An intertidal marine benthic algal vegetation and vertical distribution at Kangnung and it’s vicinity, the eastern coast of Korea was investigated to clarify the community structure and vertical distribution by quadrat method. Marine algae identified from the area were 116 species; 15 green, 31 brown and 70 red algae. The dominant species were Ulva pertusa, Codium fragile, Undaria pinnatifida, Dictyota dichotoma, Gelidium amansii, Corallina pilulifera, Carpopeltis affinis, Grateloupia filicina, Pachymeniopsis elliptica and Chondrus ocellatus at study sites. The vertical distribution of intertidal marine algae was divided into three distinct zones. They were characterized by Porphyra tenera at the upper, Enteromorpha spp. and Ulva pertusa at the middle, and Sargassum thunbergii, Hizikia fusiformis and Pachymeniopsis elliptica at the lower zones, respectively. The vertical distribution of subtidal zone algae were characterized by Laminaria spp. at 15m depth and Kjellmaniella crassifolia at 15 to 25 m depths, whereas Agarum cribrosum was dominant at the lower zone than 25 m depth.
Two species of marine algae, Fauchea spinulosa Okamura et Segawa (Rhodymeniaceae, Rhodophyceae) and Stictyosiphon soriferus (Reinke) Rosenvinge (Chodariaceae, Phaeophyceae) were newly collected from the southern coast and Cheju Island of Korea. Fauchea spinulosa was collected from subtidal zone in the insular region of the southern coast. Plants were erect from the discoidal holdfast with short stipe, pinkish to deep red, cartilageneous, dichotomously and flabellately branched, and 10-15 cm high, 5-15 mm broad. Tetrasporangia are cruciately divided and nemathecia occurred on a side of the branches. Cystocarps were mostly coronate in marginal area and spermatangia were scattered on both sides of branches. Stictyosiphon soriferus commonly occurs on muddy and sandy intertidal flat of Namhaedo located on the southern coast. Plants are epilithic, light brown, terete, 3-5 cm high, and branched heavily in irregular or alternate manner, arising from a small holdfast with rhizoidal clumps. Plurilocular sporangia were scattered in patches and slightly swollen above the cortex of the whole filament. Unilocular sporangia were not found.
Background: A red seaweed, Callophyllis japonica has been traditionally eaten in the oriental area. In a recent study, it has been demonstrated that the ethanol extract of C. japonica have antioxidant activity. However, there are few studies about the effects of C. japonica on the function of immune cells. We investigated the immunomodulatory effects of C. japonica on the function of dendritic cells, the potent antigen-presenting cells. Methods: Bone marrow-derived dendritic cells (DCs) were used and the viability was measured by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay and trypan blue exclusion test. Cytokine and nitric oxide (NO) levels were determined by using ELISA and Griess reagent, respectively. The expression levels of DC surface markers were measured by flow cytometric analysis. Results: C. japonica ethanol extract did not significantly affect the DCs viability and the IL-12 production from DCs, irrespective of the presence of lipopolysaccharide (LPS). In addition, it did not significantly change the expression of DC surface markers. However, C. japonica ethanol extract significantly inhibited the LPS-induced NO production and also increased the proliferation of allogeneic lymphocytes activated by DCs. Conclusion: Our data suggests that C. japonica ethanol extract enhances the proliferation of allogeneic lymphocytes activated by DCs which is associated with inhibition of NO production from DCs induced by LPS.
An agar-degrading Agarivorans sp. AG17 strain was isolated from the red seaweed Grateloupia filicina collected from Jeju Island. A beta-agarase gene from Agarivorans sp. AG17 was cloned and designated as agrA. agrA has a 2,985 bp coding region encoding 995 amino acids and was classified into the glycoside hydrolase family (GHF)-50. Predicted molecular mass of the mature protein was 105 kDa. His-tagged agrA was overexpressed in Escherichia coli and purified as a fusion protein. The enzyme showed 158.8 unit/mg specific activity (optimum temperature at $65^{\circ}C$ and pH 5.5 in acetate buffer) with unique biochemical properties (high thermal and pH stabilities). Enzyme produced neoagarohexaose, neoagarotetraose and neoagarobiose by degrading agar, and hydrolyzed neoagaro-oligosaccharides were biologically active. Hence the purified enzyme has potential for use in industrial applications such as the development of cosmetics and pharmaceuticals.
The red algal genus Chondrus have long been used as raw materials for carrageenan and dietary fiber in health foods. Despite the importance of genetic information in safeguarding natural seaweed resources, knowledge of the population genetics of Chondrus in the northwest Pacific is limited. In this study, genetic diversity and phylogeographic structure of 45 populations (777 specimens) of Chondrus from Korea, China, and Japan were evaluated based on mitochondrial COI-5P gene sequences, and phylogenetic relationships were confirmed based on plastid rbcL gene sequences. Molecular analyses assigned the specimens in this study to three Chondrus species: C. nipponicus, C. ocellatus, and C. giganteus; phenotype-based species classification was impossible owing to their high morphological plasticity. We found moderate intraspecific genetic diversity and a shallow phylogeographic structure in both for C. nipponicus and C. ocellatus, and low intraspecific genetic diversity in C. giganteus. Each of the three species exhibited high-level intraspecific gene flow among regions based on the most common haplotypes (CN1 for C. nipponicus, CO1 for C. ocellatus, and CG1 for C. giganteus). Our comprehensive genetic information provides insights into the phylogeographic patterns and intraspecific diversity of the economically important Chondrus species. It also highlights the need to conserve existing natural Chondrus resources through continuous monitoring of genetic diversity and phylogeographic pattern.
The objectives of this study were to develop calcium enriched menu for the aged and to investigate physicochemical characteristics and consumer acceptances of the menus. four kinds of the menu were developed with emphasis on calcium enriched food materials of red pepper leaves seasoned vegetables for menu1, shrimp for menu2, anchovy for menu3, seaweed for menu4. one portion of calorie, polysaccharide, protein, fats and calcium for the aged were calculated as 567kcal, 92.8g, 21.3g, 12.6g 223.1mg, respectively based on the recommended daily allowances for adults. Calcium content increased in the lowest value of all(p<0.05). The hunter lightness(L) values of barely rice in menu 1, radish soup in menu 1, chopped roast chicken in menu 4 and radish salad in menu 2 had significantly the highest values of all(p<0.05). The calcium enriched menus for the aged were successfully developed and these data could be used as basic informations for the improvement in health and life of the aged.
최근에 해양자원에 있는 동물, 해조류 곰팡이 세균에서 신규 잠재적인 후보약효제가 조사되어 왔다. 본 연구에서는 치료제를 탐색하기 위하여 암전이, 관절염, 만성염증 및 주름형성에 주요한 역할을 하는 기질금속단백질분해효소(s) (MMPs)를 목적효소로 이용하였다. 5종의 녹조류, 18종의 홍조류, 4종의 갈조류를 포함한 다양한 해조류가 사람피부섬유아세포 및 섬유아육종세포로부터 유래된 기질금속단백질효소에 미치는 영향을 gelatin zymography를 이용하여 조사하였다. 사람피부섬유아세포에서는 홍조류중에서 Laurencia okamurae, Polysiphonia japonica, Grateloupia lanceolate 및 Sinkoraena lancifolia에서 MMP-2 억제효과가 관찰되었다. 반면에 녹조류의 Enteromorpha compressa와 Enteromorpha linza, 갈조류의 Peltaronia bighamiae and Sargassum thunbergii에서는 MMP-2 활성화가 관찰되었다. 사람섬유아육종세포에서는 MMP-9 활성화가 갈조류인 Sargassum thunbergii, 홍조류의 Polysiphonia japonica, 녹조류의 Enteromorpha compressa와 Enteromorpha linza의 존재 하에서는 감소되었다. 본 연구에서 흥미로운 발견은 녹조류의 E. compressa와 E. linza 및 갈조류의 S. thunbergii는 정상세포에서는 MMP-2에 대하여 활성화 효과를 나타내었으나, 암세포에서는 MMP-9응 억제하는 효과를 나타낸 것이다. 이러한 결과는 녹조류의 E. compressa와 E. linza 및 갈조류의 S. thunbergii는 항암 효능을 발휘할 수 있는 성분을 함유하고 있다는 것을 암시하고 있다.
한국 서해안 어청도에서 조간대와 조하대에 생육하는 해조류의 생물량과 군집구조를 조사하기 위하여 2008년 6월부터 2009년 4월까지 계절별로 총 4회 현장 조사를 수행하였다. 조사결과 녹조류 12종, 갈조류 23종, 홍조류 66종이 출현하여 총 101종 채집되어 동정되었다. 이중 19종은 일 년 내내 출현하였다. 총 생물량의 많은 부분을 차지하고 있는 우점종은 작은구슬산호말(Corallina pilulifera), 우뭇가사리(Gelidium elegans), 지충이(Sargassum thunbergii), 톳(Sargassum fusiforme), 다시마(Saccharina japonica) 괭생이모자반(Sargassum honeri), 고사리모자반(Sargassum filicinum)이었다. 연간 평균 건중량은 $237.5gm^{-2}$이었으며, 최대값은 겨울에 $272.1gm^{-2}$, 최소값은 여름에 $204.5gm^{-2}$을 기록하였다. 조하대에서의 출현종과 생물량은 조간대에 비하여 많았다. 어청도에서 출현종수와 생물량은 투명도가 낮은 한국 서해 연안의 일반적 특성과 반대로 높은 투명도와 자연 암반의 발달로 인하여 한국 서해안의 다른 도서에 비하여 높은 것으로 보인다.
한국에서 널리 식용으로 이용되고 있는 미역, 다시마, 톳(갈조류), 파래(녹조류), 김(홍조류)의 항산화효과와 총 플라보노이드와 폴리페놀 함량과의 관련성을 알아보고자 건조 상태의 시판 해조류로부터 에탄올 추출시료를 얻은 후 $Fe^{2+}$로 유도된 지질과산화 억제율, DPPH 라디칼 소거율 및 MDA-BSA 결합 저해율을 측정하는 3가지 방법으로 항산화효과를 검정 하였고, 총 플라보노이드와 폴리페놀 함량을 측정하였다. 그 결과, 녹조류인 파래의 항산화효과가 가장 우수하였는데, 특히 지질과산화물이 단백질과 결합하는 반응을 매우 효과적으로 저해하였다. 그 다음 김과 다시마의 항산화효과가 우수하였으며, 미 역과 톳은 항산화효과가 거의 없었다. 총 플라보노이드 함량은 미역이 11.63 mg/g 건조무게로 가장 높았고, 총 폴리페놀 함량은 파래가 8.97 mg/g 건조무게로 가장 높았다. 한편, 3가지 방법에 의해 측정된 해조류의 항산화효과 모두가 총 폴리페놀 함량과 유의한 양의 상관관계를 보였으며, 특히 지질과산화억제율과 매우 높은 상관관계를 나타내어(r=0.9351, p<0.0001), 해조류에서의 항산화효과에 폴리페놀 성분이 크게 기여할 가능성을 보였다.
