Inhibitory Effect of Citrus Unshiu Peel and Cimicifuga Dahurica Extracts on Melanogenesis accroding to the Various Extraction Methods (추출조건에 따른 진피(陳皮) 및 승마(升麻) 추출물의 미백 활성효과)

  • Objectives: Excessive melanin production leads to skin pigmentation, which causes various cosmetic and health problems. Citrus unshiu Markovich peel (CS) and Cimicifuga dahurica (CD) have long been widely used as a oriental medicinal plant because of their pharmacological properties including anti-ulcer, anti-oxidant, and anti-inflammatory properties. Methods: In this study, we investigated the inhibitory effects of CS, CD extracts or CS:CD=1:2 mixture on melanogenesis according to the various extraction methods. CS and CD extracted were prepared by ethanol extraction (EE), ultrasonification extraction (USE), Supercritical extraction (SCE), reflux extraction (RE), respectively. Results: DPPH radical scavenging and tyrosinase inhibitory activity of CD extracts or CS:CD=1:2 mixture were increased in dose-dependent manners. In addition, we evaluated the effect of CS, CD extracts or CS:CD=1:2 mixture on tyrosinase activity and melanogenesis in α-melanocyte stimulating hormone-induced B16-F10 melanoma cells. CS, CD extracts or CS:CD=1:2 mixture significantly inhibited tyrosinase activity and melanogenesis at 10-200 ㎍/mL. Conclusions: Therefore, our study suggests that CS and CD extracts have potential as a safe treatment for excessive pigmentation or as a natural ingredient in cosmetics.

Extraction of Pesticide Residues in Ginseng by Microwave (Microwave를 이용한 인삼 중의 잔류 농약 추출)

  • Microwave-assisted extraction (MAE) is known as a more environmental-friendly process with economic advantages in terms of less time, less solvent, less energy and less waste than the current reflux method that is time-consuming. MAE was applied to extract three pesticides (quintozene, tolclofos-methyl and procymidone) from ginseng in a pesticide residue analysis and eliminate pesticide contaminants from ginseng prior to production of the ginseng extracts. The optimal conditions for pesticide extraction from ginseng were 50% in power (150 watts), 2 minutes, 20 mL hexane and 2.0 g sample. The results of optimal conditions were compared with those of Soxhlet method (7 hours, 150 mL hexane). The recoveries between two methods were almost same while time and amount of solvent used in MAE were significantly decreased in comparing with those in the Soxhlet methods. When the 45% ethyl alcohol as a extraction solvent was applied to eliminate pesticides from ginseng prior to the production of the ginseng extracts, it was found that 100% quintozene, 69% tolclofos-methyl and 83% procymidone were eliminated from ginseng. The data showed that MAE may be used to eliminate pesticide contaminants in ginseng prior to making the ginseng extracts.

Enhancement of Antioxidant Activities of Crataegus pinnatifida Bunge Fruit by Ultrasonification Extraction Processes (초음파추출 공정을 이용한 아가위나무 열매의 항산화 활성 증진)

  • This study was performed to investigate enhancement of antioxidant activities of Crataegus pinnatifida Bunge fruit by different extraction processes. Extracts were SE (stirrer extraction; 70% ethanol extract at $25^{\circ}C$, control), RE (reflux extraction at $60^{\circ}C$), and USE (ultrasonification for 3 h at $60^{\circ}C$ with 70% ethanol), respectively. Compared to SE, total phenolic and flavonoid contents in USE increased to 104.03% and 302.4%, respectively. USE showed the highest activity at 1.0 mg/mL concentration for 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity. Our results suggest that extracts from the ultrasonification process have relatively high antioxidative activities, and that fruit of Crataegus pinnatifida Bunge could be considered as a candidate functional antioxidant agents. These results would be useful for chemical and biological studies on natural plants and its products.

Screening of Functional Compound of Abeliophyllum distichum Nakai for Optimize Extraction Condition and Efficacy Evaluation (미선나무의 추출조건 확립 및 효능평가를 통한 기능성 탐색)

  • To evaluate optimum conditions of extract active components, extraction of Abeliophyllum distichum Nakai was performed with various ethanol and water mixture at ethanol concentration of 0, 20, 40, 60, 80 and 100%(v/v) and extraction methods. Higher polyphenol and flavonoids contents of extract achieved at 2 hours of reflux extraction in 80% ethanol. The highest DPPH free radical scavenging activity of the stem extracts by soaking method and butanol fractions of leaf extracts are 71.3% and 103.3% respectively. The results suggested that Abeliophyllum distichum Nakai can be used for valuable natural resource.

Optimization of Extraction of Marker Compounds from Red Ginsengs by Accelerated Solvent Extraction Using Response Surface Methodology (반응표면분석법을 사용한 가속 용매 추출에 의한 홍삼 지표성분의 추출 최적화)

  • A new method based on accelerated solvent extraction (ASE) combined with response surface methodology (RSM) has been developed for optimization of the extraction of ginsenoside [Rb1, Rg1, and Rg3(20S)], total phenolics, and benzopyrene in red ginseng. The RSM method, based on a five level and two variable central composite design, was employed to obtain the optimal combination of extraction conditions. In brief, ginsenosides Rb1, Rg1, and Rg3(20S) and total phenolics with undetectable benzopyrene were optimally extracted with 50% ethanol as an extraction solvent, extraction temperature of $158^{\circ}C$, extraction time of 20 min, extraction pressure of 2,500 psi, flush volume of 60%, and one extraction cycle. The contents of ginsenosides and total phenolics in red ginseng extracted by ASE under optimum conditions were significantly higher than those extracted by sonication and reflux extraction.

Comparison and validation of rutin and quercetin contents according to the extraction method of tartary Buckwheat (Fagopyrum tataricum Gaertn.) (쓴메밀 종자의 추출방법에 따른 루틴 및 퀘세틴 함량 비교)

  • The stability and accuracy of ultra-performance liquid chromatography (UPLC) used for evaluating the contents of rutin and quercetin in tartary buckwheat (Fagopyrum tataricum Gaertn.) seeds extracted by seven different extraction methods were determined. The seven extraction methods were reflux extraction (RE), ultra-sonification extraction (UE), stirrer extraction (SE), RE after UE (UE+RE), RE after SE (SE+RE), UE after SE (SE+UE), and RE with UE after SE (SE+UE+RE). Among the seven extraction methods used, RE yielded comparatively higher contents of rutin (2,277 mg/ 100 g) and quercetin (158 mg/100 g) than those yielded by other six extraction methods. The intra-day repeatability and inter-day precision of RE was 0.4-3.2% considering relative standard deviation (RSD), while accuracy was 88.8-102.4%. Therefore, RE with UPLC would be a rapid, accurate, and stable method for analyzing rutin and quercetin contents in tartary buckwheat.

Polyphenol Compound Contents and Physiological Activities in Various Extracts of the Vitex rotundifolia Stems (순비기나무(Vitex rotundifolia) 줄기 추출물의 폴리페놀 함량과 생리활성)

  • For this study, extracts of Vitex rotundifolia stems were prepared using reflux water extraction (WE), reflux ethanol extraction (EE) and hot water extract under high pressure (HWE). The extracts were investigated for the total content of phenolic compounds, antioxidant activities, and inhibitory potencies for xanthine oxidase and tyrosinase. The EE extraction method yielded the highest content of polyphenol compounds (176.34 mg/g). The electron donating abilities (EDA) were 93.46${\sim}$96.92%, when extracts were assayed at 1.0 mg/mL. The superoxide dismutase (SOD)-like activity was the highest in the WE extract (47.32% at 1.0 mg/mL). The nitrite scavenging abilities (pH 1.2) were 84.61${\sim}$88.36% and the inhibition of xanthine oxidase were over 90% at 0.5 mg/mL. Tyrosinase inhibition of HWE and WE were 57.84% and 53.47% respectively. It implies that V. rotundifolia stems have potent physiological activities and their activities were differently exhibited depending on solvent fractions.

Antioxidative Activity and Physiological Function of the Angelica dahurica Roots (백지의 항산화성 및 생리기능)

  • We analyzed the contents of polyphenol compounds, the antioxidant activity and the physiological activity to investigate the functional effects of extracts from Angelica dahurica by the reflux water extraction (RW), reflux ethanol extraction (RE) and pressure heating water extraction (PW). The content of phenolic compounds of PW was the highest at 156.30 mg/g, and those of RW and RE were 31.69 mg/g and 26.34 mg/g, respectively. The electron donating ability (EDA) were in the range of 30.56% $\sim$ 52.74% and superoxide dismutase (SOD) like activity were 10.96% $\sim$ 23.24% at 1,000 ${\mu}g/mL$. The nitrite scavenging ability of PW at pH 1.2 was 61.47%, higher than those of RW (16.81%) and RE (17.78%). The xanthine oxidase inhibitory were 90.91% and tyrosinase inhibitory rate of RE was the highest (51.71%) at the concentration of 5,000 ${\mu}g/mL$. All extracts were increased with increments of the extract concentrations.

Quantitative Analysis about Phenolic Compounds of Dendranthema Species in Korean Native Plants (국내 자생 구절초속 중 페놀화합물 함량분석)

  • Dendranthema spp are distributed in various regions in Korea. This report is the quantitative results of phenolic compounds on the 5 resources collected from all over the country. It was known that it has phenolic compounds, as an anticoagulant activities and immunoadjuvant activity of contribute to the cardiovascular effects. This was used for quantitative analysis of chlorogenic acid(CA), 3,5-Dicaffeoylquinic acid(3,5-DCA), 4,5-Dicaffeoylquinic acid(4,5-DCA) and linarin, with UPLC-PDA and LC-IT-TOF-MS. Extraction efficiency of compounds was compared by using ultrasonic extraction and reflux extraction with different extraction conditions (methods and time). In 50% MeOH extracs (30 mins) of Dendranthema zawadskii (Herb.) Tzvelev (SDZ), Dendranthema zawadskii var. yezoense (Maek.) Y.M.Lee & H.J.Choi (NDZ), Dendranthema zawadskii (Herb.) Tzvelev var. teuisectum (Kitag.) J. H. Pak (GDZ), Dendranthema careanum (H. $L{\acute{e}}v$. & Vaniot) Vorosch. (HDC) and Dendranthema zawadskii var. tenuisectum Kitag (PDZ), chlorogenic acid contents were 1.14‰, 6.95‰, 7.27‰, 1.47‰ and 2.64‰, 3,5-dicaffeoylquinic acid contents were 3.30‰, 5.60‰, 10.81‰, 2.67‰ and 1.50‰, 4,5-dicaffeoylquinic acid contents were 0.74‰, 1.93‰, 3.36‰, 0.61‰ and 0.43‰ and linarin contents were 3.90‰, 10.15‰, 0.15‰, 0.73‰ and 0.21‰, respectively.

Optimization of biodiesel production via methyl acetate reaction from cerbera odollam

  • Cerbera Odollam (sea mango) is a proven promising feedstock for the production of biodiesel due to its high oil content. Fatty acid methyl esters (FAME) were produced as the final reaction product in the transesterification reflux condensation reaction of sea mango oil and methyl acetate (MA). Potassium methoxide was used as catalyst to study its reacting potential as a homogeneous base catalyst. The initial part of this project studied the optimum conditions to extract crude sea mango oil. It was found that the content of sea mango sea mango oil was 55%. This optimum amount was obtained by using 18 g of grinded sea mango seeds in 250 ml hexane. The extraction was carried out for 24 hours using solvent extraction method. Response surface methodology (RSM) was employed to determine the optimum conditions of the reaction. The three manipulated variables in this reaction were the reaction time, oil to solvent molar ratio, and catalyst wt%. The optimum condition for this reaction determined was 5 hours reaction time, 0.28 wt% of catalyst and 1:35 mol/mol of oil: solvent molar ratio. A series of test were conducted on the final FAME product of this study, namely the FTIR test, GC-FID, calorimeter bomb and viscometer test.
