우절의 페놀성 화합물의 분리 및 동정 (Phenolic Compounds from the Node of Lotus Rhizome (Nelumbo nucifera Gaertn))

  • The node of lotus rhizome (Nelumbo nucifera, Nymphaeaceae) have been used as a traditional medicine for the remedy of hemorrhage, blood stagnancy and thirstiness. To investigate phenolic compound from the node of Nelumbo nucifera, phytochemical isolation and structure elucidation were conducted. Four phenolic compounds were isolated from aqueous methanolic extract and the structure of these compounds were identised as (+)-catechin (1), (+)-gallocatechin (2), (+)-gallocatechin (4u-8)-catechin (3) and scolpoletin (4) respectively by the analysis of spectroscopic evidences and comparisions with the data of authentic samples.

SSI effects on the redistribution of seismic forces in one-storey R/C buildings

  • In the current work, a series of seismic analyses of one-storey asymmetrical reinforced concrete (R/C) framed buildings is accomplished while the effect of soil deformability on the structural response is investigated. A comparison is performed between the simplified elastic behavior of R/C elements according to the structural regulations' instructions to the possible non-linear behavior of R/C elements under actual circumstances. The target of the time history analyses is the elucidation of the Soil-Structure Interaction (SSI) effect in the seismic behavior of common R/C structures by examining the possible elastic or elastoplastic behavior of R/C sections because of the redistribution of the internal forces by employing a realistic damage index. The conclusions acquired from the presented elastic and elastoplastic analyses supply practical guidelines towards the safer design of structures.

  • Adaptation of infections and hosts has resulted in several metabolic mechanisms adopted by intracellular pathogens to combat the defense responses and the lack of fuel during infection. Human tuberculosis caused by Mycobacterium tuberculosis (MTB) is the world's first cause of mortality tied to a single disease. This study aims to characterize and anticipate potential antigen characteristics for promising vaccine candidates for the hypothetical protein of MTB through computational strategies. The protein is associated with the catalyzation of dithiol oxidation and/or disulfide reduction because of the protein's anticipated disulfide oxidoreductase properties. This investigation analyzed the protein's physicochemical characteristics, protein-protein interactions, subcellular locations, anticipated active sites, secondary and tertiary structures, allergenicity, antigenicity, and toxicity properties. The protein has significant active amino acid residues with no allergenicity, elevated antigenicity, and no toxicity.

제주도 방선균 유래 oxazolomycin 계열 KSM-2690 B의 구조 결정과 항균활성에 관한 연구 (Structure Elucidation and Antibacterial Activity of Oxazolomycin Family KSM-2690 B Derived from Actinomycete Collected in Jeju Island)

  • KSM-2690 B (1), a peptide-polyketide hybrid compound, was discovered from an actinomycete strain (CJD 1) isolated from Dong-Baek hill on Jeju Island, Republic of Korea. The chemical structure of 1 was identified by using NMR, MS, and UV spectroscopic analyses. Careful analysis of 1D and 2D NMR data revealed that KSM-2690 (1) has an oxazole ring, a β-lactone-γ-lactam spirocycle ring, and both triene and diene structures. KSM-2690 B (1) showed inhibitory activities against E. coli at 200 ㎍/mL.

Isolation, structure elucidation and physicochemical properties of novel antibiotic polypeptide, $\varepsilon-(L-\beta-Iysine)$ polypeptide from Streptomyces sp. DWGS2

  • During the screening of material which has the antimicrobial activity against aminoglycoside-resistant bacteria, A new material $\varepsilon$-(L-$\beta$-Iysine) polypeptide from a culture medium of Streptomyces sp.(DWGS2) was isolated, and the structure and the physicochemical properties of the new material were elucidated. The new material was separated by column chromatography of the culture medium using Dowex1$\times$2, Silica gel, and Sephadex LH20 etc. The chemical structure and molecular weight were determined with the data of various NMR experiments, MALDI mass, and ESI mass experiments. The antimicrobial activity of $\varepsilon$-(L-$\beta$-Iysine) polypeptide is not only better than equal to the activity of known aminoglycoside type of antibiotics(MIC=3.125 - 6.25ug/mL) but also effective against aminoglycoside-resistant bacteria and fungi. If the mechanism of antimicrobial activity against aminoglycoside- resistant bacteria is figured out, the $\varepsilon$-(L-$\beta$-Iysine) polypeptide can be utilized for the treatment of diseases caused by aminoglycoside-resistant bacteria.

  • A new isoflavone, $(+)-4'-O-methyl-8-C-{\beta}-D-glucopyranosylgenistein$, was isolated from the aerial parts of Spartidium saharae together with the known isoflavone, $(+)-8-C-{\beta}-D-glucopyranosylgenistein$ as well as dipiperidine alkaloids, (+)-ammodendrine and (+)-N-acetyl hystrine. Details of their structure elucidation are based on chemical and spectroscopic methods. N-formyl ammodendrine was detected by GC-MS. The potential chemotaxonomic value of the alkaloid content is explored. Cytotoxic activity has been determined for both alcoholic extract and isolated compounds.

  • 포자의 전자현미경사진과 mycelium의 광학현미경사진을 찍어 strain V-8 균주의 형태학적인 특성을 조사하였을 때 방선균 계열인 streptomyces로 확인되었다. 이 균주의 배양액은 노란색을 띠고 있고 이 노란색은 430 nm에서 특징적인 흡광을 보였다. 이 물질은 유기 용매로 추출되었다. 이 물질의 자외선 흡광스펙트럼의 특징을 기존에 보고된 화합물과 비교하여 이 물질이 actinomycin계열에 속함을 발견하였다. 이 물질의 FAB mass spectrum에서 각 분획들의 주성분들의 분자의 질량 + H 이온이 1290과 1292에서 각각 관찰되었다. 이 화합물들은 분자량이 1258인 actinomycin D와는 상이한 구조를 소유하고 있는 것으로 확인되었다. 각 분획 성분들의 proton 및 carbon HIR spectra를 얻어 기존에 알려진 화합물의 자료와 비교하였을 때 actinomycin계열에 속하는 신물질로 생각되었다. 지금까지 actinomycin 계열의 항생물질이 adenosine deaminase의 효소 활성을 저해한다는 사실은 보고된바 없다. 본 연구자는 분리한 2개의 화합물에 대하여 구조 연구를 계속하고 있다.

Structure and Function of the Phytochromes: Light Regulation of Plant Growth and Development

  • Light exerts two primary roles in plant growth and development. Plants acquire all biochemical energy required for growth and propagation solely from light energy via photosynthesis. In addition, light serves as a medium through which plants recognize environmental fluctuations, such as photoperiod and presence of neighboring animals and plants. Plants therefore constantly monitor the direction, intensity, duration, and wavelength of environmental light and integrate these light signals into the intrinsic regulatory programs to achieve an optimized growth in a given light condition. Although light regulates all aspects of plant growth and developmental aspects, the molecular mechanisms and signaling cascades involved have not been well established until recently. However, recent advances in genetic tools and plant transformation techniques greatly facilitated the elucidation of molecular events in plant photomorphogenesis. This mini-review summarizes the gist of recent findings in deetiolation and suppression of shade avoidance response as classic examples of the phytochrome-mediated photomorphogenesis.

Ardimerin, a New Dimeric Lactone from the Herb of Ardisia japonica

  • A new ardimerin (1) was isolated from the herb of Ardisia japonica. Its structure was determined by the elucidation of spectroscopic data and a chemical transformation. Compound (1) showed potent radical scavenging effect on DPPH radical $(IC_{50},\;0.32\;{\m}M)$.

  • Four compounds were isolated from the culture broth of Volvariella bombycina and they were identified as ergosta-4,6,8(14),22-tetraene-3-one (1), ergosterol peroxide (2), indole-3-carboxaldehyde (3), and indazole (4) by interpretation of spectroscopic data. Among them, compound 2 exhibited melanogenesis inhibitory effect in cultured B16 mouse melanoma cells.
