An anthocyanin-deficient tomato (Lycopersicon esculentum Mill.) strain, H957, shows an unusual tolerance to low phosphorus (P). To investigate whether the tolerance originates from a tissue/cellular strength, plant tissue culture procedure was employed which facilitate to characterize the tolerance independent of morphological features. The tolerance was analyzed by comparing H957 against H883, its maternal wild type, while each explant was co-cultured on minimal P media. Comparisons were made in fresh weight, dry weight, callus and shoot formation, mineral contents, and P utilization ratios at $0-400{\cdot}\bar{I}MP$, . Growth of the two strains was severely impaired at 0 and $12.5{\cdot}\bar{I}MP.\;At\;25-200{\cdot}\bar{I}MP$, however, H957 consistently showed a greater fresh and dry weight than H883. Shoot onset of H957 was less delayed than H883 compared to optimal P conditions. H957 tissue contains an overall lower P concentration than H883. These observations indicate that H957 may tolerate to low P by its tissue or cellular strength in P utilization side from its morphology.
This study was carried out to investigate the activity of nitrogen fixation and accumulation of heavy metal and inorganic matter in Vicia amoena community at lower region in Kumho riverside, including Youngchon, Chimsangyo, Paldalgyo, Talseochon and Kumhogyo. The contents of inorganic matter and heavy metal of Kumho riverside soil increased in the down stream in each organ of the plant growing in the riverside. Generally, V. amoena community showed rapid growth of shoot and high value of Top/Root ratio. V. amoena community showed higher water content of shoot at late growth stage and higher chlorophyll content. The root nodule of V. amoena community appeared in April and increased by 0.30, 0.27, 0.24, 0.06 and 0.14 g/plant, and nitrogen fixation activity of nodule attained 20.1, 16.8, 15.4, 8.5 and 5.3 μmol·C₂H₄·g fw nodule/sup -1/·h/sup -1/ for non-contaminated area Youngchon, Chimsangyo, Paldalgyo, Talseochon and Kumhogyo, respectively, in June:. Nodule formation and nitrogen fixation activity were reduced in the down stream by the soil contamination and heavy metal accumulation and showed minimum values. at Talseochon and Paldalgyo. V. amoena showed growth adaptation against heavy metal toxicity by restricting heavy metal such as Pb, Cu, Zn, Fe from transport, and by accumulating high Ca ion in shoot, nitrogen and phosphorus in root at late growth stage than those at early one, respectively, but total heavy metal per plant showed higher values in shoot than those in root by high T/R ratio of plant growth.
The axillary buds of 15-year-old Tilia amurensis were cultured on Saito and Ide (IS), Murashige and Skoog (MS) media and woody plant medium (WPM) to establish an effective micropropagation method. Five levels of 6-benzylaminopurine (BAP) were tested. On IS medium and WPM addition of 1.0/l BAP enhanced shoot development and shoot elongation, whereas addition of 0.5/l BAP was effective on MS medium. A better results were obtained from WPM with 1.0/l BAP and MS with 0.1/l BAP. Developed shoots were subcultured on each basal media but with 0.2/l BAP, Multiple shoots were almost doubled in a month. Root formation could be enhanced at higher concentration of indole-3-butyric acid (IBA). Better rooting rate (83.3%) was achieved on a half-strength MS medium with 3.0 /l IBA. Regenerated plantlets were successfully transferred to soil. To investigate the clonal variation in shoot development and shoot elongation by axillary bud culturing, seven plus tree clones were tested, Clonal variation in tissue culturability among plus trees was recognized by the Duncan's multiple range test at the 5% level. Kang Won No. 12 showed the best response on WPM with 1.0/l BAP.
Plant regeneration from the stem tissue of Houttuynia cordata Thunberg was investigated. The medium supplemented with the combination of 2, 4-D 1 mg/L and kinetin 0. 5 mg/L was the most effective for the embryogenic callus formation. The internode segment produced more callus formation than the leaf segment. ${\frac{1}{2}}\;MS$ medium was the most effective for the embryogenic callus formation. The medium supplemented with the 1% activated charcoal produced the whole plant directly without the callus formation from the nodes. The medium supplemented with the combination of NAA 0. 2 mg/L and BA 1 mg/L was the most effective for the plant regeneration from the embryogenic callus.

Certain bacterial species associate with plant roots in soil. The plant growth-promoting rhizobacteria (PGPR) stimulate plant growth and yield in greenhouse and field. Here, we examined whether application of known bacilli PGPR strains stimulated growth and asexual reproduction in the succulent plant Kalanchoe daigremontiana. Four PGPR strains B. amyloliquefaciens IN937a, B. cereus BS107, B. pumilus INR7, and B. subtilis GB03 were applied to young plantlets by soil-drenching, and plant growth and development was monitored for three months. Aerial growth was significantly stimulated in PGPR-inoculated plants, which was observed as increases in plant height, shoot weight, and stem width. The stimulated growth influenced plant development by increasing the total number of leaves per plant. Treatment with bacilli also increased the total root biomass compared with that of control plants, and led to a 2-fold increase in asexual reproduction and plantlet formation on the leaf. Collectively, our results firstly demonstrate that Bacillus spp. promote vegetative development of K. daigremontiana, and the enhanced growth stimulates asexual reproduction and plantlet formation.
This study was conducted to regenerate shoots from internode sections(about 1mm in thickness) of Ardicia pusilla de Candolle. Internode sections were cultured on MS medium supplemented with TDZ or both TDZ and IBA. At one month after culture, primodium, which looks like protocorm like body(PLB) of orchid, appeared around swollen internodes. And then it grew and changed into the shape similar to granule of orange at two or three months after culture. At four to five months after culture, explants covered with them became a cluster, and then multiple shoots were regenerated from them. Primodia formation was the best when internode was cultured on MS medium supplemented with 0.25 $mg{\cdot}L^{-1}$ thidiazuron(TDZ) and 0.5 $mg{\cdot}L^{-1}$ indole-3-butyric acid(IBA). That internodes were cultured on MS medium supplemented with either higher concentration of TDZ than that of IBA, or equal concentration of TDZ and IBA, or TDZ only was little effective for primodia formation.
A large amount of information is currently available for somatic embryogenesis of plants. However, one common problem related to somatic embryos is that the conversion rate can be low for some species. Apical meristems are responsible for post-embryonic growth of the embryo. The low percentage observed is most likely a result of poor apical meristem development or defects in the meristem organization during somatic embryogenesis. In flowering plants, apical meristems are well developed by the late heart stage of zygotic embryo development. In conifers, such as white spruce, apical meristems are formed at the pre-cotyledon stage. Thus, apical meristem development occurs very early, prior to the maturation stage of embryo development. Once formed, meristems are stably determined. In the somatic embryo, as exemplified by white spruce, since embryo development is not synchronous, tissue differentiation including apical meristem formation occurs throughout the“maturation”stage. Different apical meristem organizations can be found among different individuals within a population. In contrast to their zygotic counterparts, the apical meristems appear not to be stably determined as their organization, as the shoot apical meristem especially, can be easily modified or disrupted. Precocious germination seldom results in functional plantlets. All these observations suggest that the conditions for somatic embryo maturation have not been optimized or are not suitable for meristem formation and development. The following strategies could improve meristem development and hence conversion: 1. Simulate in ouuio conditions to promote meristem development prior to the“maturation”treatment.2. Prevent deterioration of apical meristem organization during somatic embryo maturation.3. Promote further meristem development during embryo germination.

Callus induction from a leaf explant has been achieved in Lilium Oriental hybrid 'Casa Blanca'. The highest frequency of callus induction was obtained on MS medium supplemented with 0.5 mg/L BA and 2.0 mg/L NAA after 2 months of culture. The cultures maintained continuously without change in color and type of callus when they cultured in the dark. Plantlet regeneration with a high frequency was achieved from induced calli on the same medium. A number of shoots are formed from one cluster of callus, and bulblets developed into intact plantlets after transfer to hormone-free MS medium. No phenotypic variations were observed among regenerants. Enhancement in plantlet regeneration via callus formation would be expected to facilitate the efficiency of transformation of this Oriental hybrid 'Casa Blanca'.

This study was carried out to establish a regeneration system from leaf explant of purple sweetpotato(Ipomoea batatas L.) The optimal concentrations of plant growth regulators for callus induction and shoot formation were determined. The optimal combination for callus formation was 1$\mu$M 2,4-D 5$\mu$M BM, and highest yield of embryogenic calli were observed on Murashige and Skoog basal medium containing 0.5$\mu$M 2,4-D under light condition after 4weeks of culture. Embryogenec callus was subcultured on medium supplemented with 5$\mu$M ABA for 4 days. Subsequently, regeneration of adventitious shoots occurred when these embryogenic calli were transferred onto medium with 3∼6$\mu$M gibberellic acid. Regenerated shoots were developed into normal plantlets.
Leaf and petiole explants of kiwifruit were cultured on MT basal medium supplimented with 2,4-D, kinetin, NAA, and BA. Higher organogenic callus formation was observed on the media with NAA + BA than on the media added with 2,4-D + kinetin. Adventitious buds were formed only on media with NAA and BA. Leaf was better explant than petiole. When callus and adventitious buds were subcultured, shoot formation responsed best on medium with 0.1 mg/L NAA + 2.0 mg/L zeatin. When shoots were cultured on medium with 0.5 mg/L IAA + 0.1 mg/L BA after soaking for 1 hr at IBA solution, rooting was more effective than non-IBA treatment. Rooted shoots developed into normal plants.
