온도와 물리화학적 환경요인에 의한 상승작용의 규칙성에 관한 실험적 고찰 (Experimental Analysis on Regularities of Synergistic Interaction of Temperature with Physico-Chemical Environmental Factors)

  • 두 가지 유해인자가 동시에 작용할 때 생물체에 나타나는 영향은 길항작용에서 상승작용에 이르기까지 폭넓게 나타난다. 상승작용의 생물학적 중요성에도 불구하고 유해인자간의 복합작용을 설명할 수 있는 구체적 연구결과들은 별로 없는 실정이다. 본 연구는 고온과 기타 환경 유해인자의 복합작용에 의해 생물체에 나타내는 반응에 있어서의 규칙성을 도출해 내기 위하여 수행되었다. 고온과 이온화방사선, 자외선, 초음파 등의 동시작용에 관한 효모세포 실험결과를 분석하였으며 다른 연구자들에 의하여 수행된 바이러스, 세균포자, 동식물세포 및 포유류 배양세포에 관한 실험결과를 이용하여 도출된 규칙성을 검증하였다. 본 연구를 통하여 검증된 상승 작용에 관한 규칙성은 다음과 같이 요약될 수 있다. 방사선량률이 일정할 경우 특정 온도범위 내에서만 상승작용을 관찰할 수 있다. 방사선량률을 증가시키면 최대 상승작용을 나타내는 온도 값도 증가하게 되며, 선량률을 낮추면 온도값도 따라서 낮아지게 된다. 방사선 조사시의 온도가 일정할 경우 특정범위의 선량률에서만 상승작용이 관찰되는데, 이 범위 내에 최대상승작용 유발선량률 값이 존재한다. 방사선 조사시의 온도를 낮추면 선량률의 값 또한 낮아지며 그 역도 마찬가지이다. 본 연구에서 밝혀진 규칙성은 상승작용의 신개념 적립에 실마리를 제공할 것이며 특히 생물체에 대한 유해 환경 요인의 복합영향 해석과 환경재해 예방을 위한 영향평가 및 위해 평가에 유용하게 활용될 수 있을 것이다

Brassinolide와 Auxin과의 상호작용의 생물검정에 의한 평가 (Interaction between Brassinolide and Auxins on Bioassays)

  • 최근 국내에서 연구가 시작되고 있는 신규 식물생장조정제인 brassinolide류와 기존의 auxin류를 혼용하였을 경우 상호작용성을 구명하여 농업적 이용의 기초자료를 얻고자 몇가지 생물검정법을 이용하여 상호작용의 결과를 분석해 본 결과를 요약하면 다음과 간다. 1. 벼 엽신기부굴절검정에서는 두 화합물의 저농도 혼용조합에서 길항적인 반응을 보였으나, 대부분의 조합에서는 상가 또는 상승작용을 하였으며, 고농도의 혼용일수록 상승효과가 높았고, IAA가 2, 4-D에 비해 다소 혼용효과가 높았다. 2. 귀리 자엽소신장검정에서는 HBR의 농도에 관계없이 IAA 0.1ppm 이하에서는 상승작용을 하였으나 0.3ppm 이상의 혼용에서는 상가적인 반응을 나타내었다. 3. 무 하배축신장검정에서는 자엽의 유무에 의해 반응이 다르게 나타났는데, 자엽을 제거한 절편에서는 IAA 3ppm과 HBR과의 혼용조합을 제외하고는 대부분의 혼용조합이 길항적인 반응을 보였다. 4. 이상에서와 같이 검정방법간 혼용효과가 뚜렷한 차이가 있었으며, 공시재료에 따라 상호작용의 효과가 다르게 나타났는데 auxin의 함량이 비교적 높은 생장점을 가진 절편에서는 대체로 길항작용을 보인 반면, 선단부를 제거하여 내생호르몬이 불안정한 절편에서는 상가 또는 상승작용을 보였다.

Interaction between thyroglobulin and ADAMTS16 in premature ovarian failure

  • Objective: The aim of the present study was to examine whether interactions between polymorphisms in the thyroglobulin and ADAM metallopeptidase with thrombospondin type 1 motif, 16 (ADAMTS16) genes are associated with the development of premature ovarian failure (POF). Methods: A total of 75 patients with POF and 196 controls were involved in this study. We used a GoldenGate assay to genotype single nucleotide polymorphisms (SNPs). Logistic regression analysis was performed to identify POF-associated polymorphisms and synergistic interactions between polymorphisms in the thyroglobulin and ADAMTS16 genes. Results: Single gene analyses using logistic regression analysis showed no significant association between polymorphisms in the two genes and POF. In the results from interaction analyses, we found seven synergistic interactions between the polymorphisms in thyroglobulin and ADAMTS16, although there was no combination showing p-values lower than the significant threshold using the Bonferroni correction. When the AG genotype was present at the rs853326 missense SNP, the A and G alleles at the tagging SNPs rs16875268 and rs13168665 showed significant interactions (odds ratios=5.318 and 16.2 respectively; 95% confidence intervals, 1.64-17.28 and 2.08-126.4; p=0.0054 and 0.0079). Conclusion: Synergistic interactions between polymorphisms in the thyroglobulin and ADAMTS16 genes were associated with an increased risk of POF development in Korean women.

Machiavellianism in a Synergistic Tax Climate

  • This research examines the personal (i.e., Machiavellianism) and situational factors (i.e., tax climate) that are believed to be psychologically salient aspects in tax compliance. To the best of our knowledge, no research has been carried out to investigate the interaction effect of the two factors. This study uses a paper-and-pencil laboratory experiment 2x2 between-subject factorial design that involved 158 participants. The results indicate that a taxpayer who has a low Machiavellianism score or who is in a high synergistic tax climate reports a higher level of income. In the high synergistic tax climate, where tax norms apply, personal ethics do not play a significant role in tax compliance decisions. Where the synergistic relationship between taxpayer and authorities is low, personal ethics play an important role, i.e., low Machiavellians report a higher reported income than high Machiavellians do. This research contributes to the literature that deviates from the traditional model of tax compliance. Taxpayers are not always rational, but they might pay tax for reasons other than financial motives (Alm, 1991, 2018), that is, personal ethics in this study. This research implies the need for policymakers to consider other approaches rather than only relying on audits and fines.

Antibacterial Effect of Amentoflavone and Its Synergistic Effect with Antibiotics

  • Selaginella tamariscina is a traditional herb used in medicine. Phytochemical amentoflavone, a biflavonoid class of flavonoids, was isolated from the plant of Selaginella tamariscina. In this study, the antibacterial effects and combination effects of amentoflavone and conventional antibiotics such as ampicillin, cefotaxime, and chloramphenicol were investigated. These results showed that amentoflavone had a considerable antibacterial effect and synergistic interaction with antibiotics against various bacterial strains (fractional inhibitory concentration index ${\leq}$ 0.5), except for Streptococcus mutans. To study the mechanism(s) involved in the synergistic activities between amentoflavone and antibiotics, we detected hydroxyl radical formation using 3'-(p-hydroxyphenyl) fluorescein and measured the $NAD^+/NADH$ ratio by $NAD^+$ cycling assay. The results indicated that the formation of hydroxyl radical would be a cause of the synergistic effect and that this oxidative stress originated from a transient NADH depletion. This study suggests that amentoflavone synergizes with antibiotics and has potential as a therapeutic agent for antimicrobial chemotherapy.

Periplanetasin-2 Enhances the Antibacterial Properties of Vancomycin or Chloramphenicol in Escherichia coli

  • Periplanetasin-2 from cockroach exhibits broad-spectrum antimicrobial activity. The underlying antibacterial mechanisms rely on the stimulation of reactive oxygen species overproduction to induce apoptotic cell death. A promising strategy to increase the bioavailability of periplanetasin-2 involves reducing the dose through combination therapy with other antibacterials that show synergistic effects. Thus, the synergistic antibacterial activity of periplanetasin-2 with conventional antibacterial agents and its mechanisms was examined against Escherichia coli in this study. Among the agents tested, the combinations of periplanetasin-2 with vancomycin and chloramphenicol exhibited synergistic effects. Periplanetasin-2 in combination with vancomycin and chloramphenicol demonstrated antibacterial activity through the intracellular oxidative stress response. The combination with vancomycin resulted in the enhancement of bacterial apoptosis-like death, whereas the combination with chloramphenicol enhanced oxidative stress damage. These synergistic interactions of periplanetasin-2 can help broaden the spectrum of conventional antibiotics. The combination of antimicrobial peptides and conventional antibiotics is proposed as a novel perspective on treatments to combat severe bacterial infection.

  • 일년생잡초(一年生雜草) 피와 다년생잡초(多年生雜草) 너도방동산이 및 올미에 대한 perfluidone과 bifenox의 혼합처리(混合處理)에 의한 상호작용(相互作用)을 colby법(法) 또는 등효과선법(等效果線法)(Isobole)에 의(依)하여 평가(評價)하였다. 1. 피(1.0~1.5엽(葉))에 대(對)한 두 제초제(除草劑)의 상호작용(相互作用)은 상승효과(相乘效果)를 나타내었으며 상호작용지수(相互作用指數) 1의 최대(最大) 치(値)는 perfluidone과 bifenox의 조합비(組合費) 10:24.9ga.i./10a 일 때로 나타났다. 2. 너도방동산이(1.0~1.5 엽(葉))에 대해서도 두 제초제(除草劑)의 혼합처리시(混合處理時)에 상승효과(相乘效果)를 나타냈으며, perfluidone과 bifenox의 혼합비(混合比)가 10:33.3 ga.i./10a 일 때 상호작용지수(相互作用指數) I의 최대치(最大値)를 나타냈다. 3. 올미(0.5~1.0 엽(葉))에 대해서는 각(各) 단제처리시(單劑處理詩)의 효과(效果)가 매우 우수하였던 관계로 혼합처리시(混合處理時)의 그 상호작용(相互作用)은 검정(檢定)키 어려웠다.

Synergistic Effect of Flavonoids from Artocarpus heterophyllus Heartwoods on Anticancer Activity of Cisplatin Against H460 and MCF-7 Cell Lines

  • Artocarpus heterophyllus has been used as traditional medicine. This plant is one of the sources of flavonoid. Flavonoid compounds possessed a wide range of biological properties including anticancer. This study was performed to investigate the cytotoxic effect of flavonoids from A. heterophyllus on H460 and MCF-7 cell lines. The interaction of flavonoids and cisplatin against tested cancer cells was also evaluated. MTT assay was used to determine the cytotoxic effect of flavonoid. Isobologram analysis was selected to evaluate the synergistic effect between flavonoid and cisplatin, their interaction was then confirmed using AO/PI staining method. Amongst of flavonoid compounds, artocarpin exhibited strong cytotoxic effect on both MCF-7 and H460 cell lines with IC50 values of 12.53 ㎍/mL (28.73 μM) and 9.77 ㎍/mL (22.40 μM), respectively. This compound enhanced anticancer activity of cisplatin against H460 and MCF-7. The combination produced a synergistic effect on H460 and MCF-7 cell lines with a combination index (CI) values of 0.2 and 0.18, respectively. The AO/PI stained demonstrated that the combination of artocarpin and cisplatin caused morphological changes that indicated apoptosis. Moreover, artocarpanone also significantly increased cytotoxic effect of cisplatin compared to its single concentration with CI below than 1. This result suggested the potency of flavonoid named artocarpin to enhance the anticancer activity of cisplatin on H460 and MCF-7 cell lines.
