Staphylococcus aureus is a well-recognized human pathogen that causes a wide range of infections as a result of its extensive virulence factors. One of these factors is Panton-Valentine leukocidin (PVL), a potent pore-forming cytotoxin that has been linked to invasive S. aureus infections. PVL is one of the important virulence factors for S. aureus and has been largely recognized as one of the markers for community-acquired methicillin-resistant S. aureus. However, the presence of PVL in methicillin-susceptible S. aureus infections is not widely reported in the literature. Thrombotic sequelae of S. aureus infections associated with PVL expression are uncommon in children. We hereby report two children with thrombotic complications associated with PVL-producing methicillin-susceptible S. aureus. Both patients responded well to antibiotic and anticoagulant therapies, and survived without any long-term sequelae.

This study was carried out the antimicrobial resistance of staphylococcus aureus isolated from bovine mastitic milk, chickens, korean native cattle, korean native goats, pigs, dog and mice in northern area of kyongbuk. The result were summarised as follows ; A total of 149 S aureus were isolated from bovine mastitic milk, chickens, korean native cattle, korean native goats, pigs, dog and mice. In 80 isolates of S aureus from bovine mastitic milk, 60% of isolates revealed resistance to penicillin and ampicillin, 19% to gentamicin, 6% to tetracycline. In 36 isolates of S aureus from chickens, 72% of isolates revealed resistance to tetracycline, 58% to penicillin and ampicillin, 42% to streptomycin, 31% to lincomycin, 25% to norfloxacin, 22% to gentamicin. In 17 isolates of S aureus from korean native cattle, 100% of isolates revealed resistance to penicillin and ampicillin, 88% to lincomycin, 76% to tetracycline. 2 MRSA were isolated from the isolates of S aureus from bovine mastitic milk and revealed multi-drug resistance.

Objective : This experimental study was performed to investigate the effect of herbal eye drops, Sean-tang and Coptidis rhizoma on Staphylococcus aureus, Staphylococcus epidermidis and Pseudomonas aeruginosa keratitis. Methods : After administering herbal eye drops (Sean-tang, Coptidis rhizoma) on Staphylococcus ausreus, Staphylococcus. epidermidis and Pseudomonas aeruginosa, I measured MIC and the size of inhibition zone. MIC was measured by dropping to $50{\mu}{\ell}$ according to density(l00%, 10%, 1%, 0.1%). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Result : On Staphylococcus aureus, Staphylococcus epidermidis, Coptidis rhizoma showed the highest anti-bacterial potency and on Pseudomonas aeruginosa, Sean-tang, Coptidis rhizoma was nor anti-bacterial potency. Conclusions : The present author think that the herbal eye drops, Sean-tang and Coptidis rhizoma can be used to cure Staphylococcus aureus, Staphyloccus epidermidis keratitis and if further study is performed, the use of the herbal eye drops will be valuable and beneficial in the clinical medicines.
Using the monomolecular film technique, we compared the interfacial properties of Staphylococcus simulans lipase (SSL) and Staphylococcus aureus lipase (SAL). These two enzymes act specifically on glycerides without any detectable phospholipase activity when using various phospholipids. Our results show that the maximum rate of racemic dicaprin (rac-dicaprin) hydrolysis was displayed at pH 8.5, or 6.5 with Staphylococcus simulans lipase or Staphylococcus aureus lipase, respectively The two enzymes interact strongly with egg-phosphatidyl choline (egg-PC) monomolecular films, evidenced by a critical surface pressure value of around $23\;mN{\cdot}m^{-1}$. In contrast to pancreatic lipases, $\beta$-lactoglobulin, a tensioactive protein, failed to inhibit Staphylococcus simulans lipase and Staphylococcus aureus lipase. A kinetic study on the surface pressure dependency, stereoselectivity, and regioselectivity of Staphylococcus simulans lipase and Staphylococcus aureus lipase was performed using optically pure stereoisomers of diglycerides (1,2-sn-dicaprin and 2,3-sn-dicaprin) and a prochiral isomer (1,3-sn-dicaprin) that were spread as monomolecular films at the air-water interface. Both staphylococcal lipases acted preferentially on distal carboxylic ester groups of the diglyceride isomer (1,3-sn-dicaprin). Furthermore, Staphylococcus simulans lipase was found to be markedly stereoselective for the sn-3 position of the 2,3-sn-dicaprin isomer.

Background: Staphylococcus aureus and S. pseudintermedius are the major etiological agents of staphylococcal infections in humans, livestock, and companion animals. The misuse of antimicrobial drugs has led to the emergence of antimicrobial-resistant Staphylococcus spp., including methicillin-resistant S. aureus (MRSA) and methicillin-resistant S. pseudintermedius (MRSP). One novel therapeutic approach against MRSA and MRSP is a peptide nucleic acid (PNA) that can bind to the target nucleotide strands and block expression. Previously, two PNAs conjugated with cell-penetrating peptides (P-PNAs), antisense PNA (ASP)-cmk and ASP-deoD, targeting two essential genes in S. aureus, were constructed, and their antibacterial activities were analyzed. Objectives: This study analyzed the combined antibacterial effects of P-PNAs on S. aureus and S. pseudintermedius clinical isolates. Methods: S. aureus ATCC 29740 cells were treated simultaneously with serially diluted ASP-cmk and ASP-deoD, and the minimal inhibitory concentrations (MICs) were measured. The combined P-PNA mixture was then treated with S. aureus and S. pseudintermedius veterinary isolates at the determined MIC, and the antibacterial effect was examined. Results: The combined treatment of two P-PNAs showed higher antibacterial activity than the individual treatments. The MICs of two individual P-PNAs were 20 and 25 µM, whereas that of the combined treatment was 10 µM. The application of a combined treatment to clinical Staphylococcus spp. revealed S. aureus isolates to be resistant to P-PNAs and S. pseudintermedius isolates to be susceptible. Conclusions: These observations highlight the complexity of designing ASPs with high efficacy for potential applications in treating staphylococcal infections in humans and animals.
Staphylococcus aureus (S. aureus) is a major pathogen that causes human pneumonia, leading to significant morbidity and mortality. S. aureus coagulase (Coa) triggers the polymerization of fibrin by activating host prothrombin, which then converts fibrinogen to fibrin and contributes to S. aureus pathogenesis and persistent infection. In our research, we demonstrate that isovitexin, an active traditional Chinese medicine component, can inhibit the coagulase activity of Coa but does not interfere with the growth of S. aureus. Furthermore, we show through thermal shift and fluorescence quenching assays that isovitexin directly binds to Coa. Dynamic simulation and structure-activity relationship analyses suggest that V191 and P268 are key amino acid residues responsible for the binding of isovitexin to Coa. Taken together, these data indicate that isovitexin is a direct Coa inhibitor and a promising candidate for drug development against S. aureus infection.

Staphylococcus aureus is one of the most common microorganisms and causes foodborne diseases. In particular, biofilm-forming S. aureus is more resistant to antimicrobial agents and sanitizing treatments than planktonic cells. Therefore, this study aimed to investigate the anti-biofilm effects of cell-free supernatant (CFS) of Saccharomyces cerevisiae isolated from cucumber jangajji compared to grapefruit seed extract (GSE). CFS and GSE inhibited and degraded S. aureus biofilms. The adhesion ability, auto-aggregation, and exopolysaccharide production of CFS-treated S. aureus, compared to those of the control, were significantly decreased. Moreover, biofilm-related gene expression was altered upon CFS treatment. Scanning electron microscopy images confirmed that CFS exerted anti-biofilm effects against S. aureus. Therefore, these results suggest that S. cerevisiae CFS has anti-biofilm potential against S. aureus strains.

Purpose: Methicillin Resistant Staphylococcus aureus (MRSA) has become a major clinical problem and one of the major nosocomial pathogen worldwide. The aim of the study was to investigate the epidemiological characteristics of genotypes of MRSA isolated in the A-hospital ICU. Methods: In the period between December 2007 and May 2008, MRSA was isolated from ICU patients and its surrounding environment. Polymerase Chain Reaction (PCR) was conducted for the detection of MRSA gene. The incidence of MRSA in the clinical isolates of Staphylococcus aureus was examined by using a multiplex PCR. The spa gene of Staphylococcus aureus encodes protein A and is used for typing of MRSA. We used sequence typing of the spa gene repeat region to study the epidemiology of MRSA at a hospital. Results: Two different genotypes of MRSA were identified with 90 isolated from the patients and its surrounding environments in the ICU. Conclusion: This study may contribute to the development of effective strategies for preventing nosocomial infections. Genotyping may have more general application for the study of MRSA epidemic outbreak in hospital and community infection.
Staphylococci are Gram-positive, facultatively anaerobic cocci, normally found on the skin andmucosal surfaces of most warm-blooded animals and often involved in a wide variety of diseases in animals.Staphylococcal infections are treated with antibiotics and, consequently, antibiotic resistance and/or acquiredresistance have developed. Staphylococcus (S.) intermedius and Staphylococcus (S.) aureus are 2 comonveterinary isolates that are frequently associated with suppurative infections. This study was undertaken toexamine antimicrobial susceptibility of S. aureus (23 isolates) and S. intermedius (160 isolates) isolatedfrom dogs in Gwangju, Korea and investigate whether the antibiotic resistance of S. aureus and S.intermedius is efected by the site of isolation, age, and sex of dogs. More isolates were isolated fromadult dogs (71.3%) than juveniles (20.5%). Antimicrobial resistance was commonly found for Penicillin,Tetracycline, Trimethoprim-Sulphamethoxazole in both Staphylococcus species. All of the isolates weresusceptible to Amoxicillin/Clavulanic Acid, Cephalothin, Oxacilin, Neomycin, and Vancomycin. Appropriateprotocol for antibiotic use and strategies to reduce antimicrobial resistance rate will be needed. Periodicsubstitution of antimicrobial agents and limitation of antibiotic use should also be considered.
During the screening for the antimicrobial agents against multidrug-resistant Staphylococcus aureus, we isolated an active compound produced by strain CNU30122. The active compound was purified from culture broth by HP-20 column chromatography, ethylacetate extraction. silica gel column and Sephadex LH-20 column chromatographies and HPLC. Based on various NMR studies including $^1H-^1H\;COSY$, $^1H-^{13}C\;COSY$ and HMBC experiments. the active compound was identified as fusidic acid.
