  • A sequential-clustered integrator based on GEAR method is developed for the purpose of dynamic simulation of chemical processes. And a single simulator structure capable of employing various integration approaches is designed and its efficiency and flexiblity is evaluated. Sequential integration method is superior to simultaneous method for the process without recycle, but simultaneous method is very powerful for the coupled process with recycle.

Simultaneous and Sequential Integration by Cre/loxP Site-Specific Recombination in Saccharomyces cerevisiae

  • A Cre/loxP-${\delta}$-integration system was developed to allow sequential and simultaneous integration of a multiple gene expression cassette in Saccharomyces cerevisiae. To allow repeated integrations, the reusable Candida glabrata MARKER (CgMARKER) carrying loxP sequences was used, and the integrated CgMARKER was efficiently removed by inducing Cre recombinase. The XYLP and XYLB genes encoding endoxylanase and ${\beta}$-xylosidase, respectively, were used as model genes for xylan metabolism in this system, and the copy number of these genes was increased to 15.8 and 16.9 copies/cell, respectively, by repeated integration. This integration system is a promising approach for the easy construction of yeast strains with enhanced metabolic pathways through multicopy gene expression.

  • This study describes process and method to establish concurrent and/or sequential schedule planning for the system design process of the EIA632 standard. For this purpose, the study suggest Process and method that are context analysis of standard process, interface definition from activity decomposition, integration of related activities, and definition of concurrent and/or sequential work flow. The proposed process and method will contribute to minimize time loss that is emerged from activities iteration.

  • This study proposes a new two-level condensation scheme for the construction of a reduced system. In the first step, the candidate area is selected for the construction of the reduced system by energy estimation in element-level. In the second step, primary degrees of freedom are selected by sequential elimination from the candidate degrees of freedom linked to the selected elements. Numerical examples demonstrate that the proposed method saves the computational cost effectively and provides a reduced system which predicts the eigenvalues accurately. Moreover, the well-constructed reduced system can present the reliable behavior of the structure under arbitrary dynamic loads comparing to that of global system. Time integration in a reduced system can save the computing time remarkably. Through a few numerical examples, the efficiency and reliability of the proposed scheme are verified.

Enhancement of β-1,3-Glucanase Activity by Sequential δ-Sequence Mediated Integration in Saccharomyces cerevisiae (출아효모에서 연속적 δ-sequence 삽입유도에 의한 β-1,3-glucanase 활성 증가)

  • Beta-1,3-glucanase is widely used in various biotechnological and industrial processes, with over-production required to enable versatile utilization. We examined the overexpression of ${\beta}$-1,3-glucanase (EXGA) from Aspergillus oryzae using ${\delta}$-sequence-mediated integration. We constructed $pRS{\delta}$-exgA and $pRS{\delta}K$-exgA plasmids for integration of the EXGA gene into various chromosomes of Saccharomyces cerevisiae. These plasmids contain the ADH1 promoter for constitutive expression, a signal sequence (exoinulinase signal sequence [INU1 s.s]) for secretory production, and a ${\delta}$-sequence for integration of ${\beta}$-1,3-glucanase. The $pRS{\delta}$-exgA plasmid was transformed into the S. cerevisiae $BY4742{\Delta}exg1$ strain, and ${\beta}$-1.3-glucanase was stably overexpressed and secreted. Another plasmid, $pRS{\delta}K$-exgA, was introduced into the S. cerevisiae $BY4742{\Delta}exg1$ (YKY082) strain, and overexpression of ${\beta}$-1,3-glucanase was examined by inducible integration under geneticin selection. The activity of ${\beta}$-1,3-glucanase increased in accordance with a rise in the geneticin concentration, with 0.8 mg/ml of geneticin suitable for overexpression of ${\beta}$-1,3-glucanase. Subsequently, $pRS{\delta}K$-exgA was repeatedly transformed for sequential ${\delta}$-integration. The activity of ${\beta}$-1,3-glucanase reached about 0.063 unit/ml/$OD_{600}$, 0.095 unit/ml/$OD_{600}$, 0.131 unit/ml/$OD_{600}$ and 0.165 unit/ml/$OD_{600}$ by the first, second, third, and fourth round of integration, respectively. According to the increase in the activity of ${\beta}$-1,3-glucanase by sequential ${\delta}$-integration, the copy number (integration rate) of the EXGA gene also increased in various chromosomes. These results suggest that recombinant ${\beta}$-1,3-glucanase activity can be sequentially increased by repeated ${\delta}$-sequence integration.

  • A sequential coupled field analysis of electromagnetic free bulging was performed by using FEM. A 2D axi-symmetric electromagnetic model based on the magnetic vector potential is proposed for the calculation of magnetic field and Lorentz's forces. The Newmark integration method is used to calculate the transient dynamic plastic deformation of sheet during free bulging. In the finite element model, the effect of sheet deformation on the electromagnetic field analysis is taken into consideration. In order to confirm the sequential electromagnetic-mechanical coupling analysis, an experiment with an electromagnetic forming apparatus was conducted. The results showed that the final bulge height of the sheet predicted from the proposed method is in good agreement with experimentally measured height.

Comparison of Methods for Stable Simultaneous Expression of Various Heterologous Genes in Saccharomyces cerevisiae (출아효모에서 다양한 이종 유전자의 안정적 동시발현을 위한 방법의 비교)

  • We compared two integration systems for stable expression of heterologous genes in Saccharomyces cerevisiae. A Candida glabrata-derived gene was used as the selective marker for the Cre/loxP system, and XYLP, XYLB, GRE3, and XYL2 genes were used as model heterologous genes and ligated into the universal pRS-CMT vector. The resulting pRS-XylP, pRS-XylB, pRS-Gre3, and pRS-Xyl2 plasmids were sequentially integrated into yeast chromosome VII by four integration processes (marker rescue and gene integration). The four introduced genes were successfully expressed. Further, the pRS-PBG2 plasmid harboring expression cassettes for the four genes was constructed for one-step integration. The four genes that were introduced were stably maintained as a gene cluster and were simultaneously expressed. The one-step integration was more effective for the simultaneous integration and expression of the four genes related to xylan/xylose metabolism. This method will enable the generation of a useful biosystem through appropriate use of gene integration methods.

Is the Peak-Affect Important in Fast Processing of Visual Images in Printed Ads?: A Comparative Study on the Affect Integration Theories

  • This study investigates how affects elicited by visual images in print ads are integrated to form a liking for the ads. Assuming a sequential rather than simultaneous processing of still-cut images, we adopt the 'think-aloud' method to capture consumers' spontaneous responses to visual images. We hypothesize that not only would consumers show mixed affects toward a still-cut visual image but that they would also integrate their serial affects heuristically rather than simply averaging the affects as suggested by the compensatory hypothesis. By comparing the effects of two contradictory affect integration hypotheses (i.e., peak-affect and mood-maintenance) with compensatory integration, using a single regression model, we found that peak-negative along with mood maintenance integration of serial affects for a print ad works best in the formation of ad liking. The results also support our initial premise that people can have mixed valence even toward a still-cut ad.

Performance Analysis of Ranging Techniques for the KPLO Mission

  • In this study, the performance of ranging techniques for the Korea Pathfinder Lunar Orbiter (KPLO) space communication system is investigated. KPLO is the first lunar mission of Korea, and pseudo-noise (PN) ranging will be used to support the mission along with sequential ranging. We compared the performance of both ranging techniques using the criteria of accuracy, acquisition probability, and measurement time. First, we investigated the end-to-end accuracy error of a ranging technique incorporating all sources of errors such as from ground stations and the spacecraft communication system. This study demonstrates that increasing the clock frequency of the ranging system is not required when the dominant factor of accuracy error is independent of the thermal noise of the ranging technique being used in the system. Based on the understanding of ranging accuracy, the measurement time of PN and sequential ranging are further investigated and compared, while both techniques satisfied the accuracy and acquisition requirements. We demonstrated that PN ranging performed better than sequential ranging in the signal-to-noise ratio (SNR) regime where KPLO will be operating, and we found that the T2B (weighted-voting balanced Tausworthe, voting v = 2) code is the best choice among the PN codes available for the KPLO mission.
