Therapeutic aptamers: developmental potential as anticancer drugs

  • Aptamers, composed of single-stranded DNA or RNA oligonucleotides that interact with target molecules through a specific three-dimensional structure, are selected from pools of combinatorial oligonucleotide libraries. With their high specificity and affinity for target proteins, ease of synthesis and modification, and low immunogenicity and toxicity, aptamers are considered to be attractive molecules for development as anticancer therapeutics. Two aptamers - one targeting nucleolin and a second targeting CXCL12 - are currently undergoing clinical trials for treating cancer patients, and many more are under study. In this mini-review, we present the current clinical status of aptamers and aptamer-based cancer therapeutics. We also discuss advantages, limitations, and prospects for aptamers as cancer therapeutics. [BMB Reports 2015; 48(4): 234-237]

An Adaptive Multi-Echelon Inventory Control Model for Nonstationary Demand Process

  • In this paper, we deal with an inventory model of a multi-stage, serial supply chain system where a single product type and nonstationary customer demand pattern are considered. The retailer and suppliers place their orders according to an echelon-stock based replenishment control policy. We assume that the suppliers can access online information on the demand history and use this information when making their replenishment decisions. Using a reinforcement learning technique, the inventory control parameters are designed to adaptively change as the customer demand pattern is altered, in order to maintain a given target service level. Through a simulation based experiment, we verified that our approach is good for maintaining the target service level.

Properties of AZO/Ag/AZO Multilayer Thin Film Deposited on Polyethersulfone Substrate

  • The AZO/Ag/AZO multilayer films were deposited on polyethersulfone (PES) substrate by using facing target sputtering methods at room temperature. The AZO/Ag/AZO multilayer films with polymer substrate had advantages, such as low sheet resistance, high optical transmittance in visible range and stable mechanical properties. From the results, the AZO/Ag/AZO multilayer films (50/12/50 nm) demonstrated a sheet resistance of 11 ${\Omega}/{\square}$ and average transmittance of 87% in visible range (wavelength of 380-770 nm). Moreover, the multilayer showed stable mechanical properties compared to the single-layered AZO sample during the bending test due to the existence of the ductile Ag metal layer.

  • The influences of substrate temperature were studied when fabricating KLN thin film on amorphous substrate using an rf-magnetron sputtering method. Investigating the vaporization temperature of the each element, the excess ratio of target and the optimum deposition conditions were effectively selected when thin filmizing a material which have elements with large difference fo vaporization temperature. In order to compensate K and Li which have lower vaporization temperatures than Nb, KLN target of composition excess with K of 60% and Li of 30% was used. KLN thin film fabricated on Corning 1737 glass substrate had single KLN phase above 58$0^{\circ}C$ of substrate temperature and crystallized to c-axis direction. The optimum conditions were rf power of 100W, process pressure of 150mTorr, and substrate temperature of $600^{\circ}C$.

  • Distribution Automation System (DAS) will provide supervision and remote control of switches and reclosers such as pole pounted switches and pad-mounted switchgears on high voltage distribution line. Kepco had developed basic function such as remote monitoring, remote control, remote measuring and remote setting at first. As a next step, Kepco has been developed diverse application programs such as single line diagram drawing program, relay coordination program, feeder reconfiguration program, over load elimination program, bad data detection program, section load management program, fault processing program and so on. Kepco is examining to develop more powerful functions for special specification of foreign distribution automation system. This paper explains what is the target for overseas DAS market.

  • We prepared Al doped ZnO/Ag/Al doped ZnO on the polymer substrate by Facing Target Sputtering (FTS). FTS featured Facing Target Sputtering featured that deposition is stable at the low pressure, it has high plasma density and suppresses the substrate damage from energetic particles. We fixed to 50nm up and down thickness of AZO layer, respectively and that of intermediate Ag layer was adjusted with deposition time. In the result, AZO/Ag/AZO multilayer thin films have much better electrical conductivity than AZO single layer thin film. As increasing the thickness of Ag layer, the transmittance decreased.

A Suboptimal Algorithm of the Optimal Bayesian Filter Based on the Receding Horizon Strategy

  • The optimal Bayesian filter for a single target is known to provide the best tracking performance in a cluttered environment. However, its main drawback is the increase in memory size and computation quantity over time. In this paper, the inevitable predicament of the optimal Bayesian filter is resolved in a suboptimal fashion through the use of a receding horizon strategy. As a result, the problems of memory and computational requirements are diminished. As a priori information, the horizon initial state is estimated from the validated measurements on the receding horizon. Consequently, the suboptimal algorithm proposed allows for real time implementation.

  • In this paper, we propose a new technique to compensate for the VCO nonlinearity using only the returned beat signal whose spectrum contains the internal reflections and the targets simultaneously. In the case of a distance measurement system using single antenna, the reflections from the circulator and the antenna are much larger than the return signal from target. The beat signal by these reflections is at much lower frequency than that of the target, and the VCO nonlinearity can be compensated leer using these signals. Indoor experiments were carried out and the results show marked improvement in the shape of range profile arid the range resolution.

  • An interacting multiple model (IMM) approach which merges two hypotheses for the situations of constant speed and constant acceleration model is considered for the tracking of maneuvering target. The inflexibility of uncertainty which lies in the kinematic constraint (KC) represented by pseudomeasurement noise variance is compensated by the mixing of estimates from two model Kalman tracker: one with KC and one without KC. The numerically simulated tracking performance is compared for the "great circular like turning" trajectory maneuver by the single model tracker with constant speed KC and two model tracker which is developed in this paper.his paper.

  • The adoption of oligonucleotide aptamer is well on the rise, serving an ever increasing demand for versatility in biomedical field. Through the SELEX (Systematic Evolution of Ligands by EXponential enrichment), aptamer that can bind to specific target with high affinity and specificity can be obtained. Aptamers are single-stranded nucleic acid molecules that can fold into complex three-dimensional structures, forming binding pockets and clefts for the specific recognition and tight binding of any given molecular target. Recently, aptamers have attracted much attention because they not only have all of the advantages of antibodies, but also have unique merits such as thermal stability, ease of synthesis, reversibility, and little immunogenicity. The advent of novel technologies is revolutionizing aptamer applications. Aptamers can be easily modified by various chemical reactions to introduce functional groups and/or nucleotide extensions. They can also be conjugated to therapeutic molecules such as drugs, drug containing carriers, toxins, or photosensitizers. Here, we discuss new SELEX strategies and stabilization methods as well as applications in drug delivery and molecular imaging.
