Asimina triloba contains many kinds of annonaceous acetogenins known to have toxicity and/or activity. In this study, analysis of acetogenins was carried out on the extract and its solvent fractions of A. triloba leaves using a liquid chromatography mass spectroscopy(LC-MS). In the extract, the ethanolic extract was significantly higher acetogenin contents than hot water extract. In the analysis of solvent fraction from the ethanolic extract, n-hexane, ethylacetate, and aqueous fraction were contained 85.17~90.92%, 8.96~14.52% and 0.12~0.35% of total acetogenin in the extract. Based on these results, the analytical method is useful for comparing annonaceous acetogenins contents in plant samples. Moreover, when research or product development aiming at the physiological activity of acetogenin such as anticancer activity using A. triloba leaves is performed, a fractionation using a nonpolar organic solvents and alcohol extract may be useful methods. This method can also be used for the purpose of eliminating toxicity as well.
This study was carried out to investigate whether Chrysanthermum zawadskii var. latilobum Kitamura (C. zawadskii) extracts has an inhibitory effect against the mutagenicity by cigarette smoke condensates (CSC). C. zawadskii was extracted with 70% ethanol and the yield was 18.5%. We further fractioned 70% ethanol extract sequentially to diethylether, chloroform, dichloromethane, and aqueous water, and gained the yield of 17.5%, 5.6%, 5.8%, 32.8% and 35.5%, respectively. In the Ames test, there was no mutagenic effect of crude extract and its solvent fractions up to 2 mg/plate toward Salmonella typhimurium TA 98 with or without S-9 mix metabolic activations. On the contrary, the crude extract showed an inhibitory activity against the mutagenicity of CSC in the presence of S-9 mix metabolic activation. Diethyl ether layer among five solvent fractions showed the highest inhibitory activity. The inhibitory activity of diethyl ether fraction was also increased in a dose-dependent manner and the inhibitory rate was about 97.7% at the concentration of 1 mg/plate. In this study, we conclude that crude extract of C. zawadskii itself is potentially safe for mutagenicity, and the diethyl ether fraction has an inhibitory effect against the mutagenicity of CSC.

Background: The present study was carried out to asses whether mulberry leaves (MLs) have the potential to inhibit the mutagenic effect of cigarette smoke condensates (CSCs). Methods and Results: ML powder was extracted with 70% ethanol, and a yield of 35.1% by weight was obtained. The 70% ethanol extract of ML was further extracted sequentially using diethyl ether, chloroform, butanol, dichloromethane and water. The crude 70% ethanol extract of MLs and its solvent fractions did not show any mutagenic effect when tested at concentrations up to 1 mg/plate against Salmonella typhimurium TA98. In contrast, the crude 70% ethanol extract showed an inhibitory activity against the mutagenicity of CSCs in the presence of S-9 mixture. Among the solvent fractions, the diethyl ether fraction showed the highest inhibitory activity, which increased in a dose-dependent manner, inhibiting mutagenesis by approximately 97.1% at a concentration of 1 mg/plate. Conclusions: In this study, we found that a crude 70% ethanol extract of MLs and the diethyl ether fraction themselves are potentially not mutagenic, but inhibit the mutagenic effect of CSCs.
We investigated the inhibitory effects of solvent extracts from Ailanthus altissima in A549 human lung cancer cell. A. altissima has been recognized as a traditional healthy food due to its various biological activities against hypertension, strokes, fever, pain, neuralgia, inflammation, and cancer effects. Recently, it has been reported that the extracts of various wild vegetables show strong anti-cancer properties by induction of apoptosis. However, the mechanisms of their cytotoxicity in human lung cancer cells have been poorly understood. The present study was investigated the effects of solvent extracts from A. altissima on cell growth and apoptosis on A549 human lung cancer cells. A treatment of A. altissima inhibited the growth of A549 cells in a dose-dependent manner by inducing apoptosis. Especially, the chloroform fraction showed the highest anti-cancer effect among five kinds of fractions. And also, induction of apoptosis by chloroform fraction were associated with down-regulation of Bcl-2, and up-regulation of pro-apoptotic Bax expression. From these results, A. altissima may have a therapeutic potential in human lung cancer cells and as a functional food.

Actinidia arguta extracts obtained from three cultivars ('Sae-Han', 'Dae-Sung', and 'Chil-Bo') were assayed for their antioxidant properties and nutritional composition. Antioxidant activity of the extracts was evaluated using 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging method. Total phenolic contents of the extracts were determined by the Folin-Ciocalteu method. Vitamin C (L-ascorbic acid) content measured by a colorimetric method and reducing sugar content estimated by dinitrosalicylic acid (DNSA) method. Crude extracts from A. arguta 'Sea-Han' showed the most potent radical-scavenging activity showing 86.55% at 10 mg/ml. The DPPH radical scavenging activity of extracts and solvent fractions from Sea-Han cultivar was in decreasing order of EtOAc fraction>BuOH fraction>$CH_2Cl_2$ fraction>hexane fraction, among which EtOAc fraction showed the highest antioxidant activity (87.51 % at 5 mg/ml). Total phenolic contents in A. arguta 'Sae-Han', 'Dae-Sung' and 'Chil-Bo', were 32.93, 28.23, and 25.60 mg/g, respectively. Vitamin C contents of them were 840.57, 578.81 and 730.10 ug/g, respectively.
In this study, the biological activities of Glycyrrhiza uralensis were investigated, including antioxidative, fibrinolytic, thrombin inhibitory, and a-glucosidase inhibitory activity. The hot water extract of Glycyrrhiza uralensis was fractionated into hexane, $CHCl_3$, ethyl acetate, butanol, and water fractions, and each of these fractions were assayed individually. The water fraction showed the highest extraction yield of 19.45% (w/w). Using the DPPH method, the free radical scavenging activity was to be the strongest in the $CHCl_3$ fraction at 89.3%. Using the fibrin plate method, only the butanol fraction showed a substantial plasmin activity of 0.62 units/ml. In thrombin inhibitory activity tests, a 100-fold dilution of the ethyl acetate fraction showed the strongest activity of 46.9%. In the a-glucosidase inhibitory activity assay, a 100-fold dilution of the $CHCl_3$ fraction showed the strongest activity of 80.6%. In conclusion, the combined results of this study demonstrate that the extracts of Glycyrrhiza uralensis can be used as a material for the development of biofunctional foods for diabetics.

The solvent extracts of Oenothera laciniata, which were extracted by using several solvents with different polarities, were prepared for utility as a natural preservative. The O. laciniata extract by 80% ethanol was sequentially fractionated with n-hexane, dichloromethane, ethylacetate, and butanol. The antibacterial activities and cell growth inhibition were investigated on each strain with the different concentrations of O. laciniata extracts. Antibacterial activities were shown in ethanol, ethylacetate, and butanol fraction of O. laciniata. However n-hexane, dichloromethane and water fraction showed weak antibacterial activity against the tested microorganisms. Among the five fractions, ethylacetate fraction showed the highest antibacterial activities against microorganisms tested, such as Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella Enteritidis and Salmonella Typhimurium. The polyphenolic compounds widely occurring in the traditional medicine plants have been reported to possess high antibacterial activity. The polyphenolic compounds from ethanol, n-hexane, dichloromethane, ethylacetate, butanol, and water fraction were 63.96 mg/g, 8.49 mg/g, 28.11 mg/g, 172.64 mg/g, 114.56 mg/g, and 34.91 mg/g, respectively. There are some relationships between antibacterial activity and polyphenol content in natural plant. The ethylacetate fraction could be suitable for the development of a food preservative.
Helianthus tuberosus has been known to inhibit the growth of weeds and other plants sharing its habitat. This study was conducted to identify the allelochemicals of Helianthus tuberosus which were extracted with water and solvents. Aqueous extracts of leaf, stem, root, tuber and tuber peel of Helianthus tuberosus except tuber did not show significant differences in phytotoxicity to alfalfa seedlings. It was considered that Helianthus tuberosus contained fewer or less potential water-soluble substances that were toxic to alfalfa. Methanol extract of leaves of Helianthus tuberosus was sequentially partitioned in increasing polarity with n-hexane, ethylacetate and n-butanol. Each extract had a yield of 148, 12, 15.7 and 9.5g, respectively. Inhibitory effects on germination of alfalfa seeds treated with four fractions were not significantly different. But the significant reductions on hypocotyl length were observed for all the solvent extracts. Among the four fractions, the ethylacetate fraction showed the most significant inhibition effect on bioassay with alfalfa. Further separation of the active ethylacetate fraction by open column chromatography led to the 25 subfractions. In bioassay of each sub-fraction with alfalfa seeds, sub-fraction No. 13 showed the most inhibitory effect on seedling growth. $^1$H NMR and gas chromatography-mass spectrometry analysis revealed that sub-fraction No. 13 was the mixture of straight-chain saturated fatty acids.

Diospyros lotus has been cultivated for its edible fruits and leaves which are considered for its medicinal importance. The aim of this study was to evaluate the anti-melanogenesis of ethyl acetate (EA) fractions from D. lotus leaves in B16 cells. The order of the total polyphenol content with regard to the different solvent fractions from D. lotus leaves was EA>butanol>metahanol>chloroform>n-hexane. The major compounds of EA fraction from D. lotus leaves by HPLC analysis were myricitrin and myricetin. Cellular TYR activity and melanin content in response to treatment with 100 mg/mL of EA fraction was inhibited more strongly than group treated with arbutin. Further, EA fraction exhibited significant anti-melanogenesis effects by reducing the levels of microphthalima-associated transcription factor (MITE), inhibiting the synthesis of TYR, tyrosinase-related protein-1 (TRP-1) and TRP-2. Therefore, EA fractions from D. lotus leaves may be a good source of skin-whitening agents in the future development of medicine-based trouble skin therapy.
Twenty-four different natural food materials extracted with 80% methanol were used to investigate the inhibition of cell-mediated immediate type allergic reactions induced by compound 48/80 in rat peritoneal mast cells (RPMCs). Nine 80% methanol extracts screened at a concentration of $10\;{\mu}g/mL$ inhibited histamine release from RPMCs induced by compound 48/80. Of these, two materials (Ulmus parvifolia and Liriope platyphylla Wang et Tang) were extracted and fractionated into four different solvent types (chloroform, ethylacetate, butanol, and water), and the fractions with major anti-allergic effects were assessed. The chloroform fraction of U. parvifolia (UP) at $5\;{\mu}g/mL$ and the ethylacetate fraction of L. platyphylla Wang et Tang (LPWT) at $1\;{\mu}g/mL$ showed the greatest inhibition of histamine release induced by compound 48/80. The chloroform fraction of UP and the ethylacetate fraction of LPWT in combination showed a greater inhibition of histamine release than either fraction alone. The cAMP levels in RPMCs treated with UP and LPWT were significantly greater than in cells treated with compound 48/80 alone. Our studies suggest that extracts from UP and LPWT may alleviate immediate type hypersensitivity reactions through the increase of cAMP levels in the mast cells.
