Modulatory Effects of Chrysanyhemi Flos Pharmacopuncture on Nitric-oxide (NO) Production in Murin Macrophagy Cells

  • Objectives: Much evidence exists that herbs have effective immunomodulatory activities. Chrysanthemi Flos (CF) is effective in clearing heat, reducing inflammation, dropping blood pressure and treating headache and is used as a pharmaceutical raw material for an immune enhancer. The purpose of this study was to investigate the modulatory effect of Chrysanthemi Flos pharmacopuncture on nitric-oxide (NO) production in activating macrophages. Methods: After a murine macrophage cell line, RAW 264.7, was cultured in the presence of lipopolysaccharide (LPS), immune-modulating abilities of CF were evaluated by using NO, interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-${\alpha}$) production and phagocytic activity of macrophages. Results: CF enhanced the activities of macrophages by increasing the phagocytic activity and decreasing NO production. Especially, both LPS and CF, 200 ${\mu}g/ml$, treatment could significantly reduce the NO production, but did not change the production of IL-6 and TNF-${\alpha}$. Conclusion: The results of this study indicate that CF may be of immunomodulatory value, especially for adverse diseases due to increased NO production. It may have potential for use as immunoenhancing pharmacopuncture.

The effects of Hyunggaeyungyo-tang of suppression of iNOS production on RAW 264.7cell (Mouse cell에서 형개연교탕(荊芥連翹湯)의 iNOS 생성 억제 효과)

  • Background and Objectives : The aim of this study was to investigate anti-inflammatory and anti-oxidant effects of Hyunggaeyungyo-tang(HYT) on RAW 264.7 cells. Material and Methods : Two types of experiments were implemented for this study: first, the experiment to study the anti-oxidant effect of HYT using riboflavin; second, in vitro experiment to investigate the suppression of NF-${\kappa}$B activation using RAW 264.7 cells (I${\kappa}$B kinase and induce nitric oxide synthase mRNA expression) Results : 1. The anti-oxidant effects of HYT was dose-dependantly increased. 2. The RAW 264.7cells were treated with LPS for 1 hours prior to the addition of indicated concentrations(0.4,-1.0mg/$m\ell$) of HYT, and the cells were further incubated for 24 hours. The LPS-induced IKK & iNOS mRNA expression were dose-dependantly decreased in HYT treated RAW 264.7cells. Conclusion : The results suggest that HYT is significantly effective in the treatment of inflammation through the suppression of NF-${\kappa}$B activation and iNOS production.

Anti-inflammatory Effect of the Cirsium japonicum var. ussuriense 70% Ethanolic Extract in RAW264.7 Cells by Heme oxygenase-1 Expression (엉겅퀴 70% 에탄올 추출물의 RAW264.7 세포에서 Heme oxygenase-1 발현을 통한 항염증 효과)

  • Cirsium japonicum var. ussuriense has long been used in herbal medicine for the treatment of arthritis, dyspepsia, and bleeding in Korea. In the present study, we investigated anti-inflammatory effects of C. japonicum var. ussuriense against lipopolysaccharide(LPS)-induced activation in RAW264.7 cells by the expression of heme oxygenase (HO)-1. The 70% EtOH extract of the aerial parts of C. japonicum var. ussuriense (CJE), showed the potent anti-inflammatory effects on LPS-induced inflammation in RAW264.7 cells. The anti-inflammatory effect of CJE was demonstrated by the suppression of pro-inflammatory mediators, including pro-inflammatory enzymes (inducible nitric oxide synthase and cyclooxygenase-2). Furthermore CJE induced HO-1 expression through nuclear translocation of nuclear factor E2-related factor 2 (Nrf2) and increased HO activity in RAW264.7 macrophages. The effects of CJE on LPS-induced NO and $PGE_2$ productions were partially reversed by an HO-1 inhibitor, tin protoporphyrin (SnPP). Therefore, it is suggested that CJE-induced HO-1 expression plays a role of the resulting anti-inflammatory effects in macrophages. These results suggest that CJE may be a promising candidate for the treatment of inflammatory diseases.

Effect of Water Extract of Sparasis crispa on the Expression of TNF-α, iNOS and IL-1β Genes in RAW 264.7 Cells (꽃송이버섯 추출물이 RAW 264.7 세포에서 TNF-α, iNOS, IL-1β 유전자 발현에 미치는 영향)

  • Purpose : The purpose of this study was to examine the anti-inflammatory effects of Sparassis crispa (SC). SC is a well-known traditional herbal remedy and its mushroom is used for treatment of inflammation. Many diseases that are increasing recently have characteristics of inflammatory diseases. Researchers are finding bioactive substances from natural products that can promote treatment and prevention of inflammation. We investigated the effect of water extracted from SC on the expression of effector genes involved in the function of RAW 264.7 cells. Methods : Effects of RAW 264.7 cells on cell viability, antioxidation, and mRNA expression were examined using water extracts from SC. A 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was performed to determine the effect of water extracts from SC on cell viability in RAW 264.7 cells. Inflammation of RAW 264.7 cells induced by lipopolysaccharide (LPS) treatment and expression levels of inflammatory cytokine TNF-α, iNOS and IL-1β gene were analyzed using quantitative reverse transcription PCR (qRT-PCR) analysis. Results : The MTS assay was performed on RAW 264.7 cells after treatment with various concentrations of water extracts of SC. Treatment of RAW 264.7 cells with water extracts from SC and LPS at a concentration of 0.125, 0.5 mg/㎖ for twenty four hours promoted mRNA expression of TNF-α, iNOS and IL-1β. Conclusion : MTS assay was applied to RAW 264.7 cells after various concentrations of water extracts of SC. Through experimental demonstration of anti-oxidant and anti-inflammatory effects of water extracts from SC, we suggest that SC is a valuable material for the prevention and treatment of various inflammatory diseases.

  • Acontii Tuber is very toxious material, but In oriental medicine frequently used for emergency state like shock, coma, and severe pain and author study about Acontii Tuber in Bang-Yak-Hap-Pyun(方藥合編). Bang-Yak-Hap-Pyun(方藥合編) is widely used for clinical herbal prescpition book. Acontii Tuber has alkaloid. this chemical compound make toxious state like arrythmia, heart arrest, weakness, numbness, itching. And using this material must be careful. In Bang-Yak-Hap-Pyun(方藥合編) Acontii Tuber is used 1.2g ~ 12g. and it decrease pain, inflammation, increase activity of heart, and Acontii Tuber' commom use is severe pain disease. Raw Acontii Tuber(生附子) is rarely used, mostly roasted Acontii Tuber. In case of Acontii Tuber toxic state, in oriental medicine Gam-du-tang(甘豆湯) & Go-sam(苦蔘) is used, in western medicine atropine & lidocane is used.

Anti-inflammatory Effects of Ethanol Extract of Chinese Medicinal Plants in Yanjin on LPS-stimulated RAW 264.7 Macrophages (LPS로 자극한 RAW 264.7 세포에서 중국 연변에 자생하는 약용 식물 에탄올 추출물의 항염증 효과 연구)

  • Objectives : This study was fulfilled to investigate nominee materials as anti-inflammatory agent from ethanol extract of Chinese medicinal plants in Yanjin. Among the 20 candidates, we selected most effective one, the ethanol extract of Cicuta virosa L. (CVL). The mechanism underlying the anti-inflammatory effects of CVL is not clearly identified as yet. Accordingly, we clarified the anti-inflammatory effects of CVL and its underlying molecular mechanisms in LPS-stimulated RAW 264.7 macrophages. Methods : RAW264.7 macrophages were incubated with CVL (12.5, 25, or $50{\mu}M$) and/or lipopolysaccharide (LPS) ($1{\mu}g/m{\ell}$). Cytotoxicity was determined using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide assay and the level of nitric oxide (NO) production was measured with Griess reagent. The prostaglandin $E_2$ ($PGE_2$) production was measured with enzyme immunoassay kits and the protein expression of inducible nitric oxide synthase (iNOS) was determined using Western blot analysis. Results : Among the 20 ethanol extract of Chinese medicinal plants of Yanjin tested, CVL significantly reduced the production of NO in a dose-dependent manner via inhibition the protein expressions of iNOS without cytotoxicity on the LPS-stimulated RAW 264.7 macrophages. In addition, CVL also effectively declined the production of $PGE_2$ in LPS-simulated RAW 264.7 macrophages. Conclusions : Taken together, these data presented in this study demonstrate that CVL possesses anti-inflammatory activity by suppressing the production of pro-inflammatory mediators NO and $PGE_2$, and pro-inflammatory protein iNOS expression in LPS-stimulated RAW 264.7 macrophages.

Anti-inflammatory Effect of Anemarrhenae Rhizoma on Collagen Induced Arthritis - a Model for Rheumatoid Rrthritis in DBA/1J Mice and Cytokine Production in Raw264.7 Cells (지모의 collagen 유발 관절염에 대한 소염 효과 - DBA/1J mouse 에서의 병태 관찰 및 RAW264.7에서의 cytokine 분비측정 -)

  • In order to examine anti-inflammatory effect of Anemarrhenae Rhizoma (AR) alcohol extract on rheumatoid arthritis, the present study investigated the viability and TNF-${\alpha}$ production in Raw264.7 cells treated with AR and collagen induced arthritis in DBA/1J mice which were orally administered with AR prior to immunization. The results are as follows: AR extract at 20 and 50${\mu}g$/ml inhibited the viability of Raw264.7 by 35% and 79%, respectively. AR showed a significant decrease in TNF-${\alpha}$ levels from Raw264.7 cells treated with LPS. AR administration significantly decreased arthritic index in DBA/1J mice immunized with bovine collagen type II. AR administration significantly decreased spleen weights obtained from mice in 6 weeks after immunization. AR administration significantly decreased serum anti-type II collagen antibody levels compared with control group. AR administration decreased serum IL-6 levels compared with control group but it did not reach statistical significance.

A Study on the Direction of Research for Pharmacopuncture through the Analysis on the Current Status of Chinese Herbal Injections (중약주사제 실태현황 분석을 통한 국내 약침 연구 방향 모색)

  • Objectives : This study was conducted to suggest new alternative methods to improve pharmacopuncture and Korean medicine research by analyzing the injection route, pharmacological effect, and status studies of Chinese herbal injections. Methods : 130 types of marketed and licensed Chinese herbal injection were searched from National Medical Products Administration (NMPA) of China. CNKI, PubMed, EMBASE, and the 2020 edition of the Chinese Pharmacopoeia were used to collect additional information. 'Herbal injection' and 'Chinese herbal injection' were used as keywords. All data were collected mainly on the treatment of Chinese herbal injection. But data which were not related to the relevant research or Chinese herbal injection were excluded. Results : Intramuscular injection accounted for more than half of the single injection route (51%). Acupoint and intramuscular injections accounted for 55% of dual injection routes. Acupoint, intravenous, and intramuscular injections accounted for the largest proportion (76%) of the multiple routes of injections. As for the pharmacological effect, injection for cardiovascular diseases accounted for 29%. About the number of raw herbal materials, single herbal material was the most common. Twelve intervention studies all tested intravenous injections, and half of them investigated cardiocerebrovascular diseases. All were given by intravenous injection. In the side effect section, the most common symptoms were nausea and vomiting. Conclusions : Through the results, it is expected to be used for research and development of new pharmacopuncture and herbal medicine.

  • Protaetia Brevitarsis Seulensis(white grub) has been traditionally used as medicinal stuff to treat blood stasis, occlusion of menstruation, tetanus and liver cancer in Asian countries (Korea, Japan, China, Taiwan, India and Myanmar). Especially, Donguibogam, which is traditional korean medicinal book, described the white grub as traditional medicine to treat hepatic diseases and vascular disorders. The white grub has been considered as highly nutritional food. The major constituents of white grub are rich in protein, healthy fats, iron, calcium. Recent studies announced that white grub has hepatoprotective effect and anti-microbacterial effect. However, the immuno-enhancing effect of white grub extracts in RAW 264.7 macrophage cells has not been studied yet. In this study, the various concentrations of white grub extract were examined to find immuno-enhancing effects on RAW 264.7 cells. Cytotoxicity was determined by MTT assay and immuno-enhancing effect of white grub extract was investigated by measuring nitric oxide (NO) production compared with only lipopolysaccharide (LPS) treatment. White grub extracts (0.001 - 10 mg/ml) did not show cytotoxicity. Additionally, white grub extracts (0.001 - 1mg/ml) had Immuno-enhancing effect on RAW 264.7 cells compared with only LPS treated group. These results might be provided proof to develop beneficial immuno-enhancing material for human health.
