Characterization of Growth Inhibition and Isolation of a Copper-Resistant Rhizobacterium, Alcaligenes sp. KC-1 (Cu 내성 근권 세균 Alcaligenes sp. KC-1의 분리 및 생장특성)

  • In this study, A bacterium with an ability to resist toxic heavy metals was isolated from reeds in wetland. The isolated strain was identified to Alcaligenes sp. KC-1 by 16S rDNA sequencing. Heavy metals such as Pb, $Cr^{6+}$, Cd, Zn and Cu were supplied to media. The ecotoxic treat of the heavy metals on the growth of strain KC-1 was performed when the isolated strain Alcaligenes sp. KC-1 cultured with Cu ranging from 0 mM to 20 mM. It showed the resistance of $EC_{50}$(7.34 mM) and cell growth ($OD_{600\;nm}$ : 0.83 after 42 hours) when it was cultured in Cu.

Joint Toxic Action of Bifenthrin and Prothiofos Mixture for the Control of Insectivcide-Resistant Diamondback Moth, Plutella xylostella L. (살충제 저항성 배추좀나방 방제를 위한 Bifenthrin과 Prothiofos 혼용의 연합작용)

  • Mixture study of bifenthrin and prothiofos was conducted to control insecticide resistant diamondback moth (DBM), Plutella xylostella, which had shown 581, 18, 19, 11 fold resistance to fenvalerate, cypermethrin, furathiocarb, and prothiofos in Chinju strain, respectively and 38 and 9 fold resistance to fenvalerate and furathiocarb in Seosang strain, respectively. Optimal mixture ratios of bifenthrin and prothiofos was selected against Seosang strain of DBM by leaf dipping method in laboratory and by field test. Maximum co-toxicity coefficient by dipping method was shown at a ratio of 1:50 mixture of bifenthrin 1EC and prothiofos 50EC in active ingredient(a.i.) by 243.2 and then suddenly decreased. The mixed formulation from the dipping test was examined at the cabbage field of Seosang, Hamyang. Although insecticides were reduced to half, control efficacy was similar to farmer's conventional method. In addition, the mixture combination was also more effective against Chinju DBM population which had very high level of resistance to several representative insecticides. Mixture methods of insecticides will be effective countermeasures to the resistance problem of pests.

  • Community-associated Methicillin-Resistant Staphylococcus aureus (CA-MRSA) is notorious as a leading cause of soft tissue infections. Despite several studies on the Agr regulator, the mechanisms of action of Agr on the virulence factors in different strains are still unknown. To reveal the role of Agr in different CA-MRSA, we investigated the LACΔagr mutant and the MW2Δagr mutant by comparing LAC (USA300), MW2 (USA400), and Δagr mutants. The changes of Δagr mutants in sensitivity to oxacillin and several virulence factors such as biofilm formation, pigmentation, motility, and membrane properties were monitored. LACΔagr and MW2Δagr mutants showed different oxacillin sensitivity and biofilm formation compared to the LAC and MW2 strains. Regardless of the strain, the motility was reduced in Δagr mutants. And there was an increase in the long chain fatty acid in phospholipid fatty acid composition of Δagr mutants. Other properties such as biofilm formation, pigmentation, motility, and membrane properties were different in both Δagr mutants. The Agr regulator may have a common role like the control of motility and straindependent roles such as antibiotic resistance, biofilm formation, change of membrane, and pigment production. It does not seem easy to control all MRSA by targeting the Agr regulator only as it showed strain-dependent behaviors.

Study on the Chlorine-Resistant Bacteria Isolated from Water Pipe Network (상수도관망에서 분리한 잔류염소 내성균에 관한 연구)

  • The free residual chlorine of tap water samples, collected from 266 faucets on the water pipe network in Daegu City, was between 0.1 and 0.79 mg/L. On microorganic tests, general bacteria and the coliform goup were not detected and thus the tap water was turned out to be fit to drink. In particular, samples of which free residual chlorine was 0.1 mg/L and over were cultured in R2A agar media at $25^{\circ}C$ for 7 days, and as a result heterotrophic bacteria were detected in 65.9% of samples; (1). The closer tap water got to the faucet from the stilling basin, the lower residual chlorine concentration became but the more the bacterial count became. And, more bacteria were detected in the R2A agar medium than in the PCA medium. (2). In the case of separated strains, most colonies were reddish or yellowish. 16S rRNA sequence was identified as Methylobacterium sp. and Williamsia sp., and yellow strain was identified as Sphingomonas sp., Sphingobium sp., Novosphingobium sp., Blastomonas sp., Rhodococcus sp. and Microbacterium sp. White strain was identified as Staphylococcus sp. (3). Sterilized tap water in polyethylene bottles was inoculated with separated strain and was left as it was for 2 months. As a result, bio-film was observed in tap water inoculated with Methylobacterium sp. and Sphingomonas sp. It was found that heterotrophic bacteria increased when free residual chlorine was removed from tap water in the water pipe network. Thus, there is a need to determine a base value for heterotrophic bacteria in order to check the cleanliness of tap water in the water pipe network.

Combination of berberine and ciprofloxacin reduces multi-resistant Salmonella strain biofilm formation by depressing mRNA expressions of luxS, rpoE, and ompR

  • Bacterial biofilms have been demonstrated to be closely related to clinical infections and contribute to drug resistance. Berberine, which is the main component of Coptis chinensis, has been reported to have efficient antibacterial activity. This study aimed to investigate the potential effect of a combination of berberine with ciprofloxacin (CIP) to inhibit Salmonella biofilm formation and its effect on expressions of related genes (rpoE, luxS, and ompR). The fractional inhibitory concentration (FIC) index of the combination of berberine with CIP is 0.75 showing a synergistic antibacterial effect. The biofilm's adhesion rate and growth curve showed that the multi-resistant Salmonella strain had the potential to form a biofilm relative to that of strain CVCC528, and the antibiofilm effects were in a dose-dependent manner. Biofilm microstructures were rarely observed at $1/2{\times}MIC/FIC$ concentrations (MIC, minimal inhibition concentration), and the combination had a stronger antibiofilm effect than each of the antimicrobial agents used alone at $1/4{\times}FIC$ concentration. LuxS, rpoE, and ompR mRNA expressions were significantly repressed (p< 0.01) at $1/2{\times}MIC/FIC$ concentrations, and the berberine and CIP combination repressed mRNA expressions more strongly at the $1/4{\times}FIC$ concentration. The results indicate that the combination of berberine and CIP has a synergistic effect and is effective in inhibiting Salmonella biofilm formation via repression of luxS, rpoE, and ompR mRNA expressions.

Characterization and Frequency of Vancomycin Resistance in Staphylococcus aureus Isolated in Korea (국내에서 분리된 포도상구균의 Vancomycin 내성빈도 및 특성)

  • The vancomycin, one of the family of glycopeptide antibiotics, inhibits the synthesis of bacterial cell wall peptidoglycan and has been widely used against gram-positive bacterial infections, especially for a treatment of methicillin resistant S. aureus infection. However, clinical isolate which was intermediately resistant to vancomycin (Mu50: MIC 8 $\mu\textrm{g}$/ml) was isolated in recent years. In this study we performed vancomycin susceptibility test with the increment method and population analysis with clinical isolates S. aureus. Also we did several kinds of tests with three selected isolates (s129: MIC 7 $\mu\textrm{g}$/ml, s134: MIC 7 $\mu\textrm{g}$/ml, s135: MIC 8 $\mu\textrm{g}$/ml) to find out possible mechanism of vancomycin resistance. As a result, the prevalence of vancomycin resistant S. aureus isolates among S. aureus strains resistant to methicillin was 23.3% (25/107). The vancomycin resistances of isolated strains of S. aureus were between those of Mu5O and Mu3 strains. By PCR analysis, none of the isolates with decreased vancomycin susceptibility contained known vancomycin resistant genes such as vanA, vanB, vanC1, vanC2, and vanH. Major bands of 81 kDa, 58 kDa, 33 kDa, 28 kDa were demonstrable in whole cell lysates by SDS-PAGE from all three isolates as well as reference strains. And especially,45 kDa protein was overproduced in Mu50 strains. Among them increased production of NAD$^{+}$-linked-$_{D}$-lactate dehydrogenase (dnLDH) were detected from one clinical strain (s135) and Mu5O strain. From these data, we suggest that the mechanism of vancomycin resistance in these isolates are distinct from that in enterococci.

Population of Pesticide Resistant Strains in Cultivated Soils of Honam Area and Degradation of Chlorothalonil in soil (호남지역 농경지 토양에 분포된 농약내성균의 밀도와 살균제 Chlorothalonil의 분해)

  • To obtain the basic information for degradation of remaining pesticide accumulated in cultivated soil of Honam area, the resistant bacterial strains were investigated in Chlorothalonil(TPN). Mancozeb, Bentazone, and Butachlor levels of 100, $500{\mu}g\;ml^{-1}$, and degradation of TPN by TPN-resistant bacteria in sterilized soil was studied under TPN levels 0, 10, 50 and $100{\mu}g\;g^{-1}$. A number of resistance strains were decreased with higher at concentration level of pesticide, and were higher in greenhouse than upland or paddy soil. The resistance of bacteria was strong in other of Bentazone> Butachlor> TPN> Mancozeb. The percentage of bacterial strains of resistance for pesticides isolated from the cultivated soil were the highest in Acinetobacter spp. and Corynebacterium spp., and the lowest in Moraxella spp. A number of TPN-resistant strains were the highest at the TPN level of $10{\mu}g\;g^{-1}$, and 5 days after strains inoculation, and were higher in Pseudomonas spp. TD-25 than TC-23 or strains in non-sterilized soil. The degradation of TPN was fast in order of strain TD-25>strain in non-sterilized soil >TC-23.

Prevalence and Antimicrobial Resistance of Enterococus faecalis and Enterococcus faecium Isolated from Beef, Pork, Chicken and Sashimi (시판 축산물 및 수산물에서 Enterococcus faecalis와 Enterococcus faecium 분포 및 항생제 감수성에 관한 연구)

  • In this study, a total of 256 samples of retail raw meats (beef, pork and chicken) and sashimi were investigated for the presence of Enterococcus faecalis and Enterococcus faecium. We isolated a total of 117 E. faecalis and E. faecium from the samples, with contamination rates ranging from 18.8% for sashimi samples to 68.8% of chicken samples. E. faecalis was the predominant species recovered from all of the retail raw meats beef (42.2%), pork (42.2%), chicken (65.6%) and sashimi (12.5%). Among 117 isolates, 61 isolates (52.1%) were resistant to tetracycline, 32 isolates (27.4%) were resistant to erythromycin, 23 isolates (19.7%) were resistant to chloramphenicol, 16 isolates (13.7%) were resistant to ripampin, 10 isolates (8.5%) were resistant to gentamycin, 9 isolates (7.7%) were resistant to ciprofloxacin and 1 isolate (0.9%) was resistant to ampicillin and penicillin G. No resistance to amoxicillin + clavulanic acid and vancomycin was observed. Although no strain was resistant to vancomycin, the vanB gene was observed in 9 of 117 of Enterococcus (7.7%) demonstrating potential risk of vancomycin-resistant Enterococcus (VRE). Our results indicate that E. faecalis and E. faecium were highly prevalent in retail raw meats, but most strains were sensitive to tested antibiotics.

Effects of Coinoculation by Bradyrhizobium japonicum with other Rhizobacteria and of Antibiotic Treatment on Soybean Growth and Nodule Formation (근류균(根瘤菌)과 타(他) 근권미생물(根圈微生物)과의 혼합접종(混合接種) 및 항생제처리(抗生劑處理)가 대두(大豆)의 생장(生長)과 근류형성(根瘤形成)에 미치는 영향(影響))

  • The Neomycin resistant gene in transposon 5 (Tn5) was introduced into a mutant of Bradyrhizobium japonicum RJB 6 $str^rnal^r$ by conjugation. This marked strain was used in coinoculation of soybean with Pseudomonas fluorescens(Ps-3) and Bacillus subtilis(BCAC-4) and in antibiotic treatment of soybean for studying rate of nodule formation and growth promoting effect on soybean plant. When the marked strain RJB 6 $str^rnal^rneo^r$ with Ps-3, BCAC-4, and Ps-3 plus BCAC-4 was coinoculated into two soils, the recovery rate of the marked strain was 8.5, 4.3 and 2.9 percent in soybean cultivated soil, and 10.3. 6.0 and 5.0 percent in soybean uncultivated soil. The best growth promoting effect of coinoculation on soybean plant was found with the marked strain plus Ps-3. When the marked strain was inoculated into soybean with antibiotic treatment, the rate of nodule formation in soybean cultivated soil was between 12.5 and 25.4 percent, while that in soybean uncultivated soil was between 23.7 and 43.2 percent. The highest rate of nodule formation with the marked strain was found in streptomycin 1000 ppm treatment plot.

Cross Resistance and Point Mutation of the Mitochondrial Cytochrome b of Bifenazate Resistant Two-spotted Spider Mite, Tetranychus urticae (Bifenazate 저항성 점박이응애에 대한 교차저항성과 미토콘드리아 cytochrome b의 점돌연변이)

  • Two-spotted spider mite, Tetranychus urticae was collected from the rose greenhouse in Chilgok, Gyeongbuk Province in December 2000. This population has been selected for ten years with bifenazate (over 450 times), and increased 855.9 fold in resistance as compared with susceptible strain (S). Cross resistance of bifenazate resistant (BR) strain to eight miticides was investigated. The BR strain exhibited high and low cross resistance to acequinocyl (614.0 fold) and to chlorfenapyr (9.1 fold), respectively. Against fenazaquin (0.3 fold) and fenpyroximate (0.1 fold), however, showed the strain negatively correlated cross resistance. Each strain collected in Choeng-ju (CJ), Kang-jin (KJ), and Chung-ju (CUJ) showed 5.5-, 964.5-, and 21.8-fold resistance to bifenazate, respectively. The detoxifying enzymes of the BR strain showed 1.6-fold activity in cytochrome $P_{450}$-dependent monooxygenase ($P_{450}$) as compared with susceptible one. By comparing the mitochondrial cytochrome b (cytb) sequence, G126S point mutation was detected in the BR and KJ strains.
