The phylogenetic potential of orbicules in angiosperms (소구상체(Orbicules)의 계통분류학적 검토)

  • The distribution of orbicules was investigated for eleven taxa of six genera in Lamiaceae and four taxa of three genera in Verbenaceae using scanning electron microscopy. A literature survey to evaluate the phylogenetic potential of the orbicules and their possible correlations with tapetum types was also conducted. The orbicules are consistently absent in all investigated taxa of Lamiaceae, while small orbicules of an average size of less than $1{\mu}m$ are densely distributed in Verbenaceae. In fact, orbicules appear consistently in 123 of 150 angiosperm families when investigated in at least one species. Thus, the distribution patterns of orbicules could be a useful diagnostic character in angiosperms. In addition, orbicules occur in 84% taxa of the secretory tapetum type, while they are commonly absent in the amoeboid tapetum type (ca. 80%). The presence of orbicules may be correlated with the secretory tapetum type. However, the study of orbicules is restricted in 150 families and the tapetum type within these families can be applied for 92 families out of a total of 416 angiosperm families. Thus, further investigation of orbicules is necessary in extended taxa to address the questions pertaining to orbicules.

Variation of Seed Component Contents in Wild Soybean(Glycine soja Sieb. & Zucc.) (야생콩(Glycine soja Sieb. & Zucc.)의 종실성분 함량변이)

  • Studies were conducted to develop new breeding resources using wild soybean (Glycine soja Sieb. & Zucc.) species. In 132 wild soybeans, the average crude protein content was $45.4\%$, ranged from $37.4\%\;to\;50.2\%$ and average crude oil content was $9.3\%$ showing lower than cultivated soybean. The average total amino-acids content was $19,214\;{\mu}g/g$, containing $252\;{\mu}g/g$ of methionine and $103\;{\mu}g/g$ of cysteine, a sulfur containing amino-acid. In ratio of individual amino-acids, the glutamic acid, methionine and cystein content was $16.1\%,\;1.3\%,\;and\;0.5\%$, respectively. The correlation between total amino-acid and sulfur containing amino-acid was not significant. Total isoflavone concentration was, on average, $1,105\;{\mu}g/g$ in wild soybean germplasm. As a result of comparison with total isoflavone and genestein contents, significant positive correlations were observed between total isoflavone and genistein concentration.

Characterization and Composition of Ammonia-Oxidizing Bacterial Community in Full- Scale Wastewater Treatment Bioreactors (실규모 하수처리 생물반응기에서 발견되는 암모니아산화균 군집조성 및 특징)

  • Ammonia-oxidizing bacteria (AOB) are chemolithoautotrophs that play a key role in nitrogen removal from advanced wastewater treatment processes. Various AOB species inhabit and their community compositions vary over time in the wastewater treatment bioreactors. In this study, a hypothesis that operational and environmental conditions affect both the community compositions and the diversity of AOB in the bioreactors was proposed. To verify the hypothesis, the clone libraries based on ammonia monooxygenase subunit A were constructed using activated sludge samples from aerobic bioreactors at the Pohang, the Palo Alto, the Nine Springs, and the Marshall wastewater treatment plants (WWTPs). In those bioreactors, AOB within the Nitrosomonas europaea, N. oligotropha, N.-like, and Nitrosospira lineages were commonly found, while AOB within the N. communis, N. marina, and N. cryotolerans lineages were rarely detected in the samples. The AOB community structures were different in the bioreactors: AOB within the N. oligotropha lineage were the major microorganisms in the Pohang, the Palo Alto, and the Marshall WWTPs, while AOB within the N. europaea lineage were dominant in the Nine Springs WWTP. The correlations between the AOB community compositions of the wastewater treatment bioreactors and their operational (HRT, SRT, and MLSS) and environmental conditions (temperature, pH, COD, $NH_3$, and $NO_3{^-}$) were evaluated using a multivariate statistical analysis called the Redundancy Analysis (RDA). As a result, COD and $NO_3{^-}$ concentrations in the bioreactors were the statistically significant variables influencing the AOB community structures in the wastewater treatment bioreactors.

Seasonal Variations in Soil Solution Chemistry under Larix leptolepis, Pinus Koraiensis, and Quercus mongolica stands (일본잎갈나무, 잣나무, 신갈나무 임분 토양수 성질의 계절적 변이)

  • To investigate seasonal variations of soil solution chemistry, samples were collected from zero tension lysimeters in O, A, and B horizons of 26-year-old Larix leptolepis. Pinus koraiensis and Quercus mongolica stands in Kwangju, Kyunggi Province from September, 1996 through June. 1999. Potassium, $Mg^{2+}$, $Cl^-$, and $No_3\;^-$ concentrations increased in October and November concurrent with inputs of fresh litterfall and twigs. Sodium, $Ca^{2+}$, $K^+$, and $Mg^{2+}$ concentrations increased in March and April concurrent with the yellow sand effect. Potassium concentration showed the highest variation among ions, and $Cl^-$ concentration showed positive correlations with $K^+$ and $Mg^{2+}$ concentrations. Soil solution pH decreased while $Al^{3+}$ and $K^+$ concentrations increased during the study period. If our data reflects long term trends. then $Al^{3+}$ concentration in the O horizon will reach the toxic level (0.180 meq/l) within 10-20 years depending on species.

A Meta-analysis on the Effect of Forest Thinning on Diameter Growth and Carbon Stocks in Korea (국내 산림의 간벌에 따른 직경 생장량 및 탄소 저장량 변화에 관한 메타 분석)

  • With results from previous Korean studies on forest thinning, we conducted a meta-analysis on the effect of thinning on diameter at breast height (DBH) growth and carbon (C) stocks (tree, litter layer, coarse woody debris (CWD), and soils) in Korean forests. Thinning increased the DBH growth and the C stocks in soils by 39.2% and 12.8%, respectively, while it decreased the C stocks in tree by 30.9%. In contrast, thinning had no significant effect on the C stocks in litter layer and CWD. The DBH growth and the C stocks in tree showed significant correlations with thinning intensity and recovery time. The C stocks in litter layer correlated with recovery time while those in CWD and soils did not show significant correlation neither with thinning intensity nor with recovery time. Regression models of the DBH growth and the C stocks in tree were developed to quantify the effect of thinning intensity and recovery time. An integration of the regression model of the tree C stock into forest carbon models is expected to be essential to quantify the effect of thinning on the C stocks in litter layer, CWD, and soils. We also suggested expansion of study species, long-term and frequent monitoring, and investigation on understory vegetation in order to elucidate changes in Korean forests following thinning practices.

Correlation Analysis Between Terpene and Meteorological Factors at Artificial Coniferous Forest of Mt. Moodeung (무등산 인공 침엽수림에서 테르펜과 기상인자의 상관성분석)

  • There are many factors to influence the amount of terpene at the forest. However, it is poorly understood whether the amount is altered by meteorological factors. In order to study correlation analysis between terpene amount and meteorological(i.e., temperature, humidity, wind speed, solar radiation) and environmental factors(i.e., oxygen, carbonb dioxide) at the artificial coniferous forest of Chamaecyparis obtusa (site 1) and Cryptomeria japonica (site 2) nearby the $1^{st}$ reservoir at Mt. Moodeung, the research was executed during April to November in 2011. Forest density at site 1 was 1,692 trees/ha, being occupied with 87.2% of Chamaecyparis obtusa, higher than 925 trees/ha at site 2. Carbon dioxide at site 1 was in the range of 385~410 ppm in June, similar to at Anmyundo(395 ppm in 2010). Solar radiation has positive correlation with ambient temperature and inversely negative with relative humidity. Main species of terpene released were ${\alpha}$-pinene, camphene, ${\beta}$-pinene, cymene, ${\delta}$-limonene and camphor at two sites and terpene was more effluent at spring and summer than at fall. The large amount of terpene was emitted in the afternoon than in the morning under the influence of the wind speed and the topographical property. The terpene amount has positive correlation with relative humidity and oxygen, and negative with wind speed and soil temperature. Because correlations of ${\alpha}$-pinene and other terpene materials showed statistically significant within p=0.01. ${\alpha}$-pinene could be suggested as the basic material in explaining the amount of other terpene materials.

Evaluation of Heavy Metal Contents in the Floras Derived from Granite and Coal Bearing Shale Areas in Keumsan (금산의 화강암 및 함탄질 셰일 지역 토양내 식물체의 중금속 함량 특성)

  • Three different floras(M. sinsinsis, A. vulgaris, Robinia pseudo-acacia) were collected from the granite(GR) and coal bearing shale area(CB) and analysed for their heavy metal elements with the representative soils. Regardless of the flora species, the CB were high in average contents. Among the correlation relationships, the CB were more distinctive than the GR, and the A. vulgaris showed higher correlations than the M. sinsinsis. In the same soils, the A. vutgaris showed high contents than the M. sinsinsis and Robinia pseudo-acacia, and the M. sinsinsis were high relative to the Robinia pseudo-acacia. In the comparisons of the flora, root parts were high in most of the elements except for Zn. In the soils, the CB were high in most of elements while As and Mo showed different contents between the GR and CB. In the comparison between soil and flora, soils of the GR were high in the V and Sc contents and low in Zn and Cu, while those of the CR were high in the Cr, V and Sc contents, and low in the Zn contents, Comparing with the soil contents, the M, sinsinsis in the GR were similar to Co and V contents while, in the CB, the M. sinsinsis were similar to the Ni, Cr, Co, Zn, Mo contents, and the Robinia pseudo-acacia were similar to the Ni, Zn, Cu contents. Overall results suggested that the M. sinsinsis and A. vulgaris should be eligible for the bioremediation of the soils polluted by heavy metal such as the CB.

Environment and Vegetation Characteristics of Aconitum austrokoreense Koidz. habitats (세뿔투구꽃의 자생지 식생과 환경특성)

  • The habitat characteristics of Aconitum austrokoreense Koidz. were investigated to provide basic data for conservation and restoration. The altitude ranged from 260m to 728m with inclinations of $4-39^{\circ}$. As a result of vegetation survey within natural populations, a total of 147 taxa were identified inside 20 quadrates in 7 natural habitats. The importance value of Aconitum austrokoreense was 23.0% based on the coverage and frequency of the herbaceous layer and sat is more dominant. Species diversity was 1.23, and dominace and evenness were found to be 0.22 and 0.77, respectively. The soil type were silty loam (four sites), sandy loam (two sites) and loam (one site). The average field organic matter was 7.83% and the total nitrogen and pH were 0.35, and 5.69. Correlation analysis between environmental factors and population characteristic showed the correlations between chlorophyll content and slope, flowering rate and soil humidity. Conservation on habitats conditions is important for stable maintenance the flora and the surrounding area including habitats has to be designated a protection area for habitats conservation.

An Evaluation of Plant Growth Promoting Activities and Salt Tolerance of Rhizobacteria Isolated from Plants Native to Coastal Sand Dunes (해안사구의 토착식물로부터 분리된 근권세균의 내염능과 식물성장촉진능 평가)

  • Coastal sand dunes are important for ecosystems due to the variety of rare species that can be found in this kind of habitat, and the beautiful landscapes they create. For environmental remediation, a potential strategy is phytoremediation using the symbiotic relationship of plants and microbes in the rhizosphere, which has proven ecologically sound, safe, and cost effective. Ninety-five colonies were isolated from the rhizosphere soil (RS) or rhizoplane (RP) of Rorippa islandica, Rumex crispus, Artemisia princeps var. orientalis, Lilium sp Stellaria media, and Gramineae. These colonies were then tested for plant growth promoting activities (PGPAs) such as 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity, and siderphores synthesis ability. In addition, salt tolerance was evaluated at 4% and 8% salt concentrations. It was observed that amongst the test subjects about 50% of the strains had a high resistance to salinity. Many of them could produce indole-3-acetic acid (IAA) IAA (in RS 13.9% and in RP 7.6%), exhibited ACC deaminase activity (55.8% in RS and 36.6% in RP), and could synthesize siderphores (62.7% in RS and 50% in RP). Correlation coefficient analyses were carried out for the three kinds of plant growth promoting abilities (PGPA) and salt tolerance. A positive correlation was found between an ability to synthesize siderphores and ACC deaminase activity (r=0.605, p<0.037). Similarly, positive correlations were noted between salt tolerance and ACC deaminase activity (r=0.762, p<0.004, r=0.771), and salt tolerance and an ability to synthesize siderphores (r=0.771, p<0.003).

Metabolomic analysis of perilla seeds harvested from Korea and China (국내산 및 중국산 들깨 종자의 대사체 분석)

  • A comprehensive comparison of fatty acid and metabolite profiles in 8 species of perilla seeds harvested from Korea and China was carried out to understand the correlation between cultivation conditions and the quality of perilla seeds. Metabolomic studies revealed that the perilla seeds were distinct from each other, based on the partial least squares -discriminant analysis (PLS-DA) score plots; 23 compounds including metabolites, fatty acids, and phytosterols that predominantly contributed to this interspecies difference were identified. Changes in the growth environment affected these compounds. In particular, phenolic compounds were affected by rainfall and temperature. Moreover, temperature and altitude showed negative and positive correlations, respectively, to oleic acid. Although more studies on the effect of various environmental factors on cultivation of perilla plants as well as their seeds are needed, the present results provide information that would be useful in identifying perilla seeds produced in specific regions and in the cultivation of good quality perilla.
