Production of Tropane Alkaloids by Two-stage Culture of Scopolia parviflora Nakai Adventitious Root

  • Scopolia parviflora Nakai, a rare and endangered species, is the sole plant producing tropane alkaloids (TA) among the Korean native species. In order to enhance TA productivity the SP72 root line was selected by screening 100 of root line, and the optimal culture media for root growth and TA production were investigated with the SP72 roots. Based on the several media, SH and 2B5 medium were determined as growth medium and White and NN medium as production medium. Among the four combinations of two-stage culture, 2BN (2B5 as growth medium plus NN as production medium) showed more enhanced root growth and TA production as compared with production media of White and NN medium and growth media of SH and 2B5 medium, respectively. However, bubble column bioreactor (BCB) cultures applying two-stage culture did not reveal the effective results despite of the each successful operation of two-stage culture in conical flasks and BCB cultures.

  • Main theme of this study is the acceptance phase of western architectural culture in modern east-north asia through a series of actual survey and typological analysis of existing church buildings in modern Korea. China. Japan. For this study, 126 modern church buildings of Korea, China, Japan are selected. Also, main subject of this study has deal with the side of culture - thought which background of christianity acceptance and acceptance mind of acceptance main-body, culture adaptation and transformation based on characteristic culturalization of Korea, China, Japan. Especially, I would consistently tried to see the acceptance phase of western architectural culture according to differential acceptance mind of acceptance main-body based on characteristic culturalization of three countries. Conclusively, in the point of view of western architectural culture acceptance phase, it could be said that Korean church architecture had characterized as a blending phase of the two eclectic architectural culture. And in the case of china, as a coexistance phase of the two architectural culture with more western style-oriented. On the other hand, Japanese church architecture had characterized as a syncretism phase of the both traditional and western architectural culture, though it is eclectic style-oriented. Therefore, it is thought that different acceptance mind of western culture had main factors caused of differential acceptance phase, when the two architectural culture, the traditional and the western, encounters.

  • Cloth culture is one of the basic unit of constructing man's culture in a life of human being. One of the fundamental elements to construct cloth culture is the silk-fabric from the ancient time-Sam Han period B.C 2∼4 Korea. This study is to survey the growing aspects of the silk-fabric influence Goguryo & Bakje dynasty. It is an attempt to manifest that how silk-fabric influence through a trade or as means of currency between the two nations at that time. through a trade or as means of currency between the two nations at that time. To certify I use material such as San guo zhi, Hou Han han shu from China Samkuksaki Dong-kuk-tong-gam from Korea and NiHon Shoki from Japen and I concerned the cultural relations of politics economics and socity of the two dynastics so as to prove a relations of politics economics and socity of the two dynastics so as to prove a relative relation between the two. It is reached to the conclusion that silk fabric had been a constituent role at cloth culture at that time of Goguryo & Bakje. It is pointed out that silk-fabric had been an important role to construct the culture of social economic and politic through a trade and as a precious currency.

  • The purpose of this article is to comparatively depict the organizational effectiveness of two rail firms based on the cognitive response of employees(group of drivers). To attain this purpose, several aspects of current personnel and organization management system and organizational culture are surveyed through questionaire and analyzed by SPSS. The questionaire includes two parts of items. First, it includes items on personnel-related factors such as evaluation system, job, wage, role, leadership and job satisfaction. Second, it includes items on organizational culture model suggested by Rousseau. The major findings are : 1) In general, the cognitive response of drivers in two firms concerning personnel-related aspects is to a certain degree negative. 2) The cognitive response of drivers in two firms concerning culture-related aspects is also awfully negative. 3) Job satisfaction of drivers in the firm A is highly related to role and leadership factors in terms of personnel-related aspects, on the one hand, and to achievement factor in terms of culture, on the other hand. In case of the firm B, job satisfaction of drivers is highly related to personnel evaluation factor as well as role and leadership factors in terms of personnel-related aspects, on the one hand, and to affiliation, power, and bureaucracy factors in terms of culture, on the other hand.

  • This study highlights the homogeneity in the heritage of the costume cultures between the Gulf of Aden, a part of the Horn region of Africa and the Arab region of West. Asia. Specifically, a cross-cultural perspective is used to examine the similarities between the two regions based on their (1) geographic living cultures and (2) the use of a costume accessory called Burga-a face veil. The current trend in research on the traditional costume culture of Africa mostly ties art with the traditional costume culture and examines its from an aesthetic or animalism perspective rather than from a cross cultural Perspective. Compared to Previous research in this area this study used literature reviews and Pictorial analysis to analyze costume cultures from a cross-cultural perspective. The Burga, which is worn in the low lands of the Horn region, which is located between the West Asia and African continents, as well as the Arab region, shows a reciprocal cultural exchange between the two regions. Similarities can be found in the shape, design, decorative elements, and the way the Bursa was worn in two regions. Although the Burga as a face veil is only a small part of a costume, it reaffirms the similar living cultures and geographic characteristics of the two regions. As the living culture environment becomes similar around the world, this study should help with cross culture negotiations as well as forther the development of traditional costume research.

Analysis on Organization Performance Based on Hospital Culture (기독병원과 일반병원의 조직문화 특성에 따른 조직성과 분석)

  • Our study attempt is to see organizational performance according to the differences between types of hospital cultures. To determine theoretical relationship between the organizational culture and the performance, we select two hospitals in Pusan which are different in the purposes and shapes of establishment. We surveyed their members on a questionnaire based on the type of the organizational culture of the two institutions, analyzed, and review its organizational effectiveness. For the survey with questionnaires, which had been preliminary studied to raise its validity, question forms were distributed to 528 persons in April, 1999 based on the self-responses and recollected within 48 hours. The recollection rate was high(89.96%) and the quantity of questionnaires used for our final analysis was 430(81.44%). The Cronbach Coefficient Alpha of the questionnaires was 0.742. Regarding statistical techniques for analysis of the written materials, dispersion analysis(ANOVA) was adapted to test the organizational effectiveness of the two hospitals having the different organizational cultures, and Pearson Correlation was applied to determine correlations was among all variables. T-test was performed to test organizational effectiveness based on the differences in the extent of sharing the culture, organizational committment and work satisfaction between the two health institutions. From our analysis, we obtain the following conclusions. First, concerning with organizational culture of the two hospital, one of which is a christian hospital and the other is a private foundation hospital, the former is conservative and human-oriented but the latter focuses on renovation and accomplishment. Second, the private establishment has a relatively higher organizational effectiveness that the religious hospital as a result of analyzing the extent of sharing culture, organizational committment and work satisfaction. Third, it has been found that the correlations between the extent of the sharing culture and the organizational committment, the extent and work satisfaction, and the committment and the satisfaction are respectively positive influencing organizational effectiveness, especially work satisfaction. Fourth, cultural factors by which the christian hospital is affected more positively including human relations among its members, belief, its idea of establishment, tradition, work responsibility, power, and wage. On the other hand, factors such as director's leadership, personnel management, wage, hospital regulations and department managers' management ability have been seen as negative influences in order. And fifth, for the private foundation hospital human relations among its members, wages, work responsibility, director's leadership and department managers' management ability were positive in their sequence while wages, personnel management, hospital regulations, welfare and department managers' management ability were considered as negative influences in order. As these results of this study, the higher extent of sharing organizational culture, the more increasing in both organization committment and work satisfaction, the higher the effectiveness. Although it was somewhat difficult to generalize the results whose subjects were the two hospitals only, it was obvious that organizational culture was an important influential factor of organizational effectiveness. It is questionable that the extent of sharing organizational culture, organizational committment and work satisfaction as variables affecting the effectiveness have their validity, but this study has its significance in that it provided an approaching to evaluate the organizational culture of individual hospitals making allowances for such variables related to the general activities in its hospital. We hope the results of the study could be useful for the managerial strategies of the institutions.

  • Currently, two-dimensional culture is the main keyword in Chinese contents market. In China, many capital and economic giants have entered into two-dimensional cultural market because the number of users and the interest of the public for two-dimensional culture are increasing day by day. The reason for the rise of the two-dimensional culture into the mainstream culture is not only the switching of the mainstream consumer class but also Two-Dimensional Culture forms a huge industrial chain covering many areas including movies, performances and theme parks in addition to ACGN (Animation, Comic, Game and light Novel), the second is that more people are participating in Two-Dimensional Culture due to the specific audience with high social environment and loyalty and the third is looking for the role of the users in Two-Dimensional cultural content from three characteristics, such as strong cultural prevalence. This study analyzed the Chinese two-dimensional cultural environment through PEST(Political, Economic, Social and Technological analysis) In addition, as the ecological environment and communication method of Two-Dimensional Culture users changed, the users became active participants and the second producers of contents. I examined the way of using the users' contents through the phenomenon of 'Barrage'. Therefore, this study discussed the appropriate development path for the user-oriented contents industry in the Chinese Two-Dimensional cultural environment.

Effects of Trophoblastic Vesicle and Estradiol-$17\beta$ on the Development in Vitro of Rabbit Embryos (Trophoblastic Vesicle과 Estradiol-$17\beta$의 첨가가 가토배의 발달에 미치는 영향)

  • This experiment was conducted to determine the effects of trophoblastic vesicles (TV) and estradiol-17$\beta$ on the development in vitro of rabbit embryos. Thirty matured female rabbits were treated with PMSG followed by HCG injection and mating. Embryos were recovered with D-PBS (Dulbecco's Phosphate Buffered Saline) after superovulation, and normally developed to two-to four-cell embryos were used in the subsequent in vitro culture. Basal medium was Medium-199 su, pp.emented with 1.5% bovine serum albumin. Embryo on Day 5 after mating (Day 0) was cut into two or three pieces to remove the embryonic disc. Each piece of tissue was cultured for 24 hours at 37$^{\circ}C$ in 0.5 mlMedium-199 in 5% CO2. During culture, peices of trophoblastic tissue changed into spherical vesicles which were used for co-culture. These spheres were called trophoblstic vesicles. Two-to four-cell embryos were cultured for 4 days in Medium-199 in the absence or presence of trophoblastic vesicle, and two-to four-cell embryos cultured with varing concentration (0, 0.1, 1, 10ng/ml) of estradiol-17$\beta$ for 4 dyas. Culture vessels used were watch glass for coculture with trophoblastic vesicles and micortube for estradiol-17$\beta$ infusion. Compared with the Medium-199 alone as basal culture medium, more blastocysts (46.7% vs 15.1%; P<0.01) and morulae (84.4% vs 56.6%; P<0.05) were developed in the co-culture with trophoblastic vesicles. Estradiol-17$\beta$ infused in culture medium was not effective for embryo development to blastocysts (78.3% in control, 50.0% in 0.1ng/ml, 61.5% in 1ng/ml and 64.4% in 10ng/ml) and also to morulae (91.3% in control, 84.2% in 0.1ng/ml, 92.3% in 1ng/ml and 91.1% in 10ng/ml). Compared with the watch glass culture mehotd, more (P<0.01) blastocysts were developed in microtube culture (78.3% vs 56.6%) and more (P<0.01) morulae in microtube culture (91.3% vs 56.6%).

Developing Proposals for Korean Traditional Culture Festivals (전통문화축제의 내실화방안 연구)

  • Since 1995, the Local autonomy era has opened, local festivals have been increasing throughout the country. Traditional culture festivals are also increasing. Performing traditional culture festivals is effective for maintenance and transmission of traditional culture as well as creation and nurturing of local culture. They are also effective promoting local economy. Depending on realization level of character, place and costume based on historic research, traditional culture festivals are classified as the "Representation event" or the "Reenactment event". But there are two problems. The first is unexacting classification method for festival types made by Central government yearly. The second is unable to meet one of two goals which are protecting traditional culture and stimulating local economy throughout increasing tourists. The purpose of this study is to suggest improvement of classification method for festival types, and to offer two suggestions for ensure successful local festivals. First of all, I suggest advanced classification method for festival types. For more sophisticated collection process of national festival status, local government department should ensure purpose of festivals, and central government department should add 3 steps to existing process. For example professional committee for judgement of festival types should be founded for consulting of local and central government department. The second suggestion is reinforcement of historic research for the Representation of traditional culture event. The representation of traditional culture should focus on protection of tradition, and could be perfect by continuous historic research. The last suggestion is cooperation with local governments each other for the Reenactment of traditional culture event. The Reenactment of traditional culture should focus on promoting local economy by increasing tourists. So local governments who have similar traditional events should cooperate to get preventing loss of resource and overcome weakness of promoting.

Comparative Analysis of Callus Induction and Plant Regeneration Rates Using One-step and Two-step Cultures for Rice Anther Cultivation (벼 약배양 1단계 및 2단계 배양을 이용한 캘러스 유도 및 식물 재부화율 비교 분석)

  • Anther cultivation for crop breeding is a method of rapid production of homozygosities by greatly reducing the time required for at least six generations to develop new varieties using conventional breeding methods. This technique of producing anther culture provides an opportunity to obtain more green plants from a methodological point of view, and the techniques that save time and effort in anther culture are also important because they increase the efficiency of culture. This study compared the callus induction rate and green plant regeneration rate of a one-step and a two-step culture that differ in their culture media and culture methods. One-step culture allows callus induction and plant regeneration in one medium, whereas two-step culture requires induction and plant regeneration in two different media. In this study, we compared the callus induction and plant regeneration rates of rice anthers as one-step and two-step cultures. The callus formation rate was 13.0% for one-step cultures and 8.6% for two-step cultures, so the rate was 4.4% higher for one-step cultures than for two-step cultures. The plant regeneration rate was 1.0% in one-step cultures and 3.0% in two-step cultures, so the regeneration rate was three times higher for the two-step cultures than for one-step cultures. This suggests that the two-step cultures are more efficient than the one-step cultures for haploid production.
