Background: Mitigation of primary water stress corrosion cracking (PWSCC) is a significant issue in the nuclear industry. Advanced nickel-based alloys with lower susceptibility have been adopted, although they do not seem to be entirely immune from PWSCC during normal operation. With regard to structural integrity assessments of the relevant components, an accurate evaluation of crack growth rate (CGR) is important. Methods: For the present study, the extended finite element method was adopted from among diverse meshless methods because of its advantages in arbitrary crack analysis. A user-subroutine based on the strain rate damage model was developed and incorporated into the crack growth evaluation. Results: The proposed method was verified by using the well-known Alloy 600 material with a reference CGR curve. The analyzed CGR curve of the alternative Alloy 690 material was then newly estimated by applying the proven method over a practical range of stress intensity factors. Conclusion: Reliable CGR curves were obtained without complex environmental facilities or a high degree of experimental effort. The proposed method may be used to assess the PWSCC resistance of nuclear components subjected to high residual stresses such as those resulting from dissimilar metal welding parts.
Zinc is an essential micronutrient with crucial roles in multiple facets of biological processes. Dysregulated zinc homeostasis impairs overall immune function and resultantly increases susceptibility to infection. Clinically, zinc supplementation is practiced for treatment of several infectious diseases, such as diarrhea and malaria. Recent focus on zinc as a beneficial element for immune system support has resulted in investigation of the immunomodulatory roles of zinc in a variety of immune cells. Besides its classical role as a cofactor that regulates the structural function of thousands of proteins, accumulating evidence suggests that zinc also acts, in a manner similar to calcium, as an ionic regulator of immune responses via participation as an intracellular messenger in signaling pathways. In this review, we focus on the role of zinc as a signaling molecule in major pathways such as those downstream of Toll-like receptors-, T cell receptor-, and cytokine-mediated signal transduction that regulate the activity and function of monocytes/macrophages and T cells, principal players in the innate and adaptive immune systems.
본 연구는 염색체 1, 2, 4, 7, 8, 9 및 21번 염색체의 DNA probe를 이용하여 2Gy의 방사선을 조사한 후 DNA 양을 감안한 기대치와 관찰치의 차이를 비교함으로서 각 염색체의 방사선에 대한 감수성을 평가하여 궁극적으로 방사선 피폭시 생물학적 선량계로서 FISH기법의 타당성을 평가하고자 하였다. 1번 및 4번 염색체의 경우 상호전좌와 이동원 염색체의 관찰치가 기대치보다 더 높게 나타났으며 이와 반대로 2, 7, 8 및 9번 염색체의 경우 상호전좌와 이동원염색체의 관찰치 모두 기대치보다 낮게 나타났다. 2번 및 4번 염색체의 경우 1번 염색체보다 더 많은 acentric fragment의 빈도를 나타내었다. 1, 2, 및 4번 염색체 3종을 조합했을 때 상호전좌의 경우 관찰치와 기대치는 세포 100개당 25.5 및 25.40으로 차이가 없었으며 이동원염색체의 경우 13.25 및 13.2로 역시 거의 차이가 없게 나타났다. 따라서 본 연구결과 방사선 피폭시 발생하는 염색체이상 빈도는 염색체마다 DNA 양에 비례해 나타나지 않을 수 있어 각 염색체마다 방사선 감수성에 차이가 있을 것으로 판단된다. 또한 방사선 피폭시 생물학적 선량계로서 1, 2 및 4번을 동시에 관찰 할 경우 염색체 FISH 법을 활용하기 위하여 적절한 염색체 조합이라고 판단된다.
2008년 3월, 서울의 상수원으로 이용되는 한강에서 분리된 Enterococcus 76주를 동정하고 이 균주들에 대한 항생제 감수성과 항생제 내성유전자 구조분석, 교차접합, PFGE 및 MLST를 실시하였다. 분리 균주 중 E. casseliflavus는 25주이었으며, E. faecalis 및 E. hirae가 각각 4주 및 1주이었다. 항생제 감수성을 조사한 결과 15주가 vancomycin에 대해 내성을 나타내었으며 E. faecium 11주와 E. casseliflavus 4주가 VRE인 것을 알 수 있었다. VRE를 대상으로 vanA 유전자 검출을 통해 6주의 E. faecium가 vanA를 가지는 VREF임을 밝혔으며, vanA가 포함된 transposon인 Tn1546 구조를 분석한 결과 Km36과 Km37은 Tn1 type, Km20과 Km38은 Tn2 type 그리고 Km 39와 Km40은 Tn3 type으로 나타났다. PFGE 결과 6주의 VREF 중 Km36과 Km37은 동일한 subtype을 나타내었으며, 나머지 4주는 각기 다른 subtype을 나타냈다. VREF 6주를 대상으로 MLST를 실시한 결과 3주는 ST78, 나머지 3주는 각각 ST18, ST192 및 ST230로 나타났다. 이들의 clonal complex는 모두 병원에서 분리되는 CC17로부터 파생된 것으로 파악되었으며 4주는 CC78에, 나머지 2주는 각각 CC18과 CC192에 포함된 것을 확인할 수 있었다.
임상가검물에서 생화학 검사와 항생제 감수성 검사를 통하여 45주의 Staphylococcu aureus를 분리하여 역학적 연구를 위한 형별 분류로써 항생제 감수성 검사, bacteriophage typing,효소중합 연쇄반응 등을 실시하였으며, methicillin 내성과 관련이 있는 mecA 유전자를 검출 및 역학적 변별력이 있는 가변지역의 중합효소증폭반응을 실시하여, 약제 내성과 관련된 구조 유전자의 발현 양상을 비교하여 특정 유전자 배열의 상동성을 밝히고자 하였다. 45주의 Staphylococcus aureus중에서 mecA 유전자 양성주는 30주였으며, 그 중에서 26주가 methicillin에 내성을 보였다. 약제 내성양상에 따라 9개의 antibiogram으로 분류되었으며, SA6을 제외한 균주에서 penicillin, oxacillin, gentamicin 및 chloramphenicol 에서는 높은 내성을 보였으며, SAl3, SAl4 및 SA27에서는 rifampin에 내성을 보였다. 27주에서 bacteriophage 형별 분류가 가능하였으며, Iytic group III가 12주로 가장 많았다. mecA 유전자와 mec관련 가변부위의 polymerase chain reaction을 실시한 결과 모든 methicillin resistant Staphylococcus aureus 에서는 533 bp의 증폭 band가 관찰되었으나, methicillin 감수성 균주에서는 증폭된 band가 관찰되지 않았다. mec관련 가변부위의 polymerase chain reaction에서는 200 bp에서 600 bp사이에 분포하여 4개의 유형으로 분류되었으며, 410bp인 C형이 10균주로 가장 많았다. C형 가변부위의 DNA sequence에서 40 bp가 반복되는 dru sequence를 관찰할 수 있었으며, 이러한 dru sequence는 4 unit가 직접적으로 중복됨을 알 수 있었다.
복합원산지제품 수용에 관한 지금까지의 많은 연구들이 다양한 시사점들을 제시해옴으로써 학문적인 기여를 해왔지만, 소비자의 구매성향에 따라 태도의 변화를 살펴본 연구는 매우 드물게 진행되어왔다. 본 연구는 기존 연구의 한계점을 보완하고자, 소비자의 구매성향을 합리적 소비성향, 브랜드 추구성향, 타인의식 소비성향으로 분류하여 고관여 복합원산지제품에 대한 수용과정이 어떻게 나타나는지를 파악해보고자 하였다. 또한 기존 연구의 측정방법을 보완하기 위하여 본 연구는 제품에 대한 복합원산지정보가 노출되기 전후의 상황을 구분함으로써, 소비자들의 수용행동을 보다 정교하게 분석하고자 하였다. 이를 위해 266명의 소비자들을 대상으로 설문조사를 실시하였으며, 구조방정식모형 분석을 통해 통계분석을 실시하였다. 연구모형의 검증결과, 소비자의 구매성향 가운데 타인의식 소비성향은 복합원산지제품의 순응과 반감에 모두 유의미한 영향을 미친다는 것을 알 수 있었다. 아울러, 복합원산지제품에 대한 반감수준은 순응수준보다 더욱 강력한 파급효과를 지닌다는 것을 확인할 수 있었다. 이상의 연구결과를 바탕으로 본 연구는 복합원산지제품 가운데 고관여제품을 판매하는 기업들을 대상으로 실무적으로 유용한 시사점을 제시하고자 하였다.
The effects of biological pretreatment on the Japanese red pine Pinus densiflora, was evaluated after exposure to three white rot fungi Ceriporia lacerata, Stereum hirsutum, and Polyporus brumalis. Change in chemical composition, structural modification, and their susceptibility to enzymatic saccharification in the degraded wood were analyzed. Of the three white rot fungi tested, S. hirsutum selectively degraded the lignin of this sortwood rather than the holocellulose component. After eight weeks of pretreatment with S. hirsutum, total weight loss was 10.7%, while lignin loss was the highest at 14.52% among the tested samples. However, holocellulose loss was lower at 7.81 % compared to those of C. lacerata and P. brumalis. Extracelluar enzymes from S. hirsutum showed higher activity of ligninase and lower activity of cellulase than those from other white rot fungi. Thus, total weight loss and changes in chemical composition of the Japanese red pine was well correlated with the enzyme activities related with lignin- and cellulose degradation in these fungi. Based on the data obtained from analysis of physical characterization of degraded wood by X-ray Diffractometry (XRD) and pore size distribution, S. hirsutum was considered as an effective potential fungus for biological pretreatment. In particular, the increase of available pore size of over 120 nm in pretreated wood powder with S. hirsutum made enzymes accessible for further enzymatic saccharification. When Japanese red pine chips treated with S. hirsutum were enzymatically saccharified using commercial enzymes (Cellulclast 1.5 L and Novozyme 188), sugar yield was greatly increased (21.01 %) compared to non-pre treated control samples, indicating that white rot fungus S. hirsutum provides an effective process in increasing sugar yield from woody biomass.

Prostate adenocarcinoma is one of the leading causes of cancer related mortality in men but still limited knowledge is available about its associated functional SNPs including rs1042522 (Pro72Arg). The present study was undertaken to explore the association of this SNP with susceptibility to prostate adenocarcinoma along with its structural and functional impacts in the Pakistani population in a case-control study. Three-dimensional structure of human TP53 with Pro72Arg polymorphism was predicted through homology modeling, refined and validated for detailed structure-based assessment. We also carried out a HuGE review of the previous available data for this polymorphism. Different genetic models were used to evaluate the genotypes association with the increased risk of PCa (Allelic contrast: OR=0.0.34, 95%CI 0.24-0.50, p=0.000; GG vs CC: OR=0.17, 95%CI 0.08-0.38, p=0.000; Homozygous: OR=0.08, 95%CI 0.04-0.15, p=0.000; GC vs CC: OR=2.14, 95%CI 1.01-4.51, p=0.046; Recessive model: OR=0.10, 95%CI 0.05-0.18, p=0.000; Log Additive: OR=3.54, 95%CI 2.13-5.89, p=0.000) except the Dominant model (OR=0.77, 95%CI 0.39-1.52, p=0.46). Structure and functional analysis revealed that the SNP in the proline rich domain is responsible for interaction with HRMT1L2 and WWOX. In conclusion, it was observed that the Arg coding G allele is highly associated with increased risk of prostate adenocarcinoma in the Pakistani population (p=0.000).
The innate immune recognition is based on the detection of microbial products. Toll-like receptors (TLRs) located on the cell surface and the endosome senses microbial components and nucleic acids, respectively. Chicken TLRs mediate immune responses by sensing ligands from pathogens, have been studied as immune adjuvants to increase the efficacy of vaccines. Single nucleotide polymorphisms (SNPs) of TLR3 and TLR4 genes in chicken were associated with resistance and susceptibility to viral infection. In this study, SNPs of chTLR3 and chTLR4 genes were retrieved from public database and annotated with chicken reference genome. Three-dimensional models of the chTLR3 and chTLR4 proteins were built using a Swiss modeler. We identified 35 and 13 nsSNPs in chTLR3 and chTLR4 genes respectively. Sorting Intolerant from Tolerant (SIFT) and Polymorphism Phenotyping v2 (Polyphen-2) analyses, suggested that, out of 35 and 13 nsSNPs, 4 and 2 SNPs were identified to be deleterious in chTLR3 and chTLR4 gene respectively. In chTLR3, 1 deleterious SNP was located in ectodomain and 3 were located in the Toll / IL-1 receptor (TIR) domain. Further structural model of chTLR3-TIR domain suggested that 1 deleterious SNP be present in the B-B loop region, which is important for TIR-TIR domain interactions in the downstream signaling. In chTLR4, the deleterious SNPs were located both in the ectodomain and TIR domain. SNPs predicted for chTLR3 and chTLR4 in this study, might be related to resistance or susceptible to viral infection in chickens. Results from this study will be useful to develop the effective measures in chicken against infectious diseases.
Several species of Gram-positive bacteria have cell wall peptidoglycan (syn. murein) in which not all of the sugar moieties are N-acetylated. This has recently been shown to be a secondary effect, caused by the action of a peptidoglycan N-acetylglucosamine deacetylase. We have found that the opportunistic pathogen Listeria monocytogenes is unusual in having three enzymes with such activity, two of which remain in the cytoplasm. Here, we examine the enzyme (PgdA) that crosses the cytoplasmic membrane and is localized in the cell wall. We purified a hexa-His-tagged form of PgdA to study its activity and constructed a mutant devoid of functional Lmo0415 (PgdA) protein. L. monocytogenes PgdA protein exhibited peptidoglycan N-acetylglucosamine deacetylase activity with natural substrates (peptidoglycan) from both L. monocytogenes and Escherichia coli as well as the peptidoglycan sugar chain component N-acetylglucosamine, but not with N-acetylmuramic acid. As was reported recently [6], inactivation of the structural gene was not lethal for L. monocytogenes nor did it affect growth rate or morphology of the cells. However, the pgdA mutant was more prone to autolysis induced by such agents as Triton X-100 and EDTA, and is more susceptible to the cationic antimicrobial peptides (CAMP) lysozyme and mutanolysin, using either peptidoglycan muramidases or autolysis-inducing agents. The pgdA mutant was also slightly more susceptible than the wild-type strain to the action of certain beta-lactam antibiotics. Our results indicate that protein PgdA plays a protective physiological role for listerial cells.
