Prediction of Changes in Habitat Distribution of the Alfalfa Weevil (Hypera postica) Using RCP Climate Change Scenarios (RCP 기후변화 시나리오 따른 알팔파바구미(Hypera postica)의 서식지 분포 변화 예측)

  • Climate change can affect variables related to the life cycle of insects, including growth, development, survival, reproduction and distribution. As it encourages alien insects to rapidly spread and settle, climate change is regarded as one of the direct causes of decreased biodiversity because it disturbed ecosystems and reduces the population of native species. Hypera postica caused a great deal of damage in the southern provinces of Korea after it was first identified on Jeju lsland in the 1990s. In recent years, the number of individuals moving to estivation sites has concerned scientists due to the crop damage and national proliferation. In this study, we examine how climate change could affect inhabitation of H. postica. The MaxEnt model was applied to estimate potential distributions of H. postica using future climate change scenarios, namely, representative concentration pathway (RCP) 4.5 and RCP 8.5. As variables of the model, this study used six bio-climates (bio3, bio6, bio10, bio12, bio14, and bio16) in consideration of the ecological characteristics of 66 areas where inhabitation of H. postica was confirmed from 2015 to 2017, and in consideration of the interrelation between prediction variables. The fitness of the model was measured at a considered potentially useful level of 0.765 on average, and the warmest quarter has a high contribution rate of 60-70%. Prediction models (RCP 4.5 and RCP 8.5) results for the year 2050 and 2070 indicated that H. postica habitats are projected to expand across the Korean peninsula due to increasing temperatures.

Effects of Oxygen Consumption in the Giant Grouper Epinephelus lanceolatus by Water Temperature and Photoperiod (수온 및 광조건에 따른 대왕바리 Epinephelus lanceolatus의 산소소비 특성)

  • This study investigated the effects of water temperature and photoperiod on oxygen consumption (OC) in the giant grouper Epinephelus lanceolatus. OC rate in the giant grouper at 17, 22, 27 and $32^{\circ}C$ were $61.7{\pm}0.4$, $72.2{\pm}0.6$, $102.9{\pm}0.8$ and $141.7{\pm}1.0mg\;O_2/kg/h$, respectively, indicating a linear increase in OC with water temperature. Photoperiod was regulated in accordance with the light (06:00~18:00 h, L) and dark (18:00~06:00 h, D) phases of the diel cycle (12L/12D), with a water temperature of at 17, 22, 27 and $32^{\circ}C$. OC rates during the light and dark phases were $62.7{\pm}0.4$, $62.5{\pm}0.3mg\;O_2/kg/h$, respectively, at $17^{\circ}C$. No significant differences were observed between the light and dark phases (P>0.05). OC raters during the light and dark phases were $74.8{\pm}0.7$, $69.6{\pm}0.6mg\;O_2/kg/h$ at $22^{\circ}C$, $107{\pm}1.2$, $98.0{\pm}0.7mg\;O_2/kg/h$ at $27^{\circ}C$ and $147.6{\pm}1.1$, $135.8{\pm}0.8mg\;O_2/kg/h$ at $32^{\circ}C$, respectively, indicating that OC is higher during daylight than nighttime. Giant grouper is thought to be a diurnal fish species, because the level of oxygen consumption during light period was higher than that during dark period.

Evaluation of Water Quality after Rehabilitation of Cheonggye Stream using AGP Test (조류성장잠재력 조사를 이용한 청계천 복원 이후 수질 평가)

  • Algal growth potential (AGP) test was performed to evaluate the water quality and changes in phytoplankton communities before and after a heavy rain event at six sampling sites in Cheonggye Stream (St. 1 and 2), Jungnang Stream (St. 3 and 4), and Lower part of Han River System (St. 5 and 6) after rehabilitation of Cheonggye Stream, October 2005. To test AGP on each sampling site, cyanobacterium Microcystis aeruginosa was applied as a standard alga. Total nitrogen (TN) showed high values at Jungnang Stream, while St. 4 recorded highest values in this study. However, TN values of Cheonggye Stream and Lower Part of Han River showed similar levels. Total phosphate (TP) also showed high values at Jungnang Stream, while St. 4 recorded highest. However, TP in Cheonggye Stream were extremely low levels. Although chlorophyll-${\alpha}$ (chi-${\alpha}$) contents before the rain event were similar through the sampling sites, chl-${\alpha}$ after the rain increased dramatically at Jungnang Stream and Lower part of Ban River. In particular, after the rain, TP was the highest at St. 4, where Cyclotella sp. dominated the phytoplankton community. When compared with control, AGP values before the rain were comparatively low in all sites, while those after the rain highly increased with the dose-dependently of field water added, due perhaps to the increased nutrients by rainfall. Similar results were observed in Cheonggye Stream. Therefore, for the aesthetic fostering for the citizens, although Cheonggye Stream was Presently being sustained by treated water supply, they have a potential of outbreak of phytoplankton by the increased nutrients supply when a heavy rain comes.

Oxidative Stress-dependent Structural and Functional Regulation of 2-cysteine Peroxiredoxins In Eukaryotes Including Plant Cells (산화 스트레스에 의존한 식물 및 진핵세포 2-시스테인 퍼록시레독신의 기능 조절)

  • Peroxiredoxins (Prxs) are ubiquitously distributed and play important functions in diverse cellular signaling systems. The proteins are largely classified into three groups, such as typical 2-Cys Prx, atypical 2-Cys Prx, and 1-Cys Prx, that are distinguished by their catalytic mechanisms and number of Cys residues. From the three classes of Prxs, the typical 2-Cys Prx containing the two-conserved Cys residues at its N-terminus and C-terminus catalyzes $H_2O_2$ with the use of thioredoxin (Trx) as an electron donor. During the catalytic cycle, the N-terminal Cys residue undergoes a peroxide-dependent oxidation to sulfenic acid, which can be further oxidized to sulfinic acid at the presence of high concentrations of $H_2O_2$ and a Trx system containing Trx, Trx reductase, and NADPH. The sulfinic acid form of 2-Cys Prx is reduced by the action of sulfiredoxin which requires ATP as an energy source. Under the strong oxidative or heat shock stress conditions, 2-Cys Prx in eukaryotes rapidly switches its protein structure from low-molecular-weight species to high-molecular-weight protein structures. In accordance with its structural changes, the protein concomitantly triggers functional switching from a peroxidase to a molecular chaperone, which can protect its substrate denaturation from external stress. In addition to its N-terminal active site, the C-terminal domain including 'YF-motif' of 2-Cys Prx plays a critical role in the structural changes. Therefore, the C-terminal truncated 2-Cys Prxs are not able to regulate their protein structures and highly resistant to $H_2O_2$-dependent hyperoxidation, suggesting that the reaction is guided by the peroxidatic Cys residue. Based on the results, it may be concluded that the peroxidatic Cys of 2-Cys Prx acts as an '$H_2O_2$-sensor' in the cells. The oxidative stress-dependent regulation of 2-Cys Prx provides a means of defense systems in cells to adapt stress conditions by activating intracellular defense signaling pathways. Particularly, 2-Cys Prxs in plants are localized in chloroplasts with a dynamic protein structure. The protein undergoes conformational changes again oxidative stress. Depending on a redox-potential of the chloroplasts, the plant 2-Cys Prx forms super-molecular weight protein structures, which attach to the thylakoid membranes in a reversible manner.

Attenuation of Oxidative Stress-Induced HepG2 Cellular Damage by Cirsiumjaponicum Root Extract (HepG2 세포에서 대계 추출물에 의한 산화적 스트레스 유발 세포 손상의 억제)

  • The root of Cirsium japonicum var. maackii (Maxim.) has long been used in traditional medicine to prevent the onset and progression of various diseases and has been reported to exert a wide range of physiological effects, including antioxidant activity. However, research on its effects on hepatocytes remains scarce. This study used the human hepatocellular carcinoma HepG2 cell line to investigate the antioxidant activity of ethanol extract of C. japonicum root (EECJ) on hepatocytes. Hydrogen peroxide (H2O2) was used to mimic oxidative stress. The results showed that EECJ significantly reverted the decrease in cell viability and suppressed the release of lactate dehydrogenase in HepG2 cells treated with H2O2. Moreover, an analysis of changes in cell morphology, flow cytometry, and microtubule-associated protein light chain 3 (LC3) expression showed that EECJ significantly inhibited HepG2 cell autophagy induced by H2O2. Furthermore, it attenuated H2O2-induced apoptosis and cell cycle disruption by blocking intracellular reactive oxygen species and mitochondrial superoxide production, indicating strong antioxidant activity. EECJ also restored the decreased levels of intracellular glutathione (GSH) and enhanced the expression and activity of superoxide dismutase and GSH peroxidase in H2O2-treated HepG2 cells. Although an analysis of the components contained in EECJ and in vivo validation using animal models are needed, these findings indicate that EECJ is a promising candidate for the prevention and treatment of oxidative stress-induced liver cell damage.

  • In construction projects, the design information, which should contain accurate product information in a systematic way, needs to be applicable through the life-cycle of projects. However, paper-based 2D drawings and relevant documents has difficulties in communicating and sharing the owner's and architect's intention and requirement effectively and building a corporate knowledge base through on-going projects due to Tack of interoperability between specific task or function-oriented software and handling massive information. Meanwhile, computer and information technologies are being developed so rapidly that the practitioners are even hard to adapt them into the industry efficiently. 3D modeling capabilities in CAD systems are enormously developed and enables users to associate 3D models with other relevant information. However, this still requires a great deal of efforts and costs to have all the design information represented in CAD system, and the sophisticated system is difficult to manage. This research focuses on the transition period from 2D-based design Information management to 3D-based, which means co-existence of 2D and 3D-based management. This research proposes a model of a compound system of 2D and 3D-based CAD system which presents the general design information using 3D model integrating with 2D CAD drawings for detailed design information. This research developed an integrated information management system for design and specification by associating 2D drawings and 3D models, where 2D drawings represents detailed design and parts that are hard to express in 3D objects. To do this, related management processes was analyzed to build an information model which in turn became the basis of the integrated information management system.

Diapause and Voltinism in Ostrinia furnacalis (Lepidoptera: Crambidae) in Suwon, and Larval Instar Sensitivity to Diapause Induction (수원 지방에서 조명나방 휴면과 발생 세대수 및 휴면유도에 대한 유충의 민감성)

  • The diapause induction season in Ostrinia furnacalis (Lepidoptera: Crambidae) was estimated in Suwon. Three batches of adult generations were observed, the first one from early May to early July, the second from early or mid-July to early or mid-August, and the third from mid-August to October. In outdoor larval rearing, colony rearing occurring from mid-July to mid-August produced both non-overwintering and overwintering larvae, whereas late-reared colonies produced only overwintering larvae. Larvae collected during July and August in maize fields produced both non-overwintering and overwintering larvae, whereas late-collected larvae produced only overwintering larvae. The results indicated that O. furnacalis has a bi- or trivoltine complex life cycle in this area. In the laboratory, when larvae of all instars within 9 h after molting were first treated to a diapause induction condition (11:13 h = light:dark photoperiod and 20℃), almost all larvae were induced to diapause. However, when similar treatments were conducted age-specifically for the 5th instar larvae, diapause induction rates in 3- and 4-day-old larvae of the 5th instar decreased. In contrast, when larvae were subjected to the diapause induction treatment only during the periods from the hatching stage to the 2nd, 3rd, and 4th instar, almost all larvae were not induced to diapause. The results suggest that the early age of the 5th larval instar is the last stage for sensitivity to diapause induction stimuli. In the diapause-induced larvae, hemolymph trehalose content increased and body supercooling points dropped, compared with those in non-diapause larvae.

Bioenergy and Material Production Potential by Life Cycle Assessment in Swine Waste Biomass (전과정 평가에 의한 양돈 바이오매스의 물질 및 에너지 자원화 잠재량 연구)

  • As a result of the growing livestock industry, varieties of organic solid and waste biomass are be generated in swine breeding and slaughtering stages. Anaerobic digestion is a promising alternative for the treatment of livestock waste biomass, as well as for the material recovery and energy production. Objectives of this study were to analyze the biochemical methane potential of swine waste biomasses that were generated from swine pen and slaughterhouse and to investigate the material recovery and methane yield per head. As pig waste biomass, swine slurry, blood, intestine residue, and digestive tract content were collected for investigation from pig farmhouse and slaughterhouse. The $B_{th}$ (Theoretical methane potential) and $B_0$ (Biochemical methane potential) of swine slurry generating in swine breeding stage were 0.525 and $0.360Nm^3\;kg^{-1}-VS_{added}$, the ratio of degradation ($B_0/B_{th}$) was 68.6%. $B_{th}$ of blood, intestine residue, and digestive tract content were 0.539, 0.664, and $0.517Nm^3\;kg^{-1}-VS_{added}$, and $B_0$ were 0.405, 0.213, and $0.240Nm^3\;kg^{-1}-VS_{added}$, respectively. And the ratio of degradation showed 75.1, 32.1, and 46.4% in blood, intestine residue, and digestive tract content. Material yield of swine waste biomass was calculated as TS 73.79, VS 46.75, TN 5.58, $P_2O_5$ 1.94, and $K_2O$ $2.91kg\;head^{-1}$. And methane yield was $16.58Nm^3\;head^{-1}$. In the aspect that slaughterhouse is a large point source of waste biomass, while swine farmhouse is non-point source, the feasibility of an anaerobic digestion using the slaughtering waste biomass need to be assessed in the economical aspect between the waste treatment cost and the profitable effect by methane production.

A Study on the Types of Tree Management in Modern Palace Using Photographs and Expert Interviews (사진과 전문가 인터뷰를 통해 추론한 근대 궁궐의 수목관리 판단 연구)

  • The Florence Charter, established in 1981 by the ICOMOS-IFLA International Commission on Historic Gardens, considered the historic garden as a plant-dominated architectural composition, a perpetual balance between the artist and the artisan's desire to remain unaltered in perpetuity. Trees, the main component of the garden, require continuous management as they repeat their creation and calling according to the cycle of the seasons and accordingly, it is suggested that management to maintain the appearance of trees in Korean palaces was an indispensable element. Although it is an important matter to historically verify the tree management techniques of the palaces of the past, research has been difficult due to the absence of historical records and the disconnection of life due to the Japanese colonial period. In addition, according to the view that trees were not generally managed in the palace, research on palace tree management techniques has not been conducted so far. Therefore, this study aimed to examine whether or not the tree management of the palaces was performed in the past through expert interviews based on photographs taken in modern times. With the identification of tree species and pruning through in-depth interviews with experts using photographs taken in the modern period, the results are as follows. First, it was found that the shapes of trees and leaves could be identified through photographs and by observing the phenomena caused by planting and pruning, it was possible to estimate whether or not the tree management was implemented in the modern palace. Second, as a result of in-depth interviews with 8 experts in 4 fields, it was possible to determine the tree species and pruning status, purpose, and method. There was no significant difference in opinions between groups, and the evidence was clearly presented. Third, the type of management of trees in the palace was mainly found to be determination of the types of tree, removal of hazardous factors of trees, and management of lower vegetation and through the photographs of Jondeokjeong and Gwanwanjeong, it was confirmed that the trees were managed even before the Japanese colonial era, considering the time of filming. Based on the photographs taken, it was possible to estimate whether the trees were managed before the Japanese colonial period through expert interviews. However, it could not be clarified due to lack of historical materials as to whether it was carried out on its own according to the circumstances of the times or by the Japanese Empire. Still, in this study, evidence was obtained to refute the view that trees of the palace were not managed in the past through the collected data. Expert opinions supporting this view were collected to make the conclusion. In addition, based on the general theory of pruning, an empirical review of expert opinions was conducted to secure the reliability of the research results.

Evaluation of $^{14}C$ Behavior Characteristic in Reactor Coolant from Korean PWR NPP's (국내 경수로형 원자로 냉각재 중의 $^{14}C$ 거동 특성 평가)

  • This study has been focused on determining the chemical composition of $^{14}C$ - in terms of both organic and inorganic $^{14}C$ contents - in reactor coolant from 3 different PWR's reactor type. The purpose was to evaluate the characteristic of $^{14}C$ that can serve as a basis for reliable estimation of the environmental release at domestic PWR sites. $^{14}C$ is the most important nuclide in the inventory, since it contributes one of the main dose contributors in future release scenarios. The reason for this is its high mobility in the environment, biological availability and long half-life(5730yr). More recent studies - where a more detailed investigation of organic $^{14}C$ species believed to be formed in the coolant under reducing conditions have been made - show that the organic compounds not only are limited to hydrocarbons and CO. Possible organic compounds formed including formaldehyde, formic acid and acetic acid, etc. Under oxidizing conditions shows the oxidized carbon forms, possibly mainly carbon dioxide and bicarbonate forms. Measurements of organic and inorganic $^{14}C$ in various water systems were also performed. The $^{14}C$ inventory in the reactor water was found to be 3.1 GBq/kg in PWR of which less than 10% was in inorganic form. Generally, the $^{14}C$ activity in the water was divided equally between the gas- and water- phase. Even though organic $^{14}C$ compound shows that dominant species during the reactor operation, But during the releasing of $^{14}C$ from the plant stack, chemical forms of $^{14}C$ shows the different composition due to the operation conditions such as temperature, pH, volume control tank venting and shut down chemistry.
