Background: This study represents the first report that the anti-obesity activity of ethanol extracts of Aronia melanocarpa can be enhanced through ultrasonification at a frequency of 120 kHz at $60^{\circ}C$ (UE). Methods and Results: The amounts of cyanidin-3-O-galactose (cya-gal), a major anthocyanin in A. melanocarpa were higher by up to 402.4 mg/100 g, as compared with 221.4 mg/100 g and 322.1 mg/100 g, for hot water at $100^{\circ}C$ and 70% ethanol at $80^{\circ}C$ respectively. This result should cause the higher antioxidant activities of the UE than extract of hot water and ethanol in DPPH free radical scavenging. It was confirmed that the high antioxidant activity of UE could play an important role in inhibiting the production of proteins related to adipocyte differentiation, such as peroxisome proliferator activated receptor-${\gamma}$ (PPAR-${\gamma}$) and sterol regulatory element binding protein 1 (SREBP1). Conclusions: Ultrasonification at a frequency of 120 kHz at $60^{\circ}C$ should result in better anti-obesity activity than that observed using other processes. It was also observed for the first time that the anti-obesity activity of A. melanocarpa was associated with its antioxidant activity, possibly due to the higher elution of intact cya-gal, owing to efficient low temperature ultrasonification extraction. These results could also be applied to improve other biological activities of medicinal herbs that contain many types of heat-labile bioactive substances.
Objectives : The purpose of this study is to evaluate the anti-lipogenic effect of Sobuncheong-eum on non-alcoholic fatty liver disease in free fatty acid induced cellular model. Methods : HepG2 cells were treated with palmitate for 24h to overload intracellular triglyceride (TG) content in the presence or absence of Sobuncheong-eum extract. After palmitate treatment, Intracellular TG content was measured with TG assay kit. Several lipogenesis related markers, including AMP-activated protein kinase (AMPK), sterol regulatory element-binding transcription factor-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), and fatty acid synthase (FAS), were assessed using Western-blot analyses and RT-PCR. Results : Palmitate markedly increased intracellular TG in HepG2 cells, and which were alleviated by coadministered Sobuncheong-eum extract. Sobuncheong-eum extract activated AMPK, which plays a key role in reducing hepatic lipid accumulation, and reduced lipogenic fators, SREBP-1c, ACC, and FAS. Conclusions : Taken together, it is conceivable that Sobuncheong-eum has an potential to alleviate steatosis, and which may be mediated by activating AMPK at least in part.
Objectives : Phyllostachys pubescens and Scutellaria baicalensis are considered to be effective in promoting blood circulation in traditional medicine. In this study, we examined whether a mixture of P. pubescens leaves and S. baicalensis root (BS21) had any anti-obesity, anti-hyperlipidemia, or anti-hyperuricemia effects and the possible mechanisms of action. Methods : We examined the effects of BS21 in high-fat diet (HFD)-induced obese mice. Mice were fed HFD with BS21 (75, 150, or 300 mg/kg) or Garcinia cambogia extracts (245 mg/kg) as a positive control for 8 weeks. At the end of 8 weeks, body weight, liver and adipose weight, adipocyte size, plasma lipid profiles, adipokine and uric acid levels, and adipose tissue expression levels in obesity and uric acid production-related genes were examined. Results : BS21 decreased body weight gain, white adipose tissue, liver weight, adipocyte size, and liver triglyceride accumulation. It also reduced levels of plasma glucose, triglycerides, non-esterified fatty acids, total cholesterol, low-density lipoprotein cholesterol, alanine transaminase, leptin, and uric acid. In contrast, BS21 increased adiponectin levels. Furthermore, BS21 decreased the expression levels of adipogenesis-related genes, such as peroxisome proliferator-activated receptor γ, sterol regulatory element binding protein-1c, and fatty acid synthase, as well as xanthine oxidoreductase, which is involved in uric acid production. Conclusions : These results suggest that BS21 may exert anti-obesity, anti-hyperlipidemia, and anti-hyperuricemia effects in HFD-induced obese mice by regulating the expression of xanthine oxidoreductase and adipogenesis-related genes.
Objectives : Although the anti-obesity effect of Schizandrae Fructus water extract has been demonstrated, its underlying mechanism is still unclear. Therefore, we aimed to evaluate the anti-obesity effect of Schizandrae Fructus water extract through the p-AMP-activated protein kinase (p-AMPK), sirtuin1 (Sirt1), and peroxisome proliferator-activated receptor-gamma coactivator 1α (PGC-1α) signaling in 60% high-fat diet (HFD)-induced obese mouse model. Methods : Male C57BL/6 mice were divided into four groups. The Normal group was fed a normal diet and the obese groups were fed 60% HFD. Except for the Control group, the GG group was supplemented with 0.5% Garcinia gummigutta and the SCW group was supplemented with 0.5% Schizandrae Fructus water extract. After 6 weeks, obesity-related biomarkers in serum were measured and the expressions of protein for lipid-related factors in liver tissue were analyzed by western blot. Results : Treatment with SCW significantly down-regulated body weight compared to the Control group. SCW down-regulated levels of triglyceride and total cholesterol in serum and significantly increased p-AMPK, Sirt1, and PGC-1α in liver tissue. In addition, the expressions of fatty acid oxidation-related proteins such as peroxisome proliferator-activated receptor α (PPARα), carnitine palmitoyltransferase 1A (CPT-1A), uncoupling protein 1 (UCP1), and uncoupling protein 3 (UCP3) were significantly up-regulated. However, fatty acid synthesis-related proteins including sterol regulatory element-binding protein-1 (SREBP-1), phospho-Acetyl-CoA Carboxylase (p-ACC), and fatty acid synthase (FAS) were significantly down-regulated. Conclusions : Taken together, SCW treatment showed anti-obesity effect by regulating both fatty acid oxidation-related and fatty acid synthesis-related proteins through AMPK/Sirt1/PGC-1α signaling in 60% HFD-induced obese mice.
BACKGROUND/OBJECTIVES: Non-alcoholic fatty liver disease (NAFLD) is a leading cause of chronic liver disease and is closely associated with metabolic syndrome. In the present study, we observed the effect of ethanol extract of Allium fistulosum (EAF) on NAFLD and have suggested the possibility of using EAF as a natural product for application in the development of a treatment for NAFLD. MATERIALS/METHODS: The preventive effect on hepatic lipid accumulation was estimated by using an oleic acid (OA)-induced NAFLD model in vitro and a Western diet (high-fat high-sucrose; WD)-induced obese mouse model. Animals were divided into three groups (n = 7): normal diet group (ND), WD group, and WD plus 1% EAF group. RESULTS: EAF reduced OA-stimulated lipid accumulation in HepG2 cells in the absence of cellular cytotoxicity and significantly blocked transcriptional activation of sterol regulatory element-binding protein 1 and fatty acid synthase genes. Subsequently, we investigated these effects in vivo in mice fed either ND or WD in the presence or absence of EAF supplementation. In comparison to the ND controls, the WD-fed mice exhibited increases in body weight, liver weight, epididymal fat weight, and accumulation of fat in hepatocytes, and these effects were significantly attenuated by EAF supplementation. CONCLUSIONS: Allium fistulosum attenuates the development of NAFLD, and EAF elicits anti-lipogenic activity in liver. Therefore, EAF represents a promising candidate for use in the development of novel therapeutic drugs or drug combinations for the prevention and treatment of NAFLD.

In this study, we aimed to investigate the effects of 8 weeks of treatment with a combination of evogliptin and leucine, a branched-chain amino acid, in mice with high-fat diet (HFD)-induced diabetes. Treatment with evogliptin alone or in combination with leucine reduced the body weight of the mice, compared to the case for those from the HFD control group. Long-term treatment with evogliptin alone or in combination with leucine resulted in a significant reduction in glucose intolerance; however, leucine alone did not affect postprandial glucose control, compared to the case for the mice from the HFD control group. Furthermore, the combination of evogliptin and leucine prevented HFD-induced insulin resistance, which was associated with improved homeostasis model assessment for insulin resistance, accompanied by markedly reduced liver fat deposition, hepatic triglyceride content, and plasma alanine aminotransferase levels. The combination of evogliptin and leucine increased the gene expression levels of hepatic peroxisome proliferator-activated receptor alpha, whereas those of the sterol regulatory element-binding protein 1 and stearoyl-CoA desaturase 1 were not altered, compared to the case in the HFD-fed mice (p<0.05). Thus, our results suggest that the combination of evogliptin and leucine may be beneficial for treating patients with type 2 diabetes and hepatic steatosis; however, further studies are needed to delineate the molecular mechanisms underlying the action of this combination.
FCEE를 3T3-L1 전지방세포의 분화과정 중에 처리한 후, FCEE가 지방의 축적에 미치는 영향을 확인하였다. FCEE($200{\mu}g/mL$)를 세포에 처리한 후 Oil Red O 염색법과 AdipoRed 형광염색법을 이용한 지방 축적과 세포내 중성지방 함량을 측정한 결과, 무처리한 지방세포와 비교하여 각각 10.2%, 13.7% 유의하게 감소하는 것을 확인하였다. 이러한 결과에 대한 기작을 확인하고자 포도당 유입량과 유리 글리세롤 방출량을 측정하였다. FCEE($200{\mu}g/mL$) 처리 시 지방세포와 비교하여 포도당 유입량이 36.6% 유의적으로 감소하였으며, 유리 글리세롤 방출량도 8일째에 유의적으로 감소하였다. 이는 지방세포의 분화과정 중에 FCEE의 처리로 분화가 억제되어 지방구의 형성이 억제되고 배양액으로 배출되는 유리 글리세롤의 총량이 감소한 것으로 판단된다. 이러한 결과가 지방세포 분화의 억제에 의한 것인지 판단하기 위해 지방 형성 관련인자의 mRNA 발현량을 측정하였다. FCEE($200{\mu}g/mL$)의 처리로 AMPK mRNA 발현량은 지방세포와 비교하여 3배 증가하였으며, SREBP-1c, $C/EBP{\alpha}$와 $PPAR{\gamma}$ mRNA 발현량은 각각 0.6, 0.6 및 0.8배 감소하는 것을 확인하였다. 이상의 결과들로부터 FCEE는 지방 합성을 억제하는 활성을 보유하고 있는 바, 향후 항비만 기능성 식품소재로 활용될 수 있을 것으로 사료된다.
지방전구세포가 지방세포로 분화하는 과정에서 아선약 추출물의 영향을 확인하고자 분화유도제를 처리하여 3T3-L1 세포에서 아선약의 세포독성, 지방축적 억제 효과, triglyceride 억제 효능, 지방분화 관련 단백질들의 발현을 확인하였다. 아선약 추출물을 최대 $200{\mu}g/mL$ 농도까지 세포에 처리하여 세포독성을 측정한 결과 아선약 추출물은 $100{\mu}g/mL$ 이하의 농도에서는 세포에 독성을 유발하지 않는 것으로 확인되었다. Oil red-O 염색을 통해 지방축적률을 확인한 결과 아선약 추출물에 의한 농도 의존적인 지방축적의 감소 효과가 있었다. Triglyceride 생성량 역시 농도 의존적인 감소가 확인되었다. 아선약에 의한 지방분화의 감소가 어떠한 기작에 의해 조절되는지 확인하고자 관련 지표단백질인 $PPAR{\gamma}$와 SREBF-1 그리고 $C/EBP{\alpha}$의 발현량 변화를 확인해본 결과 $100{\mu}g/mL$ 농도에서 유의성 있게 감소하였다. 이상의 결과를 종합해볼 때 아선약 추출물은 지방세포의 분화억제와 분화된 지방세포의 지방축적을 저해함으로써 항비만 효과를 유도할 것으로 기대된다.
Background: This study investigated the anti-obesity effect of the flavonoid rich fraction (FRF) and its constituent, rutin obtained from the leaf of Morus alba L., on the lipid accumulation mechanism in 3T3-L1 adipocyte and C57BL/6 mouse models. Methods and Results: In Oil Red O staining, FRF (1,000 ㎍/㎖) treatments showed inhibition rate of 35.39% in lipid accumulation compared to that in the control. AdipoRedTM assay indicated that the triglyceride content in 3T3-L1 adipocytes treated with FRF (1,000 ㎍/㎖) was reduced to 23.22%, and free glycerol content was increased to 106.04% that of the control. FRF and its major constituent, rutin affected mRNA gene expression. Rutin contributed to the inhibition of Sterol regulatory element binding protein-1c (SREBP-1c) gene expression, and inhibited the transcription factors SREBP-1c, peroxisome proliferator-activated receptor gamma (PPAR-γ), CCAAT/enhancer binding protein α (C/EBPα), fatty acid synthase (FAS) and acetyl-CoA carboxylase (ACC). In addition, the effect of FRF administration on obesity development in C57BL/6 mice fed high-fat diet (HFD) was investigated. FRF suppressed weight gain, and reduced liver triglyceride and leptin secretion. FRF exerted potential anti-inflammatory effects by improving insulin resistance and adiponectin levels, and could thus be used to help counteract obesity. The mRNA expressions of PPAR-γ, FAS, ACC, and CPT-1 were determined in liver tissue. Quantitative real-time PCR analysis was also performed to evaluate the expression of IL-1β, IL-6, and TNF-α in epididymal adipose tissue. Compared to the control group, mice fed the HFD showed the up-regulation in PPAR-γ, FAS, IL-6, and TNF-α genes, and down-regulation in CPT1 gene expression. FRF treatement markedly reduced the expression of PPAR-γ, FAS, IL-6, and TNF-α compared to those in HFD control, whereas increased the expression level of CPT1. Conclusions: These results suggest that the FRF and its major active constituent, rutin, can be used as effective anti-obesity agents.
최근 연구에서 활성 산소 및 비만에 대한 유전자 발현이 비만과 노화를 촉진시키는 원인으로 주목 받고 있으며, 이를 통해 노화와 비만의 억제 체계를 규명하려는 다양한 방법들이 연구되고 있다. 본 연구에서는 우리나라 전국 각지의 산야지에 자생하며 농가에서 재배하기도 하는 국화과에 속하는 식물인 참취(Aster scaber Thunb.) 메탄올 추출물의 항산화력과 3T3-L1 전구지방세포의 지방세포형성과정에 미치는 영향을 확인하였다. 참취 메탄올 추출물에 존재하는 총 폴리페놀 및 플라보노이드 함량은 각각 $57.07{\pm}2.17$, $54.62{\pm}2.24{\mu}g/mg$으로 나타났다. 시료의 DPPH, ABTS radical 소거 활성을 측정한 결과 $RC_{50}$값이 각각 $22.24{\pm}0.40$, $53.19{\pm}1.61{\mu}g/mL$로 우수한 항산화 효과를 나타냈다. 참취의 메탄올 추출물이 3T3-L1 전구지방세포에서 분화유도 물질(IBMX, DEXA, Insulin)과 함께 처리했을 때 지방구의 형성을 농도 의존적으로 감소시켰다. 또한 이들은 지방세포의 증식 및 분화되는 과정에서 발현되는 adipogenic transcription factor 및 관련 유전자의 단백질 발현과 mRNA 발현을 유의적으로 감소시키는 것을 확인하였다. 이상의 결과로 참취 메탄올 추출물을 이용하여 항비만에 우수한 효능을 가지는 기능성 식품 소재로서의 개발이 가능할 것으로 생각된다.
