Sclerotium Rot of Mungbean (Phaseolus radiatus L.) Caused by Sclerotium rolfsii in South Korea (Sclerotium rolfsii에 의한 녹두 흰비단병)

  • Sclerotium rot was observed on mungbean (Phaseolus radiatus L.) plants cultivated in the exhibition field of Gyeongsangnam-do Agricultural Research and Extension Services in September 2015. The progression of rot was initially observed as water-soaked lesions on several parts of the affected plant. Severely infected plants were blighted and eventually died. White mycelial mats spread over the lesions and numerous sclerotia formed on stems near the soil line. The sclerotia were globoid in shape, 1~3 mm in size, and white to brown in color. The optimum temperature for mycelial growth and sclerotia formation on potato dextrose agar (PDA) was $30^{\circ}C$ and the hyphal width was $4{\sim}8{\mu}m$. Typical clamp connections were observed on the hyphae of fungus grown on PDA. For molecular identification, the complete internal transcribed spacer (ITS) ribosomal DNA (rDNA) of the causal fungus was sequenced and analyzed. Based on the mycological characteristics, ITS rDNA sequence analysis, and pathogenicity to host plants, the fungus was identified as Sclerotium rolfsii. This is the first report of Sclerotium rot on mungbean caused by S. rolfsii in Korea.

Characteristics of Mycelial Growth and Fruit Body Development of White Pleurotus ostreatus 'Miso' (흰색느타리버섯 '미소'의 균사배양 및 자실체 생육 특성)

  • 'Miso', a new variety of oyster mushroom was developed for the bottle culture at the Chungnam Agricultural Research & Extension Services. Its mycelium grew rapidly with $8.4{\sim}8.6\;mm/day $ at $25{\sim}28^{\circ}C$ on PDA medium. The optimum pH of the mycelial growth was pH 5.0. It took 24 days for the primitive primordium formation after inoculation on pine sawdust media mixed with 20% wheat bran. Fruiting body color was white, and the shape of pileus was convex-umbonate. In the bottle culture, the yield was 115.7 g per 850 ml bottle. Stipe length was about 54 mm, the number of stipe per bottle was 18.1, the size of pileus was 28 mm, and gill was crowded. The moisture content of 'Miso' was lower than that of Jangan No.5 and Wonhyeong No.1, and contents of protein, ash and sugar of 'Miso' were higher than those of other varieties, and tannin acid content of 'Miso' was lower than that of others. On the basis of AFLP analysis, the 'Miso' was distinct not only from Wonhyeong No.1, but also from their closest relative, oyster mushrooms.

Occurrence and Genesis of Amphiboles in Hornblende Gabbro in Guwoonri, Hwacheon and in Otanri, Chuncheon (화천 구운리와 춘천 오탄리 일대에 분포하는 각섬석 반려암체내에 존재하는 각섬석류의 산출양상 및 생성작용)

  • Jurassic hornblende gabbo intrusives are distributed in Otanri, Chuncheon, and in Guwoonri, Hwacheon located in the northern part of Gyeonggi Massif. The intrusives composed mainly of amphiboles and plagioclase can be divided into two distinct rocks depending on the shape of amphiboles: (i) subspherical amphibole gabbro which has subspherical amphibole phenocryst as a major mafic phase(Sag); (ii) prismatic amphibole gabbro which has prismatic amphiboles as a principal mafic mineral(Pag). Subspherical amphiboles in Sag have higher Cr content and higher Mg($Mg+Fe^{2+}$) ratio relative to the prismatic amphiboles in Pag. This is indicative of conversion of pyroxene into amphibole with pyroxene pseudomorph. Oxygen isotopic results of plagioclase and amphibole separated from the hornblende gabbro suggest that theses minerals have experienced oxygen isotopic exchange with relatively heavy-$^{18}O$fluid for a long period, and magmatic fluid has been involved in the formation of subspherical amphiboles. Amphiboles in hornblende gabbro are composed of distinct species of pargasite, magnesiohornblende, actinolite, which formed at different stages.

Growth of ZnO Film by an Ultrasonic Pyrolysis (초음파 열분해법를 이용한 ZnO 성장)

  • ZnO was deposited on sapphire single crystal substrate by an ultrasonic pyrolysis of Zinc Acetate Dehydrate (ZAH) with carrying Ar gas. Through Thermogravimetry-Differential Scanning Calorimetry(TG-DSC), zinc acetate dihydrate was identified to be dissolved into ZnO above $380^{\circ}C$. ZnO deposited at $380-700^{\circ}C$ showed polycrystalline structures with ZnO (101) and ZnO (002) diffraction peaks like bulk ZnO in XRD, and from which c-axis strain ${\Sigma}Z=0.2\%$ and compressive biaxial stress$\sigma=-0.907\;GPa$ was obtained for the ZnO deposited $400^{\circ}C$. Scanning electron microscope revealed that microstructures of the ZnO were dependent on the deposition temperature. ZnO grown below temperature $600^{\circ}C$ were aggregate consisting of zinc acetate and ZnO particles shaped with nanoblades. On the other hand the grain of the ZnO deposited at $700^{\circ}C$ showed a distorted hexagonal shape and was composed of many ultrafine ZnO powers of 10-25 nm in size. The formation of these ulrafine nm scale ZnO powers was explained by the model of random nucleation mechanism. The optical property of the ZnO was analyzed by the photoluminescence (PL) measurement.

  • The purpose of this study was to improve the culturability of 'IR 36', a indica type rice cultivar using DNA marker associated with the ability of plant regeneration in anther and seed culture. The culturability of 6 rice cultivars and 2 indica/japonica lines ('MGRI 036', 'MGRI 079') were investigated in anther and seed culture. The culturability of 3 japonica rice cultivars were much higher than tongil and indica rice cultivars, and 'MGRI 036' and 'MGRI 079' has high culturability with 20% regenerability, also. 34 $BC_2F_4$ 4 lines were selected by marker screening using RZ400 among 90 $BC_2F_4$ lines derived from a cross between 'MGRI 079' and 'IR 36'. The frequency of callus formation of 10 $BC_2F_4$ lines were higher than 'IR 36' in anther culture among selected 34 $BC_2F_4$ lines. The ability of plant regeneration of 10 lines were higher than 'IR 36' in the seed culture among selected 34 $BC_2F_4$ lines. A promising line, $BC_2F_4$-28, was selected to have better culturability in the anther and seed culture among selected 34 $BC_2F_4$ lines. The heading date and grain shape of the $BC_2F_4$-28 was similar to 'IR 36'. Using the RZ400 DNA marker associated with the culturability will be useful method for improving of indica rice culticvar's culturability in rice breeding program.

The Characteristics of Biopellet Produced Upon Reactor Configuration in UASB System (UASB 공법에 있어서 반응조의 형상변화에 따른 입상슬러지의 특성에 관한 연구)

  • Physicochemical and morphological characteristics of biopellets produced in "control" and modified UASB reactor were investigated to compare the reactor performance with regard to the hydrogen partial pressure. The characteristics of biopellet produced in modified UASB reactor operated with high hydrogen partial pressure were better than those of "control" reactor operated with relatively lower hydrogen partial pressure, therefore the hydrogen partial pressure effected greatly on the formation and stability of the biopellet. Furthermore, pellets from the UASB system with modified settler showed a better settleability and biomass holding capacity. The chemical composition of biopellet was distinctively different from that of common bacterial formula, $C_5H_7O_2N$. Biopellets was composed the large fraction of nitrogen in comparison with common anaerobic microbes. These results implicated the existence possibility of polypeptide-type extracellular polymer. The morphological characterization with SEM showed that microorganisms observed at surface of biopellet produced in modified UASB reactor operated with high $P_{H_2}$ condition were very similar in shape and size to the Methanobrevibactor arboriphilus-$H_2$ utilizing methanogen. The microorganisms was distinguished from those of "control" reactor operated with low $P_{H_2}$ condition. From these results, it could be explained the hydrogen partial pressure effects on pelletization mechanism.

Mycelial and cultural characteristics of a new high-yield Pleurotus pulmonarius cultivar 'Santari' for bottle culture (다수성 병재배용 산느타리 신품종 '산타리'의 균사배양 및 생육특성)

  • Oyster mushrooms are one of the most popular edible mushrooms in Korea. The 'Santari' cultivar bred in this study, which belongs to the species Pleurotus pulmonarius, is a new oyster mushroom cultivar for bottle culture. It was bred by mating monokaryons isolated from 'GMPO20404' and 'Hosan'. The optimum temperature for 'Santari' mycelial growth was $26-29^{\circ}C$ on PDA medium, and the temperatures for primordium formation and for growth of the fruit body on sawdust medium were $22^{\circ}C$ and $20^{\circ}C$, respectively. It took 34 days to complete the spawn run, 3 days to form primordia, and 3 days to finish fruit body growth in the bottle culture. The fruit body pilei were round in shape and brownish, whereas the stipes were long, thick, and white. The yield per bottle of 'Santari' was 172 g/1,100 mL, which was 43% higher than that of the reference cultivar ('Hosan'). The springiness, cohesiveness, gumminess, and brittleness of the stipe tissue were 87%, 82%, 193 g, and 16 kg, respectively. These physical property values of 'Santari' were lower than those of the control cultivar, except for the cohesiveness.

Study on Formation of FePd Nano-dot Using Agglomeration of Fe/Au Bilayer (Fe/Au 이중층의 응집현상을 이용한 FePd 나노 점 형성에 관한 연구)

  • [ $L1_0$ ]phase FePd nano-dot structures were successfully fabricated on self-organized Fe/Au bilayers. With atomic force microscopy, it is determined that surface morphologies of initially flat Fe/Au bilayer films were agglomerated and transformed their shape into nano-dots structures with increasing annealing temperature. With this bilayer as a template, FePd multilayers were deposited at various temperatures, i.e. $300^{\circ}C$, $350^{\circ}C$, $400^{\circ}C$, and $450^{\circ}C$. Surface morphologies of FePd superlattice had a near resemblance to self-organized bilayer. According to X-ray diffraction results, it is confirmed that $L1_0$ superlattice structures of FePd were obtained from samples which were annealed above $350^{\circ}C$. Results of X-ray photoelectron spectroscopy depth-profile analysis showed that chemical composition is identical to deposition sequence. As a result, without additional etching processes, fabrication of chemically ordered FePd superlattice nano-dots was achieved.

Engineering Geological Implications of Fault Zone in Deep Drill Cores: Microtextural Characterization of Pseudotachylite and Seismic Activity (시추코어 단층대에서의 지질공학적 의미: 슈도타킬라이트의 미세조직의 특징과 지진활동)

  • It is not rare that pseudotachylite, dark colored rock with glassy texture, is recognizable in deep core samples drilled up to 900 m from the surface. Pseudotachylite with widths varying few to 20 cm is sharply contacted or interlayered with the host rocks composed of Jurassic granite and Precambrian amphibolite gneiss, showing moderately ductile deformation or slight folding. Pseudotachylite occurring at varying depths in the deep drill core are slightly different in texture and thickness. There is evidence of fault gouge at shallower depths, although brittle deformation is pervasive in most drill cores and pseudotachylite is identified at random depth intervals. Under scanning electron microscope (SEM), it is evident that the surface of pseudotachylite is characterized by a smooth, glassy matrix even at micrometer scale and there is little residual fragments in the glass matrix except microcrystals of quartz with embayed shape. Such textural evidence strongly supports the idea that the pseudotachylite was generated through the friction melting related to strong seismic events. Based on X-ray diffraction (XRD) quantitative analysis, it consists of primary minerals such as quartz, feldspars, biotite, amphibole and secondary minerals including clay minerals, calcite and glassy materials. Such mineralogical features of fractured materials including pseudotachylite indicate that the fractured zone might form at low temperatures possibly below $300^{\circ}C$, which implies that the seismic activity related to the formation of pseudotachylite took place at shallow depths, possibly at most 10 km. Identification and characterization of pseudotachylite provide insight into a better understanding of the paleoseismic activity of deep grounds and fundamental information on the stability of candidate disposal sites for high-level radioactive waste.

  • Efficient plant regeneration system of birdsfoot trefoil (Lotus corniculatus L.) was development. The factors affecting the somatic embryo formation, its proliferation and regeneration capacity of leaf and stem explants of Empire cultivar was investigated. The highest number of somatic embryos was obtained on B5 medium supplemented with 1 mg/L NAA and 1 mg/L BA. Depending on different explants, highest frequency of embryogenic callus and regeneration were observed in Empire with leaf explants. The response from stem explants was slower and callus induction was less than that from leaf explants. Regenerated shoots formed complete plantlets in on 1/2 MS medium supplemented with 1 mg/L IBA. Regenerated plants were morphologically uniform with normal shape and growth pattern.
