  • LMTT (Linear Motor-based Transfer Technology) is horizontal transfer system in the maritime container terminal for the port automation. The system is driven by PMLSM (Permanent Magnetic Linear Synchronous Motor) that is consists of stator modules on the rail and shuttle car. This paper investigates the effect of the drop height of container on impact reaction force of the Impact Absorber (IA) in shuttle car for LMTT. The results of this investigation are obtained from detailed finite element analysis for various parameters, such as the spring coefficient, the drop height of container.

Effects of Cloned Genes on the Stability of Shuttle Vectors between Escherichia coli and Corynebacterium glutamicum (Escherichia coli와 Corynebacterium glutamicum간의 shuttle vectors의 C. glutamicum에서의 안정성에 대한 클론된 유전자의 영향)

  • Escherichia coli/Corynebacterium glutamicum shuttle vectors, pECCG1 and pECCG2 were constructed by joining a 3.00 kb cryptic plasmid pCB 1 from C. glutamicum and a 3.94 kb plasmid pACYC 177 from E. coli. By trimming unessential parts and introducing mulitiple cloning site into the plasmid pECCG 1, a plasmid pECCG122(5.1kb) was constructed. All the shuttle vectors were stably maintained in C. glutamicum up to about 40 generations irrespective of kanamycin addition in the medium. Threonine operon (homoserine dehydrogenase/homoserine kinase) and dapA gene (dihydrodipicolinate synthetase) of C. glutamicum were cloned into the plasmid pECCG122, and the resultant plasmids were designated pTN31 and pDHDP19, respectively. They were used to study the effect of cloned foreign gene on the stability of the plasmid pECCG122. Plasmids pTN31 and pDHDP19 were segregated rapidly from C. glutamicum when cultured in the medium without kanamycin. In medium with $50\mu${\g/ml} of kanamycin, their segregation rates were much slower than those in medium without kanamycin, but the danamycin addition didn't guarantee the complete maintenance of the plasmids in C. glutamicum.

  • In this study, the program to investigate the multiple body interaction effects between a floating platform and a shuttle tanker considering the coupled effect of hull (FPSO) with mooring lines and risers was developed. The coupled analysis program, which is called WINPOST-MULT using the hydrodynamic analysis results by WAMIT, was made. For the verification of WINPOST-MULT by means of numerical experiments, two multiple-body models of an FPSO-FPSO and an FPSO-shuttle tanker system are adopted. With the FPSO-FPSO model and a two-mass-spring system to idealize two identical bodies for the 100-year storm wave condition in GOM, the numerical simulations were performed to investigate the interaction effects between two identical bodies. For the more reality, the coupled analysis for the FPSO-shuttle tanker model in the tandem arrangement was carried out in the consideration of the environmental condition of the West Africa Sea as a rather mild condition. Through the case studies with interaction effect and without interaction effect by the iteration method and the combined method, it is verified that the program is a very useful tool for the analysis of the interaction problem of multiple-body system and the coupled problem of the hull/mooring/riser.

Effect of Design Parameters of Modulating Valve and Hydraulic Clutch on Shift Quality of a Power Shuttle Transmission (모듈레이팅 밸브 및 유압 클러치의 설계 변수가 전후진 파워시프트 변속기의 변속 품질에 미치는 영향)

  • This study was conducted to investigate the effect of design parameters of modulating valve and hydraulic clutch on the shift quality of a power shuttle transmission using a computer simulation. Computer simulation models of a hydraulic control system and a power shuttle drive train were developed and verified by an experimental power train in a laboratory. The software EASY5 was used for the modeling and simulation of the power shuttle transmission. Results of the study were summarized as follows: For a good shift quality. it is required to reduce the transient torque transmitted to the output shaft of the transmission as much as possible. This may be achieved by reducing the modulating time and clutch pressure. It was found that the design parameters most significantly affecting the modulating time and clutch pressure were the spring constant and displacement of a load piston of the modulating valve, and the spring constant and damping of the clutch piston. The modulating time decreased as the spring constant increased and increased as the displacement of the load piston decreased. The transient torque decreased as the modulating time increased. However their relationships were not always linear. As the damping decreased, both the modulating pressure and time decreased, which also resulted in a decrease in the transient torque. The spring constant of the clutch piston affected the modulating time and the peak transient torque. As the spring constant of the clutch increased, the peak transient torque decreased.

  • Recently there have been many interests on tangible sport games that can recognize the motions of players. In this paper, we propose essential technologies required for tangible games, which are methods for swing motion recognition and the calculation of shuttle cock's trajectory. When a user carries out a badminton swing while holding a smartphone with his hand, the motion signal generated by smartphone-embedded acceleration sensors is transformed into a feature vector through a Daubechies filter, and then its swing type is recognized using a k-NN based method. The method for swing motion presented herein provides an advantage in a way that a player can enjoy tangible games without purchasing a commercial motion controller. Since a badminton shuttle cock has a particular flight trajectory due to the nature of its shape, it is not easy to calculate the trajectory of the shuttle cock using simple physics rules about force and velocity. In this paper, we propose a method for calculating the flight trajectory of a badminton shuttle cock in which the wind effect is considered.

  • According as the quantity of goods transported by ship is increasing about $7\%$ per year, a port environment is rapidly changing. To meet this situation successfully, the development of the next generation port loading and unloading system (LMTT) is studied. A Frame of shuttle car for LMTT(Linear Motor-based Transfer Technology) consist of three parts which are outer beam inner beam and cross boom. Outer boom supports a container and inner boom is a framework and cross boom reinforces outer and inner boom. In this study, we carried out the finite element analysis for the effect of cross boom on the strength and stiffness qf the frame according to the number if cross beam leading position of container, the distance ratio if inner boom from center.

Nonlinear Motion Analysis of FPSO and Shuttle Tanker in a Tandem Configuration (탠덤 배치된 FPSO와 셔틀탱커의 비선형 운동 해석)

  • FPSO and shuttle tanker are connected to each other by a mooring hawser and a loading hose through which cargo oil is off-loaded. Even in mild sea-state. environmental loads can cause unstable large drift motions between two vessels in tandem off-loading operations, which may result in collision incidents. Accordingly. the analysis on the relative motion between two vessels due to the environmental loads should be investigated in initial design stage. In this study, the low speed maneuvering equation is employed to simulate nonlinear motions of FPSO and shuttle tanker. Low frequency wave drift forces including hydrodynamic interactions between two vessels are evaluated by near field approaches. Current loads are determined by mathematical model of MMG and wind loads are calculated by employing the wind spectrum according to the guidelines of API-RP2A. Mooring forces produced by turret mooring lines and a flexible hawser are modeled quasi-statically by catenary equations. The effect of environmental loads that affect nonlinear motion is investigated through variation in their magnitudes and the nonlinear motions between FPSO and shuttle tanker are simulated under wave, current and wind in time domain.

  • Transgenic animal and cell line models which are recently developed in toxicology field combined with molecular biological technique, are powerful tools for studying of mutation in vivo and in vitro, respectively. The Big Blue mutagenesis assay system is one of the most widely used transgenic systems. Especially, for the study of direct acting mutagens, Big Blue cell line is very useful and powerful to evaluate mutagenicity because the mutation frequency and mutationspectrlun showed no distinct differences between cell line and animal. The Big Blue cell lines carry stably integrated copies of lambda shuttle vector containing lac I gene as a mutational target. These lambda shuttle vectors are useful for various mutagenesis related studies in eukaryotic system due to their ability to be exposed mutagen and then transfer a suitable target DNA sequence to it convenient organism for analysis. We tried to assess the mutagenic effect of 4-NQO with Big Blue cell line. After the treatment of 4-NQO, genomic DNA was isolated and lambda shuttle vector was packaged by in Vitro packaging and then these were plated on bacterial host in the presence of X-gal to screen mutation in the lac I. We determined MF as a ratio of blue plaques versus colorless plaques and now undergoing the mutation spectrum of 4-NQO in lac J gene sequence.

L-Lysine Production by Amplification of the ddh Gene in a Lysine-producing Brevibacterium lactofementum. (Brevibacterium lactofermentum에서 ddh 유전자의 증폭에 의한 L-Lysine의 생산)

  • The ddh gene encoding meso-DAP-dehydrogenase (DDH) involved in the dehydrogenase pathway is essential for high-level lysine production in Brevibacterium lactofermentum. To investigate the effect of the ddh gene amplification on lysine production by B. lactofementum, we constructed two E. coli -B. lactofermentum shuttle vector, pEB1 and pEB2. The recombinant plasmids, pRK1 and pRK2, carrying the ddh gene were introduced into B. lactofermentum by electroporation. The specific activity of DDH by amplification of the ddh gene was increased 7-fold, and also L-Lysine production of B. lactofermentum strains harboring recombinant plasmids were 18∼20% higher than that of the control.
