To investigate impediments to plasmid transformation in Brevibacterium flavum BF4 and B. lactofermentum BL1, cell surface barriers were determined by measuring growth inhibition whilst enzymatic barriers were determined by comparing DNA methylation properties. B. lactofermentum was more sensitive to growth inhibition by glycine than B. flavum. Release of cellular proteins during sonication was more rapid for B. lactofermentum than for B. flavum. Plasmid DNA (pCSL 17) isolated from B. flavum transformed recipient $McrBC^+$ strains of Escherichia coli with lower efficiency than $McrBC^-$. McrBC digestion of this DNA confirmed that B. flavum contain methylated cytidines in the target sequence of McrBc sequences but B. lactofermentum contained a different methylation pattern. DNA derived from the B. lactofermentum transformed recipient $EcoKR^+$ strains of E. coli with lower efficiency than $EcoKR^-$, indicating the presence of methylated adenosines in the target sequence of EcoK sequences. The present data describe the differences in the physical and enzymatic barriers between two species of corynebacteria and also provide some insight into the successful foreign gene expression in corynebacteria.

Pigs are considered an ideal source of human disease model due to their physiological similarities to humans. However, the low efficiency of in vitro embryo production (IVP) is still a major barrier in the production of pig offspring with gene manipulation. Despite ongoing advances in the associated technologies, the developmental capacity of IVP pig embryos is still lower than that of their in vivo counterparts, as well as IVP embryos of other species (e.g., cattle and mice). The efficiency of IVP can be influenced by many factors that affect various critical steps in the process. The previous relevant reviews have focused on the in vitro maturation system, in vitro culture conditions, in vitro fertilization medium, issues with polyspermy, the utilized technologies, etc. In this review, we concentrate on factors that have not been fully detailed in prior reviews, such as the oocyte morphology, oocyte recovery methods, denuding procedures, first polar body morphology and embryo quality.
Objective: Specific genomic sites can be recognized and permanently modified by genome editing. The discovery of endonucleases has advanced genome editing in pigs, attenuating xenograft rejection and cross-species disease transmission. However, off-target mutagenesis caused by these nucleases is a major barrier to putative clinical applications. Furthermore, off-target mutagenesis by genome editing has not yet been addressed in pigs. Methods: Here, we generated genetically inheritable α-1,3-galactosyltransferase (GGTA1) knockout Yucatan miniature pigs by combining transcription activator-like effector nuclease (TALEN) and nuclear transfer. For precise estimation of genomic mutations induced by TALEN in GGTA1 knockout pigs, we obtained the whole-genome sequence of the donor cells for use as an internal control genome. Results: In-depth whole-genome sequencing analysis demonstrated that TALEN-mediated GGTA1 knockout pigs had a comparable mutation rate to homologous recombination-treated pigs and wild-type strain controls. RNA sequencing analysis associated with genomic mutations revealed that TALEN-induced off-target mutations had no discernable effect on RNA transcript abundance. Conclusion: Therefore, TALEN appears to be a precise and safe tool for generating genomeedited pigs, and the TALEN-mediated GGTA1 knockout Yucatan miniature pigs produced in this study can serve as a safe and effective organ and tissue resource for clinical applications.
H3N8 equine influenza virus (EIV) causes respiratory diseases in the horse population, and it has been demonstrated that EIV can transmit into dogs owing to its availability on receptors of canine respiratory epithelial cells. Recently, we isolated H3N8 EIV from an EIV-vaccinated horse that showed symptoms of respiratory disease, and which has a partially truncated nonstructural gene (NS). However, it is not clear that the NS-truncated EIV has an ability to cross the host species barrier from horses to dogs as well. Here, we experimentally infected the NS-truncated H3N8 EIV into dogs, and monitored their clinical signs and viral load in respiratory organs to determine the virus's transmissibility.

Prions cause neurodegenerative diseases in animals and humans. The scrapie prion protein (PrPSc) is the major-possibly only-component of the infectious prion and is generated from the cellular isoform (PrPC) by a conformational change. Limited proteolysis of PrPSc produces an polypeptide comprised primarily of residues 90 to 231, which retains infectivity. The three-dimensional structure of rPrP(90-231), a recombinant protein resembling PrPC with the Syrian hamster (SHa) sequence, was solved using multidimensional NMR. Low-resolution structures of rPrP(90-231), synthetic peptides up to 56 residues, a longer (29-231, full-length) protein with SHa sequence, and a short here further structure refinement of rPrP(90-231) and dynamic features of the protein. Consideration of these features in the context of published data suggests regions of conformational heterogeneity, structural elements involved in the PrPC\longrightarrowPrPSc transformation, and possible structural features related to a species barrier to transmission of prion diseases.

In this research, we further developed the one-step seed mediated method to synthesize gold nanoparticles (GNPs) and control their resulting shapes to obtain hexagonal, triangular, rod-shaped, and spherical gold nanostructures. Our method reveals that the reaction kinetics of formation of GNPs with different shapes can be controlled by the rate of addition of ascorbic acid, because this is the critical factor that dictates the energy barrier that needs to be overcome. This in turn affects the growth mechanism process, which involves the adsorption of growth species to gold nanoseeds. There were also observable trends in the dimensions of the GNPs according to different rates of addition of ascorbic acid. We performed further analyses to investigate and confirm the characteristics of the synthesized GNPs.
Dielectric barrier discharge plasma is a new technique in water pollutant degradation, which that is characterized by the production of chemically active species such as hydroxyl radicals, ozone, hydrogen peroxide, etc. If dissolving of plasma gas generated in the plasma reaction has increased, it is possible to increase the contaminant removal capacity. In this study, the improvement on the dissolving performance of plasma gas was evaluated by the indirect method measuring the overall oxygen transfer coefficient. Experiments were conducted to examine the effects of nozzle type, distance from water surface, air supply rate and liquid circulation rate. The experimental results showed that the $K_{La}$ value of the 3-prong nozzle is 2.67 times higher than the diffuser. The order of $K_{La}$ value with nozzle type ranked in the following order: 3-prong nozzle (inner diameter, less 1 mm) > circular nozzle (inner diameter, 1.5 mm) > ellipse nozzle (short diameter 1 mm, long diameter 2.5 mm) > circular nozzle (inner diameter, 3 mm). Optimal liquid circulation rate was appeared to be 1.7 L/min, the value of $K_{La}$ was 0.510 1/min. The value of $K_{La}$ with increasing air supply rate was revealed in the form of an exponential such as $K_{La}=0.3581e^{0.2919^*air\;flow\;rate}$.
Microbial digestion of feed in the rumen involves a sequential attack culminating in the formation of fermentation products and microbial cells that can be utilized by the host animal. Most feeds are protected by a cuticular layer which is in effect a microbial barrier that must be penetrated or circumvented for digestion to proceed. Microorganisms gain access to digestible inner plant tissues through damage to the cuticle, or via natural cell openings (e.g., stomata) and commence digestion from within the feed particles. Primary colonizing bacteria adhere to specific substrates, divide to form sister cells and the resultant microcolonies release soluble substrates which attract additional microorganisms to the digestion site. These newly attracted microorganisms associate with primary colonizers to form complex multi-species consortia. Within the consortia, microorganisms combine their metabolic activities to produce the diversity of enzymes required to digest complex substrates (e.g., cellulose, starch, protein) which comprise plant tissues. Feed characteristics that inhibit the microbial processes of penetration, colonization and consortia formation can have a profound effect on the rate and extent of feed digestion in the rumen. Strategies such as feed processing or plant breeding which are aimed at manipulating feed digestion must be based on an understanding of these basic microbial processes and their concerted roles in feed digestion in the rumen.

Organic light emitting diode (OLED) is considered as the next generation flat panel displays due to its advantages of low power consumption, fast response time, broad viewing angle and flexibility. For the flexible application, it is essential to develop thin film encapsulation (TFE) to protect oxidation of organic materials from oxidative species such as oxygen and water vapor [1]. In many TFE research, the inorganic film by atomic layer deposition (ALD) process demonstrated a good barrier property. However, extremely low throughput of ALD process is considered as a major weakness for industrial application. Recently, there has been developed a high throughput ALD, called 'spatial ALD' [2]. In spatial ALD, the precursors and reactant gases are supplied continuously in same chamber, but they are separated physically using a purge gas streams to prevent mixing of the precursors and reactant gases. In this study, the $Al_2O_3$ thin film was deposited by spatial ALD process. We characterized various process variables in the spatial ALD such as temperature, scanning speed, and chemical compositions. Water vapor transmission rate (WVTR) was determined by calcium resistance test and less than $10-^3g/m^2{\cdot}day$ was achieved. The samples were analyzed by x-ray photoelectron spectroscopy (XPS) and field emission scanning electron microscope (FE-SEM).
Non-thermal plasma has attracted medical researchers, since they showed higher apoptosis rate in cancer cells than normal cells. However, it is hard to conclude general cancer cell specific effect because comparison between normal and cancer cell activities after plasma treatment have not been reported yet. This research proposes a comparison of Dielectric Barrier Discharge (DBD) plasma effect on three normal cells lines and three cancer cells lines. We measured cell number, mitochondria activity (MTS assay) and amount of hydrogen peroxide (H2O2) for three days. The results show that the number of cancer cells decreased more than normal cells following of exposure time. On the other hand, mitochondria activities and amounts of H2O2 increased following of exposure time. In addition, we found that DBD plasma exposure on cell suspension in media and media only illustrated no difference in mitochondria activity, H2O2 quantity, and cell number. Thus, we can confirm higher apoptosis rate in cancer cells which is related to the reactive oxygen species (ROS) generated by DBD plasma. The related molecular mechanisms were investigated further.
