Enzymatic Synthesis of 2-Phenoxyethanol Galactoside by Whole Cells of ${\beta}$-Galactosidase-Containing Escherichia coli

  • We investigated whether ${\beta}$-galactosidase (${\beta}$-gal)-containing Escherichia coli cells could transfer a galactose to 2-phenoxyethanol, resulting in 2-phenoxyethanol galactoside (PE-Gal). PE-Gal was confirmed by liquid chromatography-mass spectrometry. In addition, we also confirmed that a galactose molecule was covalently bonded with PE during thin-layer chromatography analysis of the ${\beta}$-gal hydrolysate of PE-Gal. The yield for PE-Gal synthesis was about 37.5% (weight basis), which was about 7-8 times greater than that of a previous report. In addition, the concentration of ${\beta}$-gal (0.96 U/ml) used in this PE-Gal synthesis was about 20 times less than that in a previous report.

  • The identification of eicosapentaenoic acid (EPA) by Pseudomonas sp. CH-414 were investigated under the optimal culture conditions. The maximum dry cell weight and phospholipid production showed about 10 mg/ml and 0.6 mg/ml, respectively, at the 48 hr cultivation. The phospholipid form produced by Pseudomonas sp. CH-414 was elucidated as a phosphatidyt ethanolamine by thin layer chromatography. EPA was contained about 15% in the. extractable lipid, and the amount of EPA was about 83 $\mu $g/ml from the culture broth. Also myristic, palmitic, palmitoleic, stearic and oleic acids were identified with gas chromatography.

  • Dried roots of Paeonia japonica were extracted with dichloromethane, methanol and water aerially. Silica gel column chromatography and thin layer chromatography were used to separate the fractions with antimicrobial activities, and mass spectrometry was used to determine the mass. Dichloromethane extract showed the highest antimicrobial activity. Dichloromethane extract from Paeonia japonica could be a candidate for a new antimicrobial agent against MRSA.

Use of Lycopene, an Antioxidant Carotinoid, in Laying Hens for Egg Yolk Pigmentation

  • The possibility of lycopene affecting egg yolk pigmentation was studied with lycopene diets containing 0, 4, 8, and $12{\mu}g/g$ meal, respectively. The addition of lycopene above $4{\mu}g/g$ meal significantly improved yolk color after four days of supplementation. The transfer of lycopene into egg yolk was confirmed by thin layer chromatography, and high-performance liquid chromatography-mass spectrometry (HPLC-MS). The deposition rate of lycopene into egg yolk was approximately 2%, which was quantitatively determined using a HPLC with a UV detector. The result indicates that lycopene is a good candidate for egg yolk pigmentation and for making functional eggs.

Study on the molecular weights of radioprotective ginseng proteins by HPLC method

  • Partialy purified ginseng proteins were either treated with sodium dodecyl sulfate (SDS) and .betha.-mercaptoethanol to denature the proteins or not, and subjected to Thin Layer Chromatography (TLC) and High Performance Liquid Chromatography (HPLC) to compare the components of each fraction. Standard proteins of known molecular weights (MW) were also either treated with SDS and .betha.-mercaptoethanol or not, and subjected to HPLC to obtain regression lines for MW determination. From the retention times obtained from samples in eiether case by HPLC, the MW were estimated as following in SDS treated condition, Gl fraction showed three peaks each with MW of above 100, 000, 51, 000 and 19, 000. Gll showed one original peak with MW of 21, 000 and Gll, two peaks each with MW of 19, 000 and 14, 000. On the other hand, in non-SDS treated condition, GI fraction showed two peaks each with MW of above 200, 000 NA 52, 000. Gll showed one original peak with MW of 41, 000 and Gill, three peaks each with MW of 28, 000, 19, 000 and 14, 000.

  • As the preservation law of soil environment has reinforced, several soil remediation projects have been performing for railroad sites. One of the main sources of soil contamination is the leakage of diesel from locomotives or underground storage tank. Also, the lubricant used to maintain turnouts causes railroad soil contamination. The purpose of this study was to develop the analysis and the remediation method for lubricant-contaminated soil. The lubricant in the contaminated soil was analyzed qualitatively and quantitatively by TLC (Thin Layer Chromatography) and GC (Gas Chromatography), respectively. The organic pollutants were removed from the soil using microorganisms degrading lubricant. Hereafter it will be necessary to apply this bioremediation method in the railroad field.

A Study on Saccharides in Honey via HPLC (HPLC에 의한 벌꿀의 당성분에 관한 연구)

  • Saccharides in Korean honey have been analyzed by traditional wet chemical methods and several chromatographic technics gas, paper, column and thin layer chromatography. A simple, rapid and versatile method for the separation and determination of saccharides via high performance liquid chromatography were used eluting with $H_2O : CH_CN$ at 25:75, at a flow rate of 1.0ml/min. Acasia honey contained 36.8% fructose, 31.4% glucose, 1.7% sucrose and bush clover honey held 34.4% fructose, 32.3% glucose and 30% sucrose. Difference on the regional distribution were not found in the quantity of saccharides (42 species). The quality of inferior honey was assumed to contain much maltose, sugar and glucose than common honey.

  • In spite of the radical increase of chemical poisoning cases caused by prevalent of oranic chlorine like endosulfan, the study of detection of chemical poisoning has seldom been attempted; the study of poisoning in teeth, which is more secure physiochemically, and in all the oral tissues has been even less attempted. The author has administered endosulfan into the month of a rabbit, causing its death, and tried to detect endosulfan in the teeth, jaws, and oral tissues through forensic chemical and histopathological tests. The result obtained is as follows : 1. By thin layer chromatography, endosulfan was detected in the jews, dental hard tissues, dental pulp, blood, and the liver. 2. By gas chromatography, it was also detected in the jaws, dental hard tissues, dental pulp, blood, and the liver. 3. The congestion and hemorrhage in buccal mucosa, palatal mucosa, and dental pulp was detected.

Antimicrobial and Cytotoxic Activity of Di-(2-ethylhexyl) Phthalate and Anhydrosophoradiol-3-acetate Isolated from Calotropis gigantea (Linn.) Flower

  • A phytochemical study on the flower of Calotropis gigantea (Linn.) using silica gel column chromatography and preparative thin layer chromatography, led to the first time isolation of Di-(2-ethylhexyl) phthalate (compound 1) and anhydrosophoradiol-3-acetate (compound 2). The structures of these compounds were confirmed by spectroscopic analyses (IR, HRTOFMS and NMR). The antibacterial and antifungal activities of ethyl acetate extract, compound 1 and compound 2 were measured using the disc diffusion method. Ethyl acetate extract and compound 1 presented better results than compound 2. The minimum inhibitory concentrations (MICs) of the extract and compounds were found to be in the range of $16{\sim}128{\mu}g/ml$. The cytotoxicity ($LC_{50}$) against brine shrimp nauplii (Artemia salina) were also evaluated and found to be 14.61 ${\mu}g/ml$ for ethyl acetate, 9.19 ${\mu}g/ml$ for compound 1 and 15.55 ${\mu}g/ml$ for compound 2.

Studies on Availability of Components in Extract of Scutellariae Radix by Boiling water Method (전탕(煎湯)에 의한 황금(黃芩)의 성분(成分) 이행율(移行率)에 관한 연구(硏究) I)

  • In order to improve the qualities of boiling extract the availability of standard compounds was investigated in extract of Scutellariae Radix. The standard compounds used baicalin(1), baicalein(2), and wogonin(3). The availabilities were analyzed by thin layer chromatography (TLC) and high performance liquid chromatography(HPLC) on $C_18$ column. The extract of RS-4(W/V=10g/100m1) showed the highest availabilities as 7.95% of baicalin(1), 1.04% of baicatein(2), and 0.31% of wogonin(3).
