Effect of Yeongyupaedog-san on Cytokine Levels of Mouse Th1/Th2 Cells and Anti-allergic Activity in Ovalbumin-sensitized Allergic Inflammation Model (연교적패독산(連翹敗毒散) 물 추출물(抽出物)의 마우스 Th1/Th2 사이토카인 조절(調節)에 의한 항알레르기 효과)

  • This study was to evaluate the effect of Yeongyupaedog-san (YGPDS) on mouse Thl and Th2 cells' differentiation and ovalbumin (OVA)-induced allergic inflammation. The proliferation of mouse CD4 T cells and the secretion of Th1/Th2 cytokines under the influence of YGPDS extract were measured as well as the amount of ${\beta}-hexosaminidase$ in RBL-2H3 cells and the levels of $TNF-{\alpha}$ and 1L-6 secretion in Raw264.7 cells. BALB/c mice were orally administered with YGPDS extract and simultaneously inoculated with OVA to induce allergic reaction and measure the level of total IgE, OVA-specific IgE and the production of IFN- g, IL-4, IL-5 by the spleen cells. When mouse CD4 T cell were stimulated with anti-CD3 and anti-CD28 for 48 hours in various concentrations of YGPDS extract, it increased proliferation of CD4 cells by 11% in $100\;{\mu}g/^{ml}$ concentration but it showed an inhibition by 37% at $200\;{\mu}g/^{ml}$ CD4 T cells under Th1/Th2 polarizing conditions for 3 days with YGPDS resulted in mild decrease of IFN- g in Thl cells and significant decrease of IL-4 in Th2 cells at $500\;{\mu}g/^{ml}\;and\;100\;{\mu}g/^{ml}$ by 18% and 21%, respectively. YGPDS extract had a dose-dependent inhibitory effect on antigen-induced release of ${\beta}-hexosaminidase$ in RBL-2H3 cells. Treatment of YGPDS extract on LPS stimulated Raw 264.7 cells showed dose-dependent decrease in TNF-n production. Oral administration of YGPDS extract on OVA-induced allergic mice showed an inhibitory effect on the levels of total serum IgE and OVA-specific IgE by 25% and 34% , respectively. Culture of spleen cells with OVA resulted in significant increase of IFN- g by 44% and significant decrease of IL-4 and IL-5 by 56%, and 24%, respectively. The results show that YGPDS does not strongly induce mouse T cells to transform into Thl or Th2 but it has an anti-allergic effect in vitro, and that it also corrects the unbalance between the reactions of Th cells in allergic diseases.

The Effect of Crude Saponins of Korean Red Ginseng against Airway Inflammation and Airway Hyperresponsiveness Induced by Diesel Exhaust Particles in Mice (생쥐에서 디젤배기가스 입자에 의한 기도염증과 기도 과민성에 미치는 홍삼 조사포닌의 영향)

  • The objective of this study was to investigate the effect of crude ginseng total saponins (CGS) against airway inflammation (AI) and airway hyperresponsiveness (AH) induced by diesel exhaust particles (DEP) in mice. AI and AH were induced by the intratracheal instillation with 0.1 $mg/m{\ell}$ of DEP suspension once a week for 10 weeks combined with ovalbumin (OVA) sensitization. Mice were also treated orally with 75 $mg/m{\ell}$ of CGS, 5 days a week for 10 weeks. Oral CGS treatment decreased in the level of serum immunoglobulin (IgE) and histamine increased by DEP and OVA, and declined respiratory resistance. It also dropped an enhanced infiltration of eosinophils in the bronchoalveolar lavage fluid (BALF) of mice, and an increased T helper type 2 cell derived cytokine levels such as of interleukin (IL)-4, IL-13 and IL-5 in the BALF. However, it did not influence T helper type 1 cytokine such as interferon-gamma in the BALF. These results indicate that CGS may alleviate allergen-related AI and AH in mice and may play an important role in the modulation of asthmatic inflammation.

  • Sopungsan (SS) is a traditional Korean decoction used for the treatment of dermatitis. The aim of this study is to confirm whether or not SS has a preventive effect on the development of atopic dermatitis in dinitrochlorobenzene-applied Nc/Nga mice. SS was administered orally to Nc/Nga mice, which led to the remarkable suppression of the development of dermatitis, as determined by a histological examination and the serum IgE levels. Moreover, SS inhibited the production of thymus- and activation-regulated chemokine (TARC) and its mRNA expression in a keratinocyte cell line, HaCaT, which had been stimulated with tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interferon-${\gamma}$ (IFN-${\gamma}$). Activation of the nuclear factor-${\kappa}B$ (NF-${\kappa}B$) or activator protein-1 (AP-1) is one of key steps in the signaling pathways mediating induction of TARC. In this study, SS selectively suppressed NF-${\kappa}B$ activation which may be essential for TARC expression in $TNF-{\alpha}/IFN-{\gamma}$ treated keratinocytes. The inhibitory effect of SS on NF-${\kappa}B$ activation and TARC production might be associated with the anti-dermatitic effects of SS.

Effect of Brine Mineral Water on TMA-Induced Contact Hypersensitivity Reaction in the Mouse Model (TMA로 유도된 접촉성 과민 반응 마우스 질환모델에서 해양성 광천수 도포시의 치료효과)

  • Effect of brine mineral water(BMW) on contact hypersensitivity reaction(CHR) was estimated using trimellitic anhydride (TMA) induced CHR in the mouse. BMW exhibited potent inhibitory activity on TMA induced CHR. BMW treatment suppressed the ear swelling, and attenuated hyper-activated lymph nodes stimulated by TMA challenge, thereby reduced their weight. The immunological index was analyzed after BMW administration in CHR. The level of serum IgE was significantly down regulated after BMW treatment. Furthermore, BMW showed inhibitory property of cytokine production. BMW treatment suppressed not only Th2 type cytokine, IL-4 but also pro-inflammatory cytokines such as TNF-${\alpha}$ and IL-6. From the histological analysis, the inflammatory context appeared by atopic dermatitis lesion after challenging with TMA are diminished by BMW treatment. Our results suggest that BMW may attenuate the development of CHR, and can be available for functional ingredient.

Topical Application of S1P2 Antagonist JTE-013 Attenuates 2,4-Dinitrochlorobenzene-Induced Atopic Dermatitis in Mice

  • Sphingosine-1-phosphate (S1P) and its receptors have been implicated in atopic dermatitis. S1P2 was found to function as a proallergic receptor, while its antagonist JTE-013 was found to suppress allergic asthma in mice. Topical application of JTE-013 has not been investigated in an in vivo model of atopic dermatitis. Therefore, the therapeutic potential of JTE-013 topical application was evaluated by the use of a 2,4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis mouse model. DNCB-induced inflammation and mast cell accumulation in skin tissues were significantly suppressed by topical JTE-013 treatment in BALB/c mice. DNCB-induced increase of lymph nodes sizes and elevated inflammatory cytokines (IL-4, IL-13, IL-17, and IFN-γ) in lymph nodes were also significantly reduced by the JTE-013 treatment. Elevated serum levels of IgE were significantly suppressed by the topical treatment of JTE-013. In summary, the topical treatment of JTE-013 S1P2 antagonist suppressed DNCB-induced atopic dermatitis symptoms and immune responses. These results suggested JTE-013 as a potential therapeutic agent for atopic dermatitis.

Effects of Hanghaedan on the Cytokine Secretory Function of Monocytes of Atopic Dermatitis Patients (항해단(沆瀣丹)이 아토피 피부염 환자 단핵세포의 Cytokine 분비능에 미치는 영향)

  • Objetives The late research tendency on atopic dermatitis is mainly focused on the experiment of unbalance of cytokine released from T cell subtype. Merhods As there's no experiment with herbal medicine using Hanghaedan(HHD), we isolated and cultured monocytes of atopic dermatitis pateints (AD) peripheral blood, for knowing the influence of HHD on the secretory function of IL-4, IL-5, $IFN-{\gamma}$. Results 1. In AD group, HHD exhibits statistically significant inhibitory effect on IL-4(p<0.05). 2. In AD group, HHD exhibits statistically significant inhibitory effect on IL-5(p<0.05). 3. In AD group, HHD exhibits statistically significant inhibitory effect on $IFN-{\gamma}$ (p<0.01). 4. Sample group comparing with the cytokine concentration rate value of AD patients and that of NAD patients IL-4 of AD patients exhibits statistically significant decrease(p<0.05). 5. In AD group, there is no significant relation between serum IgE and cytokines. 6. In AD group, there is no significant relation between total eosinophil and IL-5. Conclusions Effects of Hanghaedan on the cytokine(IL-4, IL-5, $IFN-{\gamma}$) secretory function of monocytes of atopic dermatitis patients is recognized.

A study on the alteration of general characteristics and therapeutic effect of GamiYeoldahansotang(加味熱多寒少湯) in patients with Atopic Dermatitis (가미열다한소탕(加味熱多寒少湯) 투여후 아토피 피부염 환자의 임상상 변화에 대한 연구)

  • Background : Atopic Dermatitis is thought to be a common and chronic relapsing inflammatory skin disease that probably results from allergic reaction. Because it make some serious problems in children, it is needed to treat and at least subside symptoms. Objective : The aim of this study was to evaluate the efficacy of GamiYeoldahansotang(加味熱多寒少湯) for treating Atopic Dermatitis and survey the general characteristics in children with Atopic Dermatitis. Method : Atopic Dermatitis with typical clinical symptoms were included in this study. Forty two patients were treated with hot water extract of GamiYeoldahansotang(加味熱多寒少湯) for four weeks. Clinical evaluation were made by Jacob T scoring system before and after treatment. Total Serum IgE, Eosinophil count were also conducted. Results : Reduction in body surface area was not observed. But significant reductions of severity scores before and after adminstration of GamiYeoldahansotang(加味熱多寒少湯) were observed in all of three groups: mind, moderate, severe. specially severe intensity group was very statistically significant.(p<0.05) Conclusion : We speculate that GamiYeoldahansotang(加味熱多寒少湯) has some therapeutic effects in mitigating the symptoms of Atopic Dermatitis.

Zanthoxylum Piperitum Attenuates the Allergic Inflammation in vivo and in vitro

  • Natural products have received revived interest via traditional remedies or alternative medicine used for the treatment of various diseases. Zanthoxylum piperitum (ZP) has been utilized in traditional medicine for various medicinal purposes. The present study was conducted to evaluate whether ZP modulates allergic inflammation both in vivo and in vitro. We examined the pharmacological effects of ZP on 2, 4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis (AD) symptoms in mice. Additionally, in order to clarify the anti-inflammatory mechanisms of ZP, we elucidate the effect of ZP on the expression levels of inflammatory cytokines and nuclear factor-κB (NF-κB) in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated human mast cells (HMC-1). The results demonstrated that ZP attenuated AD clinical symptoms such as erythema, edema and dryness as well as histamine and IgE serum levels in DNCB-induced AD model mice. Additionally, ZP suppressed the expression of inflammatory cytokines and activation of NF-κB in AD-like skin lesions and stimulated HMC-1. These results provide experimental evidence that ZP may be useful candidate for treating allergic inflammation including AD.

Kimura Disease Misdiagnosed as Chronic Submandibular Sialadenitis : A Case Report (만성 악하선염으로 오인된 Kimura씨 병 1례)

  • Kimura disease is an uncommon chronic inflammatory disorder of unknown etiology. Clinically, patients present nontender subcutaneous swelling in the head and neck region. Peripheral eosinophilia, an elevated serum IgE are also present. The clinical course of Kimura disease is often progressive, and the main problem with treatment is disease recurrence. Treatment options in the recurrent cases range from observation to surgical excision, steroid therapy, and radiotherapy. We report a case of recurrent Kimura disease, initially thought to be chronic submandibular sialadenitis, along with the appropriate review.

Inhibitory Effects on the Type I hypersensitivity and Inflammatory Reaction of Sogunjung-tang (소건중탕이 알레르기에 미치는 영향)

  • Sogunjung-tang(SGJT) has been used for the treatment of general weakness, digestive organ disease and so on. This study was carried out for the purpose of knowing the inhibitory effect on the Type I hypersensitivity and Inflammatory reactions of SGJT. The reasults were obtained as follows: SGJT(0.1, 0.5, 1, 2mg/g) concentration of dependently inhibited compound48/80 induced anaphylaxis reaction in mice. SGJT(2mg/g) also inhibited permeability of evans blue into peritoneal cavity in mice. SGJT reduced IgE, CRP, WBC and Platelets on egg albumin induced hypersensitivity. but serum NO was grown. According to above results, SGJT may be beneficial in the type I hypersensitivity and Inflammatory reactions by inhibition of histamine release from mast cells.
