Fungal Strain Improvement for Cellulase Production Using Repeated and Sequential Mutagenesis

  • A fungal strain producing a high level of cellulase was selected from 320 fungal isolates and identified as Aspergillus sp. This strain was further improved for cellulase production by sequential treatments by two repeated rounds of $\gamma$-irradiation of $Co^{60}$, ultraviolet treatment and four repeated rounds of treatment with N-methyl-N'-nitro-N-nitrosoguanidine. The best mutant strain, Aspergillus sp. XTG-4, was selected after screening and the activities of carboxymethyl cellulase, filter paper cellulase and $\beta$-glucosidase of the cellulase were improved by 2.03-, 3.20-, and 1.80-fold, respectively, when compared to the wild type strain. After being subcultured 19 times, the enzyme production of the mutant Aspergillus sp. XTG-4s was stable.

Selection of a L-Lysine-Overproducing Strain of the Red Seaweed Porphyra suborbiculata (Rhodophyta) through Mutation and Analog Enrichment

  • An improved strain of the red seaweed Porphyra suborbiculata containing an increased amount of the essential amino acid L-lysine was obtained through mutation and analog enrichment. Mutagenesis using a 10% lethal dose of ultraviolet irradiation and an enrichment culture with the L-lysine analog aminoethyl-L-cysteine (AEC) was repeated to select the most productive strain using monospores of P. suborbiculata. The concentrations of AEC required to produce 50 and 100% inhibition of survival were 60 and 115 mM in the parent strain, and 72 and 135 mM in the selected AEC-resistant strain, respectively. The AEC-resistant strain, L130, produced 1.74-fold more lysine compared to its parent strain. Thus, mutagenesis with analog enrichment shows promise for selecting seaweed strains that can overproduce this essential amino acid.

  • For decades, traditional mutation breeding technologies using spontaneous mutation, chemicals, or conventional radiation sources have contributed greatly to the improvement of crops and microorganisms of agricultural and industrial importance. However, new mutagens that can generate more diverse mutation spectra with minimal damage to the original organism are always in need. In this regard, ion beam irradiation, including proton-, helium-, and heavier-charged particle irradiation, is considered to be superior to traditional radiation mutagenesis. In particular, it has been suggested that ion beams predominantly produce strand breaks that often lead to mutations, which is not a situation frequently observed in mutagenesis induced by gamma-ray exposure. In this review, we briefly describe the general principles and history of particle accelerators, and then introduce their successful application in ion beam technology for the improvement of crops and microbes. In particular, a 100-MeV proton beam accelerator currently under construction by the Proton Engineering Frontier Project (PEFP) is discussed. The PEFP accelerator will hopefully prompt the utilization of ion beam technology for strain improvement, as well as for use in nuclear physics, medical science, biology, space technology, radiation technology and basic sciences.

  • The constant rate of strain (CRS) consolidation test has been widely used to evaluate consolidation characteristics of soils instead of the standard Incremental Loading Test. In practical problems, after the ground improvement, the condition of the soil is over-consolidated. Therefore, it is important to determine the recompression indices and the coefficient of consolidation(or the coefficient of swelling) of unloading-reloading cycle to predict the settlement behavior. However, since standard testing procedures or studies related with strain rate are insufficient especially in unloading-reloading cycle, it is difficult to predict the settlement field behavior accurately from the CRS consolidation test results in spite of its lots of strengths. The several CRS consolidation tests were performed changing the unloading strain rate from 0.2%/hr to 20%/hr with vertical drainage condition using the reconstituted kaolinite sample. For the reconstituted kaolinite sample in CRS consolidation test, the recompression indices are insensitive to the strain rate. It is revealed that the coefficient of consolidation of reloading is affected by the developed pore pressure during unloading. Additionally, the test should be conducted in the positive pore pressure ratio range (3~15%) to obtain the reasonable coefficient of consolidation in the whole range(loading, unloading and reloading).

Strain-specific Detection of Kimchi Starter Leuconostoc mesenteroides WiKim33 using Multiplex PCR

  • Leuconostoc spp. are generally utilized as kimchi starters, because these strains are expected to have beneficial effects on kimchi fermentation, including improvement of sensory characteristics. Here, we developed a detection method for verifying the presence of the kimchi starter Leuconostoc mesenteroides WiKim33, which is used for control of kimchi fermentation. A primer set for multiplex polymerase chain reaction was designed based on the nucleotide sequence of the plasmids in strain WiKim33, and their specificity was validated against 45 different strains of Leuconostoc spp. and 30 other strains. Furthermore, the starter strain consistently tested positive, regardless of the presence of other bacterial species in starter kimchi during the fermentation period. Our findings showed that application of a strain-specific primer set for strain WiKim33 presented a rapid, sensitive, and specific method for detection of this kimchi starter strain during natural kimchi fermentation.

  • This research aims to evaluated the structure improvement and new product development of valve tester. A valve tester was redesigned for structure improvement and new product development using 3-D design program CATIA. In addition, behavior analysis was conducted on the modeled valve tester using the ANSYS program. The total deformation, stress and strain were obtained by the internal pressure change. This result was applied to the new product development of valve tester.

Improvement of FK506 Production in the High-Yielding Strain Streptomyces sp. RM7011 by Engineering the Supply of Allylmalonyl-CoA Through a Combination of Genetic and Chemical Approach

  • FK506, a widely used immunosuppressant, is a 23-membered polyketide macrolide that is produced by several Streptomyces species. FK506 high-yielding strain Streptomyces sp. RM7011 was developed from the discovered Streptomyces sp. KCCM 11116P by random mutagenesis in our previous study. The results of transcript expression analysis showed that the transcription levels of tcsA, B, C, and D were increased in Streptomyces sp. RM7011 by 2.1-, 3.1-, 3.3-, and 4.1-fold, respectively, compared with Streptomyces sp. KCCM 11116P. The overexpression of tcsABCD g enes in Streptomyces sp. RM7011 gave rise to approximately 2.5-fold (238.1 μg/ml) increase in the level of FK506 production compared with that of Streptomyces sp. RM7011. When vinyl pentanoate was added into the culture broth of Streptomyces sp. RM7011, the level of FK506 production was approximately 2.2-fold (207.7 μg/ml) higher than that of the unsupplemented fermentation. Furthermore, supplementing the culture broth of Streptomyces sp. RM7011 expressing tcsABCD genes with vinyl pentanoate resulted in an additional 1.7-fold improvement in the FK506 titer (498.1 μg/ml) compared with that observed under non-supplemented condition. Overall, the level of FK506 production was increased approximately 5.2-fold by engineering the supply of allylmalonyl-CoA in the high-yielding strain Streptomyces sp. RM7011, using a combination of overexpressing tcsABCD genes and adding vinyl pentanoate, as compared with Streptomyces sp. RM7011 (95.3 μg/ml). Moreover, among the three precursors analyzed, pentanoate was the most effective precursor, supporting the highest titer of FK506 in the FK506 high-yielding strain Streptomyces sp. RM7011.

Strain Improvement for Enhanced Production of Streptokinase and Streptodornase in Streptococcus sp.

  • Strain improvement for the enhanced production of streptokinase and streptodornase in Streptococcus sp. ATCC 12449 was performed. Strain UB111, a hyperproductive mutant which was isolated by use of nitrosoguanidine and selection of colonies with large clear zones on DNase test agar plates supplemented with $1{\%}$ glucose and $0.5{\%}$ ammonium chloride, produced about 3 fold more streptokinase and streptodornase than the wild type when tested in shake flask fermentations. The enhanced production of both streptokinase and streptodornase was achieved by cultivating the mutant in a pH-controlled fermentor containing fermentation medium enriched with yeast extract ($2.1{\%}$). Under these conditions, the mutant produced 7300 units/ml of streptokinase and 800 units/ml of streptodornase.

  • This study is aimed to diminish the errors which created during transforming from 3D shape created with CAD program to STL file formation. The length strain which created on X-Y axises at STL file transforming is diminished by using the relation between common shape error and shrinkage rate. As the result of study, we have confirmed the length strain in accordance with facetres value. Also, the shape error is compensated with shrinkage rate by error of length strain. so, we could diminish to shape error by several tens micrometer.
