of Retinal Pigment Epithelial Cells on Hesperidin/PLGA Films)

  • 망막색소상피(retinal pigment epithelium, RPE)는 시기능을 유지하는데 중요한 역할을 하여 RPE의 퇴화는 여러 망막변성질병을 유발한다. 현재 이에 대한 효과적인 치료법이 부족하여 세포 이식에 적합한 지지체를 제작하기 위해, 생분해성 고분자인 PLGA와 항염증, 항산화 작용 등의 기능이 있는 헤스페리딘을 이용하여 하이브리드 필름을 제조하였다. ARPE-19를 파종한 후, MTT 분석법을 이용하여 세포 증식률을 확인하고, 세포의 부착 및 세포 형태를 SEM을 통하여 확인하였다. 또한 RPE 세포의 특이적 유전자 발현정도를 확인하기 위하여 RT-PCR을 수행하였고, RPE65의 발현을 확인하기 위해 AEC 면역화학적 염색을 실시하였다. 그 결과, 헤스페리딘/PLGA 필름은 PLGA보다 RPE 세포의 부착, 증식 및 표현형 유지가 우수함을 확인하였고, 이를 통해 헤스페리딘/PLGA 필름의 망막재생을 위한 조직공학적 담체로써 응용 가능성을 확인할 수 있었다.

아토피양 접촉피부염 유발 NC/Nga mouse에서 도고 온천수에 첨가한 게르마늄 성분의 효과 (Effect of DOGO phreatic water containing germanium on Atopic Dermatitis in NC/Nga mouse)

  • Objectives : The purpose of this study is to investigate the effects of DOGO phreatic water containing germanium on Atopic Dermatitis in NC/Nga mouse. Methods : We made DOGO phreatic water added germanium. After making atopic dermatitis caused by sensitizing NC/Nga mouse to DNCB(dinitrochlorobenzene), we made mouse swim in tanks each filled with distilled water, tap water, DOGO phreatic water, DOGO phreatic water(added germanium) for 30minutes everyday. 3weeks later, we analyzed skin clinical score, total IgE levels(by ELISA), WBC differential counting(Neutrophils, Monocytes), absolute cell number of $Neutrophil^+Gr-1^+$, CCR3 mRNA expressions(by Real-time PCR), IL-4, IFN-${\gamma}$ production levels(by ELISA), histologic test(by H&E staining, toluidine blue staining). Results : The results of making NC/Nga mouse induced atopic dermatitis swim in tanks filled with DOGO phreatic water(contain germanium) are as follows. 1. Skin clinical scores were decreased significantly in comparison to control group. 2. Total IgG levels were decreased significantly in comparison to control group. 3. WBC differential counting(Neutrophils, Monocytes) were decreased significantly in comparison to control group. 4. Absolute cell number of $Neutrophil^+Gr-1^+$ were decreased significantly in comparison to control group. 5. CCR3 mRNA expressions were decreased significantly in comparison to control group. 6. IL-4, IFN-${\gamma}$ production levels were decreased significantly in comparison to control group. 7. The epithelial tissue thickness, leucocytes infiltration, erythema, edema, excoriation, scaling, mast cells infiltrations in dorsal skin were decreased in comparison to control group. Conclusions : These results indicate that DOGO phreatic water(contain germanium) can be used for helping treat atopic dermatitis.

Dosimetric Evaluation of Low-Dose Spillage Volumes for Head and Neck Cancer Using Intensity-Modulated Radiation Therapy and Volumetric Modulated Arc Therapy Treatment Techniques

  • Purpose: This study was designed to investigate the dosimetric difference between intensity-modulated radiation therapy (IMRT) and volumetric modulated arc therapy (VMAT) in head and neck cancer (HNC). The study primarily focuses on low-dose spillage evaluation between these two techniques. Methods: This retrospective study involved 45 patients with HNC. The treatment plans were generated using the IMRT and VMAT techniques for all patients. Dosimetric comparisons were performed in terms of target coverage, organ-at-risk (OAR) sparing, and various parameters, including conformity index, uniformity index, homogeneity index, conformation number, low-dose volumes, and normal tissue integral dose (NTID). Results: No significant (P>0.05) difference in planning target volume coverage (D95%) was observed between IMRT and VMAT plans for supraglottic larynx, hard palate, and tongue cancers. A decrease in dose volumes ranging from 1 Gy to 30 Gy was observed for VMAT plans compared with those for IMRT plans, except for V1Gy and V30Gy for supraglottic larynx cancer and V1Gy for tongue cancer. Moreover, decreases (P<0.05) in NTID were observed for VMAT plans compared with that for IMRT plans in supraglottic larynx (4.50%), hard palate (12.80%), and tongue (7.76%) cancers. In contrast, a slight increase in monitor units for VMAT compared with those for IMRT in supraglottic larynx (0.46%), hard palate (2.54%), and tongue (7.56%) cancers. Conclusions: For advanced-stage HNC, both IMRT and VMAT offer satisfactory clinical plans. VMAT offers a conformal and homogeneous dose distribution with comparable OAR sparing and higher dose falloff outside the target volume than IMRT, which provides an edge to reduce the risk of secondary malignancies for HNC over IMRT.

조직 등가물질 두께 증가에 따른 디지털 엑스선 영상의 해상도 분석에 관한 연구 (A Study on the Resolution Analysis of Digital X-ray Images with increasing Thickness of PMMA)

  • 디지털 래디오그라피 시스템에서 발생하는 산란선은 신호의 증가를 가져오는 장점도 있지만 피사체를 투과한 엑스선 영상의 해상도 저하, 노이즈 증가로 궁극적인 검출능이 감소된다. 공간주파수 도메인에서 해상도를 평가하기 위한 변조전달함수(modulation-transfer function, MTF)에서 간접적으로 산란선을 측정하는 방법은 제로-주파수에 해당하는 변조전달함수 값이 강하되는 정도로 간주할 수 있다. 본 연구는 환자 조직 등가물질로 폴리메틸메타아크릴레이트(polymethyl methacrylate, PMMA)를 사용하였으며 다양한 두께에 대한 변조전달함수를 획득하여 산란선이 해상도에 미치는 영향을 정량화하였다. PMMA 두께 증가에 따라 엑스선 영상 신호는 35 ~ 83%까지 감소되는 것이 관찰되었고, 이는 PMMA에 엑스선이 흡수 혹은 산란되는 양상으로 판단되며 이러한 결과는 변조전달함수를 저하시키는 것과 동시에 산란선 비율을 증가시키는 결과로 나타났다. PMMA에 의한 변조전달함수 저하를 보정하기 위한 방법은 간접변환방식 검출기에서 발생되는 빛 퍼짐 경향까지 절단하여 변조전달함수 값의 상승을 가져왔다. 보다 합리적인 방법으로 경계확산함수(edge-spread function, ESF) 혹은 선확산함수(line-spread function, LSF)에서의 피팅, 제로 값으로 채우는 처리 등이 이루어져야 할 것으로 사료된다.

3T3-L1 지방전구세포와 고지방 식이로 유도된 비만 HR-1 마우스 피부에 도포한 한약 추출 복합물의 항비만 효과 및 안전성 평가 (Anti-obesity Effects and Safety of the Mixture of Herbal Extracts in 3T3-L1 Cells and HR-1 Mice Fed a High Fat Diet)

  • The aim of this study was to investigate whether a novel formulation of an herbal extracts has an inhibitory effect on obesity. To determine its anti-obesity effects, we performed anti-obesity-related experiments in vitro and in vivo. Thus, our present study was carried out to evaluate the anti-obesity effect of herbal extracts using a high fat diet (HFD)-induced obese mouse model and 3T3-L1 adipose cells. The effects of each herbal extracts on lipid accumulation in 3T3-L1 cells were examined using Oil Red O staining. Results showed that treatment with each herbal extracts at $10{\sim}100{\mu}g/ml$ had no effect on cell morphology and viability. Without evidence of toxicity, herbal extracts treatment decreased lipid accumulation compared with the untreated adipocytes controls as shown by the lower absorbance of Oil Red O stain. Futhermore, compared with control-differentiated mature adipocytes, each herbal extracts significantly inhibited lipid accumulation in mature 3T3-L1 adipocytes. In the HFD-fed obese mice, body weight, liver weight and white adipose tissue weights were significantly reduced by mixture of herbal extracts administration in mouse skin. Futhermore, we found that mixture of herbal extracts administration suppressed serum triglyceride (TG), and total cholesterol (TCHO) in HFD-induced obese mouse model. The mixture of herbal extracts of permeability was estimated by measuring the transepithelial electrical resistance (TEER) value in pig skin. The optimized formulations of herbal extracts (Test 3 formulation) showed skin permeation. However, test 1 formulation containing essential oil as enhancer showed maximum skin permeation. After confirming the enhanced skin permeability, in vivo studies were performed to assess whether skin irritation potential on the basis of a primary irritation index (PII) in rabbit skin. Reactions were scored for erythema/edema reactions at 24 h, 48 h and 72 h post-application. It was concluded that the test 1 formulation was not irritation (PII = 0). The present study suggests that the test 1 formulation might be of therapeutic interest with respect to the treatment of obesity.

타액 연구의 최신 지견과 임상 응용 (Trends in Saliva Research and Biomedical Clinical Applications )

  • 타액선의 기능과 타액의 구성성분은 개체의 건강 상태를 반영하는 지표가 될 수 있다. 타액에서 바이러스와 미생물, 호르몬, 면역 및 대사산물 등을 검출하는 미량원소 분석기술이 발달함에 따라, 전신건강의 진단, 평가, 예방 분야에서 타액의 활용가능성이 높아지고 있다. 진단 검체로써 타액은 혈액에 비해 채취 방법이 비 침습적이어서 환자의 불편감이 적고 비 전문가에 의한 검체 수집이 가능할 뿐 아니라 채취과정 중 감염 위험성이 낮다는 점에서 장점이 있다. 이러한 이유로 스트레스, 마이크로바이옴, 유전학 및 후생유전학 분야의 연구에 있어 타액 내 단백질, 유전물질이나 각종 생체표지자 등을 활용하는 방법이 주목받고 있다. 또한 전신 건강에 대한 빅데이터 수집 연구와 관련하여 타액을 효율적으로 활용, 보관하기 위한 인체 자원 은행의 필요성이 강조되고 있으며, 조직공학과 접목하여 타액선 재생연구도 활발히 진행되고 있다. 검체 채취법이나 보관, 활용 방법의 표준화를 비롯하여 해결해야 할 과제가 남아있으므로, 본 리뷰에서 타액 및 타액선에 관한 최근의 연구 동향을 알아보고 미래 발전 방향에 대하여 검토해 보고자 하였다.

도시·건축형태와 미기후의 관계에 대한 관찰 연구 (An Observation Study of the Relationship of between the Urban and Architectural Form and Microclimate)

  • 본 연구는 도시·건축형태가 도시공간 내 미시기후에 미치는 영향을 도출하고자 했다. 본 연구에서는 도시 내 미시기후에 영향을 미치는 도시·건축형태의 요소로 도시형태 및 도시조직, 건축물 형태 및 특성 등을 선정했다. 분석을 위해 2017년 8월에 기상청에서 서울시 내에 설치한 23개의 AWS 설치 지점 도시건축의 특성이 명확하게 구분되는 6곳을 선정하여 연구진의 AWS 장비를 이용하여 실측을 했다. 공간의 범위는 AWS 설치 지점으로부터 반경 500m로 한정하여, 미시기후 및 도시·건축형태 요소들을 조사했다. 분석결과, 가로의 방향, 건축물 폭, 건축물 깊이, 건축물 높이, 지형의 경사도 및 향, 교통량 등은 미시기후인 국지풍속, 일사량, 국지온도 등에 영향을 미치고 있었다. 본 연구의 결과는 폭염의 피해 저감 및 그에 따른 시민 개개인의 건강 관리에 도움을 줄 것으로 기대된다. 또한, 도시의 온도를 낮춤으로써 건축물 에너지 부하 저감 효과를 거둠으로써 건축물 에너지 소비량 감소의 효과를 통해 보다 건강하고, 지속가능한 도시 조성에 기여할 것으로 기대된다.

Comparison of One- and Two-Region of Interest Strain Elastography Measurements in the Differential Diagnosis of Breast Masses

  • Objective: To compare the diagnostic performance and interobserver variability of strain ratio obtained from one or two regions of interest (ROI) on breast elastography. Materials and Methods: From April to May 2016, 140 breast masses in 140 patients who underwent conventional ultrasonography (US) with strain elastography followed by US-guided biopsy were evaluated. Three experienced breast radiologists reviewed recorded US and elastography images, measured strain ratios, and categorized them according to the American College of Radiology breast imaging reporting and data system lexicon. Strain ratio was obtained using the 1-ROI method (one ROI drawn on the target mass), and the 2-ROI method (one ROI in the target mass and another in reference fat tissue). The diagnostic performance of the three radiologists among datasets and optimal cut-off values for strain ratios were evaluated. Interobserver variability of strain ratio for each ROI method was assessed using intraclass correlation coefficient values, Bland-Altman plots, and coefficients of variation. Results: Compared to US alone, US combined with the strain ratio measured using either ROI method significantly improved specificity, positive predictive value, accuracy, and area under the receiver operating characteristic curve (AUC) (all p values < 0.05). Strain ratio obtained using the 1-ROI method showed higher interobserver agreement between the three radiologists without a significant difference in AUC for differentiating breast cancer when the optimal strain ratio cut-off value was used, compared with the 2-ROI method (AUC: 0.788 vs. 0.783, 0.693 vs. 0.715, and 0.691 vs. 0.686, respectively, all p values > 0.05). Conclusion: Strain ratios obtained using the 1-ROI method showed higher interobserver agreement without a significant difference in AUC, compared to those obtained using the 2-ROI method. Considering that the 1-ROI method can reduce performers' efforts, it could have an important role in improving the diagnostic performance of breast US by enabling consistent management of breast lesions.

Comparative Analysis of the Physical and Biochemical Properties of Light-cure Resin-modified Pulp Capping Materials

  • This study compared the solubility, water absorption, dimensional stability, release of various ions (hydroxyl, calcium, sulfur, strontium, and silicon), and cytotoxicity of light-cured resin-modified pulp-capping materials. Resin-modified calcium hydroxide (Ultra-blendTM plus, UBP), light-cured resin-modified calcium silicate (TheraCal LCTM, TLC), and dual-cure resin-modified calcium silicate (TheraCal PTTM, TPT) were used. Each material was polymerized; solubility, 24-hour water absorption, and 30- day dimensional stability experiments were conducted to test its physical properties. Solubility was assessed according to the ISO 6876 standard, and 24 hours of water absorption, 30 days of dimensional stability were assessed by referring to the previous protocol respectively. Eluates at 3 and 24 hours and on 7, 14, and 28 days were analyzed according to the ISO 10993-12 standard. And the pH, Ion-releasing ability, cell proliferation rate, and cell viability were assessed using the eluates to evaluate biochemical characteristics. pH was measured with a pH meter and Ion-releasing ability was assessed using inductively coupled plasma atomic emission spectrometry (ICP-AES). Cell proliferation rate and cell viability were assessed using human dental pulp cells (hDPCs). The former was assessed by an absorbance assay using the CCK-8 solution, and the latter was assessed by Live and Dead staining. TPT exhibited lower solubility and water absorption than TLC. UBP and TPT demonstrated higher stability than TLC. The release of sulfur, strontium, calcium, and hydroxyl ions was higher for TLC and TPT than for UBP. The 28-day release of hydroxyl and silicon ions was similar for TLC and TPT. TLC alone exhibited a lower cell proliferation rate compared to the control group at a dilution ratio of 1 : 2 in cell proliferation and dead cells from Live and Dead assay evaluation. Thus, when using light-cure resin-modified pulp-capping materials, calcium silicate-based materials can be considered alternatives to calcium hydroxide-based materials. Moreover, when comparing physical and biochemical properties, TPT could be prioritized over TLC as the first choice.
