Weekly Variation of Phytoplankton Communities in the Inner Bay of Yeong-do, Busan (부산 영도 내만에서 식물플랑크톤 군집의 주간 변동 특성)

  • To understand the temporal variation of phytoplankton communities in a coastal area, the biomass and diversity were weekly investigated in the inner bay of Yeong-do, Busan. In the study area, chlorophyll a concentration ranged from 0.43~7.58 mg m-3 during the study, indicating the study area was in mesotrophic or eutrophic status. The fractions of chlorophyll a occupied by large phytoplankton (> 3 ㎛ diameter) exhibited an average of 80% of total chlorophyll a in this study. Among the large phytoplankton, while Bacillariophyta was the most dominant in spring and summer, Cryptophyceae prevailed in the fall and winter. On the contrary, in the picophytoplankton community less than 3 ㎛ in diameter, Mamiellophyceae was the most dominant in most seasons, Cryptophyceae was relatively high with an average of 17.7 ± 17.6% throughout the year, but seasonal variations were large. Dinophyceae rarely occupied a higher fraction up to 60.4% of the picophytoplankton community. By weekly monitoring at a coastal station for 13 months, it is suggested that phytoplankton communities in coastal waters could be changed on a short time scale. If data are steadily accumulated at the time-series monitoring site for a long time, these will provide important data for understanding the long-term dynamics of phytoplankton as well as the impact of climate and environmental changes.

The Validation of a Commercial Testosterone RIA Test Kits (Testosterone RIA 검사 kit 별 유효성 비교평가)

  • Purpose Testosterone is a steroid hormone synthesized by the Leydig cells of the testes in men, and by the adrenal cortex and ovaries in women. Testosterone production is regulated by luteinzing hormone secreted by the anterior pituitary gland. In this experiment, the effectiveness of testosterone radioimmunoassay (RIA) kits produced by three companies was evaluated and compared in case the production of testosterone kits was stopped or supply problems occurred. Materials and Methods In October 2021, samples were collected from the patients (n=49) who requested the testosterone RIA test. The experiment was conducted by dividing the patient's sample into low concentration (1.0 ng/mL or less), medium concentration (2.0-4.0 ng/mL) and high concentration (6.0 ng/mL or more). The Testosterone RIA test compared and evaluated the validity of Company A kits used in this hospital and those of Company B and C used in other hospitals. The precision, sensitivity, recovery, linearity and correlation were evaluated for each kit. The testosterone RIA test was carried out in accordance with the insert kit manual for each manufacturer. Results As a result of measuring the precision of the intra assay, the Coefficient of Variation (CV) value of the company A kit was high at 11.4% only in the low concentration sample, and in the case of the company B and C kits, the CV value was less than 10% at low, medium, and high concentrations. In the inter-assay precision measurement, the CV value was less than 15% in both A and C kits, but in the case of the B kit, the CV value exceeded 15% at low and medium concentrations. Sensitivity was 0.13 ng/mL for company A, 0.01 ng/mL for company B, and 0.01 ng/mL for company C, and the linearity of all three kits showed excellent linearity. In the case of recovery rate, all of the A, B, and C company kits showed results that were out of 90-110%. In the case of correlation test, when compared with the company A kit currently use in here, the correlation coefficient (R2) value for the company B kit was 0.9508, and for the company C kit was 0.9352 Conclusion As a result, there was a slight difference in precision at the low concentration sample. The correlation test showed an excellent correlation coefficient. However, it was difficult to secure samples of various concentrations because there were not many tests of testosterone requested at this hospital. So, additional experiments should be carried out by acquiring samples of various concentrations on each laboratory later.

Optimization of the Indole-3-Acetic Acid Production Medium of Pantoea agglomerans SRCM 119864 using Response Surface Methodology (반응표면분석법을 활용한 Pantoea agglomerans SRCM 119864의 Indole-3-acetic acid 생산 배지 최적화)

  • In this study, we optimized the composition of the indole-3-acetic acid (IAA) production medium using response surface methodology on Pantoea agglomerans SRCM 119864 isolated from soil. IAA-producing P. aglomerans SRCM 119864 was identified by 16S rRNA gene sequencing. There are 11 intermediate components known to affect IAA production, hence the effect of each component on IAA production was investigated using a Plackett-Burman design (PBD). Based on the PBD, sucrose, tryptone, and sodium chloride were selected as the main factors that enhanced the IAA production at optimal L-tryptophan concentration. The predicted maximum IAA production (64.34 mg/l) was obtained for a concentration of sucrose of 13.38 g/l, of tryptone of 18.34 g/l, of sodium chloride of 9.71 g/l, and of L-tryptophan of 6.25 g/l using a the hybrid design experimental model. In the experiment, the nutrient broth medium supplemented with 0.1% L-tryptophan as the basal medium produced 45.24 mg/l of IAA, whereas the optimized medium produced 65.40 mg/l of IAA, resulting in a 44.56% increase in efficiency. It was confirmed that the IAA production of the designed optimal composition medium was very similar to the predicted IAA production. The statistical significance and suitability of the experimental model were verified through analysis of variance (ANOVA). Therefore, in this study, we determined the optimal growth medium concentration for the maximum production of IAA, which can contribute to sustainable agriculture and increase crop yield.

Growth Characteristics of Tomatoes Grafted with Different Rootstocks Grown in Soil during Winter Season (대목 종류에 따른 저온기 토경재배에서의 토마토 생육 특성 분석)

  • Cultivation of tomatoes in Korea grown in soil covers 89% of the total area for tomato cultivation. Tomatoes grown in soil often encounter various environment stresses including not only salt stress and soil-borne diseases but also cold stress in the winter season. This study was conducted to comparatively analyze the performance of rootstocks with cold stress by measuring the growth, yield, and photosynthetic efficiency in tomatoes grown in soil. The rootstocks were used 'Powerguard', 'IT173773', and '20LM' for the domestic rootstock cultivars and 'B-blocking' for a control cultivar. The tomato cultivar 'Red250' was used as the scion and the non-grafted tomatoes. Stem diameter, flowering position, leaf length, and leaf width were investigated for the growth parameters. The stem diameter of the non-grafted tomatoes decreased by 15% compared to the grafted tomatoes at 80 days after transplanting when exposed to low temperatures of 9-14℃ for 14 days. The leaf length and width of the non-grafted tomatoes were the lowest with 42.4 cm and 41.8 cm at 80 days after transplanting. The total yield per plant was the highest in tomato plants grafted on 'Powerguard' with 1,615 g and lowest in non-grafted tomatoes with 1,299 g. As the result of measuring the chlorophyll fluorescence parameters, PIABS and DI0/RC, which mean the performance index and dissipated energy flux, 'Powerguard' was the highest with 3.73 in PIABS and the lowest with 0.34 in DI0/RC, whereas non-grafted tomatoes was the lowest with 2.62 in PIABS and the highest with 0.41 in DI0/RC at 80 days after transplanting. The stem diameter has positive correlation with PIABS, while it has negative correlation with DI0/RC. The results indicate that can be analyzed by chlorophyll fluorescence parameters can be used for analyzing the differences in the growth of tomato plants grafted on different rootstocks when exposed to cold stress.

  • Asking to change the cropping patterns to save the labor and capitals in paddy rice cultivation, the study was intended to know the weed problems under the various possible cultural systems; namely, direct seeding (in broadcast and row), machine transplanting and hand transplanting. Under the conditions as weedy check plots, paddy yields were significantly variated among cropping systems, and the functions of panicle No. and spikelet No. to the yield were neglected, among others. However, the yield and yield components were narrowed among cropping systems, and the function of spikelets number per area was comparatively improved to the others.

Studies on the Flowering and Maturity in Sesame 1. Flowering Habit by Different Plant Types (참깨 개화, 등숙에 관한 연구 -제 1 보 참깨 초형에 따른 개화특성에 관한 연구-)

  • This experiment was performed to investigate the flowering habit of sesame (Sesamum indicum L.). Sesame varieties tested could be classified into 8 different plant types by their morphological traits such as capsule shape, capsule setting habit and branching types among sesame gene pool of Crop Experiment Station, ORD. The first flower was appeared at the lowest node on main stem. Flowers were appeared progressively toward the tip of the main stem and also toward the tips of branches. The interval of flowering for a node was about one day, but 3 to 8 days for the flowers on the tips. Side flowers started at 4 to 5 nodes lower than those of center flower at the same day. Flowers were beared 2 by 1 node on the middle part of flower setting node (7-9) in mono capsule setting habit in spite of its normal is 1 by 1 node on the other nodes. Flowers were beared opposite direction on each node of stem and flowering toward the tip of main stem composed of cross shape between nodes and spiral, reverse of clockwise direction. We called this habit as cross spiral flowering order and cross spiral phyllotaxis. The first flower on branches was appeared when center flower on the 5th node of main stem began to flower. The branches produced at higher nodes on main stem showed larger flowering periods and more number of flowers than that at lower parts. BTB (Branch, Tricapsule, Bicarpels, 4 Loculi) type showed three capsule setting habits and same flowering period both on main stem and branches while BTQ (Branch, Tricapsule, Quadricarpels, 8 Loculi) type showed three capsule setting habit on main stem and mono-capsule setting habit on branches. In BTQ type, the period of flowering was much shorter on branches than on main stem. Branching type was considered more promising than non branching type for the breeding of early maturing high yielding variety because branching type has the advantage of bearing a lot of flowers in comparatively short flowering period.

Popping Mechanism and Shape Moulding Factor of Popcorn (튀김옥수수의 파열방향 및 튀김형태 결정요인)

  • Popped popcorn generally have a regular popping direction and typical shape. But the reason and mechanism are not clear yet. This experiment was carried out to investigate the shape moulding factor of popped popcorn. Pericarp thickness of tip-cap section of kernels is slightly thicker than that of top section and this fact provides the important information to the reason. Popping starts when the moisture pressure of heated popcorn is increased and reaches at the critical pressure. Therefore, in the same moisture pressure conditions, top sections are bursted first because their pericarp section is thinner than that of tip-cap section. At the very moment tip-cap sections pull down the top sections of peri carp as bi-metal does. So kernels which removed tip-cap section showed the irregular popping shape because they lost the tip-cap pericarp function. How-ever, kernels which removed embryo showed the typical popping shape but their popping volume was small due to emition and shortage of critical moisture pressure. But kernels which removed the whole pericarp and top pericarp were not popped at all because moisture was entirely emitting out of kernels. These results suggest that the shape moulding factor of popped popcorn is the pericarp thickness differences between the top and tip-cap section of kernels.

  • Because red ginseng was exported in large quantities to the Qing Dynasty in the 19th century, a large-scale ginseng cultivation complex was established in Kaesong. Sibyunje (時邊制), a privately led loan system unique to merchants in Kaesong, made it possible for them to raise the enormous capital required for ginseng cultivation. The imperial family of the Korean Empire promulgated the Posamgyuchik (包蔘規則) in 1895, and this signaled the start of the red ginseng monopoly system. In 1899, when the invasion of ginseng farms by the Japanese became severe, the imperial soldiers were sent to guard the ginseng farms to prevent the theft of ginseng by the Japanese. Furthermore, the stateled compensation mission, Baesanggeum Seongyojedo (賠償金 先交制度), provided 50%-90% of the payment for raw ginseng, which was paid in advance of harvest. In 1895, rising seed prices prompted some merchants to import and sell poor quality seeds from China and Japan. The red ginseng trade order was therefore promulgated in 1920 to prohibit the import of foreign seeds without the government's permission. In 1906-1910, namely, the early period of Japanese colonial rule, ginseng cultivation was halted, and the volume of fresh ginseng stocked as a raw material for red ginseng in 1910 was only 2,771 geun (斤). However, it increased significantly to 10,000 geun between 1915 and 1919 and to 150,000 geun between 1920 and 1934. These increases in the production of fresh ginseng as a raw material for red ginseng were the result of various policies implemented in 1908 with the aim of fostering the ginseng industry, such as prior disclosure of the compensation price for fresh ginseng, loans for cultivation expenditure in new areas, and the payment of incentives to excellent cultivators. Nevertheless, the ultimate goal of Japanese imperialism at the time was not to foster the growth of Korean ginseng farming, but to finance the maintenance of its colonial management using profits from the red ginseng business.

Diversification of Rice Quality for Processing. Physicochemical Characteristics and Inheritance of Floury Endosperm Mutants (특수 가공용 미질개발 : 분상질배유 돌연변이 계통의 이화학적특성과 유전)

  • This study was carried out to assess the agronomic characters and physicochemical properties of floury and chalky-endosperm mutant lines induced by chemical mutagen treatment to rice varieties, Hwacheongbyeo and IR24. Linkage analysis of a floury-endosperm gene was carried out using linkage testers. The grain size of brown rice of the mutants was smaller than that of the original varieties. The l, 000-grain and 1$\ell$ weight were lighter in the mutants compared with those in the original varieties. The compound starch granules in the endosperm cell of the mutants showed a loosely-packed crystalline structure. Amylose contents in mutants ranged from 16.9 to 28.5%. Crude protein contents of the mutants were not significantly different from the original rice variety, Hwacheongbyeo, but white core mutant(line 47106) derived from IR24 showed higher protein(l1.32%) compared with IR24(8.30%). The mutants showed slightly harder gel characteristics, and much lower viscosity in Amylograph than original varieties. Steamed rice-cakes from mutant lines showed greater volume than those from original varieties. During the process of alcohol fermentation, Brix in the mutants(especially floury mutants) decreased faster and the alcohol production after 10-day fermentation was much greater in the mutants than in the original varieties. Three different gene loci for floury endosperm characteristics were identified from the allelism test among mutant lines, and the genes were tentatively symbolized as flo-a, flo-b and flo-c, respectively. A floury gene, flo-a, was linked with lg(liguleless) gene in the linkage group N, with R.V. 5.76$\pm$1.72%.

Species Composition and Vegetation Structure of Abies koreana Forest in Mt. Jiri (지리산 구상나무림의 종조성 및 식생구조)

  • This study set up 49 survey areas with an area of about 400 square meters in Abies koreana natural habitat to identify the species composition and vegetation structure of the A. koreana forest in the Mt. Jiri Nation Park, conducted field surveys using phytosociological methods, and performed the cluster analysis using the Two-Way Indicator Species Analysis (TWINSPAN) and Table manipulation. Subsequently, species composition analysis using the importance value, species diversity analysis, DBH analysis, sapling analysis, and similarity analysis was conducted by each cluster type. The cluster analysis classified the A. koreana forest in Mt. Jiri into five clusters, A, B, C, D, and E. The forest was divided into two clusters, Magnolia sieboldii-Dryopteris crassirhizoma-Sasa borealis and Betula ermanii-Solidago virgaurea-Calamagrostis arundinacea. The former was classified as type A and B by Cornus controversa-Hydrangea macrophylla, and the latter was classified as type E, a typical community, and a Sorbus commixta-Rhododendron mucronulatum cluster. And the S. commixta-R. mucronulatum cluster was divided into C type and D type by Picea jezoensis-Ligularia fischeri and Ainsliaea acerifolia. Through vegetation analysis, the importance value of A. koreana, Quercus mongolica, Acer pseudosieboldianum, Fraxinus sieboldiana, and B. ermanii was highly expressed in the A. koreana forest in Mt. Jiri. Regarding species diversity, the results were similar to those reported in other studies of A. koreana forests in Mt. Jiri. The analysis of diameter at breast height (DBH) showed that A. koreana dominated all layers, and the growth of saplings was also good, indicating that the dominance of A. koreana is expected to continue for a while. However, when considering the value of biodiversity that is expected to increase and threats caused by climate change, systematic preservation and management are required to respond to various threats based on continuous monitoring.
