상승된 이산화탄소와 온도 그리고 한발 영향에 따른 감자의 군락 형태와 무기영양 변화 (Impact of Elevated Carbon Dioxide, Temperature, and Drought on Potato Canopy Architecture and Change in Macronutrients)

  • 본 연구는 옥외환경조절 시설인 SPAR챔버에서 향후 기후변화가 지속 될 경우 상승된 이산화탄소와 온도 그리고 한발에 따른 감자의 군락 형태와 무기영양 변화에 대한 결과는 다음과 같다. 1. AT+2.8C700는 상승된 $CO_2$와 고온에 의하여 초장과 엽면적 그리고 건물중이 상당히 증가 하였고, AT+2.8C700DS는 한발의 영향으로 상당히 감소하였다. 2. 수확기에서 잎은 AT+2.8C700은 ATC450에 비하여 질소와 인산이 감소한 반면 칼륨, 칼슘 그리고 마그네슘 농도가 증가하였다. 괴경은 질소, 인산 그리고 칼륨이 감소하였으며, 칼슘과 마그네슘은 변화가 없었다. 반면, AT+2.8C700DS는 한발의 영향으로 무기영양 농도가 상대적으로 다른 처리비하여 낮았지만, 마그네슘 농도만 증가하였다. 3. $CO_2$와 온도 상승을 보면 AT+2.8C700는 ATC450에 비하여 질소 농도의 감소로 26% 단백질 함량이 감소를 하였다. 한발을 받은 AT+2.8C700DS보다 AT+2.8C700는 단백질 함량이 20%가 감소 하였다. 4. 따라서 상승된 $CO_2$ 농도와 한발은 무기영양의 변화뿐만 아니라 광합성 관련된 당과 아미노산 같은 2차 대사물질도 변화와 인간의 단백질 섭취에도 영향을 줄 것이다.

Corynespora Leaf Spot of Balsam Pear Caused by Corynespora cassiicola in Korea

  • Corynespora leaf spot occurred severely on balsam pear (Momordica charantia) at Changwon, Gyeongnam province in Korea in November and December 2003. The causal fungus isolated from infected leaves of the plants grew well on potato dextrose agar showing gray to brown color. Solitary or catenary conidia of the fungus were obclavate to cylindrical in shape, and pale olivaceous brown or brown in color. The number of isthmus pseudosepta ranged from 4 to 20 and measured 36~186${\times}$8~19 ${\mu}m$ in size. Conidiophores were pale to light brown in color and measured 94~648${\times}$3~8 ${\mu}m$ in size. Optimal temperature for mycelial growth was $30{\circ}C$. On the basis of mycological characteristics and pathogenicity, the fungus was identified as Corynespora cassiicola (Berk. & Curt.) Wei. This is the first report on the corynespora leaf spot of M. charantia caused by C. cassiicola in Korea.

Occurrence and Analysis of Apple Blotch-like Symptoms on Apple Leaves

  • Apple blotch-like symptoms (ABLS) were observed on 'Fuji' apple leaves in Cheongsong, Gunwi and Yeongcheon apple orchards located in Gyeongbuk Province during 2010-2014. Characteristics of ABLS were yellowing, brown spots on leaves, and defoliation, similar to apple blotch diseased (ABD) leaves, which are infected with Marssonina coronaria. It is difficult to differentiate by eye between ABLS and ABD, which has led to misdiagnosis and overuse of fungicides. The present study was conducted to investigate the cause of ABLS using stereomicroscopy, culture isolation, cross-sectional analysis of leaves, and PCR. No acervuli were found on the surface of ABLS leaves and no growth was observed on potato dextrose agar (PDA) plates in culture. Furthermore, cross-sectional analysis revealed similar results, and mycelia were absent in ABLS leaves. By contrast, all these characteristics were present in ABD leaves. Furthermore, no fungi or viruses were detected in ABLS leaves by PCR, suggesting that the disease is not caused by these agents. These findings suggest that ABLS might be a physiological disorder in plants that is distinct from ABD.

금속 가공유 취급 공정에서 엔도톡신 발생 및 노출 특성 고찰 (Comprehensive Review of Endotoxin Level Reported in Metalworking Operations)

  • Objectives: The aim of this study is to comprehensively summarize endotoxin levels reported in operations using metalworking fluids(MWFs). Methods: An extensive literature review was conducted of studies reporting endotoxin levels in processes using metalworking fluids. Keyword search terms included 'metalworking fluids', 'machining fluids', 'metalworking operation', 'machining operation' and 'endotoxin', which were used in combination. Results: A total of ten manuscripts were found to report on airborne endotoxin levels from metalworking operations in the automobile industry. Polycarbonate(PC), polyvinyl chloride(PVC) and mixed cellulose ester(MCE) were used to collect airborne endotoxin. Limulus Amebocyte Lysate was mainly used to quantify endotoxin amount. The levels of airborne endotoxin reviewed varied considerably, ranging from < $4EU/m^3$ to $790EU/m^3$, which was found to be far lower than those from cotton and potato processing plants, sawmills, and poultry farms. Several studies assumed that exposure to endotoxin could be a causative agent of respiratory diseases. Conclusions: Inhalation endotoxin exposure levels reported from metalworking operations were found to be lower than those from industries handling organic materials, even though it could be considered as a possible cause for several respiratory diseases.

Fitness is Recovered with the Decline of Dimethachlon Resistance in Laboratory-induced Mutants of Sclerotinia sclerotiorum after Long-term Cold Storage

  • After four years of cold storage, dimethachlon resistance of two laboratory-induced resistant Sclerotinia sclerotiorum isolates SCG7 and LA50 declined by 99.5% and 98.9%, respectively, and cross resistance to iprodione and procymidone also declined dramatically. Along with the decline of fungicide resistance, osmotic sensitivity to sodium chloride and glucose decreased tremendously; mycelial growth rate, sclerotia number and weight per potato dextrose agar (PDA) plate increased on average by 118.6%, 85. 5% and 64.5%, respectively; and virulence to detached leaves of oilseed rape increased by 72.7% on average. Significant negative correlations were detected between dimethachlon resistance levels and mycelial growth rate on PDA (r = -0.980, P = 0.021), and between resistance levels and lesion diameters on detached leaves of oilseed rape plants (r = -0.997, P = 0.002). These results have profound implications for assessing the potential risk for resistance development to dicarboximide fungicides in S. sclerotiorum.

감귤저장시 부패균 증식억제를 위한 식물유래 천연항균제 탐색

  • 감귤의 저온저장 중 부패방지를 위한 천연항균제 개발의 일환으로 감귤의 저온저장 중 과피로부터 분리한 곰팡이를 MEA와 PDA 배지에서 배양하면서 분생자병과 분생자의 형태적 특성을 조사한 결과 플라스크형의 phialide, 분생자병의 branching type이 simple type이며, conidial head가 columnar shape 등의 특징을 갖는 Penicillium sp. CF-301로 동정되었다. 또한 국내 자생식물을 채집하여 유기용매로 추출한 후 향균활성을 조사한 결과 측백나무 열매와 주목나무를 제외한 8종의 식물 추출액이 감귤부패균에 대하여 향균활성을 보였으며, 특히 측백나무 즙액은 8mm의 투명환을 보여 가장 높은 항균활성을 나타내었다.

  • Juglans regia L. 나무밑에는 다른 식물이 잘 자라지 못하고 있다. 그래서 그 임상토양, 수용추출액, 빗물 등을 써서 작물을 심어 본 결과 발아와 생장이 심하게 억제되었다. 그 억제물질을 찾기 위해서 Juglans regia L. 나무의 수용추출액과 빗물을 종이크로마토그래피법으로 분석하여 ferulic acid, p-coumaric acid, caffeic acid, vanillic acid, p-hydroxybenzoic acid, chlorogenic acid, gallic acid를 분리확인했으며, 이 물질들이 억제작용에 관계한 것으로 추정한다.

Occurrence of Blossom Blight of Chrysanthemum boreale Caused by Didymella chrysanthemi

  • Black blights attacked the blossom and flower buds of wild chrysanthemum (Chrysanthemum boreale) in the experimental field in Hamyang in 1998. The infection rate of the disease on the plant ranged from 4.0 to 91.8%. The pathogen isolated from the infected flower buds produced numerous conidia in pycnidia. The pycnidia, which were immersed into the petals, emerged through the epidermis by short ostiolate neck. Conidia had 0-3 septate (mostly uniseptate) and were 10-27.5 $\times$5-7.5 ㎛ in size. The fungus produced pseudothecia on potato dextrose agar (PDA), and uniseptate ascospores produced in asci were 10$\times$2.7 ㎛ in size. The pathogen also produced pycnidia and pycnidiospores on PDA after 4 weeks in the dark condition. The conidia produced on PDA were smaller than those from infected plants. Based on the examined mycological characteristics, the fungus was identified as Didymella chrsanthemi.

Structure function relationships amongst the purple acid phosphatase family of binuclear metal-containing enzymes

  • The purple acid phosphatases comprise a family of binuclear metal-containing enzymes. The metal centre contains one ferric ion and one divalent metal ion. Spectroscopic studies of the monomeric, ${\sim}$36 kDa mammalian purple acid phosphatases reveal the presence of an Fe(III)Fe(II) centre in which the metals are weakly antiferromagnetically coupled, whereas the dimeric, ${\sim}$110 000 kDa plant enzymes contain either Fe(III)Zn(II) or Fe(III)Mn(II). The three dimensional structures of the red kidney bean and pig enzymes show very similar arrangements of the metal ligands but some significant differences beyond the immediate vicinity of the metals. In addition to the catalytic domain, the plant enzyme contains a second domain of unknown function. A search of sequence databases was undertaken using a sequence pattern which includes the conserved metal-binding residues in the plant and animal enzymes. The search revealed the presence in plants of a 'mammalian-type' low molecular weight purple acid phosphatase, a high molecular weight form in some fungi, and a homologue in some bacteria. The catalytic mechanism of the enzyme has been investigated with a view to understanding the marked difference in specificity between the Fe-Mn sweet potato enzyme, which exhibits highly efficient catalysis towards both activated and unactivated phosphate esters, and other PAPs, which hydrolyse only activated esters. Comparison of the active site structures of the enzymes reveal some interesting differences between them which may account for the difference. The implications fur understanding the physiological functions of the enzymes will be discussed.

Characterization and Genetic Diversity of Benzimidazole-resistant and -sensitive Monilinia fructicola Isolates in Korea

  • To characterize benzimidazole-resistant and -sensitive Monilinia fructicola populations, the fungal isolates were obtained from peach plants showing brown rot and bloosom blight. Benzimidazole-sensitive isolates did not grow on potato dextrose agar(PDA) amended with $\geq1.0{\mu}g$ active ingredient(a.i.)/ml of the fungicides. However, benzimidazole-resistant isolates grew on PDA regardless of the tested concentrations of fungicides. Benzimidazole-resistant isolates did not grow on diethofencarb-PDA, but sensitive isolates grew on the same PDA. In the nucleotide sequences of $\beta$-tubulin gene, only codon 198(GAG: glutamic acid), a target site for benzimidazole, was replaced with GCG(alanine) in all of the resistant isolates, and this substitution seems to play an important role in the development of resistance. Other interesting codons such as 165(GCT), 200(TTC), and 241(GCT) were not changed among the isolates. Benzimidazole-resistant and -sensitive isolates were clustered clearly in random amplified polymerphic DNA analyses and the results revealed that low levels of genetic diversity between benzimidazole-sensitive and -resistant isolates of M. fructicola in the investigated regions.
