  • Promoting job growth and quality has been central to developing countries to reduce poverty for decades. In this regard, Nepal is not unique in its need to develop effective TVET programme as it is considered showing positive association with workforce development and productivity. While the copious academic literature has highlighted upon staff training programme in general education, it encounters lack of adequate attention in the field of TVET education. The purpose of the study is to identity and prioritise the professional development needs for the teachers of TVET sector in Nepal. A sample(N=317) of Nepalese teachers from seven provinces were asked to answer the scale. The Borich needs assessment model was employed to assess the perceived level of importance and present preferred training modules among 29 teaching subjects. Moreover, a quadrant analysis using 2×2 matrix was used as a diagnostic tool to denote overriding training topics. The result shows that following competence skills constructs were perceived to be in high need of enhancement as well as in existence of discrepancies: 'occupational characteristics of TVET trainer', 'training needs analysis and curriculum development', 'curriculum development based on national competence standard(NCS)', and 'Institution management evaluation'.

  • A DNA profile database was constructed to investigate the genetic relatedness of 72 germplasm samples of Pyrus and related cultivars using microsatellite markers. Three P. pyrifolia, four P. commus, and one P. betulifolia cultivars with different morphological traits were screened using 387 pairs of microsatellite primers. A core set of 11 primer pairs was selected to obtain 133 polymorphic amplified fragments meeting three criteria: high polymorphism information contents (PIC), high repeatability, and distinct allele patterns. The number of alleles per locus ranged between 4 and 22. Average PIC was 0.743 (range: 0.557 - 0.879). Cluster analysis using the unweighted pair - group method with arithmetical average (UPGMA) separated the 72 pear cultivars and germplasm samples into four major groups: Chinese, European pears, and a cluster of 55 Asian pears that could be reclassify into two subcluster, I - $1^{st}$ and II - $2^{nd}$, according to pedigree information. Almost all of the cultivars were discriminated by 11 microsatellite marker genotypes. The microsatellite DNA profile database may be utilized as tool to verify distinctness, uniformity, and stability between candidate cultivar, and to verify in the distinctness of existing cultivars.

  • The development of random amplified polymorphic DNA (RAPD) and expressed sequence tag-derived simple sequence repeats (EST-SSRs) provided a useful tool for investigating Korean ginseng genetic diversity. In this study, 18 polymorphic markers (7 RAPD and 11 EST-SSR) selected to assess the genetic diversity in 31 ginseng accessions (11 Korean ginseng cultivars and 20 breeding lines). In RAPD analysis, a total of 53 unique polymorphic bands were obtained from ginseng accessions and number of amplicons ranged from 4 to 11 with a mean of 7.5 bands. Pair-wise genetic similarity coefficient (Nei) among all pairs of ginseng accessions varied from 0.01 to 0.32, with a mean of 0.11. On the basis of the resulting data, the 31 ginseng accessions were grouped into six clusters. As a result of EST-SSR analysis, 11 EST-SSR markers detected polymorphisms among the 31 ginseng accessions and revealed 49 alleles with a mean of 4.45 alleles per primer. The polymorphism information content (PIC) value ranged from 0.06 to 0.31, with an average of 0.198. The 31 ginseng accessions were classified into five groups by cluster analysis based on Nei's genetic distances. Consequently, the results of ginseng-specific RAPD and EST-SSR markers may prove useful for the evaluation of genetic diversity and discrimination of Korean ginseng cultivars and breeding lines.

  • Life Cycle Assessment(LCA) is an environmental assessment tool for evaluating environmental burdens associated with products, processes and activities from the raw material acquisition stage to the end-of-life stage. End-of-life stage as well as other processes requires a reliant database in order to increase the confidence in the LCA results. In this study, the flow of Personal Computer(as PC) in the end-of-life stage was examined and the database of two scenarios has been established, i.e. one is part reuse and the other is no part reuse, in the end-of-life phase of PC. Also, key environmental issues were identified by carrying out LCA on a PC in the end-of-life phase for eight environmental impact categories. The 'ozone layer depletion' contributes the highest environmental impact due to generation of $Cl_2$ gas during the incineration of waste plastics. In addition, the scenario 1(part reuse) is more environmentally sound than the scenario 2(no part reuse) when comparing two scenarios.

  • The conventional design of the information system architecture based on the personal experience of information systems has been acted as a limit in progress utilizing appropriate resource allocation and performance improvements. Architecture design depending on personal experience makes differences in variance of a designer's experience, intellectual level in related tasks and surroundings, and architecture quality according to individual's propensity. After all these problems cause a waste of expensive hardware resources. At working place, post-monitoring tools are diversely developed and are running to find the bottleneck and the process problems in the information operation. However, there are no simulation tools or models that are used for expecting and counteracting the problems at early period of designing architecture. To solve these problems we will first develop a simulation model for designing information system architecture in a pilot form, and will verify validity. If an error rate is found in the permissible range, then it can be said that the simulation reflects the characteristic of information system architecture. After the model is developed in a level that can be used in various ways, more accurate performance computation will be able to do, getting out of the old way relying on calculations, and prevent the existence of idle resources and expense waste that comes from the wrong design of architecture.
