A Genome-Scale Co-Functional Network of Xanthomonas Genes Can Accurately Reconstruct Regulatory Circuits Controlled by Two-Component Signaling Systems

  • Bacterial species in the genus Xanthomonas infect virtually all crop plants. Although many genes involved in Xanthomonas virulence have been identified through molecular and cellular studies, the elucidation of virulence-associated regulatory circuits is still far from complete. Functional gene networks have proven useful in generating hypotheses for genetic factors of biological processes in various species. Here, we present a genome-scale co-functional network of Xanthomonas oryze pv. oryzae (Xoo) genes, XooNet (www.inetbio.org/xoonet/), constructed by integrating heterogeneous types of genomics data derived from Xoo and other bacterial species. XooNet contains 106,000 functional links, which cover approximately 83% of the coding genome. XooNet is highly predictive for diverse biological processes in Xoo and can accurately reconstruct cellular pathways regulated by two-component signaling transduction systems (TCS). XooNet will be a useful in silico research platform for genetic dissection of virulence pathways in Xoo.

Experimental Research of Piece-Mold Casting: Gilt-Bronze Pensive Bodhisattva

  • We have tried the experimental research of lost-wax casting to reconstruct Gilt-Bronze Pensive Bodhisattva; preliminary and reconstruction experiment based on ancient texts. Main object to reconstruct is Korean National Treasure No.83, Gilt-Bronze Pensive Bodhisattva (Maitreya), then we measure alloy ratio and casting method based on the scientific analysis. Other impurities were removed from the base metal components(copper : tin : lead) and their ratio was set to 95.5 : 6.5 : 3 where the ratios for tin and lead were increased by 2.5% each. The piece-mold casting method was used, and piece-mold casting experiments were carried out twice in this study but supplementary research on piece-mold casting was necessary. The microstructure was confirmed to be typical cast microstructure and the component analysis result was similar to that of the prior study. Analysis of the chemical composition is confirmed to copper, tin, lead, and zinc, and the chemical composition of the matrix was 87.8%Cu-7.5%Sn-2.7%Pb-2.1%Zn, and similar to previous experimental research. Also resulted in the detection of small impurity in Zn. Analysis of the mould revealed that the mould was fabricated by adding quartz and organic matter for structural stability, fire resistance, and air permeability. We expect that our research will contribute to provide base data for advanced researches in future.

Authenticated Quantum Secret Sharing using GHZ state swapping (GHZ 상태 교환을 이용한 인증된 양자 비밀 공유)

  • We propose a quantum secret sharing protocol which can authenticate more than half of members using GHZ state swapping. The Trusted Third Party, Trent can authenticate all members using previously shared ID among Trent distributing his message and the members wanting to reconstruct the message. Authenticated members can reconstruct a secret message through GHZ swapping. Moreover, this protocol is efficient to expand the number of members to arbitrary number n, so it is a close quantum secret sharing protocol to classical secret sharing protocol.

A data fusion method for bridge displacement reconstruction based on LSTM networks

  • Bridge displacement contains vital information for bridge condition and performance. Due to the limits of direct displacement measurement methods, the indirect displacement reconstruction methods based on the strain or acceleration data are also developed in engineering applications. There are still some deficiencies of the displacement reconstruction methods based on strain or acceleration in practice. This paper proposed a novel method based on long short-term memory (LSTM) networks to reconstruct the bridge dynamic displacements with the strain and acceleration data source. The LSTM networks with three hidden layers are utilized to map the relationships between the measured responses and the bridge displacement. To achieve the data fusion, the input strain and acceleration data need to be preprocessed by normalization and then the corresponding dynamic displacement responses can be reconstructed by the LSTM networks. In the numerical simulation, the errors of the displacement reconstruction are below 9% for different load cases, and the proposed method is robust when the input strain and acceleration data contains additive noise. The hyper-parameter effect is analyzed and the displacement reconstruction accuracies of different machine learning methods are compared. For experimental verification, the errors are below 6% for the simply supported beam and continuous beam cases. Both the numerical and experimental results indicate that the proposed data fusion method can accurately reconstruct the displacement.

Total Urethra and Penile Shaft Reconstruction with Combined Pedicled Anterolateral Thigh Flap and Radial Forearm Free Flap after Total Penectomy

  • Total reconstruction of the penis (TPR) represents a challenge for urologists and plastic surgeons, especially when urethral length is severely reduced. We here describe, for the first time in an oncologic scenario, a double flap phalloplasty using a pedicled anterolateral thigh (ALT) flap for penile reconstruction and a radial forearm free flap (RFFF) for complete neourethra and glans reconstruction following penile amputation. A 48-year-old patient came to our department following a total penectomy with inferior urethral derivation. The indication for a double flap phalloplasty was posed as only way to fully reconstruct the urethra on its length avoiding possible complications of single flap reconstruction using tube-into-tube technique. Both flaps healed uneventfully with no neourethral strictures or fistulas described. At 18 months follow-up, the patient was extremely satisfied with the aesthetic result and was able to void in standing position. We think that a double free tissue transfer for TPR should be considered, particularly when a urethral length > 14 cm needs to be reconstructed. While the pedicled ALT can be used to reconstruct a proper penile shaft with an easily concealed scar, the RFFF can provide adequate neourethra length with satisfactory sensory recovery at the neoglans.

Analysis of Federal Expenses to Restore, Repair, Reconstruct, or Replace Disaster Damaged Roads and Bridges in the U.S.

  • In the U.S., the state, local, tribe, and territorial governments seek funding from the federal government through the Public Assistance program to carry out these recovery works. In this paper historic public assistance data between 1998 and 2021 have been analyzed to derive several insights such as: types of disasters causing the most damage, states requiring more support, net present value of the federal expense etc. This paper has found that the states requiring more support from the federal government are not always the states suffering the maximum losses from the disasters. It has also found that the net present value of the federal expense between 1998 and 2020 to restore, repair, reconstruct, or replace disaster damaged roads and bridges across the U.S. is $15 billion in 2021 values. Moreover, this paper has tested the correlation between the states' public assistance funds requirements and the existing condition and performance of roads and bridges as revealed by the American Society of Civil Engineer's infrastructure grade card. It has found a weak correlation between these two. The outcomes of this paper can be used by the decision makers to analyze the viability of any possible alternative to the exiting public assistance program. The insights can also help in better decision making in pre-disaster preparation and post-disaster funds allocation.

Consequence of Synthetic Bone Substitute Used for Alveolar Cleft Graft Reconstruction (Preliminary Clinical Study)

  • Background The outcome of alveolar grafting with synthetic bone substitute (Osteon III) in various bone defect volumes is highlighted. Methods A prospective study was accomplished on 55 patients (6-13 years of age) with unilateral alveolar bone cleft. Osteon III, consisting of hydroxyapatite and tricalcium phosphate, is used to reconstruct the defect. Alveolus defect diameter was calculated before surgery (V1), after 3 months (V2), and finally after 6 months (V3) postsurgery. In the t-test, a significant difference and correlation between V1, V2, and V3 are stated. A p-value of 0.01 is considered a significant difference between parameters. Results The degree of cleft is divided into three categories: small (9 cases), medium (20 patients), and large (26 cases).The bone volume of the clefted site is divided into three steps: volume 1: (mean 18.1091 mm3); step 2: after 3 months, volume 2 resembles the amount of unhealed defect (mean 0.5109 mm3); and the final bone volume assessment is made after 6 months (22.5455 mm3). Both show statistically significant differences in bone volume formation. Conclusion An alloplastic bone substitute can also be used as a graft material because of its unlimited bone retrieval. Osteon III can be used to reconstruct the alveolar cleft smoothly and effectively.

Reconstruction of a Severe Open Tibiofibular Fracture using an Ipsilateral Vascularized Fractured Fibula with a Thoracodorsal Artery Perforator Free Flap

  • The Gustilo IIIB tibiofibular fractures often result in long bone loss and extensive soft tissue defects. Reconstruction of these complex wounds is very challenging, especially when it includes long bone grafts, because the donor site is limited. We describe our experience using a set of chimeric ipsilateral vascularized fibula grafts with a thoracodorsal artery perforator free flap to reconstruct the traumatic tibia defects. A 66-year-old male suffered a severe comminuted tibia fracture and segmented fibula fracture with large soft tissue defects as a result of a traffic accident. He also had an open calcaneal fracture with soft tissue defects on the ipsilateral side. All the main vessels of the lower extremity were intact, and the cortical bone defect of the tibia was almost as large as the fractured fibula segment. We used an ipsilateral vascularized fibula graft to reconstruct the tibia and a thoracodorsal artery perforator flap to resurface the soft tissue, using the distal ends of peroneal vessels as named into sequential chimeric flaps. After 3 weeks, the calcaneal defect was reconstructed with second thoracodorsal artery perforator free flap. Reconstruction was successful and allowed rapid rehabilitation because of reduced donor site morbidity.

An Approximate Reconstruction of NPT for Synchronized Data Broadcasting (동기화된 데이터방송을 위한 근사적인 NPT 재구성 기법)

  • DVB-MHP recommends that NPT(normal play time) be used as the times of stream events. NPT is the local time within an event(TV program). But we found that commercial transport stream (TS) generators and middlewares for DVB-MHP settop boxes are not ready to support the use of NPT by applications. In particular, TS generators do not create NPT reference descriptors needed to reconstruct NPT at the TV receiver. This situation is undesirable because program providers cannot experiment with the idea of synchronized applications. So we have implemented a TS generator that inserts NPT reference descriptors to TS and MyGetNPT API to approximately reconstruct NPT. STC (system time clock) is needed to reconstruct NPT, but Xlets are not allowed to read it. So, we approximate STC by using PCR (program clock reference) and the Java system tune. In this method, the stream generator extrats PCRs from an existing TS and inserts them into null TS packets in the form of MPEG sections, which can be read by Xlets. Because PCRs are displaced into new positions in TS, their values should be adjusted based on the time intervals between the original positions and the new positions. We implemented a synchronized application by using our TS generator and MyGetNPT API, where the task of stream events are to display graphic images. We found that graphic images are displayed where 240 ㎳ from their intended time, where 240ms is a human tolerance for the synchronization skew between graphic image and video.

The Applicability of Stable Isotope Analyses on Sediments to Reconstruct Korean Paleoclimate (우리나라의 고기후 복원을 위한 습지 퇴적물의 안정동위원소 분석 가능성 연구)

  • Stable isotope analyses on lake or wetland sediments are useful to reconstruct paleoclimate. Organic and inorganic carbonates obtained from lake sediment are isotopically analyzed to get oxygen and carbon isotopic ratios. Oxygen isotope ratios can be used to quantitatively and qualitatively reconstruct paleo-temperature or humidity while carbon isotope ratios be used to reveal environmental changes around the lake or human impacts on the area. Peat mosses in peat bogs are nice samples for the carbon isotope analysis, which derives paleo-temperature and paleo-atmospheric $CO_2$ changes. In coastal area, the reconstruction of past sea-level is possible because terrestrial originated organic matter is carbon isotopically different from marine originated organic matter. Also, scientists can do research on Asian Monsoon based on the fact that $\delta^{13}C$ of C3 plants and C4 plants are consistently different each other and that they are distributed differently with respect to salinity. In Korea, paleoenvironmental studies using stable isotopes are not popular yet because of low academic interests on the methodology and difficulties of obtaining proper sediment samples. Interesting results can be produced to answer paleoenvironmental questions of Korea if scientists isotopically analyze sediment cores from a paleo-lake such as Hanon in Jeju island, peat bogs such as Mujechi-Neup and Yong-Neup, and coastal wetlands.
