Isolation and Identification of Rumen Fungus and Its Produced Conjugated Linoleic Acid (반추위에서 분리한 곰팡이의 Conjugated Linoleic Acid 생산과 ITS-1 영역의 염기서열 해석)

  • The aim of this study was to isolate and identify men fungus which produces conjugated linoleic acid. IS-13 fungus hydrogenated conjugated linoleic acid and trans-11 vaccenic acid within 12 hr after addition of linoleic acid. The homology of IS-13 rumen fungus was compared with internal transcribed spacer 1 region (ITS1)sequences of twenty three men fungi. The length of ITS1 region of IS-13 isolate was 218 bp. IS-13 isolate has the most similar sequence (98% matched) with Orpinomyces species according to maximum-likehood and distance matrix results. The result supported that IS-13 isolate belonged to Orpinomyces genus.

Comparison of Measurement Methods for Head-related Transfer Function(HRTF) (머리전달함수 측정법의 실험적 비교)

  • Three methods(the stepped sine method, the statistical method(random excitation method) and the maximum-length sequence(MLS) method) for head-related transfer functions(HRTFs) are experimentally compared in view point of accuracy and efficiency. First, the stepped sine method has high signal-to-noise ratio, but low efficiency. Second, the statistical method is fast measurement speed, but weak to noise than the other methods. Finally, the MLS method shows both good efficiency and high signal-to-noise ratio, but it needs additional software or equipment such as MLS signal generator. For comparison of measurement accuracy, HRTFs of KEMAR dummy are measured for various azimuths and elevations. Error norms for magnitude and phase of HRTFs are defined and calculated for the measured HRTFs. The calculated error norms show that the methods give similar results in magnitude and phase except a little phase difference in the MLS method.

Molecular cloning and expression analysis of a ferritin H subunit from rock bream, Oplegnathus fasciatus (돌돔 ferritin H 유전자의 클로닝과 발현 분석)

  • Ferritin is an evolutionarily conserved protein that plays an important role in iron storage and detoxification. In this study, the gene encoding a ferritin H subunit homologue (RbFH) was cloned from rock bream (Oplegnathus fasciatus) and analyzed at the expression. The full-length ferritin H cDNA was 1162 bp long and contained an open reading frame (ORF) of 531 bp that encoded 177 amino acid residues with a predicted molecular mass of 20.8 kDa. The 5' UTR was 297 bp in length, and the 3' UTR 298 bp, and preceded by a 5'-untranslated region that contains a putative Iron Regulatory Element (IRE). The deduced amino acid sequence of RbFH shares extensive sequence identities with the H ferritins of a number of fish species and contains the ferroxidase center that is preserved in ferritin H subunits. Examination of tissue specific expression indicated that RbFH expression was most abundant in PBLs, RBC, liver and muscle.

Prediction of DO Concentration in Nakdong River Estuary through Case Study Based on Long Short Term Memory Model (Long Short Term Memory 모델 기반 Case Study를 통한 낙동강 하구역의 용존산소농도 예측)

  • In this study, we carried out case study to predict dissolved oxygen (DO) concentration of Nakdong river estuary with LSTM model. we aimed to figure out a optimal model condition and appropriate predictor for prediction in dissolved oxygen concentration with model parameter and predictor as cases. Model parameter case study results showed that Epoch = 300 and Sequence length = 1 showed higher accuracy than other conditions. In predictor case study, it was highest accuracy where DO and Temperature were used as a predictor, it was caused by high correlation between DO concentration and Temperature. From above results, we figured out an appropriate model condition and predictor for prediction in DO concentration of Nakdong river estuary.

Quantitative Trait Loci for Stem Length in Soybean Using a Microsatellite Markers (콩에서 Microsatellite 마커를 이용한 양적형질 유전자의 분석)

  • Identification of individual quantitative trait loci (QTL) is a prerequisite to application of marker-assisted selection for stern length. Two simple sequence repeat (SSR)-based linkage maps were constructed from recombination inbred line populations between cross of Keunolkong and Shinpaldalkong. Two parents used differed greatly in stem length, which were 30.57 cm and 49.75 cm in Keunolkong and Shinpaldalkong, respectively. Using the constructed maps, regression analysis and interval mapping were performed to identify QTLs conferring stem length. Four QTLs for stem length on linkage groups (LG) F, J, N and O were identified in the Keunolkong ${\times}$ Shinpaldalkong population and they totally explained 37.83% of variation for stem length. In the population, two major QTLs on LG J and O conditioning 14.25% and 10.68% of the phenotypic variation in stem length were determined and two QTLs with minor effect were detected on LG F and N. Identification of QTLs for stem length and mapping individual locus should facilitate to describe genetic mechanisms for stem length in different population. SSR markers tightly linked to QTLs for stem length allow to accelerate the elimination of deleterious genes and selection for desirable recombinants at early stage in crop breeding programs.

Cloning and Expression of the Duck Leptin Gene and the Effect of Leptin on Food Intake and Fatty Deposition in Mice

  • Leptin is the adipocyte-specific product of the obese gene and plays a major role in food intake and energy metabolism. Leptin research was mainly focused on mammalian species, but understanding of leptin and its function in poultry is very poor. In this study, the duck leptin gene was amplified using the reverse transcription-polymerase chain reaction (RT-PCR) from duck liver RNA. The cDNA fragment was inserted into the pET-28a expression vector, and the resulting plasmid was expressed in Escherichia coli BL21 (DE3). Experimental mice were given an intraperitoneal injection of 10 mg/kg leptin dissolved in phosphate buffered saline (PBS), while the control mice were injected with PBS. The effect of leptin on food intake, body weight and fatty deposition in mice was detected. Sequence analysis revealed that duck leptin had a length of 438 nucleotides which encoded a peptide with 146 amino acid residues. The sequence shares highly homology to other animals. The coding sequence of duck leptin was 84 and 86% identical to human and pig leptin nucleotides sequence. Highest identity was with the rat coding sequence (95%). The identity of the amino acid sequence was 84, 82 and 96% respectively compared to that of the human, pig and rat. Results of SDS-PAGE analysis indicated that a fusion protein was specifically expressed in E. coli BL21 (DE3). The purified product was found to be biologically active during tests. Continuous administration of recombinant duck leptin inhibited food intake. Despite the decrease of food intake, leptin significantly induced body weight and fatty deposition. These changes were accompanied by a significant down-secretion of plasma glucose, cholesterol, triglyceride and insulin levels in mice. The observations provide evidence for an inhibitory effect of leptin in the regulation of food intake and for a potential role of duck leptin in the regulation of lipogenesis.

Mining and analysis of microsatellites in human coronavirus genomes using the in-house built Java pipeline

  • Microsatellites or simple sequence repeats are motifs of 1 to 6 nucleotides in length present in both coding and non-coding regions of DNA. These are found widely distributed in the whole genome of prokaryotes, eukaryotes, bacteria, and viruses and are used as molecular markers in studying DNA variations, gene regulation, genetic diversity and evolutionary studies, etc. However, in vitro microsatellite identification proves to be time-consuming and expensive. Therefore, the present research has been focused on using an in-house built java pipeline to identify, analyse, design primers and find related statistics of perfect and compound microsatellites in the seven complete genome sequences of coronavirus, including the genome of coronavirus disease 2019, where the host is Homo sapiens. Based on search criteria among seven genomic sequences, it was revealed that the total number of perfect simple sequence repeats (SSRs) found to be in the range of 76 to 118 and compound SSRs from 01 to10, thus reflecting the low conversion of perfect simple sequence to compound repeats. Furthermore, the incidence of SSRs was insignificant but positively correlated with genome size (R2 = 0.45, p > 0.05), with simple sequence repeats relative abundance (R2 = 0.18, p > 0.05) and relative density (R2 = 0.23, p > 0.05). Dinucleotide repeats were the most abundant in the coding region of the genome, followed by tri, mono, and tetra. This comparative study would help us understand the evolutionary relationship, genetic diversity, and hypervariability in minimal time and cost.

Spirometra decipiens (Cestoda: Diphyllobothriidae) Collected in A Heavily Infected Stray Cat from the Republic of Korea

  • Morphological and molecular characteristics of spirometrid tapeworms, Spirometra decipiens, were studied, which were recovered from a heavily infected stray cat road-killed in Eumseong-gun, Chungcheongbuk-do (Province), the Republic of Korea (=Korea). A total of 134 scolices and many broken immature and mature proglottids of Spirometra tapeworms were collected from the small intestine of the cat. Morphological observations were based on 116 specimens. The scolex was 22.8-32.6 mm (27.4 mm in average) in length and small spoon-shape with 2 distinct bothria. The uterus was coiled 3-4 times, the end of the uterus was ball-shaped, and the vaginal aperture shaped as a crescent moon was closer to the cirrus aperture than to the uterine aperture. PCR amplification and direct sequencing of the cox1 target fragment (377 bp in length and corresponding to positions 769-1,146 bp of the cox1 gene) were performed using total genomic DNA extracted from 134 specimens. The cox1 sequences (377 bp) of the specimens showed 99.0% similarity to the reference sequence of S. decipiens and 89.3% similarity to the reference sequence of S. erinaceieuropaei. In the present study, we report a stray cat heavily infected with S. decipiens identified by mitochondrial cox1 sequence analysis and morphological examinations of the adult worms.

An Experimental Study on the Strength of Composite-to-Aluminum Hybrid Single-Lap Joints (복합재-알루미늄 단일겹침 하이브리드 체결부 강도 특성 실험 연구)

  • Strength and failure of composite-to-aluminum rivetted, bonded, and rivet/bonding hybrid single-lap joints were investigated by experiment. A total of 82 joint specimens were tested with 3 different overlap lengths and 2 types of stacking sequence. FM73m adhesive film and NAS9308-4-03 rivet were used for hybrid joints. While failure loads of the bonded and hybrid joints increased as the overlap length increased, failure loads of the rivetted joints were not affected by the overlap length. Effect of the stacking sequence was not remarkable in the simple bonded or rivetted joints. Failure loads of the hybrid joints, however, showed the maximum of 30% difference depending on the stacking sequence. Major failure mode of the bonded and hybrid joints was the delamination of the composite adherend and failure mode of riveted joints was the rivet failure with local bearing.

Design of T-N2SCD Detection Model based on Time Window (타임 윈도우 기반의 T-N2SCD 탐지 모델 구현)

  • An intrusion detection technique based on host consider system call sequence or system call arguments. These two ways are suitable when system call sequence or order and length of system call arguments are out of order. However, there are two disadvantages which a false positive rate and a false negative rate are high. In this paper we propose the T-N2SCD detection model based on Time Window in order to reduce false positive rate and false negative rate. Data for using this experiment is provided from DARPA. As experimental results, the proposed model showed that the false positive rate and the false negative rate are lowest at an interval of 1000ms than at different intervals.
