무지개송어, Oncorhynchus mykiss 자어의 난황흡수 및 초기성장에 관한 연구 (A Study on the Yolk Absorption and Early Growth in the Alevin of Rainbow Trout, Oncorhynchus mykiss)

  • 무지개송어, Oncorhynchus mykiss 난황자어의 부상기 이전까지의 부화일수에 따른 전장(TL)성장식은 $TLt=2.7e^{-1.24{\cdot}e^{-0.11t}}(r^{2}=0.66)$, 전중(TW)성장식은 $TWt=1.8e^{-2.03{\cdot}e^{-0.11t}}(r^{2}=0.66)$, 체세포중량(SW)성장식은 $SWt=1.8e^{-5.41{\cdot}e^{-0.13t}}(r^{2}=0.83)$로 나타났다. 부화일수 경과에 따른 난황장, 난황고, 난황부피는 감소를 나타내는 직선식이였다. 전중-전장, 체세포중량-전장, 난황장-전장, 난황고-전장, 난황부피-전장, 난황중-전장, 난황중-전중, 난황중-난황고 및 난황중-난황장의 성장형질간의 상대성장식은 부화일 경과에 따라 난황홉수 경향을 반영하였다.

해수산 무지개송어(Oncorhynchus mykiss) 프레임 육포의 관능 특성 (Sensory Characterization of Fish Jerky Produced from Frame Muscle of the Sea Rainbow Trout Oncorhynchus mykiss)

  • This study investigated the sensory characteristics (texture, odor, taste and color) of jerky produced from ground sea rainbow trout (SRT) Oncorhynchus mykiss frame muscle (FM). The hardness of the ground SRT-FM jerky was 453.9±91.0 g/cm2, which was lower than that of commercial animal jerky (893.5±404.6 g/cm2) and commercial fish jerky (1,394.4±363.5 g/cm2). The difference in the hardness values of the ground SRT-FM jerky and commercial animal jerky was not significant. The volatile basic nitrogen content of the ground SRT-FM jerky was 48.3±1.6 mg/100 g, which was higher than that of commercial fish jerky (21.6±6.2 mg/100 g) and commercial animal jerky (18.2±6.3 mg/100 g). However, the fish odor of the ground SRT-FM jerky was masked by the presence of various additives. The hydrophilic and lipophilic browning indices of the ground SRT-FM jerky were higher than those of the commercial jerky. The total taste value of the ground SRT-FM jerky was 169.0, and the major amino acids were glutamic acid and aspartic acid. These results suggest that ground SRT-FM jerky would be acceptable to consumers.

해수산 무지개송어(Oncorhynchus mykiss) 프레임 육포의 위생적 및 영양적 특성 (Nutritional and Microbiological Characterization of Fish Jerky Produced Using Frame Muscle of the Sea Rainbow Trout Oncorhynchus mykiss)

  • Large quantities of by-products—such as fish frame, head, skin and viscera—are generated during seafood processing, and these by-products are not utilized extensively. Therefore, sea rainbow trout Oncorhynchus mykiss (SRT) frame muscle (FM) jerky was prepared by grinding SRT-FM, followed by mixing with seasoning, forming and drying. The nutritional and microbiological characteristics of the SRT-FM jerky were then investigated. The proximate composition of the SRT-FM jerky was 19.1% moisture, 38.7% crude protein, 7.9% crude lipid and 4.4% ash. The viable bacterium count of the SRT-FM jerky was 3.9 log CFU/g, and Escherichia coli and Staphylococcus aureus were not detected. The total amino acid content of SRT-FM jerky was 37.3 g/100 g, and the major amino acids were aspartic acid, glutamic acid, leucine and lysine. Based on the recommended daily intake of fish jerky (100 g), the most abundant mineral was potassium. The fatty acid composition of the SRT-FM jerky was 26.2% saturated acid, 34.5% monoenoic acid and 39.3% polyenoic acid, and the major fatty acids were 16:0, 18:1n-9, 18:2n-6, 20:5n-3 and 22:6n-3. These results suggest that SRT-FM jerky has high nutritional value.

해수 사육 무지개송어(Oncorhynchus mykiss)에서 분리된 Vibrio anguillarum의 특성 분석 (Characteristics of Vibrio anguillarum Isolated from Seawater Cultured Rainbow Trout Oncorhynchus mykiss in Korea)

  • From 2014 to 2017, mortalities of seawater-cultured rainbow trout Oncorhynchus mykiss, were observed in the Goheung and Jeju areas of Korea, with Vibrio anguillarum (seven strains: RT1, 2, 3, 4, 5, 6, and 7) identified as the etiological agent. The phenotypic (based on API 20NE, API ZYM, and E-test kits), serotypic (slide agglutination tests with O1, O2, O3, O4, and O7 antisera), and genotypic (16S rRNA and ompU sequencing) characteristics of the seven RT strains were analyzed and compared to those of seven additional V. anguillarum stains (SF, isolated from sweet fish; FM, isolated from flathead mullet; ATCC43305; ATCC43311; ATCC43307; ATCC43308; and KCTC2711). The phenotypes of the RT strains showed variance, while the slide agglutination tests of the RT1-7, SF, and FM strains all showed positive reactions with serotype O1 antiserum. The 16S rRNA and ompU sequences of the RT1-7, SF, and FM strains were affiliated with V. anguillarum ATCC43305 (Serotype O1), but the ompU sequence of the SF strain differed from those of the RT1-7, FM, and ATCC43311 strains, including one amino acid substitution. We thus confirmed that serotype O1 V. anguillarum, with multiple phenotypes, continues to infect seawater-cultured rainbow trout in Korea.

  • Effects of muscle fluid concentrates from Styela clava on fish diets were investigated in rainbow trout(Oncorhynchus mykiss). Fish were fed on one of the isonitrogenous(48%) and isolipic(20%) feed containing 5 to 20% of muscle fluid concentrates for 8 weeks. Hematological parameters such as hemoglobin, hematocrit, albumin, glucose, total bililubin, triglyceride and glutamic pyruvate transaminase of the rainbow trout which were fed on the diets varied, but no specific trend became apparent. However, glutamic oxaloacetic transaminase value was significantly higher than that of normal fishes. Therefore, concentrated ascidian fluid which is normally discarded, can be combined with commercial diets for the normal growth of rainbow trout.

  • Carbonic anhydrase is essential for the cellular transportation of hydrogen and bicarbonate ions and plays a key role in a wide variety of physiological processes. Rainbow trout, Oncorhynchus mykiss is an important freshwater fish in aquaculture industry and is known to be one of the most susceptible species to environmental contamination. In this study, carbonic anhydrase was detected in the kidney and intestine of rainbow trout. Carbonic anhydrase was isolated from cytosolic proteins and identified by using SDS-PAGE, isoelectric focusing, and immunohistochemical methods. A specific protein band with molecular weight of 30 kDa and pI of 7.0 was detected by Western blotting. The immunohistochemical results showed that carbonic anhydrase was located at various cells in the kidney and intestine of rainbow trout.

Effects of Recombinant Aquaporin 3 and Seawater Acclimation on the Expression of Aquaporin 3 and 8 mRNAs in the Parr and Smolt Stages of Rainbow Trout, Oncorhynchus mykiss

  • This study aimed to examine the role of two aquaporin isoforms (AQP3 and AQP8) in response to the hyperosmotic challenge of transitioning from freshwater (FW) to seawater (SW) during parr and smoltification (smolt) using the rainbow trout, Oncorhynchus mykiss. We examined the changes in the expression of AQPs mRNAs in the gills and intestine of the parr and smolt stages of rainbow trout transferred from FW to SW using quantitative real-time PCR in an osmotically changing environment [FW, SW, and recombinant AQP3 (rAQP3) injection at two dosage rates]. Correspondingly, AQPs were greater during smoltification than during parr stages in the rainbow trout. Plasma osmolality and gill $Na^+/K^+$-ATPase activity increased when the fish were exposed to SW, but these parameters decreased when the fish were exposed to SW following treatment with rAQP3 during the transition to seawater. Our results suggest that AQPs play an important role in water absorbing mechanisms associated with multiple AQP isoforms in a hyperosmotic environment.

  • 무지개송어, 산천어, 무지개송어와 산천어간의 잡종 3배체를 대상으로 성비조사와 형태학적 계측형질을 측정하였다. 무지개송어, 산천어의 암.수 성비는 공히 유사한 비율로 나타난 반면, 잡종과 잡종 3배체에서는 수컷이 암컷에 비해 높게 나타났다(p<0.01). 잡종 3배체는 두고/두장, 등지느러미 기부장/체장 및 가슴지느러미 길이/체장의 계측형질에서는 친어로 사용된 무지개송어와 산천어의 중간을 나타낸 반면 상악장/두장의 계측형질에서는 무지개송어 쪽을 다소 닮게 나타났다. 이러한 결과들은 잡종 3배체에서 3배체화에 따른 사용된 친어들에 대비된 성비 그리고 계측형질에서의 특징을 나타내었다.

Immune gene expression following LPS exposure in the gill of rainbow trout, Oncorhynchus mykiss

  • In the present study, immune gene expression of rainbow trout, Oncorhynchus mykiss, against bacterial endotoxin (LPS) was studied in vivo. The expression of prointflammatory cytokine genes (IL-1$\beta$ and TNF-$\alpha$) and IFN-related genes (IRF-I, Mx-3) at gill was assessed by RT-PCR at different time point of day1 and day 3 post-injection. It was shown that the proinflammatory cytokine gene expression at gill was induced 1 day after LPS injection but the expression was not sustained until day 3. Meanwhile upregulated expression of IFN-related genes was found to be only at day 3 post injection, indicating indirect effect of LPS on these genes.

무지개송어의 해수순치과정에 일어나는 삼투조절세포의 미세구조 (Ultrastructural Change of Osmoregulatory Cells during Seawater Adaptation in Rainbow Trout (Oncorhynchus mykiss))

  • 양어장에서 염분농도가 각각 $5^{\circ}/_{\circ\circ}$, $10^{\circ}/_{\circ\circ}$, $15^{\circ}/_{\circ\circ}$$1.5m^3$인 사육조에서 순치 적응된 평균 체중이 130g인 smolt 단계의 치어와 870g인 성어의 무지개송어 (Oncorhynchus mykiss) 총 150마리를 공시어로 이용하여 아가미 및 신장의 미세구조의 변화를 알기 위해서 광학 및 전자현미경으로 관찰하여 얻어진 결과는 다음과 같다. 해수에 순치 적응된 무지개송어에서 아가미엽의 비대와 아가미엽 상피세포의 분리 그리고 신사구체의 수축, 신장의 응혈 등의 조직학적 변화가 나타났다. 해수에 순치 적응된 개체의 삼투압은 당수에 서식하는 개체의 삼투압보다 높게 나타나서 평균 300 m Osmol 이상으로 나타났다. 삼투압의 증가는 염분과 무기질의 배출효율을 높이기 위해서 혹은 체내의 삼투압의 균형을 맞추기 위해서 삼투압이 증가하는 것으로 사료된다. 아가미엽의 염세포가 증가하는 발생정도는 염분농도의 증가와 더불어 높게 나타났고, 아가미엽 표면은 염분농도가 높아질수록 요철상태가 심한 구조로 나타났으며, 길게 변형된 미토콘드리아가 관찰되었다. 무지개송어는 조직병리학적 변화에도 불구하고 해수에 순치 적응하는 것으로 나타났다.
