Effects of Extracellular Signaling on the Endogenous Expression of Self-Renewal-Stimulating Factor Genes in Mouse Embryonic Stem Cells

  • In order to provide the basis for developing practical mouse embryonic stem cells (mESCs) culture method, how the endogenous level of self-renewal-stimulating factor genes was altered in the mESCs by different extracellular signaling was investigated in this study. For different extracellular signaling, mESCs were cultured in 2 dimension (D), 3D and integrin-stimulating 3D culture system in the presence or absence of leukemia inhibitory factor (LIF) and transcriptional level of $Lif$, $Bmp4$ and $Wnt3a$ was evaluated in the mESCs cultured in each system. The expression of three genes was significantly increased in 3D system relative to 2D system under LIF-containing condition, while only $Wnt3a$ expression was increased by 3D culture under LIF-free condition. Stimulation of integrin signaling in mESCs within 3D system with exogenous LIF significantly up-regulated transcriptional level of $Bmp4$, but did not induce transcriptional regulation of $Lif$ and $Wnt3a$. In the absence of LIF inside 3D system, the expression of $Lif$ and $Bmp4$ was significantly increased by integrin signaling, while it significantly decreased $Wnt3a$ expression. Finally, the signal from exogenous LIF significantly caused increased expression of $Lif$ in 2D system, decreased expression of $Bmp4$ in both 2D and 3D system, and decreased expression of $Wnt3a$ in integrin-stimulating 3D system. From these results, we identified that endogenous expression level of self-renewal-stimulating factor genes in mESCs could be effectively regulated through artificial and proper manipulation of extracellular signaling. Moreover, synthetic 3D niche stimulating endogenous secretion of self-renewal-stimulating factors will be able to help develop growth factor-free maintenance system of mESCs.

천연 약용자원 추출물의 인수공통 감염 세균에 대한 in vivo 및 in vitro에서의 항균 효과 (In vivo and In vitro Antimicrobial Effects of Natural Antibiotics Present in Crude Extracts of Various Medicinal Plants)

  • Bacteria are among the most common causes of severe diseases in both plants and animals. Salmonella spp. has deleterious effects and is the cause of various transmittable diseases. Because of strains resistivity, side effects and high prices of synthetic antibiotics, it has become essential to explore safe and economical natural sources of antibiotics. In this study, growth inhibitory effects of natural antibiotics present in crude extracts of Galla rhois, Thujae semen, Paeonia japonica, and Armeniacae semen were investigated both in vivo and iv vitro. Ethanol extracts of the above-mentioned plants were prepared and tested against seven serovars of Salmonella and Escherichia coli by disc diffusion method. In addition, the antibacterial effects of the plant extracts were determined in vivo using ducks as model animals. Reverse transcription-polymerase chain reaction was performed using blood and fecal samples of control, infected, and treated groups of the ducks to determine the gene expression levels of the bacteria. Our results confirmed that the Galla rhois ethanol extract had the highest antibacterial activity among the plant extracts when they were used individually. However, the Galla rhois, Thujae semen, and P. japonica ethanol extracts showed stronger antibacterial effects against all the bacterial species used when the extracts were combined at a ratio of 3:3:2, respectively.

  • 채소 등 작물의 부패균인 Erwinia carotovora subsp. carotovora의 생육을 억제하는 길항세균을 경작지 표토층으로부터 분리하고 배양조건에 따른 억제력변화를 조사하고, 길항관련 유전자의 소재를 확인하였다. 분리균들중 가장 우수한 억제력을 갖는 S4, S14, S65 균주를 최종 선발하고 Pseudomonas 근연종으로 동정하였다. 523 배지를 사용하여 배양조건에 따른 억제력 변화를 조사한 결과 배지초기 pH를 7-8로 조정하여 $30^{\circ}C$에서 3일간 배양하였을 때 가장 높은 억제력을 보였고, 배지성분중 탄소원으로 mannitol과 sorbitol, 무기염으로 CaCl2를 첨가했을 때 효과적이었으나 질소원에 대한 감수성은 낮았다.

The effect of Korean Red Ginseng extract on rosiglitazone-induced improvement of glucose regulation in diet-induced obese mice

  • Background: Korean Red Ginseng extract (KRG, Panax ginseng Meyer) and its constituents have been used for treating diabetes. However, in diet-induced obese mice, it is unclear whether KRG can enhance the glucose-lowering action of rosiglitazone (ROSI), a peroxisome proliferator-activated receptor gamma synthetic activator. Methods: Oral glucose tolerance tests (oGTTs) were performed after 4 days of treatment with a vehicle (CON), KRG [500 mg/kg body weight (b.w.)], ROSI (3.75 mg/kg b.w, 7.5 mg/kg b.w, and 15 mg/kg b.w.), or ROSI and KRG (RK) in obese mice on a high-fat diet. Adipose tissue morphology, crown-like structures (CLSs), and inflammation were compared by hematoxylin-eosin staining or quantitative reverse transcription polymerase chain reaction. Results: The area under the glucose curve (AUC) was significantly lower in the RK group (15 mg/kg b.w. and 500 mg/kg b.w. for ROSI and KRG, respectively) than in the CON group. There was no significant difference in the AUC between the CON and the other groups. Furthermore, the AUC was significantly lower in the RK group than in the ROSI group. The expression of the Ccl2 gene and the number of CLSs were significantly reduced in the RK group than in the CON group. Conclusion: Our results show a potential enhancement of ROSI-induced improvement of glucose regulation by the combined treatment with KRG.

재조합 Vibrio parahaemolyticus 콜라겐분해효소의 분리 및 특성 분석 (Isolation and Characterization of Recombinant Vibrio parahaemolyticus Collagenase)

  • 식중독 병원균인 장염비브리오균 (V. parahaemolyticus)의 세포외 분비 효소 중 콜라겐분해효소를 발현벡터인 pET-29b에 클로닝시키고 대장균에서 발현시킨 다음, 부분정제하여 그 특성을 조사하였다. V. parahaemolyticus collagenase는 $(NH_4)_2So_4$침전, affinity adsorption, 그리고 Sephacryl S-100 gel filtration 과정을 통하여 부분정제 되었다. 이 collagenase는 73%의 회수율과 43.7의 정제도를 나타내었으며, 전기영동시 분자량은 약 35 kDa로 나타났다. 이 효소의 최적 pH 및 온도는 6~12와 $35^{\circ}C$이었고, 온도안정성 조사에서 $55^{\circ}C$까지는 90% 잔존 찬성을 유지하였으나 $60^{\circ}C$이상에서는 급격하게 효소활성이 실활되었다. 기질특이성조사에서 type I collagen과 콜라겐분해효소의 합성기질로 알려진 Z-GPGGPA에서 gelatin과 casein에 비해 높은 활성을 보이는 것으로 보아 이 효소가 진정한 콜라겐분해효소라는 것을 알 수 있었다.

Functional Identification and Expression of Indole-3-Pyruvate Decarboxylase from Paenibacillus polymyxa E681

  • Indole-3-acetic acid (IAA) is produced commonly by plants and many bacteria, however, little is known about the genetic basis involving the key enzymes of IAA biosynthetic pathways from Bacillus spp. IAA intermediates from the Gram-positive spore-forming bacterium Paenibacillus polymyxa E681 were investigated, which showed the existence of only an indole-3-pyruvic acid (IPA) pathway for IAA biosynthesis from the bacterium. Four open reading frames (ORFs) encoding indole-3-pyruvate decarboxylase-like proteins and putative indole-3-pyruvate decarboxylase (IPDC), a key enzyme in the IPA synthetic pathway, were found on the genome sequence database of P. polymyxa and cloned in Escherichia coli DH5$\alpha$. One of the ORFs, PP2_01257, was assigned as probable indole-3-pyruvate decarboxylase. The ORF consisted of 1,743 nucleotides encoding 581 amino acids with a deduced molecular mass of 63,380 Da. Alignment studies of the deduced amino acid sequence of the ORF with known IPDC sequences revealed conservation of several amino acids in PP2_01257, essential for substrate and cofactor binding. Recombinant protein, gene product of the ORF PP2_01257 from P. polymyxa E681, was expressed in E. coli BL21 (DE3) as a glutathione S-transferase (GST)-fusion protein and purified to homogeneity using affinity chromatography. The molecular mass of the purified enzyme showed about 63 kDa, corresponding closely to the expected molecular mass of IPDC. The indole-3-pyruvate decarboxylase activity of the recombinant protein, detected by HPLC, using IPA substrate in the enzyme reaction confirmed the identity and functionality of the enzyme IPDC from the E681 strain.

Synergistic Interactions of Schizostatin Identified from Schizophyllum commune with Demethylation Inhibitor Fungicides

  • Botrytis cinerea, which causes gray mold disease in more than 200 plant species, is an economically important pathogen that is mainly controlled by synthetic fungicides. Synergistic fungicide mixtures can help reduce fungicide residues in the environment and mitigate the development of fungicide-resistant strains. In this study, we screened microbial culture extracts on Botrytis cinerea to identify an antifungal synergist for tebuconazole. Among the 4,006 microbial extracts screened in this study, the culture extract from Schizophyllum commune displayed the most enhanced activity with a sub-lethal dosage of tebuconazole, and the active ingredient was identified as schizostatin. In combination with 5 ㎍/ml tebuconazole, schizostatin (1 ㎍/ml) showed disease control efficacy against gray mold on tomato leaf similar to that achieved with 20 ㎍/ml tebuconazole treatment alone. Interestingly, schizostatin showed demethylation inhibitor (DMI)-specific synergistic interactions in the crossed-paper strip assay using commercial fungicides. In a checkerboard assay with schizostatin and DMIs, the fractional inhibitory concentration values were 0.0938-0.375. To assess the molecular mechanisms underlying this synergism, the transcription levels of the ergosterol biosynthetic genes were observed in response to DMIs, schizostatin, and their mixtures. Treatment with DMIs increased the erg11 (the target gene of DMI fungicides) expression level 15.4-56.6-fold. However, treatment with a mixture of schizostatin and DMIs evidently reverted erg11 transcription levels to the pre-DMI treatment levels. These results show the potential of schizostatin as a natural antifungal synergist that can reduce the dose of DMIs applied in the field without compromising the disease control efficacy of the fungicides.

재조합 Corynebacterium glutamicum으로부터 헴첼 생산에 미치는 프로모터의 효과 (Effect of Promoters on the Heme Production in a Recombinant Corynebacterium glutamicum)

  • We published that bacterial heme was over-produced in a recombinant Corynebacterium glutamicum expressing 5-aminolevulinic acid synthase ($hemA^+$) under control of a constitutive promoter ($P_{180}$) and the heme-producing C. glutamicum had commercial potentials; as an iron feed additive for swine and as a preservative for lactic acid bacteria. To enhance the heme production, the $hemA^+$ gene was expressed under controls of various promoters in the recombinant C. glutamicum. The $hemA^+$ expression by $P_{gapA}$ (a constitutive glycolytic promoter of glyceraldehyde-3-phosphate dehydrogenase) led 75% increase of heme production while the expression by $P_{H36}$ (a constitutive, very strong synthetic promoter) resulted in 50% decrease compared with the control ($hemA^+$ expression by $P_{180}$ constitutive promoter). The $hemA^+$ expression by a late log-phase activating $P_{sod}$ (an oxidative-stress responding promoter of superoxide dismutase) led 50% greater heme production than the control. The $hemA^+$ expression led by a heat-shock responding chaperone promoter ($P_{dnaK}$) resulted in 121% increase of heme production at the optimized heat-shock conditions. The promoter strength and induction phase are discussed based on the results for the heme production at an industrial scale.

First Report of Leptosphaerulina saccharicola Isolated from Persimmon (Diospyros kaki) Tree Bark in Korea

  • A fungal strain, designated PTT-2, was isolated from the bark of the trunk of a persimmon (Diospyros kaki) tree in Cheongdo, Korea. The isolate showed morphological similarities with Leptosphaerulina saccharicola. Strain PTT-2 had more rapid growth on potato dextrose agar medium than on oatmeal agar, malt extract agar, and synthetic nutrient poor agar media, with colony sizes of 53.8 mm, 49.8 mm, 48.4 mm, and 28.1 mm after 7 days at $25^{\circ}C$ temperature, respectively. Strain PTT-2 produced ascospores, which had irregular wavy edges, oblong to ellipsoidal shape, hyaline appearance and $23.6{\times}10{\mu}m$ size. The black ascomata were developed on PDA medium, and asci were recorded. A BLAST search of the internal transcribed spacer (ITS) region, TEF1-${\alpha}$ and RPB2 gene sequences revealed that strain PTT-2 showed more than 99% nucleotide similarity with a strain of Leptosphaerulina saccharicola previously reported from Thailand. A neighbor-joining phylogenetic tree was constructed by concatenating the above-mentioned sequences, and showed that strain PTT-2 clustered in the same clade with L. saccharicola. Based on these findings, this is the first record of Leptosphaerulina saccharicola occurring in Korea.

KMS99220 Exerts Anti-Inflammatory Effects, Activates the Nrf2 Signaling and Interferes with IKK, JNK and p38 MAPK via HO-1

  • Neuroinflammation is an important contributor to the pathogenesis of neurodegenerative disorders including Parkinson's disease (PD). We previously reported that our novel synthetic compound KMS99220 has a good pharmacokinetic profile, enters the brain, exerts neuroprotective effect, and inhibits $NF{\kappa}B$ activation. To further assess the utility of KMS99220 as a potential therapeutic agent for PD, we tested whether KMS99220 exerts an anti-inflammatory effect in vivo and examined the molecular mechanism mediating this phenomenon. In 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-treated mice, oral administration of KMS99220 attenuated microglial activation and decreased the levels of inducible nitric oxide synthase and interleukin 1 beta ($IL-1{\beta}$) in the nigrostriatal system. In lipopolysaccharide (LPS)-challenged BV-2 microglial cells, KMS99220 suppressed the production and expression of $IL-1{\beta}$. In the activated microglia, KMS99220 reduced the phosphorylation of $I{\kappa}B$ kinase, c-Jun N-terminal kinase, and p38 MAP kinase; this effect was mediated by heme oxygenase-1 (HO-1), as both gene silencing and pharmacological inhibition of HO-1 abolished the effect of KMS99220. KMS99220 induced nuclear translocation of the transcription factor Nrf2 and expression of the Nrf2 target genes including HO-1. Together with our earlier findings, our current results show that KMS99220 may be a potential therapeutic agent for neuroinflammation-related neurodegenerative diseases such as PD.
