Analysis of Volatile Compounds in Bamboo and Wood Crude Vinegars by the Solid-Phase Microextracion(SPME) Method (SPME법에 의한 죽초 및 목초액 중의 휘발성 성분 분석)

  • Volatile compounds in three different kinds of crude vinegars obtained from oak (Quercus serrata), bamboo (phyllostachys) and pine (Pinus densiflora) species were analyzed by the solid-phase microextraction (SPME) method. A total of 264 peaks were detected on the chromatograms obtained from the polar (CBP 20) and the nonpolar (CBP 1) columns, which were used for analyzing the volatile compounds in these vinegars. The major volatile compounds identified by using the polar column were 2-butanone, acetic acid, guaiacol, phenol, cresols, 4-ethyl guaiacol, 4-ethyl phenol, and syringol. Using the nonpolar column, seven compounds could be identified: 1,2-dimethoxybenzyl alcohol, 1-hydroxy-2-butanone, 1-(2-furanyl)-1-propane, ethisolide, furfuryl acetate, 1,2-dimethoxybenzene, phenyl acetate. The volatile compounds were classified into five groups: phenols, neutral compounds, organic acids, esters and others. The phenols were the main component and comprised 49~65% of the volatile compounds of these vinegars. In the case of bamboo vinegar, the proportion of the phenols in the volatile compounds was lower than that of the two wood vinegars. However, the proportions of the neutral compounds and the organic acids were higher than those of the wood vinegars. Therefore, it seems that these differences of the proportions of the volatile compounds would make a certain difference of a smoke flavor between the bamboo vinegar and the wood vinegars.

A Study on Germination Characteristics through Seed Variable Immersion for Restoration of North Korea Forest (북한산림복구용 종자 변온침지 처리를 통한 발아특성 연구)

  • This study was conducted to investigate the germination characteristics through the temperature change treatment compared to the general room temperature immersion treatment of the native species, the Larix kaempferi, and the Berula platyphyllavar. japonica seeds, and to obtain basic data for the production of healthy seedlings with high productivity and efficiency. As a result, the germination rate of the larch seeds showed a significant difference according to room temperature and temperature immersion treatment, and the average germination days of the birch seeds showed a significant difference according to room temperature immersion and temperature immersion treatment. On the other hand, there was no significant difference in germinal uniformity. The results of the study are as follows. First, the results of the analysis of the fallen leaves were the highest germination rate in the treatment of 1.5 days of hot water and 1.5 days of cold water, the fastest germination rate, the lowest average germination day, and the highest germination uniformity. This is the best result in most items, and the change temperature immersion treatment of hot and cold baths was superior to the conventional room temperature immersion treatment. Second, the results of birch tree showed that the germination rate was the highest in 1.5 days of hot water and 1.5 days of cold water. The average germination day is 1.5 days of cold water 1.5 days of hot water 1.5 days, but the difference between 1.5 days of hot water and 1.5 days of cold water is 0.01 days. The highest germinated uniformity was found in 1.5 days of cold water and 1.5 days of hot water. As a result, in the case of larch and birch, the seed temperature immersion (1.5 days of hot water, 1.5 days of cold water) treatment is superior to the existing three-day immersion treatment in various analysis methods, and it is expected that productivity and efficiency can be improved at a low cost in a short period of time at the seedling production site through seeds.

Antioxidant and Antimicrobial Activities of Various Citrus Peels (감귤류 종류에 따른 과피의 산화방지 및 항균 활성)

  • To investigate the functional activity of different citrus fruit peels, antioxidant compounds in 70% ethanol extracts of mandarin, lemon, orange, and grapefruit peel powders were identified, and antioxidant and antibacterial activities were quantitated. Mandarin peel contained the highest content of total phenolic compounds and total flavonoid substances (21.46±0.12 mg GAE/g and 11.57±0.05 mg RE/g, respectively). The total phenolic compound content of the three other citrus fruits was 14.16±0.18-18.44±0.07, and their total flavonoid content was 5.51±0.10-7.46±0.09 mg RE/g. DPPH radical scavenging activity was the highest in lemon peel (87.64±0.21%), and mandarin peel displayed the best antioxidant activity with respective ABTS radical scavenging activity and FRAP measurements of 43.20±0.61% and 78.82±1.06 mM TE/g. Grapefruit peel antimicrobial activity increased with treatment time, and was the most potent among the four tested citrus species, inhibiting Staphylococcus aureus by about 4.05 log cycle. These findings demonstrate that mandarin and grapefruit peel can be used to prevent oxidation, improve food storage capabilities, and potentially preserve food quality.

  • A cDNA encoding the protein lethal (2) essential for life [l(2)efl] was cloned from Gryllus bimaculatus and named GBl(2)efl. This protein is composed of 189 amino acids, including an N-glycosylation site and 15 phosphorylation sites. Its predicted molecular mass is 21.19 kDa, with a theoretical isoelectric point of 6.2. The secondary structure of GBl(2)efl was predicted from the identification of random coils (56.08%), alpha helices (22.22%), extended strands (17.99%), and beta turns (3.7%) through sequence analyses. A homology analysis revealed that GBl(2)efl exhibited a high similarity with other species at the amino acid level, ranging from 52% to 69%. While GBl(2)efl mRNA expression was higher in the dorsal longitudinal flight muscle following a three-day starvation and in the Malpighian tubules following a one-day starvation, no changes in expression were detected in other tissues. Furthermore, tunicamycin-induced endoplasmic reticulum (ER) stress resulted in an approximately 1.8-fold higher expression in the fat body compared with the wild type.

  • 'Angelica' is one of the most traditionally consumed medicinal herbs around Northeast Asia including Korea for treatments of various diseases or health care purposes like hematopoiesis, blood circulation for women, sedative, analgesic, and a tonic medicine etc. Angelica gigas Nakai, a Korean native species of Angelica, is clearly different from the others in containing a high concentration of active ingredients like pyranocoumarines including decursin, decursinol, and decursinol angelate. These compounds have various kinds of positive effects such as anti-tumor activity including the precaution of neutropenia occurred during anticancer drug administration, improvements of metabolic disorders, menstrual irregularity, impairment of renal function, respiration improvement, cognition-enhancement, anti-inflammatory effect, anti-oxidative effect, enhancing fertility and so forth. Thus it implies incredible potentialities in future development for foods and drugs. However, certain purity-related qualities and/or overdose in food products can cause side effects like toxicities; therefore, their safety profiles should also be considered. This review focuses on the positive and negative effects of three pyranocoumarines in Angelica gigas Nakai and some possibilities and considerations for future food and drug products development.

Comparative Study on the Growth Condition of Landscape Woody Plants according to the Ground Structure - Focusing on Manseok Beach Town Complex 2, Incheon - (지반구조에 따른 수목 생육상태 비교 연구 - 인천광역시 만석비치타운 단지를 대상으로 -)

  • The purpose of this study is to compare growth condition of landscape woody plants growing on the different ground structures in apartment complex. I chose Manseok Beach Town Complex 2, in Manseok-dong, Seo-gu, Incheon which has both natural and artificial ground as a subject site. Analysis of three phases of soil showed that artificial ground had average liquid phase of 30.89%, artificial ground mounding 33.88% and natural ground 24.40%. It means that artificial ground has higher water content than natural ground despite having same earthiness. It is believed that artificial ground is not as well drained as natural ground even though it is connected to the natural ground and has a deep soil depth because of mounding. Comparative study between woody plants on natural ground and those on artificial ground demonstrated that trees on natural ground grew 40.4% compared to those on artificial ground(0.875mm more) in terms of diameter growth. Average diameter growth of trees on natural ground was 3.040mm against 2.165mm for those on artificial ground. All 19 tree species which were measured for root diameter growth showed similar or higher growth on natural ground than on artificial ground. When it comes to growth of height, arborvitae showed highest growth on natural ground, followed by Thuja occidentalis, Pinus strobus, Magnolia denudata, Diospyros kaki and Aesculus turbinata. I measured branch growth and rate of leaf adherence of Pinus strobus. Average annual rate of branch growth of woody plants on natural ground was twice as high as those on artificial ground. I could conclude that ground structure influences branch growth of Pinus strobus. Statistics analysis of tree damage demonstrated significant result, meaning that there is a difference in the average damage rate depending on structure of ground. In order to validate growth difference by planting ground, I conducted T-Test of growth of diameter, root diameter, branch and height on woody plants growing on natural and artificial ground. As a result, it is believed that there is a difference in the growth of trees depending on the ground structure. Putting all these results together demonstrates that woody plants on natural ground generally grow better than those on artificial ground, which means ground structure does have an influence on the environment of growth of trees.

  • Benthic animals are important indicators in benthic environmental quality assessment. This study investigated the environmental characteristics and the distribution pattern of benthic animals, and assessed the benthic ecosystem using AMBI (AZTI's marine biotic index) and WQI (water quality index) in the subtidal zone of Muan bay. Samplings were collected from 10 stations in the subtidal zone of Muan bay on summer. In the upper area of Muan bay, grain size was finer and organic content was higher than those of in the lower area. The pollution indicator organism such as Musculista senhousia, Theora fragilis and Lumbrineris longifolia were dominant at some stations. The benthic community was distinguished into three groups of upper, center and lower area of Muan bay, and which were coincided with the results by correlation analysis between organic matter content and benthic health assessment (WQI and AMBI). As a result of this study, the health condition of the subtidal zone in Muan bay were good. However, from the results that benthic animals were not evenly distributed, and also the opportunistic species appeared, the load of organic matter in Muan bay seems to be increasing.

Analysis of Food Sources of Pre- and Post-diet in a Bivalve Using DNA Metabarcoding (DNA metabarcoding을 이용한 이매패류 공식 전후 먹이원 분석)

  • Research on food sources through DNA metabarcoding is being used for various organisms based on high resolution and reproducibility. In the study, we investigated the difference in food sources between pre and post-starving in the three bivalve species (Anemina acaeformis, Anodonta woodiana, and Unio douglasiae) through DNA metabarcoding using 18S rRNA V9 primer. The food source of pre-starving appeared in 87 genera, 71 families, 51 orders, 35 classes, and 22 phyla. The primary food sources were the zoo and phytoplankton, including Chlamydomonadales, Euglenales, Ploima, Sphaeropleales, and Stephanodiscales. However, all zoo and phytoplankton were not observed after starving except Schizopyrenida and Rotifera. In Levin's niche breadth analysis, the Bi index of A. woodiana is 0.3, which was higher than A. acaeformis(0.14) and U. douglasiae (0.21), indicating that they feed on various food sources. The niche overlap of A. acaeformis was measured as 0.78 in A. woodiana, 0.7 in U. douglasiae showing a relative high value compared to other bivalves. The trophic level of A. acaeformis, A. woodiana, and U. douglasiae based on the food source information were investigated as 2.0, 2.0, and 2.5, respectively. The results of the previous study on the trophic level using stable isotopes showed 1.8 to 2.4 values were similar to the results of this study. These results suggest that DNA metabarcoding can be an effective analyzing tool for the gut content in the bivalves.

Cut Flower Yields and Qualities of Rosa hybrid Affected by Night Cooling in High Temperature Season (고온기 야간 냉방이 절화 장미 수량 및 품질에 미치는 영향)

  • Roses are one of the most produced flower species in the world, and cut roses are produced in the greenhouses all year round. Recently, due to the increase in the temperature in the greenhouses in summer, the quality of cut roses is seriously deteriorated, such as shortening the stem length. This study was conducted to investigate the effects of the growing seasons on the qualities of cut roses and also to test the effect of cooling at night in high temperature season on the cut flower qualities of roses. Comparing the qualities and yields of cut roses for each season, the major cut flower qualities such as flower stem length, stem diameter and fresh weight were statistically significantly decreased in roses ('Pink Beauty' and 'Pink Shine') produced in summer. The yields didn't show a statistically significant difference in both cultivars. Investigating the cut flower qualities, the flower stem length increased by 15% for 'Pink Beauty', 11% for 'Ararat', and 12% for 'Pink Shine' when treated with cooling at night in warm season than the untreated control. In addition, when treated with cooling at night in warm season, the fresh weight of all three cultivars increased by 20-30% statistically significantly than conventionally cultivated control. It is expected that cooling at night in warm season will be helpful to improve the cut flower quality deterioration in summer.

Investigation of Source Area and Material Characteristics of the Stone Properties from Stone-lined Tombs in Cheongyang Rokpyeong-ri Site, Korea (청양 록평리 유적 석곽묘 석재의 재질특성 및 산지탐색)

  • In the stone-lined tombs from Cheongyang Rokpyeong-ri site, several stone-lined tombs corresponding to the Three Kingdoms Period, Unified Silla Period and Goryeo Dynasty were excavated. In this study, the stone properties of petrological, mineralogical and geochemical characteristics from 15 stone-lined tombs were analyzed, the rock species and occupation rate were reviewed, also the provenance area was interpreted. There are five types of rocks used as members of the tombs, including black sandstone, greywacke, slate, conglomerate and gneiss. As a result of magnetic susceptibilities, the rocks of the tombs and presumed provenance are all shown in the same range, and the similarity between the tombs and the source area is high in geochemical behavior characteristics. The stone properties of the tombs are the same as the presumed provenance rocks and is distributed within a 500m around the bedrock of the tombs in the excavation site, so it seems that self-sufficiency was possible on the site without a special procurement system. These stones are interpreted to have been constructed through some trimming without any special technique, but some stone tombs have been confirmed to used homogeneous black sandstone at the bottom of the tombs, so it is believed that there were artificial selection of stones.
