Soybean (Glycine max (L.) Merr.) seed is the main source of protein and oil for human and animal. The use of soybean protein has been expanded in the food industry due to their excellent nutritional benefits. But, antinutritional and allergenic factors are exist in the raw mature soybean. Lipoxygenase, Kunitz trypsin inhibitor (KTI) protein, and ${\alpha}^{\prime}-subunit$ of 7S globulin are main antinutritional factors in soybean seed. Breeding of a new soybean strain with lacking these components is needed. The objective of this research was to select new soybean line with lipoxygenase-free, KTI-free, and ${\alpha}^{\prime}-subunit$ free (lx1lx1lx2lx2lx3lx3titicgy1cgy1 genotype). Total 434 $F_2$seeds were obtained from the cross of cultivar, "Gaechuck#2" and PI506876. Presence and absence of lipoxygenase, KTI protein, and ${\alpha}^{\prime}-subunit$ of 7S globulin was tested by SDS electrophoresis using a partial seed of each $F_2$seed. Only one $F_2$seed with lacking these three components was selected and was planted to $F_2$plant. Absence of lipoxygenase, KTI, and ${\alpha}^{\prime}-subunit$ protein was confirmed on the $F_3$seeds harvested. Selected line with lx1lx1lx2lx2lx3lx3titicgy1cgy1 genotype might be useful for soybean breeding.

Soybean proteins are widely used for human and animal feeds worldwide. The use of soybean protein has been expanded in the food industry due to their excellent nutritional benefits. But, antinutritional and allergenic factors are present in the raw mature soybean. P34 protein, referred as Gly m Bd 30K, has been identified as a predominant immunodominant allergen. The objective of this research is to identify the genetic mode of P34 protein for the improvement of soybean cultivar with a very low level of P34 protein. Two $F_2$ populations were developed from the cross of "Pungsannamulkong" ${\times}$ PI567476 and "Gaechuck2ho" ${\times}$ PI567476 (very low level of P34 protein). Relative amount of P34 protein was observed by Western blot analysis. The observed data for the progeny of "Pungsannamulkong" and PI567476 were 133 seeds with normal content of P34 protein and 35 seeds with very low level of P34 protein (${\chi}^2=1.157$, P=0.20-0.30). For the progeny of "Gaechuck#1" and PI567476, the observed data were 177 seeds with normal content of P34 protein and 73 seeds with very low level of P34 protein (${\chi}^2=2.353$, P=0.10-0.20). From pooled data, observed data were 310 seeds with normal content of P34 protein and 108 seeds with very low level of P34 protein (${\chi}^2=0.156$, P=0.50-0.70). The segregation ratio (3:1) and the Chi-square value obtained from the two populations suggested that P34 protein in mature soybean seed is controlled by a single major gene. Single gene inheritance of P34 protein was confirmed in 32 $F_2$ derived lines in $F_3$ seeds, which were germinated from the low level of P34 protein obtained from the cross of "Pungsannamulkong" and PI567476. These results may provide valuable information to breed for new soybean line with low level of P34 protein and identification of molecular markers linked to P34 locus.
Soybean [Glycine max (L.) Merr.] seed is an important dietary source of protein, oil, carbohydrates, isoflavones, and other nutrients for humans and animals. But, antinutritional factors in the raw mature soybean are exist. Kunitz trypsin inhibitor (KTI) protein and stachyose are main antinutritional factors in soybean seed. The genetic removal of the antinutritional factors will improve the nutritional value of soybean seed. The objective of this research was to breed a new yellow soybean strains (rs2rs2titi genotype) with the traits of lacking of KTI protein and low content of stachyose. Breeding population was developed from the cross of "Jinyangkong" and 15G1 parents. Presence or absence of KTI protein was detected based on Western Blot technique. Content of stachyose in mature seed was detected by HPLC. Total four new strains (603-1, 603-2, 625, and 694) with KTI protein free and low content of stachyose were selected. Four strains (603-1, 603-2, 625, and 694) have yellow seed coat and hilum. Plant height of 603-1 strain was 65 cm and 100-seed weight was 29.2 g. Plant height of 603-2 strain was 66 cm and 100-seed weight was 26.2 g. Plant height of 625 strain was 64 cm and 100-seed weight was 27.1 g. Content of stachyose for four new strains was 3.0~3.50 g/kg. Four strains selected in this research will be used to improve new yellow soybean cultivar with KTI protein free, and low content of stachyose.

Double cropping system including paddy field soybean is widely adopted nationwide, due to rise in market price and its higher income than paddy field rice. Sowing date of soybean as a second crop is being delayed depending on first crop's growth period and harvesting time. Due to the increased temperature in October and delayed first frost date, soybean could be harvested without frost damage even in late-plating. Therefore, selection of soybean cultivar which is appropriate for this environment is very important. The effect of sowing date and genotype of soybean on growth and yield was investigated for three planting dates (June 20, July 5, and July 20) with ten cultivars developed for soy-pate production, to figure out plant development and yield pattern in delayed planting. As planting date is delayed, plant height and pod number was decreased and this pattern was more clearly detected in mid-late maturity cultivars. Hundred-seed weight did not show significant changes even in late planting, due to compensations between yield components. Yield reduction of July 20 in contrast to that of June 20 showed that Nampung (9.6%) showed the least yield decline. Maximum yield was achieved from Daepung, Taekwang, and Uram among other soybean cultivars in late planting. Shortening of growth period was strongly detected in reproductive stage while length of vegetative stage was regularly maintained in both early and mid-late maturity cultivars.
The objectives of this study were to investigate the physicochemical characteristics of soybean leaves in different cultivars and to develop a soybean curd prepared with soybean leave powder as a functional food. Four cultivars (Daewonkong leaf, Daepungkong leaf, Hwangguemkong leaf, and Seoritae leaf) were selected for this experiment. A significant difference was observed in the proximate composition of soybeans leaves (p<0.05). Soybean leaves had the highest content of carbohydrates. The mineral composition and isoflavone content in soybean leaves were significantly different among the cultivars (p<0.05). In particular, Daepongkong leaf had the highest content of genistein, daidzein, and total isoflavones. This study was also conducted to determine the quality characteristics of a soybean curd developed from daepongkong leaf with various concentrations (0, 0.1, 0.2, and 0.3%) of soybean leaf powder (SLP). The soybean curd yield rate increased according to the level of SLP added, whereas the L and a color values decreased. In the sensory evaluation, intensity scores for color, after taste, leafy taste, and chewiness were highest for the 0.3% soybean curd. The soybean curd with 0.2% SLP soybean milk attained the highest overall acceptability score. These results showed that soybean leaf was preferred over soybean curd containing 0.2% soybean leaf powder.

Soybean seeds contain high amounts of isoflavones that display biological effects and isoflavone content of soybean seed can vary by year, environment, and genotype. Objective of this study was to identify quantitative trait loci that underlie isoflavone content in soybean seeds. The study involved 85 $F_2$ populations derived from Korean soybean cultivar 'Kwangkyo' and wild type soybean 'IT182305' for QTL analysis associated with isoflavone content. Isoflavone content of seeds was determined by HPLC. The genetic map of 33 linkage groups with 207 markers was constructed. The linkage map spanned 2,607.5 cM across all 33 linkage groups. The average linkage distance between pair of markers among all linkage groups was 12.6 cM in Kosambi map units. Isoflavone content in $F_2$ generations varied in a fashion that suggested a continuous, polygenic inheritance. Eleven markers (4 RAPD, 3 SSR, 4 AFLP) were significantly associated with isoflavone content. Only two markers, Satt419 and CTCGAG3 had F-tests that were significant at P<0.01 in $F_2$ generation for isoflavone content. Interval mapping using the $F_2$ data revealed only two putative QTLs for isoflavone content. The peak QTL region on linkage group 3, which was near OPAG03c, explained $14\%$ variation for isoflavone content. The peak QTL region on linkage group 5, which was located near OPN14 accounted for $35.3\%$ variation for isoflavone content. Using both Map-Maker-QTL $(LOD{\geq}2.0)$ and single-factor analysis $(P{\leq}0.05)$, one marker, CTCGAG3 in linkage group 3 was associated with QTLs for isoflavone content. Efficient in vitro regeneration system is essential for the successful crop breeding of soybean (Glycine max (L.) Merr.) using the new biotechnology. The genotype of donor plants strongly influences the establishment of tissue culture system. Therefore, the screening of genotypes with excellent tissue culture ability is very important for soybean genetic improvement. In this study, we report the tissue culture efficiency of 21 soybean cultivars belong to Korean soybean core-collection and two foreign cultivars (Jack and Maverick). The Kwangan, Anpyeong and Seonam are share close genetic relationship in 21 cultivars and these three cultivars were observed the high frequency of germination and regeneration. Furthermore, the high tissue culture abilities were also observed in the Williams 82 used in reference genome sequencing and the two foreign cultivars. The transformation of pBAtc:tRNA with bar gene was performed by Agrobacterium tumefaciens in the cultivars with high tissue culture ability. Transformation of the bar gene was identified by PCR analysis in Kwangan, Pungwon, Seonam, and Maverick. Our results provide useful information for the breeding of various soybean cultivars by plant biotechnology such as, genome editing.
This study was conducted to investigate the yield loss of soybean by initial density of Heterodera glycines in soil. A soybean cultivar named 'Cheongja5ho' was cultivated as test material in Dangjin city, Chungcheongnam-do, Korea. Each seed was planted with distance of 25 cm in 35 plots by initial density of egg of the cyst nematode on June 21st, and allowed to harvest on November 12st in 2019. As a results, the yield of soybean decreased sharply when as the initial density of eggs of cyst nematode increased. The initial density of eggs in soil affected largely on the decreased yields of the soybean by more than 20% in above 2,000 eggs, and 30% in above 6,000 eggs of the cyst. Beyond the 6,000 eggs, the yield loss was more slowly decreased compared with the above cases. Eggs of H. glycines are widely distributed in Korea, resulting in the serious soybean yield loss by H. glycines.
Soybean (Glycine max (L.) Merr.) is a high-protein oilseed crop, cultivated worldwide. Soybean seeds are abundant in various secondary metabolites with physiologically active. Mature seeds of 25 soybean cultivars with various morphological and physiological characteristics analyzed for secondary metabolites, such as carotenoid, isoflavone, and soyasaponin. These secondary metabolites showed various content by genotype in 25 cultivars. Total carotenoid content ranged from 1.23 to 33.78 mg/g and three cultivars, such as IT177645, PI90763 and IT234975 with black seed coat showed high levels of total carotenoid. Total isoflavones content ranged from 20.28 to 276.35 mg/100g and were detected high levels in Savoy, PI90763 and KLG16001. In addition, total soyasaponins content ranged from 33.12 to 246 mg/100g and were detected high levels in PI90763, PI86490 and IT234975. The PI90763 was showed abundant content in all of the carotenoid, isoflavones and soyasaponins. These results could be valuable information for the development of new soybean cultivars and regulation of secondary metabolites biosynthesis in soybean.

Changes in air temperature, CO2 concentration and precipitation due to climate change are expected to have a significant impact on soybean productivity. This study was conducted to evaluate the climate change impact on growth and development of determinate soybean cultivar in the southern parts of Korea. The high temperature during vegetative period, which does not accompany the increase of CO2 concentration, increased the canopy photosynthetic rate in soybean, but after flowering, the high temperature above the optimal ranges interrupts the photosynthetic metabolism. In yield and yield components, high temperature reduced both the pod and seed number and single seed weight, resulting in a reduction of total seed yield. On the other hand, the increase in CO2 concentration dramatically increased the canopy photosynthetic rate over the whole growth period. In addition, high CO2 concentration increased the number of pods and seeds, which had a positive effect on total seed yield. Under concurrent elevation of air temperature and CO2 concentration, canopy photosynthesis increased significantly, but enhanced canopy photosynthesis did not lead to an increase in soybean seed yield. The increase in biomass and branch by enhanced canopy photosynthesis seems to be attributed to an increase in the total number of pods and seeds per plant, which compensates for the negative effects of high temperature on pod development. However, Single seed weight tended to decrease rapidly by high temperature, regardless of CO2 concentration level. Elevated CO2 concentration did not compensate for the poor distribution of assimilations from source to sink caused by high temperature. These results show that the damage of future soybean yield and quality is closely related to high temperature stress during seed filling period.
