The antioxidant activities of two crude extracts ($CH_2Cl_2$ and MeOH) and their solvent fractions (n-hexane, 85% aq. MeOH, n-BuOH, and $H_2O$ fractions) from Corydalis heterocarpa were determined by evaluating authentic $ONOO^-$ and $ONOO^-$ generated from SIN-1 (3-morpholinsydnonimine) in vitro as well as by measuring the degree of occurrence of intracellular reactive oxygen species (ROS) and nitric oxide (NO). Scavenging activities of solvent fractions on authentic $ONOO^-$ increased in the order of n-BuOH > 85% aq. MeOH > $H_2O$ > n-hexane fractions, while those on $ONOO^-$ generated from SIN-1 increased in the order of n-BuOH > 85% aq. MeOH > $H_2O$ > n-hexane fractions. In addition, all solvent fractions effectively inhibited the intracellular ROS and NO levels. The n-BuOH fraction especially exhibited the strongest ROS scavenging effect. Further purification of n-BuOH fraction led to the isolation of cnidimoside A, which presented the potent ROS scavenging effect at $10\;{\mu}M$. From these results, extracts of C. heterocarpa and its component, cnidimoside A, were predicted to be potentially useful as ingredients for protecting against oxidation.
Samples (10 mg/mL) of wild Grifola frondosa aqueous extract and solvent fractions were examined for fibrinolytic, thrombin inhibitory, acetylcholinesterase inhibitory, and antioxidative activities to determine the biological activities. The fibrinolytic activity of the aqueous extract and solvent fractions was 0.93 and 0.73 plasmin units/mL, respectively. The thrombin inhibitory activity of the butanol extract was 79.60%. The chloroform fraction had high acetylcholinesterase inhibitory activity (85.88%). The aqueous extract had low antioxidative activity (39.81%). The aqueous fraction hydrolyzed Bβ subunits of human fibrinogen but did not show any reactivity for the γ form of the human fibrinogen. The findings indicate the potential of wild Grifola frondosa for the development of drugs and bio-functional foods to prevent cardiovascular diseases.
Phlorotannins which is oligomeric polyphenol of phloroglucinol unit were isolated from solvent fractions of methanolic extract of the brown alga, Ecklonia cava (EC). The inhibitory effects of phlorotannins from EC solvent fractions on oxidative stress were examined in human dermal fibroblasts (HDFs) related to wrinkle formation. Among the solvent fractions, phlorotannins from ethyl acetate fraction exerted the highest scavenging effect on DPPH radical, hydroxyl radical and alkyl radical analyzed by electron spin resonance (ESR) spectroscopy. The levels of intracellular reactive oxygen species (ROS) and lipid peroxidation were measured using 2',7'-dichlorofluorescin diacetate (DCFH-DA) and diphenyl-1-pyrenylphosphine (DPPP), respectively. Their levels were significantly decreased in the presence of phlorotannins from ethyl acetate fraction, compared with other fractions obtained from EC extract (P < 0.01). Furthermore, intracellular glutathione (GSH) level was significantly increased in a time dependent manner by the phlorotannins. Therefore, these results suggest that phlorotannins from EC extract could have a therapeutic potential for prevention and treatment of several diseases such as wrinkle formation related to oxidative stress.
Our study investigated the fibrinolytic, thrombin inhibitory, anti-oxidative and anti-inflammatory activities of the water extract and solvent fractions isolated from Pleurotus ferulea. Fibrinolytic activity was investigated using the fibrin plate method. Thrombin inhibitory activity was used to analyze thrombin inhibitor assay. The DPPH assay was used to estimate anti-oxidative activity. Inhibition of NO production was measured for anti-inflammatory activity in LPS-activated murine RAW 264.7 macrophage cells. An MTS assay was used to evaluate the effects of the water extract and solvent fractions isolated from Pleurotus ferulea on cell viability. Our results showed the fibrinolytic activity to be strong in the ethyl acetate fraction at 1.33 plasmin units. The ethyl acetate fraction also showed high thrombin inhibitory activity at 94.45%. The anti-oxidative activity of the water extract was 37.01% and the anti-inflammatory activity of the chloroform fraction was 98.13%. These findings suggest that Pleurotus ferulea's extract and fractions could be applicable in the development of functional foods for the treatment and prevention of cardiovascular diseases.
This study was carried out to examine the antioxidative, antimicrobial and nitrite scavenging effects of various solvent extracts and different solvent fractions from Schizandra chinensis RUPRECHT(Omija) seed. The results were as follows; The antioxidative activities using lard, soybean oil and linoleic acid were the highest in methanol, ethanol and methanol extract from omija seed, respectively. The free radical scavenging activity using DPPH method was the strongest in acetate fraction of methanol extract from seed. The methanol extract from omija seed had the strongest antimicrobial activities to L. plantarum, B. subtilis, E. coli and P. citrinum, while ethyl acetate extract had the strongest against S. aureus and S. typhimurium. The buthanol fraction from methanol extract had the highest antimicrobial activities, followed by B. subtilis, L. plantarum, E. coli, S. aureus and S. typhimurium. The nitrite scavenging ability was pH dependent, highest at pH 1.2 and lowest at pH 6.0. The buthanol fraction of methanol extract from omija seed had more effective nitrite scavenging ability than other fractions of extracts.
Objectives : The purpose of this study is to isolate tyrosinase inhibitory material from Pinellia ternata and characterize its own structure and activity. Methods : Pinellia ternata (600g) was extracted with 95% methanol (1L) at $37^{\circ}C$ for 4 days, with shaking at 250rpm. The extract was further solvent-fractionated with n-hexane, chloroform, ethylacetate and water. The active fraction was subjected to JAI recycling prep-HPLC JAIGEL GS-320 column. The structure was identified for the active peak with NMR and GC. Results : Tyrosinase was potently inhibited by 95% methanol extracts from Pinellia ternata. The $IC_{50}$ value of the extracts was estimated to be 0.05mg/ml. The extracts was divided into four solvent-fractions, and the most potent tyrosinase inhibition was found in ethylacetate layer. $IC_{50}$ value of ethylacetate fraction was 0.001mg/ml. This fraction was further purified with JAI Recycling Preparative HPLC (Model: LC 9104). The isolated compound showing inhibitory activity was characterized on its chemical structure by NMR and the compound was identified as 3,4-dihydroxybenzaldehyde. $IC_{50}$ was found to be 7.74 ${\mu}M$ which is much lower than that of kojic acid $(66.5{\mu}M)$. Conclusions : The data suggest that 3,4-dihydroxybenzaldehyde isolated and identified from Pinellia ternata is very strong inhibitor to melanin biosynthesis.
The separation of indole from a model mixture comprising four kinds of nitrogen heterocyclic compounds [indole (In), quinoline (Q), iso-quinoline (iQ), quinaldine (Qu)], three kinds of bicyclic aromatic compounds [1-methylnaphthalene (1MN), 2-methylnaphthalene (2MN), dimethylnaphthalene (DMN)], biphenyl (Bp) and phenyl ether (Pe) was examined by batch cocurrent 4 stages equilibrium extraction. The model mixture used as a raw material in this work was prepared according to the components and compositions contained in coal tar fraction (the temperature ranges of fraction: $240{\sim}265^{\circ}C$). An aqueous solution of formamide was used as a solvent. Indole was recovered more than 99% through 4 stages of the equilibrium extraction. The range of selectivity of indole in reference to DMN obtained through the 5 stages equilibrium extraction was found to be 63~118. The process for separation and recovery of indole contained in coal tar was studied by using the experimental results obtained from this work and the previous work.
In this work the antioxidant effects of browning reaction products prepared by xylose-tryrtophan reaction system were discussed. The antioxygenic brown pigments were separated by solvent extraction, and column chromatography and isolated by gel filteration. The functional groups of the brown pigments which had antioxidant activity were examined. The brown pigments extracted with methanol showed antioxidant effect and were fractionated in 5portions on DEAE-cellulose column. The elutes with methanol: acetic acid(10:30 v/v sol n(A), methanol: chloroform(95:5 v/v) sol n(C), and chloroform: acetic acid(10:30 v/v) sol n(E) only showed antioxidant activity and their compositions were 22.43, 21.51 and $34.43\%$ respectively. When each fraction on DEAE-cellulose column was reseparated on Sephadex LH-20 column, 2 fractions were obtained from portion A and C respectively. Molecular weights of A, C and E fraction of brown pigments were from 2,600 to 3,700. By elucidation of IR spectra, the pigment fractions which showed a strong antioxidant activity were tearing the indole group. It is suggested that the antioxidant function of the brown pigment is due to hydroxy and amino group. A higher activity of the brown pigment fraction E might be attributed to carboxylic acid or carboxylic ester compounds.
In this study, the activity of Takli-san for anti-oxidation, anti-wrinkle and whitening effect was verified and its applicability as a cosmetic material was confirmed. 1-1-diphenyl-2-picryl-hydrazyl radical scavenge, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenge, elastase and tyrosinase inhibitory activity were investigated by solvent fraction, and ethyl acetate fraction of water extract from Takli-san (TW-EA) showed the highest inhibitory activity. Western blot was performed to confirm anti-wrinkle and whitening effect in the cells, and it was investigeted that the TW-EA inhibition effect was superior to that of the positive contol used for the comparison test. Through the results of the experiments, the applicability as a cosmetic material of Takli-san was verified.
