A lipid-enriched strain of Botryococcus braunii (UTEX 572) was cultivated in a semi-batch aeration tank to enhance biomass as well as to develop intracellular lipids and fatty acids. A 30 day period of incubation produced 1.39 g/L of biomass and 0.31 g/L of total lipids in the biomass. The grown biomass was pre-treated using several methods to extract the total lipid content efficiently: ultrasonication was found to yield the highest percentage of lipids-namely 19.8% per biomass. Direct heating of biomass in an autoclave also showed better performance than when using only conventional solvent extraction. To enhance the biomass harvest and lipid extraction efficiency, coagulation and flocculation steps were added to the extraction process. It is noteworthy that not only the solvent type but also the solvent/biomass ratio greatly affected efficiency. In addition, the moisture content of the harvested(wet) biomass affected the efficiency significantly. This study elucidated the need for future research on optimizing this extraction process.
The change in the electronic absorption and emission energy of 7-aminocoumarin derivatives in binary solvent mixtures has been studied. The electronic transition energy along with the Stokes' shift is correlated with the orientation polarizability of the solvent as well as the empirical solvent polarity parameters $E_T$ (30). It is observed that the emission peak shift traces the change of $E_T$ (30) value very closely in binary solvent mixtures. The emission transition more strongly depends on the solvent polarity than the absorption, which indicates the dipole moment gets larger on excitation. From the dependence of the Stokes’ shift of 7-aminocoumarins with the solvent polarity parameters and the ground state dipole moment obtained by the semi-empirical calculations, the excited state dipole moment was estimated. The fluorescence lifetime change of 7-aminocoumarins in binary solvent mixtures was measured and the results are explained in terms of molecular conformation and solvent polarity. The study indicates the empirical solvent polarity $E_T$ (30) is a good measure of microscopic solvent polarity and it probes in general the non-specific solvent interactions.
When extracting semi-volatile components of herbal medicines using hot water vapor, some substances may react with water vapor or oxygen, and some volatile substances may be lost, when using an organic solvent extraction method has the disadvantage that it may contain a non-volatile material and residual organic solvent. In addition, it is inefficient to separate semi-volatile substances from herbal medicines into each single component and conduct biological activity research for each component to determine the effective ingredient, and some components may be lost in the separation process. In this study, semi-volatile substances evaporated under two pressure-reduced conditions in Chinese herbal medicines such as Cnidii Rhizoma, Aucklandiae Radix and Amomum Fructus were separated by cooling with liquid nitrogen. Those were analyzed by gas chromatography-mass spectrometry (GC-MS) to identify the components, and this method may be used to study biological activities at the cellular level. The substances separated under reduced pressure, essential oil obtained by simultaneous distillation extraction (SDE) method and substances by using solid phase micro-extraction (SPME) from Cnidii Rhizoma, Aucklandiae Radix and Amomum Fructus were analyzed by GC-MS. In the case of Cnidii Rhizoma and Aucklandiae Radix, there were some differences among the essential oil components obtained by SDE and those identified by low temperature capture (CT) and SPME method, these were believed to be produced by some volatiles reacting with water or oxygen at the boiling point temperature of water.
The huge amount of solvents used in the semi-conductor and display industry typically result in waste of valuable solvents which often form complex azeotropic mixtures. This study explored a recovery process of a quaternary waste solvent, comprising methyl 2-hydroxybutyrate, propylene glycol monomethyl ether acetate, ethyl lactate, and ethyl-3-ethoxy propionate. In this study, a novel shortcut column method with a graphical approach was exploited for the distillation column design of complex quaternary azeotropic mixtures. As a result, the proposed shortcut method and design procedure solved the complex separation paths successfully with less computational efforts while achieving all requirements for component purity.
In this study, high school science teachers' and students' various conceptions related to osmosis phenomena were compared with and analyzed in relation to the content of science textbooks used in high school science classrooms and college science courses. The questionnaires developed by the researchers were administered to science teachers and students. Differences can be found between the explanations of science textbooks on osmotic pressure and semi-permeable membranes. Many science teachers and students thought of osmotic pressure as 'membrane pressure occurred by the movement of a solvent'. Moreover, the types of teachers of semi-permeable membranes were similar regardless of their academic majors. Many of the teachers thought of a semi-permeable membrane as a membrane that 'passes small-size particles'; however, many students thought of this type of membrane as being 'selectively permeable'. Also, the salt-pickling cabbage phenomenon seemed to cause significant confusion to science teachers and students. These study results show that teachers and students possess various conceptions related to the osmosis phenomena. These different conceptions related to osmosis phenomena might cause confusion and diverse conceptions including misconceptions among teachers and students.
Three kind of ASE (Accelerated Solvent Extraction) and SPMDs (Semi-Permeable Membrane Devices) combined methods (ASE-SPMDs, ASE-accelerated SPMDs and SPMDs without extraction) and general Soxhlet-GPC were compared each other for the analysis of PCDD/FS in sediment. The average recovery rate of three types ASE and SPMDs combined methods (108.1%) were higher than that of the Soxhlet-GPC (79.5%) for three samples in each method using surrogate internal standards. The average coefficient of variation (10%, $2.1{\sim}25.2%$) for each congener of PCDD/Fs shows the reasonable results. Total PCDD/Fs concentrations after SPMDs without extraction were quite low, but those after ASE-SPMDs and ASE-accelerated SPMDs methods were close to the Soxhlet-GPC. Thus, the ASE-SPMDs and ASE-accelerated SPMDs methods are considered as the excellent pre-treatments method because they need less solvent and time without quality degradation.
The oil and concentrated protein powder from squid viscera was extracted and recovered by a semi-batch supercritical carbon dioxide ($SCO_2$) extraction system and the degree of oxidation in the extracted oil was measured in order to compare with extracted oils using organic solvents. The degree of storage in treated squid viscera by $SCO_2$ extraction was measured in order to compare with untreated squid viscera. As results obtained, it was found that the auto-oxidation of the oils using $SCO_2$ extraction occurred very slowly compared to the oils by organic solvent extraction. And the treated squid viscera by $SCO_2$ extraction was reached the point of initial rottenness slowly than untreated squid viscera.
Initial burst is one of the major challenges in protein-encapsulated microparticle systems. Since protein release during the initial stage depends mostly on the diffusional escape of the protein, major approaches to prevent the initial burst have focused on efficient encapsulation of the protein within the microparticles. For this reason, control of encapsulation efficiency and the extent of initial burst are based on common formulation parameters. The present article provides a literature review of the formulation parameters that are known to influence the two properties in the emulsion-solvent evaporation/extraction method. Physical and chemical properties of encapsulating polymers, solvent systems, polymer-drug interactions, and properties of the continuous phase are some of the influential variables. Most parameters affect encapsulation efficiency and initial burst by modifying solidification rate of the dispersed phase. In order to prevent many unfavorable events such as pore formation, drug loss, and drug migration that occur while the dispersed phase is in the semi-solid state, it is important to understand and optimize these variables.
