Magnetic $Ni_{0.7}Co_{0.3}Fe_2O_4$ nanoparticles that were prepared via the rapid combustion process were functionalized and modified to obtain magnetic $Ni_{0.7}Co_{0.3}Fe_2O_4@SiO_2-CHO$ nanocomposites, on which penicillin G acylase (PGA) was covalently immobilized. Selections of immobilization concentration and time of fixation were explored. Catalytic performance of immobilized PGA was characterized. The free PGA had greatest activity at pH 8.0 and $45^{\circ}C$ while immobilized PGA's activities peaked at pH 7.5 and $45^{\circ}C$. Immobilized PGA had better thermal stability than free PGA at the range of $30-50^{\circ}C$ for different time intervals. The activity of free PGA would be 0 and that of immobilized PGA still retained some activities at $60^{\circ}C$ after 2 h. $V_{max}$ and $K_m$ of immobilized PGA were 1.55 mol/min and 0.15 mol/l, respectively. Free PGA's $V_{max}$ and $K_m$ separately were 0.74 mol/min and 0.028 mol/l. Immobilized PGA displayed more than 50% activity after 10 successive cycles. We concluded that immobilized PGA with magnetic $Ni_{0.7}Co_{0.3}Fe_2O_4@SiO_2-CHO$ nanocomposites could become a novel example for the immobilization of other amidohydrolases.
Uranium mill tailing ponds (UMTPs) are risk source of debris flow and a critical source of environmental U and Rn pollution. The technology of microbial induced calcium carbonate precipitation (MICP) has been extensively studied on reinforcement of UMTs, while little attention has been paid to the effects of MICP on U & Rn release, especially when incorporation of metakaolin and bacillus subtilis (MBS). In this study, the reinforcement and U & Rn immobilization role of MBS -MICP solidification in different grouting cycle for uranium mill tailings (UMTs) was comprehensively investigated. The results showed that under the action of about 166.7 g/L metakaolin and ~50% bacillus subtilis, the solidification cycle of MICP was shortened by 50%, the solidified bodies became brittle, and the axial stress increased by up to 7.9%, and U immobilization rates and Rn exhalation rates decrease by 12.6% and 0.8%, respectively. Therefore, the incorporation of MBS can enhance the triaxial compressive strength and improve the immobilization capacity of U and Rn of the UMTs bodies solidified during MICP, due to the reduction of pore volume and surface area, the formation of more crystals general gypsum and gismondine, as well as the enhancing of coprecipitation and encapsulation capacity.

In head and neck radiation therapy, the thermoplastic immobilization mask used for fixing the patient's posture and reproducibility causes scattered rays by being in close contact with the skin. To investigate the increase in skin dose due to the scattered rays generated from the immobilization mask, we evaluated dose reduction by decreasing contact between face skin and immobilization mask in computerized radiotherapy planning system with CT scanned images. In addition, to confirm the reproducibility problem of the setup due to the decrease in the cover area of immobilizing, the difference of each setup was confirmed using DRR and CT images. As the mask area covered for immobilizing was reduced, the dose on the skin surface significantly decreased, and it was confirmed that there was no significant difference in reproducibility even if the entire face was not covered and fixed.
Backgrounds : Two stress pathways, the hypothalamic-pituitary-adrenal (HPA) axis and the sympathetic nervous system, regulate immune system responses through release of corticosteroids, norepinephrine and epinephrine. respectively. These neuromediators act on immune cells via specific receptors on their surface to modulate the production of key regulatory cytokines. Objectives : To evaluate the preventive effects of oriental medicine Hyeolbuchukyeo-tang (HC) on stress. Methods : Mice were divided into three groups: nounal group, control group under immobilization stress and HC group which received Hyeolbuchukyeo-tang (HC) under immobilization stress. following sacrifice, their splenocytes were isolated and splenocyte surface markers were determined. The brains were removed and mRNA determined. In vivo, we separated RNA Iron cultured macrophages (RAW264.7). Results : In our study, immune functions were decreased in stress due mainly to changes of various neuromediators, cytokines and macrophage activities, and the treatment of HC increased those expressions. Conclusions : In summary, the present study documents the anti-stress effects of HC through stress-immune regulation.
Over the years, nanostructures have been developed to enable to support enzyme usability to obtain highly selective and efficient biocatalysts for catalyzing processes under various conditions. This review summarizes recent developments in the nanostructures for enzyme supporters, typically those formed with various inorganic materials. To improve enzyme attachment, the surface of nanomaterials is properly modified to express specific functional groups. Various materials and nanostructures can be applied to improve both enzyme activity and stability. The merits of the incorporation of enzymes in inorganic nanomaterials and unprecedented opportunities for enhanced enzyme properties are discussed. Finally, the limitations encountered with nanomaterial-based enzyme immobilization are discussed together with the future prospects of such systems.
The purpose of the present study was to determine whether the scuticocidal activity of olive flounder (Paralichthys olivaceus) head-kidney leucocytes can be enhanced by stimulation with polyinosine-polycytosine [poly (I:C)]. The growth of Miamiensis avidus was not affected by exposure to unstimulated or poly (I:C)-stimulated leucocytes alone, heat-inactivated immune serum alone, or unstimulated leucocytes plus heat-inactivated immune serum. However, leucocytes stimulated with poly (I:C) showed clearly high scuticocidal activity against M. avidus in the presence of heat-inactivated immune serum. Furthermore, numerous poly (I:C)-stimulated leucocytes occupied the surface of scuticociliates in the presence of the heat-inactivated immune serum, which led to lysis of scuticociliates. These results suggest that both of the stimulation of leukocytes and the immobilization of scuticociliates are necessary to kill scuticociliates by leukocytes.
RGDF immobilized micro-patterned surface was developed to detect the functional state of platelets. Using photolithographic technology, an RGDF micro-patterned surface was prepared on silicon wafer. Platelet adhesion to this surface was observed by fluorescence microscopy after staining platelets with mepacrine. Nonactivated platelets pretreated with $PGE_1$ interacted incompletely with the RGDF micro-patterned surface, whereas activated platelets treated with ADP interacted with the surface extensively. These results show that the distinct selectivity of an RGDF-immobilized micro-patterned surface can be used to detect the functional state of platelets.
BACKGROUND: Oyster shell(OS) is alkaline with pH 9.8, porous, and has high concentration of $CaCO_3$. It could be used as an alternative of lime fertilizer to immobilize cadmium(Cd) in heavy metal contaminated arable soil. Therefore, this study has been conducted to compare effects of calcium(Ca) materials [OS and $Ca(OH)_2$] on Cd extractability in contaminated soil and determined mechanisms of Cd immobilization with OS. METHODS AND RESULTS: Both Ca materials were added at the rates of 0, 0.1, 0.2, 0.4, and 0.8% (wt Ca wt-1) in Cd contaminated soil and the mixtures were incubated at $25^{\circ}C$ for 4 weeks. Both Ca materials increased pH and negative charge of soil with increasing Ca addition and decreased 1N $NH_4OAc$ extractable Cd concentration. 0.1 N HCl extractable Cd concentration markedly decreased with addition of OS. 1 N $NH_4OAc$ extractable Cd concentration was related with pH and net negative charge of soil, but not with 0.1 N HCl extractable Cd concentration. We assumed that Cd immobilization with $Ca(OH)_2$ was mainly attributed to Cd adsorption resulted from increase in pH-induced negative charge of soil. Scanning electron microscope (SEM) images and energy dispersive spectroscopy(EDS) analyses were conducted to determine mechanism of Cd immobilization with OS. There was no visible precipitation on surface of both Ca materials. However, Cd was detected in innerlayer of OS by EDS analyses but not in that of $Ca(OH)_2$. CONCLUSION: We concluded that Cd immobilization with OS was different from that with $Ca(OH)_2$. OS might adsorbed interlayer of oyster shell or have other chemical reactions.
We have expressed extracellular poly(3-hydroxybutyrate) (PHB) depolymerase of Ralstonia pickettii T1 on the Escherichia coli surface using Pseudomonas OprF protein as a fusion partner by C-terminal deletion-fusion strategy. Surface display of depolymerase was confirmed by flow cytometry, immunofluorescence microscopy and whole cell hydrolase activity. For the application, depolymerase was used as an immobilized catalyst of enantioselective hydrolysis reaction for the first time. After 48 h, (R)-methyl mandelate was completely hydrolyzed, and (S)-mandelic acid was produced with over 99% enantiomeric excess. Our findings suggest that surface displayed depolymerase on E. coli can be used as an enantioselective biocatalyst.
This research was conducted to develop a quick and sensitive method of detecting carbamate residues using the immobilization of antibody-antigen interactions with surface plasmon resonance (SPR). We have used commercialized surface plasmon resonance equipment (Biacore 3000). The antibody used for the immunoassay was specific for glutathione-s-transferase (GST) and the antigens included several carbamate pesticides (carbofuran, carbaryl, and benfuracarb). When antigens were applied to the protein GST, the detection limit was 2 ng/mL of carbamate pesticide. The fabricated protein GST maintained its activity for over 200 measurements. Thus we determined that the SPR biosensors could detect the specific reversible binding of a reactant in solution to a binding partner immobilized on the surface of the sensor and allow real-time detection and monitoring.
