In this study, we investigated antimicrobial and cytotoxicity effects of Undaria pinnatifida Sporophyll, which using methanol, dichloromethane and ethanol were extracted and fractionated into four different types: methanol (UPMM), hexane (UPMH), butanol (UPMB) and aqueous (UPMA). The antimicrobial activity was increased in proportion to its concentration by the paper disc method. Among the solvent fractions, UPMM and UPMB showed relatively strong antimicrobial activities in the order. Among various partition layers, the methanol partition layer (UPMM) was showed the strongest cytotoxic effects on all cancer cell lines. We also observed quinone reductase (QR) induced effects in all fraction layers of UP on HepG2 cells. The QR induced effects of UPMH on HepG2 cells at $320\mu g/mL$ concentration indicated 2.36 with a control value of 1.0.
The antioxidant properties and protective effects of Inula britannica on ${H_2}{O_2}$-induced SH-SY5Y neuroblastoma cell damage were investigated. A series of solvent fractions, including hexane(Fr.H), petroleum ether, chloroform, ethyl acetate(Fr.EA), and water fraction(Fr.W), were prepared from the 70% methanol extracts of Inula britannica. Fr.W had the highest total contents of phenolics and flavonoids, followed by Fr.EA. The antioxidant properties of the fractions were also evaluated by analyzing their scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radicals, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals, and nitric oxide. Fr.W showed the strongest activities in all assays. The concentrations of Fr.W that resulted in 50% reductions of the DPPH and ABTS radicals were 20.7 ${\mu}g$/mL and 39.4 ${\mu}g$/mL, respectively. Fr.W showed the weakest cytotoxic activities on the SH-SY5Y cells, whereas it effectively protected ${H_2}{O_2}$-induced cell death, increasing cell survival by 35.0-77.0% at a concentration range of 62.5-250 ${\mu}g$/mL. In this range, Fr.W also significantly decreased intracellular ROS levels by 34-39%. Overall, the antioxidant properties of Inula britannica can contribute to rescuring neuronal cells from oxidative stress-induced cell injury.
To examine the characteristics of antioxidative compounds from Capsella bursa-pastoris, ethanol extracts were separated into five organic solvent fractions; hexane(Fr.H), diethyl ether (Fr.E), ethyl acetate(Fr.EA), butanol (Fr.B), and water(Fr.D) fractions. Fr.B showed the greatest electron donating ability and inhibitory effect on lipid peroxidation. Whereas Fr.E had the most excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. The inhibitory effect of each fraction on xanthine oxidase was also measured. Fr.E had the strongest inhibitory effect on xanthine oxidase and $IC_{50}$ was $5.65\;{\mu}g$. The results indicate that the superoxide radical scavenging activity of Fr.E is caused by the inhibitory effect on radical generating system of xanthine oxidase. The cyclic metabolism of poly(3-hydroxyhutyrate-co-3-hydroxyvalerate) synthesized from glucose by Alcaligenes sp. SH-69 in the presence or absence of new carbon substrate was investigated. In batch culture, the content and weight average molecular weight of the copolymer already stored in the cell decreased rapidly when there was no other carbon source available. After the depletion of carbon source, the amount of high molecular weight copolymer decreased more rapidly than that of low molecular weight copolymer, and as a result, average molecular weight distribution shifted to the lower value. The addition of a mixture of glucose and levulinic acid when the initial carbon substrate, glucose, was nearly depleted supported the continual increase in cell mass and the accumulation of poly(3HB-co-3HV) with high molar fraction of 3HV. However, solvent fractionation of the polymer with acetone revealed the degradation of pre-existing polyhydroxyalkanoale (PHA) in parallel with the synthesis of PHA from new carbon substrate. Even though PHAs obtained from each substrate alone were the copolymer of 3HB and 3HV, it was found that the polymer accumulated in the cells grown by sequential feeding was mainly physical mixture of two poly(3HB-co-3HV) copolymers containing different molar fractions of 3HV.
We selected 8 plants among 11 Jeju native plants to search useful natural anti-oxidants by determining the amount of total polyphenols and the various anti-oxidative effects. Ethyl acetate extracts of Castanopsis sieboldii (Makino) Hatus. and butanol extracts of Oenothera laciniata Hill showed strong DPPH free radical scavenging effect. The IC50 value of each solvent extract was 1.6 ㎍/㎖ and 2.4 ㎍/㎖, respectively. The ethyl acetate fraction of Castanea crenata Siebold & Zucc exhibited strong inhibition against nitric oxide production. For the inhibition of xanthine oxidase, the ethyl acetate extracts of Castanea crenata Siebold & Zucc showed strong inhibition activity with 16 ㎍/㎖ of its IC50. The ethyl acetate extracts from Castanopsis sieboldii (Makino) Hatus showed strong superoxide scavenging effect with 7 ㎍/㎖ of its IC50. The hydroxyl radical scavenging activity of butanol extract of Castanopsis sieboldii (Makino) Hatus was 76%. Therefore, with more researches on purification and identification of active compounds, plants studied are expected to be natural sources for the functional food/cosmeceuticals with anti-oxidative properties.
This study investigated the acaricidal and repellent activities of various plant extracts and Cnidium officinale-derived materials against Haemaphysalis longicornis. Among the 24 methanolic plant extracts, C. officinale showed the highest acaricidal activity (93.3%) against H. longicornis. Platycodon grandiflorus, Phytolacca americana, Magnolia denudata, Houttuynia cordata, C. officinale, and Pueraria montana showed significant differences in their repellent activity. Consequently, the activity of solvent-fractionated materials and the derived material of the C. officinale, was investigated and a 90% acaricidal effect was observed three days after hexane fraction treatment, indicating that the repellent response was statistically significant. Butylidenephthalide, which is a C. officinale-derived compound, showed 90% acaricidal activity seven days after treatment. Additionally, all H. longicornis moved to the untreated area in the t-tube olfactometer experiment, confirming the repellent activity of butylidenephthalide. Therefore, the results of this study suggest the use of C. officinale crude extract and butylidenephthalide as potent acaricidals and repellents against H. longicornis, respectively.
The flowers of Cosmos bipinnatus were extracted with solvent made with methanol:water (4:1) and the concentrates were partitioned into ethyl acetate (EtOAc), n-butanol (n-BuOH), and water (H2O) fractions. The octadecyl silica gel (ODS) and silica gel (SiO2) column chromatographies were repeated for the EtOAc fraction to isolated of two phenolic compounds. The chemical structure of the isolated compounds were identified as benzyl O-β-ᴅ-glucopyranoside (1), and 2-phenylethyl O-β-ᴅ-glucopyranoside (2) through spectroscopic datas such as nuclear magnetic resornance, infrarad spectroscopy, and mass spectroscopy. These two compounds were first isolated from C. bipinnatus flowers through this study. To evaluate the anti-atopic activity of the two isolated compounds using a HaCaT cell line induced by ultraviolet light, several experiments were conducted and neither both compounds showed toxicity in the concentration range of 1 to 1,000 ㎍/mL. In the results of anti-atopic activity through Thymus and activation regualted chemokine (TARC) assay, both compounds showed dose-dependent TARC inhibitory activity. In particular, compound 1 showed significant activity even in a low concentration range of 10 ㎍/mL, and in different concentration ranges. Also compound 1 showed higher inhibitory activity than other compound, confirming that the anti-atopic activity was the most excellent. Based on these results, it is considered that it can be used as a functional cosmetic material.

In the present study, we prepared hot water extracts and the subsequent organic solvent fractions of methanol extracts of Houttuynia cordata (HC) and Lespedeza cuneata (LC), and investigated their anti-inflammatory effects on lipopolysaccharide-stimulated RAW264.7 cells. Among the treated samples, hexane, chloroform, and ethyl-acetate fractions of HC and LC inhibited nitric oxide (NO) production in a dose-dependent manner, and decreased inducible nitric oxide synthase (iNOS) protein expression. And, we analyzed the flavonoid contents of the ethyl-acetate fraction of HC and LC, and chose apigenin for the further experiments because apigenin was one of flavonoids commonly found in HC and LC. Apigenin dramatically inhibited NO production in a dose-dependent manner without affecting cell viability and decreased iNOS and cyclooxygenase-2 (COX-2) expression. In addition, apigenin suppressed the phosphorylation of p38 and Jun N-terminal kinase (JNK) indicating that apigenin exerts anti-inflammatory activity via the mitogen-activated protein kinase (MAPK) signaling pathway. Subsequently, we conducted RNA-sequencing analysis to detect differentially expressed genes upon apigenin treatment. Among the down-regulated genes, four cytokine genes (interleukin (IL)-1α, IL-1β, IL-6, and colony stimulating factor 2 (CSF2)) were selected for the further analysis, and the reduction of their expression by apigenin was confirmed with quantitative real-time polymerase chain reaction. Overall, our results suggest that Houttuynia cordata and Lespedeza cuneata have the anti-inflammatory effects and apigenin can be the one of key molecules responsible for their anti-inflammatory activities.
This research has been carried out to find out the highest effect on insecticidal, fungicidal and herbicidal activities in leaves, stem and tuber extracts of yacon (Smallanthus sonchifolius) and extraction methods such as water, boiling water and methanol. Characteristics of potential herbicidal components among extraction methods were investigated by solvent fractions such as hexane, chloroform, ethyl acetate, butanol, and water. Generally, methanol extract was the best on inhibition of germination rate, plant height, and root length in cucumber and barley. On the other hand, the inhibition effect on growth in cucumber and barley was the best in tuber among plant parts of yacon. Inhibition of germination rate, plant height, and root length in cucumber and barley in solvent fractions was the best in water fraction, but there were no differences in other fractions. Digitaria sanguinalis L. and Solanum nigrum L. by 5 and 10% extractions of yacon tuber were controlled by more than 70~80% and 95~100%, respectively. However, there was no inhibition effect on foliar treatment in cucumber and barley as affected by 5 and 10% extractions of yacon tuber. Mortality of green peach aphid (Myzus persicae Sulzer) was 50% at 3 days after treatment of 5% extracts of yacon leaves. Mortality of brown plant hopper (Nilaparvata lugens Stal) was only 24% in 5% extracts of stems and leaves with midrib, but was 57% in 5% extracts of leaves without midrib. There was no fungicidal effect on anthracnose (Colletotrichum truncatum), wilt disease (Fusarium oxysporum), verticillium wilt (Verticillium dahliae), bacterial blight (Xanthomonus oryzae) in 5% extracts of yacon leaves.
Sweetpotato shoot tops (leaves, tips and petioles) are known to be very useful parts as vegetables because of their high nutritive values and great biomass yield. In this study, the phenolic compound contents, antibacterial activity, mutagenic activity, and antimutagenic activity were investigated in sweetpotato tips that were 10-15cm of shoot top including stems, petioles and tender leaves after sprout of storage roots. The study was done by extracting sweetpotato tips with 80% ethanol and the ethanol fraction was re-extracted with hexane, chloroform, ethyl acetate, butanol and water. In ethyl acetate and butanol fractions, total phenolic compounds contained 95. 6mg/g extract and 69.3 mg/g extract, respectively, The antibacterial activity was measured using the paper disk method with concentrations of 1, 2, 5 and 10 mg/disk of butanol and ethyl acetate fractions against L. monocytogenes and S. Typhimurium strains. Higher doses of solvent extracts showed the higher antibacterial activities. In addition, 5, 10 and 20 mg/mL of the extracts were tested to determine the antibacterial activity in liquid culture. The sweetpotato leaf extract by ethyl acetate showed 1 log reduction compared to control after 24 hrs on Listeria monocytogenes, but 20 mg/ml of butanol extract completely inhibited the growth of the pathogen after 12 hrs. The extracts from ethyl acetate or butanol on Salmonella Typhimurium did less than 1 log reduction during cultivation compared to control. The numbers of S. Typhimirium TA98 and TA100 revertant colonies were 29-33 and 159-188 CFU/plate, respectively, indicating that solvent extracts were no mutagenic activity. The antimutagenic test was performed by adding direct mutagen 2-NF and MMS, and butanol and ethyl acetate showed antimutagenic effect. Thus, this study showed that sweetpotato tips had high phenolic contents and both antimicrobiol and antimutagenic properties. Sweetpotato tips would be good nutritive source because of their high nutrient content without any toxicity in consuming.
