Disease monitoring of cultured rainbow trout and coho salmon in Gangwon province in 2021 (2021년 강원도 양식 무지개송어 및 은연어 비법정전염병 모니터링)

  • Disease including parasite, bacteria and virus cause serious mortality to salmonid fish in the aquaculture. In this study, we investigated the current disease status of the rainbow trout (Oncorhynchus mykiss) and coho salmon (Oncorhynchus kisutch) in Yanayang, Pyeongchang, Jeongseon and Yeongwol of Gangwon province in 2021 and performed molecular characterization of those pathogen. For parasites, Ichthyophthirius multifiliis was observed at 2 farms. For bacteria, we identified Aeromonas sobria from kidney of rainbow trout using phylogenetic analysis of gyrB gene. A. salmonicida were isolated from necrosis site of gill cover and fin in coho salmon and necrotic lesion of fin in rainbow trout. Phylogenetic analysis using vap gene indicated that A. salmonicida isolated in this study were clustered with previously reported A. salmonicida subsp. salmonicida isolates. For virus, JRt-Nagano type of infectious haematopoietic necrosis virus was detected in rainbow trout, but infectious pancreatic necrosis virus and Oncorhynchus masou virus were not detected. These results provide useful information for the prevention of disease spread and transmission when cultivating new species such as Atlantic salmon in Korea.

  • With increasing recognition on conservation of endangered species in Taiwan, one of the major conservation projects is the habitat restoration of Formosan Landlocked Salmon which is major threatened by check dams in the channel for their blockading pathway to upstream and causing the problems of population isolation and close-blood mating. By creating an opening in the central dam body appropriately, partial removal dams can provide pathway for the fish for the better upstream channel habitat. Four check dams at Gau-Shan Creek were remodeled between April of 1999 and September of 2002 with information supported from model experiments under certain hydraulic condition of field environment. Based on the follow-up investigation, the channel morphology of observation sections is in stable condition and the total number of Formosan Landlocked Salmon in this creek increased promptly at the reach containing partial-removed dams.

  • Chum salmon의 알이 부화하는 동안 물곰팡이의 감염으로부터 보호하기 위해 알을 10 mg/L 염소농도의 하이포아염소산나트륨(NaOCl) 용액에 매일 15분간 수정란에서부터 발안기까지 처리하였다. 감염된 알의 수 및 발안기 알의 수는 부화 23일째 관찰하였다. 전체의 알에서 감염된 알의 비율은 대조구(11.3;59.3%)보다 NaOCl 처리한 실험구(1.8; 33.4%)에서 유의적으로 낮게 나타났다(P<0.01). 전체 알에 대한 발안란의 비율은 대조구(66.1:97.5%)보다 NaOCl처리한 실험구(85.9:98.6%)에서 유의적으로 우수한 경향을 보였다(P<0.01). NaOCl의 항균 활성은 알의 생존율을 향상시킨다. 따라서 NaOCl는 chum salmon의 난에 대한 물곰팡이의 감염에 대하여 아주 유용한 항균물질이라고 판단된다.

Pinworm Infection at Salmon Ruins and Aztec Ruins: Relation to Pueblo III Regional Violence

  • The study of coprolites has been a theme of archaeology in the American Southwest. A feature of archaeoparasitology on the Colorado Plateau is the ubiquity of pinworm infection. As a crowd parasite, this ubiquity signals varying concentrations of populations. Our recent analysis of coprolite deposits from 2 sites revealed the highest prevalence of infection ever recorded for the region. For Salmon Ruins, the deposits date from AD 1140 to 1280. For Aztec Ruins, the samples can be dated by artifact association between AD 1182-1253. Both sites can be placed in the Ancestral Pueblo III occupation (AD 1100-1300), which included a period of cultural stress associated with warfare. Although neither of these sites show evidence of warfare, they are typical of large, defensible towns that survived this time of threat by virtue of large populations in stonewalled villages with easily accessible water. We hypothesize that the concentration of large numbers of people promoted pinworm infection and, therefore, explains the phenomenal levels of infection at these sites.

Molecular identification of Anisakid worm third stage larvae isolated from masou salmon Oncorhynchus masou (시마연어, Oncorhynchus masou에서 분리된 아니사키스 속 선충 3기 유충의 분자생물학적 방법을 이용한 동정)

  • Anisakid nematodes third stage larvae were isolated from the muscles of masou salmon (Oncorhynchus masou). Fish were purchased from Jumunjin fishery market in Gangneung. Four Anisakid third stage larvae were isolated from 4 fish. Molecular identification of the isolated worms was conducted by PCR-RFLP analysis of ribosomal DNA internal transcribed spacer region and direct sequencing of mitochondrial DNA cox2 gene. As results, all the tested individual worms were identified as Anisakis simplex (sensu stricto). This is the first report of molecular detection of anisakid worms in salmonid fishes in Korea.

Food Component Characteristics of Fish Frames as Basic Ingredients of Fish Gomtang (생선 곰탕의 추출소재로서 Fish Frame의 식품학적 특성)

  • The food components of fish (skipjack tuna, yellowfin tuna, bluefin tuna, conger eel, salmon, Spanish mackerel, armored weasel-fish) frames and their hot-water extracts were investigated to explore their possibilities as resources for fish Gomatang. According to the results of volatile basic nitrogen and heavy metal, bluefin tuna and salmon frames were below the safety limits suggested by Codex Code. Thus, bluefin tuna and salmon frames appeared to be safe as basic ingredients for fish Gomtang. The major components of all hot-water extracts from fish frames were nitrogenous component. According to the results of extractive nitrogen and sensory evaluations of hot-water extracts from fish frames, the salmon frame was a good raw material as a basic ingredient of fish Gomtang. The calcium and phosphorus contents of hot-water extracts from salmon frame were 18.0 mg/100mL and 33.1 mg/100mL, respectively.

Preparation and Characteristics of Fish-frame-added Snacks (Fish-frame을 이용한 snack의 제조 및 특성)

  • Fish-frames are processing byproducts, which are left after obtaining fillets or muscle during fish processing. The fish-frame generally consists of muscle, collagen, calcium, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). We used fish-frame powder (FFP) of chum salmon and skipjack tuna to prepare and characterize snacks for human consumption with different proportions of FFP. The crude protein and lipid contents of fish-frames were 16.3 and 9.4% for chum salmon and 18.6 and 8.3% for skipjack tuna, respectively. The volatile basic nitrogen (30.6 mg/100 g) and browning index (0.393) of FFP from chum salmon were lower than those of FFP from skipjack tuna. Thus, the FFP of chum salmon was better for making snacks than that of skipjack tuna. Five snacks were prepared with 0, 10, 20, 30, and 40% (w/w) substitution ratios of FFP from chum salmon. The moisture content of the snacks decreased (33.6 to 11.5%) with increasing FFP substitution ratio, whereas crude ash (2.9 to 7.5%), protein (11.4 to 18.4%) and lipid (13.7 to 35.1%) increased. Sensory scores for the texture and taste of the snack with 30% FFP were significantly higher (p<0.05) than those for other snacks; the color and flavor scores of all snacks did not differ significantly. The major fatty acids in the snacks were 16:0 and 18:0 as saturates, 18:1n-9 as monoenes, and 18:2n-6 and 18:3n-3 as polyenes. Snacks with FFP contained small amounts of EPA (0.5 to 0.8%) and DHA (1.3 to 1.8%) in the total lipid composition. The total amino acid content (16.08 g/100 g) of the snack with 30% FFP was higher than that of the snack without FFP (11.18 g/100 g), and the major amino acids were aspartic acid, glutamic acid, glycine, leucine, and lysine. The calcium and phosphorus contents of the snack with 30% FFP were 1,272 mg/100 g and 854 mg/100 g, respectively, and their ratio was the optimal range (2:1 to 1:2) for body absorption efficiency.

Inhibition of Foodborne Pathogens on Polystyrene, Sausage Casings, and Smoked Salmon Using Nonthermal Plasma Treatments (비열 플라즈마 처리를 이용한 polystyrene, 소시지 케이싱, 그리고 훈제연어에서의 식중독균 저해)

  • The effects of nonthermal plasma treatments against Salmonella Typhimurium, Escherichia coli O157:H7, and Listeria monocytogenes living on polystyrene (PS), sausage casings, and smoked salmon were investigated. Inoculated PS, casings, and salmon were treated with nonthermal plasma generated with helium (5 L/min) or with both helium (5 L/min) and oxygen (100 mL/min) at 60 Hz and 30 kV/cm for 2, 5, or 10 min. S. Typhimurium exhibited the highest sensitivity to the helium-used treatment. The greatest reduction (3.9${\pm}$0.8 log$CFU/cm^2$) was observed with L. monocytogenes on PS after the treatment with the mixed gas for 5 min. The treatment with the mixed gas inhibited L. monocytogenes on casings and salmon by 0.5${\pm}$0.3 log$CFU/cm^2$ and 1.0${\pm}$0.3 log CFU/g, respectively. Different treatment times did not result in different reductions of L. monocytogenes on both casings and salmon. The types of treatment gas and material of contamination need to be considered for evaluating the antimicrobial effects of nonthermal plasma treatments.

  • To improve functional properties and enhance application of FPC in food industry, modified salmon FPC with enzyme treatment was produced and its general properties were investigated. Salmon FPC has over $84\%$ of protein and less than $0.18\%$ of lipid. Solubilities of FPC extracted with IPA and ethanol were very poor as less than $3\%$ in every pH range. In case of enzyme : substrate ratio of 1 : 100, degree of hydrolysis significantly increased until 4 hours and then slightly increased. No considerable differences were observed in general components of hydrolysates. Results of SDS-PAGE showed one unique band in each case and their molecular weight was less than 6,500. The flow properties of hydrolysates showed newtonian flow. Whiteness of hydrolysates were higher than that of salmon FPC as $5\~7$. There was no significant differences in the amount of peptide, but that of free amino acid slightly increased from 0.17 to 0.21 mg/ml.

Antioxidant Effect of Anserine Extracted from Salmon (Oncorhynchus keta) (연어(Oncorhynchus keta) 추출 anserine의 항산화 효과)

  • Ion-exchange chromatography and ultrafiltration were used to extract anserine from salmon (Oncorhynchus keta). The salmon anserine showed DPPH radical scavenging activity in the range of 7.30% to 31.05% in a dose-dependent manner. This reducing power of salmon anserine also increased as the concentration increased. Metal chelate activity, superoxide dismutase - like activity, and thiobarbituric acid reactive substances assay showed similar results. The anserine also suppressed the increment of the peroxide value and linoleic acid during storage periods. These results suggest that salmon anserine might be useful as a natural antioxidant in various foodstuffs.
