Carbon Tetrachlorid으로 유발된 Rat의 만성 장기(간, 신장) 손상시 헛개나무열매 추출물등 6종 생약의 투여시 조직병리학적변화 (Therapeutic Effect of Hovenia dulcis Thunberg Extracts and 6 Types Herbal Extracts on $CCl_4-induced$ Chronic Organ Damages in the Rats-Liver and Kidney)

  • This study was conducted to clarify the effects of the Korean raisin (Hovenia dulcis Thunb) extracts and polyherbal extracts consisted of 6 types herbal extracts including HDT (HDTmix) on $CCl_4$ induced organ damages. Extracts were prepared by autoclave ($121^{\circ}C$, 15 psi, 3 hours) and filtered with nylon cloth and filter paper then freezing dried. Male Sprague-Dawley rats $(200^{\circ}{\ae}20g)$ were used as experimental groups, which were divided into 5 groups; intact control group (100 mg/kg), $CCl_4$ dosing group (100 mg/kg), $CCl_4$ dosing after HDT extract dosing group (100 mg/kg), $CCl_4$ dosing after HDTmix dosing group (100 mg/kg), Silymarin dosing group (5 mg/kg) after all test articles were orally dosed once a day for 28 days. They were sacrifiled under ether anesthesia. HDT extracts and HDTmix dramatically inhibits the $CCl_4$ intoxicated hepato/nephropathies with immuno-suppress changes on the spleen. They showed more dramatical protective effects on most of specific organs compared to that of Silymarin 5 mg/kg except for hepatoprotective effects in which, quite similar effects were detected. In addition, HDT extracts showed synergic effects with other types of herbal extracts because HDTmix showed more favorable protective effects on the all specific organs showing $CCl_4-related$ histopathological changes compared to HDT extracts.

혈관신생 및 이식암세포증식 억제를 통한 가미소암산의 항암작용연구 (Studies on the Antitumor Activity of Gamisoam-san via Suppressing Angiogenesis and Growth Factor Expression)

  • Gamisoamsan is a prescription originated in Soamsan which is known as an anti-cancer remedy in the traditional Korean Medicine. To enhance the synergic effects of anti-cancer activity of Soamsan, this study reconstituted the original components of Soamsan with a slight modification and produced a novel herbal remedy, namely Gamisoamsan. To investigate the effects of Gamisoamsan on anti-cancer reaction, I studied the effects of Gamisoamsan on angiogenesis via chorioallantoic membrane (CAM) assay, corneal neovascularization assay and the effects on expression of growth factor which are VEGF, TGF-β, bFGF and IMUP-1. Anti-cancer effects of Gamisoamsan was also abserved through hematological parameters, tumor volume and survival rate in mice. Gamisoamsan inhibited embryonic angiogenesis of blood vessels in CAM assay and inhibited neovascularization of ral cornea. Gamisoamsan reduced cell proliferation in HT1080 cells and IC50 was 2.18 ㎎/㎖ Gamisoamsan reduced the expression of VEGF, TGF-β, bFGF and IMUP-1 which was known as vascular growth factor and this effects of Gamisoamsan was predominant than VP-16. The treatment of Gamisoamsan decreased the CT-26 cell inoculated-tumor volume in mice colon adenocarcinoma and increased mice survival which was inoculated CT-26 cells. The results of the present study suggest that Gamisoamsan extracts has a potential anti-tumor activity and may be an useful remedy to prevent and/or treat cancer.

Concentration-dependent in vitro Anti-osteoarthritis Effects of Mixed Formula - Pomegranate Concentrate Powder: Eucommiae Cortex: Achyranthis Radix 5:4:1 (g/g) on the Primary Cultured Rat Articular Chondrocytes

  • The objective of present study is to evaluate concentration-dependent in vitro anti-osteoarthritic (OA) effects of synergic mixed formula consisted of dried pomegranate juice concentrate powder, Eucommiae Cortex aqueous extract and Achyranthis Radix aqueous extract 5:4:1 (g/g) mixture on the primary cultured rat articular chondrocytes. First, any cytotoxic effect of mixture was observed using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium Bromide) assay. Next, cyto-protective effect of test substances was evaluated by using the recombinant human interleukin $(rhIL)-1{\alpha}$ induced chondrocytes. In addition, anti-inflammatory effects were also observed on the lipopolysaccaride (LPS) treated chondrocytes through prostaglandin $E_2(PGE_2)$ productions and 5-lipoxygenase (LPO) activities, and inhibitory effects on matrix metalloproteinase (MMP)-2 and MMP-9 activities were observed on $rhIL-1{\alpha}$ treated chondrocytes with their extracellular matrix (ECM) related mRNA expressions. No obvious cytotoxic effects of mixture were demonstrated. Inflammatory damages of chondrocytes and related ECM degradations induced by treatment of LPS or $rhIL-1{\alpha}$ were significantly and concentration-dependently inhibited by pretreatment of mixture from a concentration level of 0.001 mg/ml to 1 mg/ml. In addition, mixture showed $IC_{50}$ for $rhIL-1{\alpha}-induced$ MMP-2 and MMP-9 activities as 44.01 and $162.47{\mu}g/ml$, and also showed $EC_{50}$ for $rhIL-1{\alpha}-induced$ inhibition of collagen type II, SOX9 and aggrecan mRNA expression as 8.61, 10.79 and $4.47{\mu}g/ml$, respectively. It is observed that mixture showed concentration-dependent anti-inflammatory and cytoprotective ECM preserved effects on the primary cultured rat articular chondrocytes without cytotoxicity.

  • Objectives : The effects of Jaeumkanghwatang (JEKHT) co-administration on the pharmacokinetics of tamoxifen were observed after oral combination treatment of tamoxifen 50 mg/kg with JEKHT 100 mg/kg on JEKHT 6-day repeated oral pretreated rats with 8-day repeated co-administration to confirm the effects of JEKHT co-administration on the pharmacokinetics of tamoxifen. Methods : Six days after pretreatment of JEKHT 100 mg/kg, tamoxifen 50 mg/kg was co-administered with JEKHT 100 mg/kg, once a day for 8 days within 5 min. The blood were collected at 30 min before administration, 30 min, 1, 2, 3, 4, 6, 8 and 24 hrs after end of first and last 8th tamoxifen treatment, and plasma concentrations of tamoxifen were analyzed using LC-MS/MS methods. PK parameters of tamoxifen ($T_{max}$, $C_{max}$, AUC, $t_{1/2}$ and $MRT_{inf}$) were analysis as compared with tamoxifen single administered. Results : Six-day repeated oral pretreatment of JEKHT and 8-day repeated oral co-administration of tamoxifen within 5 min did not influenced on the plasma concentrations and pharmacokinetic parameters of tamoxifen, oral bioavailability, as compared with tamoxifen single treated rats, except for some negligible effects. Conclusions : It is concluded that JEKHT did not influenced on the plasma concentrations and pharmacokinetic parameters, the oral bioavailability of tamoxifen. Therefore, it is considered that co-administration of JEKHT and tamoxifen will be provide an effective novel treatment regimen on the comprehensive and integrative medicine for breast cancer patients, if they showed favorable synergic effects on the pharmacodynamics or reduce the tamoxifen treatment related toxicity and side effects in future studies.

녹차 폴리페놀에 노출된 Imipenem 내성 Pseudomonas aeruginosa의 항균효과 및 세포반응 (Antibacterial Effects and Cellular Responses of Imipenem-resistant Pseudomonas aeruginosa Exposed to Green Tea Polyphenols)

  • 본 연구는 항생제인 imipenem에 내성이 있는 Pseudomonas aeruginosa에 대한 차 폴리페놀(TPP)과 imipenem의 살균 상승효과와 세포반응을 조사하기 위하여 수행되었다. Imipenem 내성 Ps. aeruginosa는 병원의 환자로부터 분리하였다. TPP와 imipenem을 단독으로 처리하였을 때와 병용으로 처리하였을 때의 최소억제농도(MIC)를 측정한 결과, imipenem 감수성과 내성 균주는 TPP와 imipenem을 병용처리 하였을 때, imipenem 농도가 각각 16배, 8배가 감소되는 것을 확인하였다. 또한, time-kill 조사를 통해 TPP와 imipenem의 항균효과를 조사하였으며, 병용처리 하였을 때 낮은 농도의 imipenem에서도 동일한 항균효과를 나타내는 것을 확인하였다. TPP에 의한 imipenem 감수성과 내성 균주의 스트레스 충격 단백질 발현을 조사하기 위하여 anti-DnaK와 anti-GroEL 단일항체를 이용한 Western blot을 통해 관찰하였다. 스트레스 충격 단백질인 DnaK와 GroEL은 TPP의 노출시간이 증가함에 따라 발현양이 증가하다가 감소하는 것을 확인하였으며, 유도된 DnaK와 GroEL의 분자량은 각각 70 kDa과 60 kDa으로 나타났다. TPP의 농도와 시간에 따른 세균의 LPS 증감 변화를 SDS-PAGE와 은 염색을 통하여 확인하였고, TPP와 imipenem에 노출된 세균의 세포 외부 형태변화를 주사전자현미경을 이용하여 관찰한 결과, 움푹 패이고, 주름진 표면을 가지는 것으로 관찰되었다.

전나무 정유의 항진균 효과와 유효성분의 시너지효과 평가 (Evaluation on Anti-fungal Activity and Synergy Effects of Essential Oil and Their Constituents from Abies holophylla)

  • 본 연구는 전나무 정유의 Epidermophyton floccosum, Trichophyton mentagrophytes, Trichophyton rubrum과 같은 피부사상균에 대한 항진균 활성을 구명하고 피부염 치료제로서의 전나무 정유의 잠재력을 평가하고자 하였다. 전나무 정유와 분획물, 유효성분의 표준시약을 이용하여 한천희석법, paper disc 확산법, TEM을 이용한 세포 변화 관찰 등을 통해 항진균 활성을 평가하였고, 주성분은 GC-MS로 분석하였다. 유효성분들의 시너지효과는 체커보드법을 통해 평가하였다. 그 결과, 가장 높은 활성을 나타낸 물질은 ${\alpha}$-bisabolol을 함유하고 있는 분획 G4였으며 다른 분획 보다 뛰어나게 높은 활성을 나타냈다. 체커보드법을 통해 ${\alpha}$-bisabolol과 bornyl acetate를 병용시 뛰어난시너지 효과를 나타냈고 세포 변화 관찰을 통해 분획 G4에 노출된 E. floccosum과 T. rubrum 균사는 정상세포와 달리 세포막과 세포소기관이 파괴됨이 관찰되었다. 따라서 전나무 정유와 그 구성성분들은 높은 항진균 효과를 가지고 있기 때문에 피부염 치료제의 원료로 이용이 가능할 것이라 기대되며, 특히 ${\alpha}$-bisabolol의 우수한 항진균 활성에 주목할 필요가 있다고 사료된다.

용담고삼환(龍膽苦參丸) 에탄올 추출물의 Streptococcus mutans에 대한 항균활성에 관한 연구 (Antimicrobial Effects of Ethanol Extract of Yongdamgosam-hwan against Streptococcus mutans)

  • Objectives : Yongdamgosam-hwan(YGH) has been used as a traditional medicine from old times for antiinflammatory effects. Streptococcus mutans(S. mutans) is known as a prime bacteria responsible for causing caries by forming a biofilm referred to as dental plaque on the tooth surface. But antimicrobial activity of YGH with dental disease is not sufficiently understood. This study was designed to investigate the effects of YGH ethanol extract on antimicrobial effect against Streptococcus mutans.Methods : The antimicrobial effect of YGH ethanol extract was assessed by the paper disk diffusion method and optical density method to determine minimum inhibition concentration(MIC), also observed by fractional inhibitory concentration index(FICI) and time-kill assay to figure out the synergic effect on the combination of YGH ethanol extract with antibiotics.Results : The YGH ethanol extract 500 μg was 7.5-8.5 mm diameter of clear zone of inhibition against Streptococcus mutans in a concentration-dependent manner and MIC was 250 μg/mL. The administration of the ethanol extract in combination with gentamicin and streptomycin induced a reduction of ≥4-8-fold in all tested bacteria. Furthermore, time-kill study was found that a combination of YGH ethanol extract with oxacillin and streptomycin produced a more rapid decrease in the concentration of bacteria CFU/mL than the YGH ethanol extract or antibiotics alone.Conclusions : As a result, the YGH ethanol extract has good antimicrobial effects. And the results suggest that YGH could be employed as a natural antibacterial agent in dental care products.

은교산과 Rufloxacin 병용이 호기성 Gram(+) 세균주에 대한 시험관내 항균력에 미치는 영향 (Effects of In Vitro Synergism of Eunkyo-san and Rufloxacin against 9 Strains of Aerobic Gram(+) Bacteria)

  • In order to evaluate the in vitro synergic effect of Eunkyo-san, with quinolone antibiotics, rufloxacin (RUFX), the minimal inhibitory concentration (MIC), MIC50 and MIC90 of single use of quinolones and concomitant treatment with Eunkyo-san against 9 strains of aerobic gram positive bacteria. The obtained results were as follows : In the case of aerobic gram positive bacteria, the MIC, MIC50 and MIC90 against Staphylococcus aureus, Staphylococcus aureus smith, Staphylococcus epidermidis, Staphylococcus pyogens, Streptococcus pneumoniae Type Ⅰ, Type Ⅱ and Type Ⅲ was significantly decreased in concomitant treated groups with Eunkyo-san compared to those of single treated groups of RUFX, respectively. However, no significant changes were demonstrated against Bacillus subtilis and Enterococcus faecalis. In conclusion, the in vitro antibacterial activity of RUFX were increased against some strains of aerobic gram positive strains, especially, pneumococcus such as Staphylococcus and Streptococcus by concomitant use of Eunkyo-san.

혼합 난연제를 함유하는 UV 경화형 Epoxy Acrylate Resin의 난연 특성 (Flame Retardancy of UV-Cured Epoxy Acrylate Resin Containing Combined Flame Retardants)

  • 대표적인 UV 경화형 수지로 알려진 epoxy acrylate에 인계, 브롬계 난연제 및 금속 수산화물을 단독 또는 혼합하여 첨가하고 함량에 따른 난연 효과를 조사하였다. 난연성은 UL-94 V(ASTM D3801-87) 테스트법을 이용하여 측정하였다. 난연제 도입에 따른 물성의 저하를 막기 위해 montmorillonite(MMT)를 난연 보조제로 첨가하여 최종 경화체의 인장 특성의 변화를 확인하였다. 단일 난연제의 경우 브롬계 난연제인 decabromo-diphenyl oxide (DECA)가 함 할로겐 인계 난연제인 2,2,2-trichloroethyl dichlorophosphate (TCEDP)보다 낮은 함량에서 우수한 난연 효과를 나타냈으며 TCEDP의 함량이 증가할수록 경화 속도와 최종 전환율이 저하됨을 확인하였다. 금속 수산화물인 $Mg(OH)_2$는 40 wt%가 첨가되어도 난연 효과가 나타나지 않았다. 특히 DECA/TCEDP 혼합계는 MMT를 도입하지 않고도 기계적 물성의 저하 없이 탁월한 난연 효과가 나타나는 것으로 확인되었다.

The Clinical Effect of Manipulation of Acupuncture to Shen-Men and Nei-Kuan on Blood Pressure of Normal Male Subjects

  • Objectives : With an assumption of traditional oriental medical theory, to evaluate the effect of tonification/sedation manipulative acupuncture on blood pressure in male normotensives. Methods : This study was a randomized cross-over trial. We enrolled healthy male normotensive subjects, and assigned them randomly to group A or group B. Group A received acupuncture therapy on Shen-Men (He-7) and Nei-Kuan (EH-6) with tonification-manipulation, while group B received it with sedation-manipulation. Blood pressure was assessed every ten minutes for 2 hours. After 3 days of washout period, the subjects were crossed over to the other manipulation, and the same procedures were performed. Results : We observed that blood pressure stimulated by the sedation-manipulation had a tendency to decrease compared to that by the tonification-manipulation. Conclusions : We suggest the sedation manipulation has more synergic effect with the inhibitory effects of Shen-Men and Nei-Kuan on the sympathetic nervous system than the tonification-manipulation.
