The Characteristics of I269S and I224S Double Mutant Horse Liver Alcohol Dehydrogenase (I269S와 I224S 이중변이 알코올 탈수소효소의 특성)

  • Ile-224 in I269S mutant horse liver alcohol dehydrogenase isoenzyme S (HLADH-S) was mutated to serine by site-directed mutagenesis in order to study the role of the residue in c oenzyme binding to the enzyme. The specific activity of the I269S and I224S mutant enzyme to ethanol was increased 6-fold and all Michaelis constants($K_a,\;K_b,\;K_p,\;and\;K_q$,/TEX>) were larger than those for the wild-type and I269S enzyme. The substitution decreased the afffinity to coenzymes and increased the specific activity of the enzyme. The mutant enzyme showed the highest catalytic efficiency for octanol among the primary alcohols. But it didn`t have activities on retinoids and 5${\beta}$-cholanic acid-3-one. From these results, it was confirmed that the hydrophobic interaction of Ile-224 residue with coenzyme was related to coenzyme affinity in ADH reaction. The substitution also affected the substrate affinities to the enzyme.

Effect of Microstructures and Sintering Additives on the Mechanical Properties of Si$_3$N$_4$ (질화규소의 기계적 특성에 미치는 미세구조 및 소결조제의 영향)

  • Four distinctive hot pressed and heat treated S${i_3}{N_4}$ceramics, S${i_3}{N_4}$-8%${Y_2}{O_3}$, S${i_3}{N_4}$-6% ${Y_2}{O_3}$-2% $A{l_2}{O_3}$, S${i_3}{N_4}$-4% ${Y_2}{O_3}$-3% $A{l_2}{O_3}$, 그리고 S${i_3}{N_4}$-1% MgO-1% Si$O_2$(in wt%), were prepared and characterized by X-ray diffraction, scanning electron microscopy, image analysis and mechanical tests. The fracture toughness of S${i_3}{N_4}$-8% ${Y_2}{O_3}$specimens containing large elongated grains showed the highest value of about 9.8MPa$m^{1/2}$. Two out of four S${i_3}{N_4}$, ceramics(S${i_3}{N_4}$-6% ${Y_2}{O_3}$-2% $A{l_2}{O_3}$and S${i_3}{N_4}$-4% ${Y_2}{O_3}$-3% $A{l_2}{O_3}$) heat treated at 200 $0^{\circ}C$retained the fracture strength of over 900MPa and fracture toughness of over 8.0MPa$m^{1/2}$. Large ${\beta}$-S${i_3}{N_4}$grains having a diameter larger than 1${\mu}$m appeared to contribute to increase in fracture toughness.

  • Let I be an ideal in a Gorenstein local ring A with the maximal ideal m and d = dim A. Then we say that I is a good ideal in A, if I contains a reduction $Q=(a_1,a_2,...,a_d)$ generated by d elements in A and $G(I)=\bigoplus_{n\geq0}I^n/I^{n+1}$ of I is a Gorenstein ring with a(G(I)) = 1-d, where a(G(I)) denotes the a-invariant of G(I). Let S = A[Q/a$_1$] and P = mS. In this paper, we show that the following conditions are equivalent. (1) $I^2$ = QI and I = Q:I. (2) $I^2S$ = $a_1$IS and IS = $a_1$S:sIS. (3) $I^2$Sp = $a_1$ISp and ISp = $a_1$Sp :sp ISp. We denote by $X_A(Q)$ the set of good ideals I in $X_A(Q)$ such that I contains Q as a reduction. As a Corollary of this result, we show that $I\inX_A(Q)\Leftrightarrow\IS_P\inX_{SP}(Qp)$.

  • The nuclear LS-energy matrix elements have been calculated with the harmonic oscillator shell model wave functions for the configurations ( $l_{i}$ $l_{i+1}$$l_{i}$ $l_{i+1}$) where 1$_1$= $l_{s}$ , $l_2$=lp, $l_3$=ld, 2s, $l_4$=1f, 2p, $l_{5}$ =1g, 2d, 3s. The resulting matrix elements are expressed in terms of both Talmi integrals $I_1$ and Slater integrals $F^{k}$ . In addition to this various sum rules are derived and applied to check the results of the calculations.ons.

Study on iodine Labelling (II) Efficient of Labelling Rose Bengal, Hippuran, and Human Serum Albumin in Small Scale

  • For efficient micro scale syntheses of Rose $Bengal-^{131}I$, $Hippuran-^{131}I$, and $H.S.A.-^{131}I$, the dependence of labelling yields on pH, on salt contents, and on the volume of buffer solution in the reaction mixtures as well as the reaction apparatus were studied. pH of 5.6 was optimum for preparation of both Rose $Bengal-_{131}I$ and Hippuran $-^{13}I$ but pH of 8.5 was optimum for preparation of $H.S.A.-^{131}I$. Salt in the reaction mixtures hindered drastically the formation of $Hippuran-^{131}I$ but it slightly increased the labelling yield of H.S.A.. The compactly closed reaction vessels were effective for preparations of both Rose $Bengal-^{131}I$and $Hippuran-^{131}I$ in small volume. Thereupon, the labelling procedures were modified to bring about higher labelling yields and better reproducibilities. By these newly established procedures, the labelling yields of Rose $Bengal-^{131}I$ and $Hippuran-^{131}I$ could be increased even with the home-produced sodium $iodide-^{131}I$ solution containing reducing agent.

C(S) extensions of S-I-BCK-algebras

  • In this paper we consider more systematically the centralizer C(S) of the set $S = {f_a $\mid$ f_a : X \to X ; x \longmapsto x * a, a \in X}$ with respect to the semigroup End(X) of all endomorphisms of an implicative BCK-algebra X with the condition (S). We obtain a series of interesting results. The main results are stated as follows : (1) C(S) with repect to a binary operation * defined in a certain way forms a bounded implicative BCK-algebra with the condition (S). (2) X can be imbedded in C(S) such that X is an ideal of C(S)/ (3) If X is not bounded, it can be imbedded in a bounded subalgebra T of C(S) such that X is a maximal ideal of T. (4) If $X (\neq {0})$ is semisimple, C(S) is BCK-isomorphic to $\prod_{i \in I}{A_i}$ in which ${A_i}_{i \in I}$ is simple ideal family of X.

  • In this paper we introduce S(X, $x_{0}$) which is a generalization of Ellis group G(X, $x_{0}$), and S-sets in S(X, $x_{0}$). In particular we cind the sufficient condition for the group A(I) of all automorphisms of I and K=Iu to be isomorphic, where I is a minimal right ideal and u is an idempotent of I.f I.

  • Let f be a function which assigns a positive integer f(v) to each vertex v $\in$ V (G), let r, s and t be non-negative integers. An f-coloring of G is an edge-coloring of G such that each vertex v $\in$ V (G) has at most f(v) incident edges colored with the same color. The minimum number of colors needed to f-color G is called the f-chromatic index of G and denoted by ${\chi}'_f$(G). An [r, s, t; f]-coloring of a graph G is a mapping c from V(G) $\bigcup$ E(G) to the color set C = {0, 1, $\ldots$; k - 1} such that |c($v_i$) - c($v_j$ )| $\geq$ r for every two adjacent vertices $v_i$ and $v_j$, |c($e_i$ - c($e_j$)| $\geq$ s and ${\alpha}(v_i)$ $\leq$ f($v_i$) for all $v_i$ $\in$ V (G), ${\alpha}$ $\in$ C where ${\alpha}(v_i)$ denotes the number of ${\alpha}$-edges incident with the vertex $v_i$ and $e_i$, $e_j$ are edges which are incident with $v_i$ but colored with different colors, |c($e_i$)-c($v_j$)| $\geq$ t for all pairs of incident vertices and edges. The minimum k such that G has an [r, s, t; f]-coloring with k colors is defined as the [r, s, t; f]-chromatic number and denoted by ${\chi}_{r,s,t;f}$ (G). In this paper, we present some general bounds for [r, s, t; f]-coloring firstly. After that, we obtain some important properties under the restriction min{r, s, t} = 0 or min{r, s, t} = 1. Finally, we present some problems for further research.

We show that there exists one or two positive solutions by using fixed-point theorem for operator on a cone. The conclusions in this paper essentially extend and improve the known results.

Genetic Variation in the Natural Populations of Korean Stewartia (Stewartia koreana Nakai) Based on I-SSR Analysis (I-SSR 분석에 의한 노각나무 천연집단의 유전변이)

  • We investigated the genetic variation in Stewartia koreana Nakai by examining 61 I-SSR amplicons in 120 individuals distributed among six natural populations in Korea. The overall percentage of polymorphic I-SSR amplicons was 81.9% and mean number of amplicons per I-SSR primer was 12.2. Levels of genetic diversity within 6 populations were similar each other[Shannon's Index $0.358{\sim}0.467$(mean: 0.407)]. The Mt. Obong population had the highest level of genetic diversity and was most distinctive from the other populations. Most variation existed among individuals within population(88.2%). Genetic differentiation among populations(${\phi}_{ST}$) was 0.118. The UPGMA dendrogram based on the genetic distance failed in showing decisive geographic relationships.
