  • The objective of this study was to evaluate the anti-melanogenic effects of Hizikia fusiforme (HF) fractions in ${\alpha}$-melanocyte stimulating hormone-induced B16F10 mouse melanoma cells. Ethanol extractions of Hizikia fusiforme (EEHF) were subjected to fraction by using dichloromethane (CFHF), ethyl acetate (EAFHF), butanol (BFHF), and water (WFHF). EEHF, CFHF, and EAFHF inhibited tyrosinase activity and melanin synthesis in B16F10 mouse melanoma cells in a dose-dependent manner. The melanin contents were inhibited by 40.5% and 33.2% in response to treatment with 50 ${\mu}g/ml$ of EEHF and CFHF, respectively. In addition, tyrosinase activities showed a 53.3% and 54.1% reduction in treatment with 50 ${\mu}g/ml$ of EEHF and CFHF. Western blotting analysis showed that EEHF, CFHF, and EAFHF inhibited tyrosinase, TRP-1, TRP-2, and MITF expression in a dose-dependent manner. In conclusion, these findings indicate that ethanol and dichloromethane fractions of Hizikia fusiforme, which inhibit melanin synthesis and tyrosinase activity, are effective skin-whitening agents.

  • Crude polysaccharide fractions of commercially available grape wines (red wine, white wine and wild grape wine) were prepared by evaporation and ethanol precipitation to confirm and identify anti complementary polysaccharides in the wines. When these fractions were evaluated for their anti complementary activity, crude polysaccharide fractions of red wine (RW-0) and wild grape wine (WGW-0) showed higher anti-complementary activities than those of white wine (WW-0). RW-0 and WW-0 were further fractionated into RW-1, WW-1 as high-molecular fractions, and RW-2, WW-2 as low-molecular fractions through gel permeation column chromatography on Sephadex G-75. RW-1 had the most potent activity with the highest carbohydrate content (91.3%). Anti-complementary activity of red wine was higher than that of white wine, suggesting that active polysaccharides such as pectin and hemicellulose are mainly distributed in the grape skin which is removed during white wine making. In addition, high molecular fractions, RW-1 and WW-1 with high contents of carbohydrate and high yields showed higher activities than those of low molecular fractions, RW-2 and WW-2.

  • Angelica tenuissima, also known as Ligusticum tenuissimum, is classified as a food-related plant and has been used as traditional medicines treating headache and anemia in Asia. However, its anti-melanogenic effect has not been reported in detail. When the extract of Angelica tenuissima (ATE) was prepared by the extraction with 70% EtOH at $80^{\circ}C$ (final yield = 22%), the contents of decursin and Z-ligustilide in ATE were determined 0.06% and 8.43%, respectively. Total flavonoid and phenolic content in mg ATE were $5.52{\pm}0.07{\mu}g$ quercetin equivalents and $237.27{\pm}13.24{\mu}g$ gallic acid equivalents, respectively. Antioxidant capacity of ATE determined by DPPH and ABTS assay was increased with a dose dependent manner up to $1000{\mu}g/m{\ell}$. The amount of melanin synthesis followed by ${\alpha}-melanocyte$ stimulating hormone on B16F10 cells were significantly reduced in the presence of ATE (250 to $1000{\mu}g/m{\ell}$, p<0.05). ATE (125 to $1000{\mu}g/m{\ell}$, p<0.05) suppressed the tyrosinase activity but did not show any significant effect on ${\alpha}-glucosidase$ activity at the same condition. Taken together, ATE possesses tyrosinase inhibitory potential with significant antioxidant capacities. These effects of ATE might be involved in suppression of melanin synthesis, at least, in B16F10 cells. The anti-melanogenic potential of ATE will provide an insight into developing a new skin whitening product.

  • The incidence of Anthracnose causing severe damage to the foliage and fruit in watermelon has increased in some major watermelon producing areas in Korea. To develop anthracnose resistant line, 'AU-Producer' having resistant gene to anthracnose was selected from germplasm and crossed with high quality line '920533'. Following the initial cross, backrossing and disease screening were performed to select resistant lines that produced high yields with excellent quality fruit. As a results of these procedure, a mid maturing watermelon line, 'Hangyeol' with resistance to anthracnose (Colletotrichum orbiculare) race 1 and 3 was developed at the National Institute of Horticultural and Herbal Science, Rural Development Administration (RDA). It has red flesh and commonly produces fruit with clear stripes on skin between 5.3 to 10.1 kg. Average soluble solid contents are ranged from 9.8 to 11.8$^{\circ}$Bx. The yield and quality of 'Hangyeol' is comparable to or better than those harvested from the popular commercial cultivars. The achievement of this experiment could contribute to provide the resistant parents in an anthracnose resistance breeding program in watermelon.

  • Anthracnose is a major limiting factor of the watermelon production in Korea. A mid-late maturing watermelon line, 'Hanbit' with resistance to anthracnose (Colletotrichum orbiculare) race 1 and 3, was developed by the cross between anthracnose resistant cultivar 'AU-Producer' and high quality inbred line '920533' at the National Institute of Horticultural and Herbal Science, Rural Development Administration (RDA). 'Hanbit' produces 5.4 to 9.5 kg of red flesh fruits with clear stripes on skin. Average soluble solid contents are ranged from 10.4 to 11.2$^{\circ}$Bx. The yield and quality of 'Hanbit' was comparable to or better than those harvested from the popular commercial cultivars. 'Hanbit' can be cultivated in open fields and under the protected green house condition. In addition to the commercial production, it can be used as a resistant material in $F_1$ hybrid breeding program.

  • Prunus mume Sieb. et Zucc is widely distributed in East Asia (Korea, Japan, and China), and its fruit is often used as a medication and food material. However, because most previous studies have only investigated the state of Prunus mume fruit extract, studies on the various ways of processing this extract are still needed to increase its utilization. In this study, we evaluated the physicochemical properties and physiological activities of spray-dried Prunus mume vinegar powder (SPP). The sugar content, pH, total acidity, and moisture content of the SPP were 8.90 °Brix, 3.19, 1.05%, and 3.07%, respectively. The SPP exhibited significantly high antioxidant activity in terms of DPPH radical scavenging activity (65.55%), reducing power (1.48), and hydrogen peroxide scavenging activity (48.07%). In addition, the SPP remarkably decreased the cell viability of human breast MDA-MB-231 and human skin cancer SK-MEL-28 in a dose-dependent manner. The morphological results of the treatment of MDA-MB-231 cells with SPP were distorted, shrunken cell masses. Furthermore, apoptotic bodies and nuclear condensation formed in the SPP-treated MDA-MB-231 cells. The total polyphenol and flavonoid contents of the SPP were 59.58 ㎍/g (gallic acid equivalent) and 57.56 ㎍/g (quercetin equivalent). The results of this study indicate that SPP, which has antioxidant activity and anticancer effects, can be useful in the development of natural medicines and functional food ingredients.

  • The effect of night temperature was investigated on mango (Mangifera indica cv. Irwin) for setting appropriate night temperature and managing mango cultivation in a greenhouse. Different night temperatures (10, 15, and 20℃) were treated from budding to fruit development in mango trees. As night temperature increased, the initiation of flowering, full blooming, fruiting, and fruit development tended to be accelerated. There were no significant differences in the growth of shoots flushed after the fruit harvest among trees treated with different night temperatures. The lengths of the flower stalk were the shortest at 10℃ of night temperature; however, there were no significant differences between 15℃ and 20℃ of the night temperature. The length, diameter, and weight of mango fruits were higher at 15℃ and 20℃ than 10℃. Soluble solid contents and acidities showed no difference; the firmness and skin color were better as the night temperature increased. Accordingly, setting the minimum night temperatures to 15℃ or 20℃ can increase the number of fruits and produce high-quality fruits when cultivating mango in the greenhouse. Even the minimum night temperature to 15℃ can reduce heating costs in the greenhouse.

  • Mast cells containing a variety of mediators in their cytoplasmic granules are widely distributed in connective tissues and mucosal surfaces of skin, airways, and guts. Within these tissues, mast cells are involved in the pathophysiological conditions such as inflammation, self-defense, tissue-remodeling, and autoimmunity. In order to understand the functional roles of master cells in the uterus, we histologically examined the distribution and density of uterine mast cells in the different aged mice. Until 6 weeks mast cells were sparsely detected in the uterus. But at 7 weeks after birth, when estrous cycle begins, the number of mast cells within uterine tissues increased dramatically and the increment of mast cell density continued up to 32 weeks-age. After then, uterine glandular tissue degenerated gradually and density of uterine mast cell decreased. Uterine mast cells were mainly found in the myometrium and they were closely associated with smooth muscle cells, fibroblasts, and collagens, which contents were changed according to the uterine development in the myometrium. These results suggest that uterine mast cells could be involved in myometrial contractions mediated by smooth muscle cells and tissue reconstitution or remodeling during estrous cycle and parturition including the various immunological functions.

  • In this study, we investigated the antioxidant effects and active ingredients of Gynura Procumbens extracts obtained through various extraction methods for the development of natural-based cosmetics and pharmaceutical materials. The contents of compounds, total flavonoids, chlorogenic acid, and caffeic acid were compared at different concentrations, revealing the highest content of active ingredients in the 100% ethanol extract. Antioxidant activity assessed by DPPH and ABTS radical scavenging assays showed a concentration-dependent increase in antioxidant activity with the ethanol concentration. Additionally, we validated the DNA damage inhibition and anti-inflammatory effects of Gynura Procumbens extracts through UVB irradiation on Hs68 cell models. The 100% ethanol extract demonstrated significant inhibition of the expression of p-p53, γ-H2AX, iNOS, and COX-2 induced by UVB, indicating its potential in alleviating photodamage effects. Consequently, the efficient extraction of Gynura Procumbens for skin functional material development was confirmed, suggesting the suitability of ethanol or alcohol-based solvents.
