Effects of medium components on Mycelial Growth and Polysaccharide production in Liquid Culcure of Coriolus versicolor

  • This study was executed to investgate the effects of medium components on liquid culture in the flask culture of Coriolus versicolor. This work was focused on raising the mycelial growth and the polysaccharide production. In order to optimize the medium, different carbon and nitrogen sources were investgated. Glucose and yeast extract were chosen for the production of mycelia and polysaccharide as carbon and nitrogen sources, respectively, in the flask culture. For the mycelia growth and polysaccharide production, the medium contained glucose 20g/L, yeast extract 6g/L, $KH_2PO_4$ 0.46g/L, $MgSO_4.7H_2O$ 0.5g/L. The liquid culture conditions for the mycelial growth were $27^{\circ}C$, 200rpm and working volume 100mL using 250mL flask.

Capsular Polysaccharide Serotypes Among Staphylococcus aureus Isolates from Cases of Bovine Mastitis and Dogs (젖소 유방염과 개에서 분리된 황색포도상구균에 대한 Capsular Polysaccharide형의 동정)

  • 본 연구는 임상형 및 준임상형 유방염에 이환된 젖소의 유즙에서 분리된 24주의 황색포도상구균에 대한 capsular polysaccharide(CP)형을 확인하고, 1% 토끼 혈청이 함유된 serum-soft agar 배지에서 집락의 모양을 관찰하였다. 또한 동물에 따라 우세한 CP형의 차이가 있는지를 비교하고자 개에서 분리된 13주에 대하여 동일한 실험을 수행하여 다음과 같은 결과를 얻었다. 집락모양을 관찰한 결과 젖소 유래 24주 중 16주(66.7%)는 diffuse, 5주(20.8%)는 compact, 나머지 3주(12.5%)는 분류가 불가능(indeterminate)하였다. CP형 확인결과 9주(37.5%)는 type 5, 2주(8.3%)는 type 8, 13주(54.2%)는 분류가 불가능(non-typeable)하였다. 한편, 개에서 분리된 균주 중 1주(type 8)를 제외한 12주(92.3%)는 type 5로 분류되었으며, 13주 중 8주(61.5%)가 diffuse형의 집락을 보였다. 본 실험에 사용한 균주의 수가 충분하지 못한 문제가 있지만 동물에 따라 우세한 CP형이 다를 수 있으며 분류 불가능한 균주가 상대적으로 높은 비율을 차지하였는데 이는 새로운 CP형의 분류형이 필요함을 시사하는 것으로 판단된다. 또한 type 5와 type 8만을 포함한 유방염 백신은 한계가 있을 것으로 사료된다.

Efficacy of Bovine Staphylococcal Mastitis Vaccine Composed of Alpha toxin, Capsular Polysaccharide and Fibronectin Binding Protein in Lactating Cows and Heifers (비유우와 처녀우에서 황색포도구균의 alpha toxin, capsular polysaccharide와 fibronectin binding protein으로 구성된 유방염 백신의 효능)

  • capsular polysaccharide(CPS), alpha toxin과 재조합 fibronectin binding protein (FnBP)으로 구성된 유방염 백신을 개발하여 야외효능실험을 수행하였다. 이 subunit 백신은 비유우 23두와 처녀우 20두를 대상으로 10개월동안 각각 수행되었다. 비유우는 매 3주간격으로 2회 백신접종을 하였으며 대조군은 PBS를 상유방림조절 주위에 피하주사하였다. 처녀우를 대상으로 한 실험에서는 분만 8주전부터 시작하여 매 3주간격으로 2회 비유우 백신접종부위와 동일하게 접종하였다. 접종후 비유우에서는 황색포도상구균에 의한 총유선내 감염율이 대조균에 비해 유의성있게 감소하였으며 (p<0.001) 처녀우에서도 총유선내 감염이 백신 접종균(1.6%)이 대조군(11.7%)에 비해 다음 비유기동안 유의성있게 낮았다. (p<0.001). 비유기동안 백신접종한 젖소의 체세포수는 변화가 없었으며 처녀우에서는 추가접종후 백신접종군에서 대조군의 체세포수에 비해 낮았지만 통계학적으로 유의성은 없었다(p>0.05). 본 실험결과 황색포도구균에 대한 유방염 아단위 백신은 비유우와 처녀우에서 체세포수를 증가시키지 않고 총 유선내 감염율을 낮추어주었다. 하지만 본 실험은 1군데의 목장을 대상으로 하였기 때문에 향후 대규모 목장을 대상으로 하는 유방염 백신의 야외효능실험이 심도있게 이루어져야 한다고 생각된다.

Structural and Physiological Characteristics of Rhamnogalacturonan II from Fruit Wines

  • To characterize the polysaccharides which exist as soluble forms in fruit wines, crude polysaccharides were isolated from red, white, raspberry, wild grape, and pear wine, respectively. Among them, the crude polysaccharide (RW-0) in red wine showed the highest yield and considerable amounts of thiobarbituric acid (TBA)-positive materials. The pectic polysaccharide RW-2 was purified to homogeneity from RW-0 by subsequent size-exclusion chromatography using Sephadex G-75 and its structure was characterized. RW-2 consisted of 14 different monosaccharides which included rarely observed sugars in general polysaccharides, such as 2-O-methyl-fucose, 2-O-methyl-xylose, apiose (Api), 3-C-carboxy-5-deoxy-L-xylose (aceric acid, AceA), 3-deoxy-D-manno-2-octulosonic acid (Kdo), and 3-deoxy-D-lyxo-2-heptulosaric acid (Dha). Methylation analysis indicated that RW-2 comprised at least 20 different glycosyl linkages such as 3,4-linked fucose, 2,3,4-linked rhamnose, 3'-linked apiose, and 2,3,3'-linked apiose, being characteristic in rhamnogalacturonan II (RG-II). High performance size-exclusion chromatography indicated that RW-2 mainly comprised RG-II of higher molecular weight (12,000), and that the changes of molecular weight to apparent 7,000 under less than pH 2.0 were observed. These analyses indicated that the higher molecular weight polysaccharide in RW-2 was mainly present as a RG-II dimer.

  • A polysaccharide with antithrombin activity in Manda$^{(R)}& (PAM) was purified via procedures comprising three major steps, i.e. fractional precipitation with ethanol, anion exchange chromatography, and gel permeation chromatography. PAM showed a symmetrical peak on size exclusion HPLC, as assessed by refractive index, and behaved as a single band on cellulose acetate electrophoresis. The average molecular mass was estimated to be 222 kDa by gel filtration. PAM was found to be a sulfated heteropolysaccharide that contains sulfate group (20.5%, w/w) and uronic acid moiety (7.1 %, w/w) in addition to neutral sugar consisting of fucose, xylose, mannose, galactose, and glucose in a molar ratio of 1.00 : 0.35 : 0.28: 0.22 : 0.15. This polysaccharide appeared to inhibit blood coagulation via the intrinsic pathway in a dose-dependent pattern. The clotting of fibrinogen by thrombin was also significantly mitigated by the presence of PAM.

  • We investigated the optimum conditions for conversion of water-insoluble components of basidiocarps of Ganoderma lucidum to water-soluble components by hydrolyzing with chitinase. We also tried it with Ganoderma luciclum residue remaining after extracting hot water-soluble components of Ganoderma lucidum. After hydrolyzing under optimum conditions (20 ppm chitinase, 2% Ganoderma lucidum or 6% Ganoderma lucidum residue, at pH 3 and at $ 35^{\circ}C$), the contents of total water-soluble components (polysaccharide or protein) were measured, and it was found that the contents of water-soluble components increased to 1.5-2.7 fold. Michaelis constant, $K_m$ and maximum rate, $V_max$ calculated by Lineweaver-Burk plot for hydrolysis of Ganoderma lucidum were 1.75% and 0.02%/min respectively and those for hydrolysis of Ganoderma lucidum residue were 53.15% and 0.53%/min respectively The protein-bound polysaccharide was isolated after hydrolysis and molecular weights were measured by Sepharose CL-4B gel filtration and compared with the molecular weights of polysaccharide before hydrolysis.

  • Polysaccharide from Sanguisorba officinalis was separated and fractionated using DEAE-Sephadex ion-exchange chromatography and Sephacry HR-200 gel filtration chromatography. One of the fraction(Fr. II) was sulfated and its anticoagulant activity was tested in vitro. Sulfation could increase the clotting time 50 times compared to unsulfated one. Fr. II was hydrolyzed and its composition was analyzed by conjugation with 7-amino-1, 3-naphthalene disulfonic acid using HPLC and electrophoresis. Arabinose and galactose were mainly composed at the ratio of 4 : 1. In addition, xylose and rhamnose were also found.

Studies on Antitumor Components of Wild Pholiota squarrosa (Fr.) Quel. (야생 비늘 버섯의 항암 성분에 관한 연구)

  • In order to investigate the antitumor components of Korean wild higher fungi, the carpo-phores of Pholiota squarrosa collected in Kaung Nung area were extracted with water by refluxing for eight hours at 80-90.deg. C. The extracts were dialized for six days against distilled water and lyophilized to produce crude polysaccharide powders. They were found to have the antitumor activity against sarcoma 180 implanted in mice. Especially, the inhibition ratio of the extract of Pholiota squarrosa was 78.7% in the dose of 20mg/kg/day for the period of ten days. The tumor in two of the ten mice was completely regressed. The components of these aqueous extracts were found to be polysaccharide and protein. The total polysaccharide was 42% by Anthrone method and the protein was 55% by Lowry-Folin method in the antitumor component of Pholiota squarrosa.

  • To investigate antitumor components in Korean higher fungi, the carpophores of Pholiota squarrosa belonging to the family Strophariaceae were collected and extracted with hot water. A protein-bound polysaccharide fraction was obtained by adding ethanol to the extract and by dialyzing through Visking tube. The fraction was examined for antitumor activity against sarcoma 180 implanted in mice. It showed an inhibition ratio of 78.7% at the dose of 20mg/kg/day. The tumor in two of the ten mice was completely regressed. The chemical analysis of the antitumor fraction by Anthrone and Lowry-Folin methods showed that it consisted of 42% polysaccharide and 55% protein. The enzyme fraction of the carpophores showed no proteolytic activity on casein.
