Influence of Abiotic Treatments on Isoflavone Accumulation in Soybean Seeds during Germination (물리적 요인이 콩 발아 중 isoflavone 함량에 미치는 영향)

  • Isoflavones, a group of secondary metabolites present in plants and especially abundant in soybeans, are garnering increasing interest because of their medicinal properties. Numerous environmental factors influence isoflavone accumulation in plants. Successful attempts to increase isoflavone contents in soybean seeds have not been widely reported. In this report, we examined an effective method for producing a high amount of isoflavones during soybean seed germination, by treating the seed with 4 abiotic factors. Soybean seeds were exposed to ultrasound (50 Hz) and heat ($40^{\circ}C$) for 30minutes, then germinated for 5 days in darkness. Another sample was subjected to water deficit during germination in darkness, and the other sample was subjected to light conditions during the germination period. The results showed that the ultrasound treatment increased isoflavone content during soybean germination. Three days after germination, the ultrasound-treated seeds presented the highest isoflavone content (6,042.1 ug/g dry weight) compared to the other treatments, which was also 1.3-fold increases over the content of the raw seed's before germination. With respect to each type of isoflavone group, aglycone (partially genistein) content was markedly higher, whereas glucoside was lower than that of the untreated samples. In other experiment, total isoflavone content after heat, water deficit, and light treatments was 5,600.0, 4,740.1, and 5,631.4 ug/g, respectively. These present work suggests that ultrasound treatment is a novel approach for improving the production of isoflavones in soybean seeds during germination.

Meaning of Basic Geometry Patterns to Ancient Koreans and Its Classification (고대 한국인이 선호한 기본도형의 의미와 유형)

  • The purposes of this study are to identify the meaning of the geometrical patterns preferred by ancient Korean peoples and to classify them into some groups by their similarity. We investigated various patterns found on clothing and relics from GoJoseon to Goguryeo period, and utilized secondary sources such as history articles, Internet materials and photo and analyzed the associations of the varied patterns found in pottery, handicrafts, and clothing with the ancient cultures. We found the letters (ㅇ, ㅁ, and ㅅ of Korean alphabet, Hangul) preferred by ancestors who worshipped nature to identify the significations attached by them to particular patterns. The results confirm the following: first, the circle pattern indicated the sun, moon, stars in the sky, a bronze mirror, and a man's face. Circles and ovals were also observed to represent the individual souls of the clan or community. Second, square patterns symbolized the land and the patterns that signified the wellbeing of family and the country. Oblique rectangles were more frequently used as they represented a double use of the triangle, a shape that implied mystic power. Third, triangle symbolized regeneration, power, and humanity. While the Neolithic Age jade remnants of hair combs appear not to be irrelevant to the process of comb-shaped pottery production of the time, many fine comb-like lines may be found on bronze mirrors. Through its review of the glorious designs inherited from and established by ancient ancestors, the present research endeavor may help in identifying the spirits and traditions of Korean history.

Ginsengenin derivatives synthesized from 20(R)-panaxotriol: Synthesis, characterization, and antitumor activity targeting HIF-1 pathway

  • Background: Ginseng possesses antitumor effects, and ginsenosides are considered to be one of its main active chemical components. Ginsenosides can further be hydrolyzed to generate secondary saponins, and 20(R)-panaxotriol is an important sapogenin of ginsenosides. We aimed to synthesize a new ginsengenin derivative from 20(R)-panaxotriol and investigate its antitumor activity in vivo and in vitro. Methods: Here, 20(R)-panaxotriol was selected as a precursor and was modified into its derivatives. The new products were characterized by 1H-NMR, 13C-NMR and HR-MS and evaluated by molecular docking, MTT, luciferase reporter assay, western blotting, immunofluorescent staining, colony formation assay, EdU labeling and immunofluorescence, apoptosis assay, cells migration assay, transwell assay and in vivo antitumor activity assay. Results: The derivative with the best antitumor activity was identified as 6,12-dihydroxy-4,4,8,10,14-pentamethyl-17-(2,6,6-trimethyltetrahydro-2H-pyran-2-yl)hexadecahydro-1H-cyclopenta[a]phenanthren-3-yl(tert-butoxycarbonyl)glycinate (A11). The focus of this research was on the antitumor activity of the derivatives. The efficacy of the derivative A11 (IC50 < 0.3 µM) was more than 100 times higher than that of 20(R)- panaxotriol (IC50 > 30 µM). In addition, A11 inhibited the protein expression and nuclear accumulation of the hypoxia-inducible factor HIF-1α in HeLa cells under hypoxic conditions in a dose-dependent manner. Moreover, A11 dose-dependently inhibited the proliferation, migration, and invasion of HeLa cells, while promoting their apoptosis. Notably, the inhibition by A11 was more significant than that by 20(R)-panaxotriol (p < 0.01) in vivo. Conclusion: To our knowledge, this is the first study to report the production of derivative A11 from 20(R)-panaxotriol and its superior antitumor activity compared to its precursor. Moreover, derivative A11 can be used to further study and develop novel antitumor drugs.

Biochemical Adaptation of the Oriental Tobacco Budworm, Helicoverpa assulta, to Host-plant Defensive Compounds (기주식물 방어물질에 대한 담배나방의 생화학적 적응)

  • Plant secondary metabolites play an important role in insect-plant interactions. Herbivorous insects have various strategies to cope with the plant defensive compounds. Polyphagous insects feed on a wide variety of plant species, and their detoxification mechanisms are more complex since they tend to respond to a large array of different plant-derived chemicals. Alternatively, oligophagous insects specialize on only a few related plant species and may be expected to have a more efficient form of adaptation. This adaptation could involve either the production of large quantities of enzymes to detoxify their defensive compounds or the sequestration of the compounds or their metabolites. The oriental tobacco budworm, Helicoverpa assulta, is a specialist herbivore, feeding on a few plants of Solanaceae, such as tobacco and hot pepper. Understanding its host-plant adaptation not provides an important insight on physiology, ecology and evolution of specialist herbivores, but also gives a clue to develop management strategies of the pest species such as H. assulta. This paper briefly reviews the specialist, H. assulta, focusing on its host range, larval associations with the host plants, and detoxification mechanisms to nicotine and capsaicin, two characteristic defensive compounds derived from its two major host plants, tobacco and hot pepper, respectively. It summarizes the relevant research over the last half century and provides a future perspective on this subject.

Studies on Korean Digital Fantasy Film -Genealogy, Case, and Social Meaning (한국 디지털 판타지 영화 연구 -계보, 표본, 그리고 사회적 의미)

  • Recently, the cinema industry faced a crisis on the rise of various media platforms such as Netflix, Amazon Prime, IPTV, and Kakao Page. The rate of film release in the theater has become ever shorter, and the secondary consumption of film through IPTV, tablet, PC, or mobile has seen a drastic increase. In the midst of this new media-geography, the most significant change in recent years would be the rise of the 'fantasy film' genre. This paper explores the conditions and characteristics of fantasy films in the way in which the genre has been constituted, and delves into particular aspects that its contents contain. This is an attempt to understand the sociology of the birth of a new genre. In this process, this paper will ask two frequently raised questions in regard to this genre. The first is to ask whether we can discern fantasy from reality, and the second is to examine whether the fantasy genre implicates certain social subversion. These two questions aim to discover how fantasy forms a relationship with reality and what this means. To do so, this paper will trace the genealogy of the fantasy film genre in Korea and analyze recent big hits such as the series as the model case of digital fantasy film. Through this exploration, this paper will be able to provide a new sociology of the fantasy film production and consumption in the 21st century Korea.

  • To participate in global carbon neutrality, the Korean government is also planning to carry out zero-energy building certification for all buildings by 2030 through the enforcement decree of the 'Green Building Support Act'. Accordingly, the government is providing various projects related to solar power generation, which are relatively close to life. In particular, roof-mounted photovoltaic power generation systems are attracting attention in terms of using unused space to produce energy without destroying the environment, but low power generation efficiency compared to other photovoltaic power generation facilities is pointed out as a disadvantage. Therefore, in this paper, to solve this problem, we propose an efficient solar panel angle variable system through research on the solar panel structure for single-axial solar tracking, and also consider the application environment of the roof-mounted solar power generation system. Suggests measures to prevent damage and secondary damage. In addition, it is judged that it is possible to control the solar panel based on ICT convergence and configure the accident prediction safety system to link the project-based education program.

Experimental Evaluation of Developed Ultra-low NOx Coal Burner Using Gas in a Bench-scale Single Burner Furnace (Bench-scale 연소로에서 가스 혼소를 통한 초 저 NOx 석탄 버너 개발 연구)

  • This study developed and tested an ultra-low NOx burner in an 80 kW combustion furnace. The experiment was conducted in an 80 kW single burner combustion furnace with changing the swirl numbers, total equivalence ratios, and primary/secondary oxidizer ratios. In this study, liquefied natural gas (LNG) was used as an auxiliary fuel to significantly reduce NOx production. In a thermal power plant, the amount of NOx generated during coal combustion is about 300 ppm. However, using the burner tested in this study, it was possible to reduce the amount of NOx generated via LNG co-firing to 40 ppm. If the input amount of the primary oxidizer is enough for the gas to be completely combusted and the gas and coal are added simultaneously, the combusted gas forms a high-temperature region at the burner outlet and volatilizes the coal. As a result, the N contained in the devolatilized coal is discharged. Therefore, when the coal is subsequently burned, the amount of NOx produced decreases because there is almost no N remaining in the coal. If a thermal power plant burner is developed based on the results of this study, it is expected that the NOx generation will be significantly lower in the early stage of combustion.

Hair Growth Effect of TS-SCLF from Schisandra chinensis Extract Fermented with Lactobacillus plantarum

  • This study investigated the hair growth effect of Schisandra chinensis extract (TS-SC) and TS-SC fermented by Lactobacillus plantarum (TS-SCLF) on human dermal papilla cells (hDPCs). The production of vascular endothelial growth factor (VEGF), insulin-like growth factor 1 (IGF-1), keratinocyte growth factor/fibroblast growth factor 7 (KGF/FGF-7) and hepatocyte growth factor (HGF), transforming growth factor beta 1 (TGF-β1) were examined. The secretion rates of VEGF and KGF/FGF-7 were high in TS-SC, and the secretion rates of IGF-1 and HGF were high in TS-SCLF. TGF-β1 was inhibited in a concentration-dependent manner in all samples. Gene expression of VEGF, IGF-1, KGF, HGF and alkaline phosphatase, relevant to hair growth, were examined. The data revealed that TS-SC and TS-SCLF successfully promoted hair growth in hDPCs. The IGF-1 gene was expressed in a dose-dependent manner in TS-SCLF. These results indicate that TS-SC and TS-SCLF fermented extract effectively promoted hair growth and gene expression relevant to hair growth in hDPCs. Used in clinical trials the test substance 'CMK-LPF01' showed a statistically significant increase in the number of hairs at 8 weeks, 16 weeks, and 24 weeks compared to before product use, and a change in hair growth, a secondary efficacy evaluation variable. Through additional research in the future, it is expected that "CMK-LPF01" can be developed as a functional material that can help alleviate symptoms of hair loss.

  • This study was carried out to determine agronomic and genetic characteristics of the production process in tropical yams(Dioscorea alata L.). To make clear the possibility of cultivation of tropical yams in the Suwon area, the growth of the aboveground parts and the enlargement of tuber of Dioscorea alata, were investigated. The aboveground parts of the plants whose sprouted mother tuber were planted in late April were grown slowly until the 70-days after planting and grown rapidly after. From about 50-days after plantation, total length of vines slowly decreased due to the burning of the branch vines of lower modes. With regard to the branching progressed to 4th for the solo yam and to 6th for purple yam. The secondary branches tended to grow better. When the formation of daughter tubers was observed at the 50-days after planting, their enlargement was very slow. The rapid growth of tubers began at the 40-days and continued to the 90-days. General components such as protein, fiber, and lipid were higher in D. opposita rather than in D. alata. Hardness of D. alata was 2696.2 while that of D. opposita was 4946.9. Lightness of D. alata was 73.99, being higher than that of D. opposita.

Analysis for Economic Induced Effect of Oil and Gas E&P Industry's Business Ecosystem (산업생태계를 고려한 석유·가스개발산업의 경제적 유발효과 분석)

  • In this analysis, from the perspective of the business ecosystem, the economic effects of the oil and gas E&P industry were estimated. Australia, Norway, and the US, which are countries that are active in oil and gas E&P projects, and Japan and Korea, which are not, were analyzed. According to the analysis results, every country has a induced production coefficient of the oil and gas E&P industry greater than 1. Norway and the US have formed an industrial ecosystem across all industries, evenly demanding intermediate inputs for the primary, secondary, and tertiary industries. In contrast, Korea and Japan have a very large input ratio from the tertiary industry and an industrial ecosystem centered on the tertiary industry. It should be considered that the high input ratio of certain industry in business ecosystem determine the scale of the induced effect. When the business ecosystem are formed with many industries evenly, and the induced coefficient is high, it is necessary to increase the amount of final demand of the industry to make the effects of coefficient to increase.
