Production of Monoclonal Antibody Against the Plasmalemma of Amoeba and its Application in Determining the Role of Membrane Components (아메바 세포막에 대한 단항체 생산 및 이를 이용한 막 조성 물질의 역할규명)

  • Monoclonal antibodies (MAbs) reacting with the plasmalemma of Amoeba proteus were produced. Specificity of the 3 MAbs was determined by transfer blotting of the SDS polvacryfamide gel. AMS antibody reacted with the mucopolysaccharide bands of the spacer gel, 220 KD and 50 KD proteins of the resolving gel. The maior glycoprotein bands (175 KD, 165 KD) and 50 KD protein of the plasmalemma were recognized by AUG antibody. A third, AMP antibody reacted with the 50 KD protein only. In immunofluorescence microscopy of the enzyme treated cells, the antigens of these MAbs were sensitive to proteases, but not sensitive to neuraminidase. In the assay of cell to substratum attachment after binding with the antibody, AMG and AMP antibodies exerted no effect, but AMS hindered the attachment and cell spreading. Thus the effective components of the plasmalemma in cell to substratum attachment appear to be the mucopolysaccharides and 220 KD protein. The membranes of latex particle infested phagosomes did not show any distinction from the plasmalemma in fluorescence microscopy. Phagosome membranes of amoebae appear to be derived from the plasma membrane without selection in terms of the antigen composition. Amoeba Proteus의 세포막과 반응하는 단세포군 항체를 생산하였다. SDS polyacrylamide gel을 transfer blotting하여 이들 항체의 반응 특이성을 조사해 본 결과 AMS 단항체는 PAS로 염색되는 spacer gel의 mucopolysaccharide 린드, resolving gel의 220 KD 및 50 KD 단백질과 반응하였으며, 세포막의 주요 당단백질인 175 KD 및 165 KD 빈드와 50 KD 단백질은 AMG 단항체에 의해서 인지되었다. 그리고 AMP단항체는 공통인 50 KD 단백질과 특이하게 반응하였다. 효소처리한 아메바의 면역형광칠미경적 조사에서 이들 항체에 대한 항원분자들은 모두 단백질분해효소에 민감하였으며 neuraminidase에 대해서는 변화가 없었다. 이들 항체를 결합시킨 아메바의 용기표면 부착 가능성을 분석한 결과 AMP 및 AMG 단항체는 아무런 영향을 미치지 못하였으며 AMS 단항체는 세포의 용기표면 부착 및 세포의 펴짐을 저해하였다. 따라서 아메바의 용기표면 부착은 mucopolysaccharide 및 220 KD 단백질에 의해서 매게되는 것으로 나타났다. 그리고 latex particle을 담고 있는 식포막은 면역형광형미경적 조사에서 세포막과 차이가 없었다. 따라서 겐포막은 항원 조성에 있어서 비 선택적으로 세포막에서 유도되는 것으로 나타났다.

Alterations in Protein and Amino acid Contents During Growth on Culture Cells of Wild Viola (Viola patrinii DC.) (야생 흰 제비꽃(Viola patrinii DC.) 배양세포의 생성과정 중 단백질 및 아미노산의 함량변화)

  • Proteins in friable and compact calli of Viola patrinii DC. were analysed. The protein contents in friable calli were lower than those in compact calli. In suspension culture, it increased to the maximum after 3 weeks culture from inoculation and decreased after 4 weeks culture. Several strong lavel signals were detected with the SDS-PAGE analysis. The polypeptides of 28, 31 and 35 KD were observed from the friable cell culture, from the compact cell culture strong band of 30 KD was determined, indicating that these polypeptides may be the major protein occurring during their cultures. Changes in amino acid contents during culture of the viola suspension cells were investigated the amino acid contents were greatest between two and three weeks culture of the viola suspension cells.

A ketogenic diet reduces body weight gain and alters insulin sensitivity and gut microbiota in a mouse model of diet-induced obesity

  • Purpose: Ketogenic diets (KDs) have anti-obesity effects that may be related to glucose control and the gut microbiota. This paper hypothesizes that KD reduces body weight and changes the insulin sensitivity and gut microbiota composition in a mouse model of diet-induced obesity. Methods: In this study, C57BL/6 male mice were assigned randomly to 3 groups. The assigned diets were provided to the control and high-fat (HF) diet groups for 14 weeks. The KD group was given a HF diet for 8 weeks to induce obesity, followed by feeding the KD for the next 6 weeks. Results: After the treatment period, the KD group exhibited a 35.82% decrease in body weight gain compared to the HF group. In addition, the KD group demonstrated enhanced glucose control, as shown by the lower levels of serum fasting glucose, serum fasting insulin, and the homeostatic model assessment of insulin resistance, compared to the HF group. An analysis of the gut microbiota using 16S ribosomal RNA sequencing revealed a significant decrease in the proportion of Firmicutes when the KD was administered. In addition, feeding the KD reduced the overall alpha-diversity measures and caused a notable separation of microbial composition compared to the HF diet group. The KD also led to a decrease in the relative abundance of specific species, such as Acetatifactor_muris, Ligilactobacillus_apodemi, and Muribaculum_intestinale, compared with the HF group. These species were positively correlated with the body weight, whereas the abundant species in the KD group (Kineothrix_alysoides and Saccharofermentans_acetigenes) showed a negative correlation with body weight. Conclusion: The current study presents supporting evidence that KD reduced the body weight and altered the insulin sensitivity and gut microbiota composition in a mouse model of diet-induced obesity.

Peptide Sequence Analysis of the CNBr-Digested 34-36 kd Sperminogen

  • Sperminogen was purified from the acid extracts of boar spermatozoa and partial peptide sequence of the 34-36 kd sperminogen was determined. Acid extracts of boar spermatozoa was gel-filtered through Sephadex G-75, and the 34-36 kd sperminogen was purified by preparative SDS-PAGE. The sperminogen bands were sliced out, and 34-36 kd sperminogen were eluted from the gel fragments and was subjected to peptide sequencing. Since the amino termini were blocked for Edman degradation method, internal amino acid sequences of the eluted 34-36 kd sperminogen were obtained from CNBr-digested peptides of sperminogen. Among several bands resolved on tricine SDS-PAGE, 14, 22 and 26 kd peptides were subjected to peptide sequencing. The ana1yzed amino acid sequences of the 26 and 22 kd peptides showed high homologies with that of the zona pellucida binding protein, Sp38, and the analyzed amino acid sequence of the 14 kd peptide showed neither sequence homology nor similarity with any known proteins.

  • We make use of the simplified Hirota's bilinear method with computer symbolic computation to study a variety of coupled potential KdV (pKdV) equations. Each coupled equation is completely integrable and gives multiple soliton solutions and multiple singular soliton solutions. The phase shifts for all coupled pKdV equations are identical whereas the coefficients of the obtained solitons are not identical. The four coupled pKdV equations are resonance free.

Understanding the importance of cerebrovascular involvement in Kawasaki disease

  • Kawasaki disease (KD) is a systemic vasculitis in infants and young children. However, its natural history has not been fully elucidated because the first case was reported in the late 1960s and patients who have recovered are just now entering middle age. Nevertheless, much evidence has raised concerns regarding the subclinical vascular changes that occur in post-KD patients. KD research has focused on coronary artery aneurysms because they are directly associated with fatality. However, aneurysms have been reported in other extracardiac muscular arteries and their fate seems to resemble that of coronary artery aneurysms. Arterial strokes in KD cases are rarely reported. Asymptomatic ischemic lesions were observed in a prospective study of brain vascular lesions in KD patients with coronary artery aneurysms. The findings of a study of single-photon emission computed tomography suggested that asymptomatic cerebral vasculitis is more common than we believed. Some authors assumed that the need to consider the possibility of brain vascular lesions in severe cases of KD regardless of presence or absence of neurological symptoms. These findings suggest that KD is related with cerebrovascular lesions in children and young adults. Considering the fatal consequences of cerebral vascular involvement in KD patients, increased attention is required. Here we review our understanding of brain vascular involvement in KD.

QT dispersion in children with Kawasaki disease

  • Background: We analyzed the changes in QT dispersion (QTd) in children with Kawasaki disease (KD), and determined the presence of repolarization abnormality in these children even in the absence of coronary artery abnormalities. Methods: Ninety-one children with KD and 20 healthy controls were enrolled in this retrospective study. Serial echocardiographic and electrocardiographic (ECG) measurements in the beginning of treatment, 2nd month and 6th month after the diagnosis were compared. Fifty-one of 91 children had at least 2 serial ECG data. The number of patients who had 3 consecutive ECG data was 23. Results: Among the 67 KD patients with no coronary artery changes, the consecutive mean QTd values were 41.86 ms, 37.84 ms, and 25.47 ms, respectively (26 ms for controls). In the analysis of changes among KD patients without coronary artery abnormalities, QTd showed a significant decrease with time (p=0.01). Especially, the 1st month and the 6th month QTd values were significantly different (p=0.028). The mean QTd values in KD patients with coronary artery changes were significantly higher than those in KD patients with no coronary artery changes at each time (1st, 2nd, and 6th month exam). Conclusion: QTd is significantly increased in children during the early stage of KD. Repolarization abnormality may exist during the acute stage of KD, regardless of the echocardiographic changes.

  • This paper investigates algebraic semantics for some weak Boolean (wB) logics, which may be regarded as left-continuous t-norm based logics (or monoidal t-norm based logics (MTLs)). We investigate as infinite-valued logics each of wB-LC and wB-sKD, and each corresponding first order extension $wB-LC\forall$ and $wB-sKD\forall$. We give algebraic completeness for each of them.

Study on the Antioxidative Activity of Human Milk (모유의 항산화능에 관한 연구)

  • The antioxidative activity of each fraction in human milk was examined using H2O2 and FeSO4-induced lipid peroxidation of mouse liver homogenate in order to elucidate the antioxidative substances of human milk. High molecular weight(~>20KD) fraction had more antioxidative effect on lipid peroxidation than low molecular weight(~ <20KD) fraction. Furthermore, the changes of antioxidative enzyme activities were estimated during lactation to study the roles of human milk. The human milk showed high activities of catalase, glutathione(GSH) peroxidase and GSH S-transferase. These results suggest that the antioxidative activities may mostly be attributed to high molecular weight fraction containing catalase, GSH peroxidase and GSH S-transferase.

Changes in the Polypeptide Patterns of Oat Root Tips Exposed to Alachlor (Alachlor에 의한 귀리 근단(根端) 분열조직(分裂組織)의 단백질(蛋白質) Pattern의 변이(變異))

  • The effect of alachlor treatment on protein synthesis was studied. Protein synthesis was inhibited by $1{\times}10^{-4}$ M and $1{\times}10^{-3}$M of alachlor 5.8% and 86.5%, respectively, while did not occur blow $1{\times}10^{-5}$M alachlor. Soluble protein of alachlor treated oat root tips was examined by polyacrylamide gel electrophoresis. The proteins extracted from oat root tips showed that they were made up of subunits blow 100 kd polypeptides by SDS-PAGE. As compared to control, high molecular proteins(above 47 kd) were inhibited of oat root treated with alachlor, while low molecular proteins(below 23 kd) were increased. Two-D gels showed that alachlor caused decrease(1-6 spots) or increase(7-10 spots) in number of polypeptides on silver staining. The intensity of some polypeptides of soluble proteins (molecular mass of 83 kd : 1, 2 spots, 70 kd : 3, 4 spots, and 47.5 kd : 5, 6 spots) decreased in alachlor treatment, whereas the intensity of other peptide bands (20 kd : 7 spot and 16 kd : 8, 9, 10 spots) increased. Oat root tip proteins present in the neutral zone are masked by diffusing of major proteins, but proteins in acid zone are resolved minor proteins.
